Abstract

Utilization of Insect Pathogen (Beauveria bassiana Bals.) to Control the Suction Pest of Cocoa (Helopeltis spp.) in Gadungan Village,The District of East Selemadeg, Tabanan RegencyCultivation of cocoa has the potential to improve the economy of farmers. However, cocoa production has decreased due to Helopeltis spp. The control of Helopeltis spp. still use synthetic chemical pesticides that are harmful to the health of farmers, consumers and environmental pollution. Therefore, it needs to be done by eco-friendly control using Beauveria bassiana which is reported to be able to control some insect pests such as Helopeltis spp. Utilization of B. bassiana is usually in the form of formulations. The research was conducted to determine the quality formulations and efficacy of B. bassiana against Helopeltis spp. obtained from Dinas Perkebunan Provinsi Bali. The research method using randomized block design with 5 treatments, 5 replications and the treatment using concentration formulation 20 grams, 15 grams, 10 grams, 5 grams to dissolve in one liter of water and the control just using water. The observed variable is the number of former suction on fruit. The identification results showed the tested formulations made from active B. bassiana which is characterized by colonies are white, round-shape and the spore structure is like grapes with a good quality because it has spore density 1,06x107 spore/ ml. B. bassiana is quite effective to infect Helopeltis spp. because it just need 4 days to kill Helopeltis spp. The test results showed the concentration of 20 grams formulation is the best concentration with the fewest number of former suction on fruit.

Abstract

Isolation and Selection of Antagonistic Bacteria to Control Stem Rot Disease on Vanilla Plant (Vanilla planifolia Andrews) by In Vitro Test. Isolation, selection, and identification of effective microbes are important steps to obtain biological agents. The perpose of this research was to get potential bacteria as controlling agents for stem rot of vanilla. The research was conducted at Plant Disease Laboratory, Faculty of Agriculture, Udayana University, Denpasar, from December 2017 to February 2018. The research comprised of isolation and identification the pathogen of stem rot of vanilla, pathogenicity test, isolation and identification of antagonistic bacteria, in vitro test of antagonistic bacteria ability to controled the pathogen of stem rot of vanilla, and hypersensitive response of antagonistic bacteria on tobacco plant. The result showed that the pathogen of stem rot on vanilla plants was identified as Fusarium oxysporum. There of 21 isolates of bacteria that has been isolated from field, there were 7 isolates could suppress of F. oxysporum growth are isolates P001, P002, P003, P004, P005, BM02, and BM03. In vitro test of antagonistic bacterial with F. oxysporum showed that isolate P002 could suppress pathogen growth with the highest percentage (98,36%), P005 (98,34%), P001 (92,78%), P003 (82,02%), P004 (80,34%), BM02 (71,80%), and BM03 (66,70%). Hypersensitive respons test showed that isolates P001, P002, P003, P004, P005, BM02, and BM03 weren’t pathogenic for plants.

Abstract

This study are aims to determine the effect of dry heat treatment (DHT) on the quality of seed beans and to determine the length of time required for dry heat treatment (DHT) in seed beans. This research has six activities, that are: 1. sampling symptomatic plants virus on long bean; 2. preparation of seed; 3. dry heat treatment (DHT); 4. test for Immunosobent Serology Enzyme-linked Assay (ELISA); 5. observations; 6. data analysis. The research were designed by completely randomized design (CRD) with six treatments and five replications. The research result show that dry heat treatment does not give adverse effects on seed germination of beans. Treatment of dry heat on a temperature of 70ºC for 60 hours most effective for inactivation of bean common mosaic virus (BCMV) on the seed beans and did not cause damage on the other elements of seed quality. Based on these studies further research on dry heat treatment to determine their effectiveness to protec plants from bean common mosaic virus (BCMV) in endemic areas, should be conducted.

Abstract

The Influence of Packages Technology to Aphid Insect (Myzus persicae Sulz) and Fruit Flies (Bactrocera doralis Comp.) on Cayenne Chilli (Capsicum frutescens L.) and Long Chilli (Capsicum annuum L.) in the Lowlands This research aims are to determine the influence and effectiveness of technology using Trichoderma’s compost, rain shelters, and pesticid eto developmentof insect pests on long chilli and cayenne chilli in the lowlands. This research was conducted from January to April 2018 in Sempidi Village, Mengwi District, Badung Regency. The research method using Randomize Block Design with the factor pattern consisted of 8 treatments and 4 replications. Parameters observed were population of aphid insect and fruit flies, and total crop yield as supporting the data. This result showed that the highest percentage of damage by aphids on the treatment of V1P0 (cayenne chilli control) was 5% and V2P0 (large chilli control) of 10% in two week after treatment. The highest percentage of fruit fly damage on V2P0 tretament was 2.77% and V1P2 is 2.28% at 1st time of harvest. The lowest percentage of aphid damage and fruit fly on treatment V2P2 (large chili with rain shelters) and V1P2 (cayenne chili with rain shelters), aphids damage were 5% and 10% on 9 week after transplanting, and fruit fly damage were 0.94% and 0.76% at 1st time of harvest. The highest yield was obtained on Trichoderma treatment on cayenne chili and large chili with total weight of 509.28gr and 3618.88gr, respectively. From the results of this research was found that the use of rain shelters technology, Trichoderma and pesticides, affect the damage of aphids, fruit flies, and crops yield.

Abstract

Affectivity of Trichoderma sp. that Added in Leaf Compost to Control Fusarium Wilt Diseases on Strawberry Plants (Fragaria sp.) in Pancasari Village Buleleng RegencyThis research purpose in order to find the best concentrations of Trichoderma sp. which is added into the leaf compost to control fusarium wilt disease on strawberry plant. This research was conducted on December 2017 to March 2018 in Plant Diseases Laboratory of Udayana University and Pancasari Village, Sukasada District, Buleleng Regency. The experimental design used a Randomized Block Design (RBD) with six treatments and four replications, for each treatment consists of ten plant units. Observation variables in this research are disease percentage, Trichoderma sp. population on the ground, plant height, yield, and the weight of plants. The results showed that Trichoderma sp. treatment which were added into leaf compost is more effective to control the fusarium wilt diseases on strawberry plants rather than control treatments. P5 is the best treatment as it has the lowest disease percentage for around 20%, stimulate the highest growth of the plants into 25.8 cm, and has the highest yields 187 g.

Abstract

Identification of the Fungal causal Agent of Main Diseases in Hydroponic Lettuce (Lactuca sativa L.) Hydroponics is an agricultural cultivation system without using soil but using water-containing nutrient solutions. Hydroponic technology is used as an alternative in plant cultivation to maintain optimal plant growth. It is completely protected from the influence of external elements such as rain, pests, climate, and others. One of the plants that are often cultivated in hydroponic technology is lettuce. However, in practice, hydroponic lettuce plants are also faced with the constraints of pathogenic fungi that cause disease in cultivated plants. This research was conducted to identify pathogenic fungi that cause disease in hydroponic lettuce plants. The results showed that the disease in hydroponic lettuce was a leaf spot disease caused by Cercospora sp. and wilt disease caused by Fusarium sp. Leaf spot disease is the main disease with a disease percentage of 41.90%.

Abstract

Identification of the causes of lute diseases in strawberry plants (Fragaria sp.) In Pancasari Village And Control Potential By Microbial Antagonists Strawberries (Fragaria sp.) Is one of the most popular fruits of the community because it has many benefits. Strawberry development in Bali is mostly done in the Bedugul area including Pancasari Village because it has a suitable climate for the development of strawberry plants. This study aims to identify the wilting disease that causes a decline in the yield of strawberry plants that reach 95% in Pancasari Village, Sukasada District, Buleleng Regency. The use of synthetic chemical pesticides is the only way that farmers do in Bedugul area to suppress the strawberry disease. Several studies have reported that unwise and excessive use of synthetic chemical pesticides can cause problems and disrupt environmental balance. Control of plant diseases using microbial antagonists is one of the alternative of environmentally friendly control The method used is isolation, identification, pathogenity test and inhibit microbe antagonistic inhibitory test by dual culture technique. Research activities included (1) sampling of symptomatic plants, (2) isolation and identification of associated fungi at root of strawberry plant, (3) isolate fungal pathogenic test (4) isolation of microbial antagonistic candidate, (5) In Vitro test of antagonistic microbial ability Controlling the pathogen of wilt disease in strawberry plants.The results showed that, mushrooms associated with rooting of strawberry plants were pathogens of the genus Verticillium sp. In Vitro antagonistic test with Verticillium sp. Showed the results in which microbial antagonists were able to suppress pathogen growth by 64%. Anatagonic microbes from the fungus Trichoderma sp. Able to suppress the growth of pathogens of wilt disease (Verticillium sp.). Trichoderma sp. Has the ability to grow quickly Trichoderma sp. Also produce various compounds or enzymes that serve to inhibit or break down the cell walls of plant pathogens.

Abstract

Detection of Potyvirus in Mosaic Symptomatic Euphorbia heterophylla in Melon Planting Area in the city of Denpasar Melon (Cucumis melo L.) is a horticultural plant which is included in the Cucurbitaceae group and is highly favored by the community. The development of melon cultivation has faced some obstacles that cause the level of consumption of melon can not be fulfilled. Viruses in plants have a wide range of hosts including several weeds that can be alternative hosts and as a source of inoculums. Potyvirus group is the largest group of plant viruses and plays an important role in causing harm. This research was conducted to determine whether weeds can potentially as an alternative host of Potyvirus so that they can be used as reference materials for academics and farmers so that they take strategies to control weeds in melon plants. Variations in symptoms found in the field include mosaics, vein banding, vein clearing, and chlorosis. The method used in this study was the Polymerase Chain Reaction (PCR). The research activities include (1) sampling and (2) detection and identification of potyviruses in weeds. The results of this study found that Euphorbia heterophylla weeds could become alternative hosts for Potyvirus. The RT-PCR technique succeeded in amplifying target DNA fragments measuring 683 bp according to the CIFOR / CIREV universal primers used in samples from Denpasar.

Abstract

ABSTRACT Effectiveness Test of Rhizobacteria as Antagonist Agents against Fusarium oxysporum f.sp. capsici Causes Fusarium Wilt Disease in Chili Pepper  Plants(Capsicum frutescens L.) Utilization of rhizobacteria as antagonist agents Fusarium oxysporum f.sp. capsici was recommended. The objectives of this research were conducted to evaluate the effectiveness of rhizobacteria as an agents of biocontrol. F. oxysporum f.sp. capsici was put in the center of PDA medium (control). Rhizobacteria as antagonistic fungus was inoculated four side onto PDA medium and then F. oxysporum f.sp. capsici was put in the center of PDA medium. Result of this research, Klebsiella pneumoniae isolates KTNA2 showed strong inhibitory activity against F. oxysporum f.sp. capsici. Percentage of inhibitory activity at 89,65%. Aplication of Pantoea agglomerans isolates GTA24, Stenotrophomonas maltophilia isolates KTTA4, Klebsiella pneumoniae isolates GSA6, and Klebsiella pneumoniae isolates KTNA2 in the open field could suppress the disease incidence about 33,33%, 43,33%, 56,67%, and 63,33% respectively. Abstract

Seed Quality Testing Several Types of Horticultural Crops Circulating in Bali This study aims to determine the physical quality, viability, and vigor seeds of several types of horticultural crops those circulating in Bali. This research was conducted at Breeding and Seed Technology Laboratory and Plant Disease Laboratory of Udayana University Faculty of Agriculture, which lasted from February to April 2015. The method used is seed testing method with duplo system and using T-Test with three replications. The types of seeds that are tested for their quality consist of seven species: mustard, long bean, eggplant, ercis, cucumber, cayenne, and cabbage. The variables observed were physical quality (moisture content), viability (seed germination), and vigor (synapticity of seed growth). The results showed that the seeds of some horticultural crops circulating in the market in Bali have a pretty good quality. This can be seen from the physical quality test of seeds in the form of moisture content, almost all seeds have a relatively safe water content for storage. The result of seed quality test of percentage of seed germination gain between 90.33% - 97.33% in laboratory test is very good because it is above the standard germination value. The result of the simultaneous vigor test grew up to 59.00% - 69.00% is a good enough vigor value because it is at a level above 50%.
