Abstract

Konfirmasi secara Morfologi Jenis Jamur Penyebab Penyakit Layu Tanaman Stroberi di Bali Indonesia Stroberi adalah salah satu buah yang sangat digemari oleh konsumen karena mengandung banyak phytochemical yang merupakan senyawa fenolik yang bermanfaat bagi kesehatan. Perkembangan stroberi di Bali mengalami beberapa kendala yang menyebabkan penurunan produksi, salah satunya adalah penyakit layu. Namun, hingga saat ini belum banyak informasi spesifik tentang agen penyebab penyakit layu tersebut khususnya di Bali. Identifikasi penyebab penyakit layu tersebut sangat perlu diakukan, oleh karena itu pada penelitian ini dilakukan identifikasi secara makroskopis dan mikroskopis terhadap jamur penyebab penyakit tersebut. Selain itu penelitian ini juga bertujuan untuk mengetahui patogenisitas jamur yang diisolasi dari tanaman stroberi bergejala layu tersebut. Hasil uji patogenisitas menunjukkan bahwa isolat jamur patogen tersebut memiliki daya patogenesitas terhadap tanaman setroberi yang diuji. Persentase indeks penyakit adalah 80% dari isolat Gobleg, 90% dari isolat Pancasari dan Candi Kuning dibandingkan dengan kontrol. Berdasarkan identifikasi morfologi secara makroskopis dan mikroskopis jamur penyebab layu pada tanaman stroberi adalah dari genus Fusarium.

Abstract

Bunchy top symptoms on banana has been reported in Bali Island since early 2011. Symptoms variation were observed in the field similar to infection of Banana bunchy top virus (BBTV). The identity of the BBTV in Bali on the basis of DNA-S nucleotide sequence has not been studied, therefore research was conducted to identify the species of BBTV infecting local banana in Bali based on sequence analysis. Research activities were initiated by collecting field samples from several local banana growing areas in Bali Island. Incidence of bunchy top disease in all locations reached 8% to 44% with disease severity ranged from 2.6% to 30%. Identification of BBTV from field samples were done by polymerase chain reaction using specific primers for BBTV (CPF/CPR) followed by sequence analysis of amplified DNA target. Specific BBTV DNA fragment was successfully amplified from 10 field samples; sequence analysis of DNA fragments showed their highest homology with BBTV. In addition the phylogenetic analysis confirmed the close relationship of  BBTV  isolates from Bali with various BBTV isolates from Indonesia and other isolates from the Asian group in GeneBank.

Abstract

Plant pests and diseases cause decreases in strawberry yield in Bali, including plant viruses. Strawberry vein banding virus (SVBV) is a virus that infects strawberry plants and is transmitted by insects. However, studies about this disease in Bali are still limited. This study aimed to detect SVBV within insect bodies and determine the insect species of its vector. Methods used included (1) sampling; (2) detection of SVBV from insect bodies using primers (SVBV F/SVBV R) and PCR; and (3) molecular identification of viruliferous insect with primers (LCO 1490 F/HCO 2198 R) using PCR and sequencing analysis. This study successfully detected SVBV DNA in an insect from three insect families associated with strawberry plants in Pancasari Village, Buleleng Regency, Bali. Results from the nucleotide sequences analysis in SVBV viruliferous insects indicate that the insect was Myzus persicae.

Abstract

Laporan Baru tentang Distribusi Jamur Entomopatogenik Aschersonia sp. yang Menginfeksi Kutu Putih di Bali. Penelitian tentang distribusi jamur entomopatogenik Aschersonia sp. dilakukan untuk mengetahui persebaran Aschersonia yang menginfeksi kutu putih di seluruh Bali. Penelitian tersebut dilakukan di Kabupaten Gianyar, Bangli dan Buleleng pada centra perkebunan jeruk dimulai tahun 2014 sampai tahun 2018. Aschersonia pertama kali ditemukan pada tahun 2014 di Desa Sekaan Kintamani Bangli dan Desa Kerta Payangan Kabupaten Gianyar, dan pada tahun 2018 ditemukan di Desa Gobleg Sukasada Kabupaten Buleleng. Distribusi jamur tersebut ditemukan di dataran tinggi dan tidak ditemukan di dataran rendah. Hal ini sangat terkait dengan keadaan iklim terutama kelembaban dan suhu. Jamur Aschersonia sangat baik tumbuh pada suhu 20°C yang berada pada dataran tinggi.

Abstract

One of the causes for the low productivity of rice in Indonesia is the occurrence of rice blast disease. Blast disease is one of the main diseases of rice plants around the world. Therefore to minimise the risk of the lost yield by blast disease, the Ministry of Agriculture of the Republic of Indonesia has the agriculture insurance program. To support these program, the research about observations of blast disease in a fixed location are needed. The objective this research is to confirm pathogens causing the blast disease in the fields, to determine the development of blast disease and its effect to the rice yield (grain weight and straw weight). The study was conducted in a fixed paddy field at Subak Uma Dalem, Sobangan Village, Mengwi District, Badung Regency, Bali Province from February 2019 to April 2019. The data showed that the disease incidence of rice blast increased every week starting from 1st week (28 DAT) until 10th week (104 ATP). Our study confirmed that the fungus caused the blast disease in the field was Magnaporthe oryzae. The higher incidence of blast disease in the field resulted reduced the rice yield by decreasing both grain and straw weights.

Abstract

Bacterial leaf blight (BLB) has become a major obstacle and a limiting factor in national rice production. Basically, this disease is caused by the bacterium Xanthomonas oryzae pv. oryzae. Therefore, to minimize the risk of disease infection to yield lost; The Ministry of Agriculture of the Republic of Indonesia has the agriculture insurance program. To support the program, the observations of disease incidence of the disease using existing method are needed. The purpose of this study was to determine the development of bacterial leaf blight on rice plants in Subak Munggu and to determine the effect of bacterial leaf blight on the yield of rice plants. Research activities included (1) isolation of pathogens from symptomatic leaves of rice plants, (2) weekly observations (3) morphological identification of pathogens (4) yields analysis. The results of this study showed the development of bacterial leaf blight in Subak Munggu, Munggu Village, Mengwi Sub-district, Badung Regency for ten weeks. The yield by calculating the weight of grains in each plot was calculated, it can be concluded that a sample that showed a high percentage of disease at the harvest time will cause the low grain yield production.

Abstract

ABSTRAKPenyakit layu stewart telah menimbulkan masalah besar bagi negara produsen jagung. Penyakit yang disebabkan oleh bakteri Pantoea stewartii subsp. stewartii ini pertama kali dilaporkan keberadaannya pada tanaman jagung di Bali yaitu pada tahun 2017. Kejadian penyakit baru pada komoditas penting seperti jagung, sangat urgen untuk dicarikan solusi pengendaliannya. Menyusun strategi pengendalian penyakit, harus mengacu pada pertanian berkelanjutan dengan mempertimbangkan keseimbangan dan keamanan lingkungan, kesehatan petani dan konsumen. Alternatif pengendalian yang yang merupakan tujuan penelitian ini adalah pemanfaatan rhizobakteria sebagai penginduksi ketahanan tanaman terhadap penyakit layu stewart. Penelitian rumah kaca dirancang menggunakan Rancangan Acak Kelompok (RAK) dengan pola faktorial dilakukan untuk menguji potensi rizobakteri sebagai agens penginduksi ketahanan tanaman jagung. Penelitian menggunakan dua faktor yaitu isolat rizobakteri (isolat TLKC, isolat CgBd dan tanpa isolat rizobakteri yaitu menggunakan air steril sebagai kontrol) dan cara apilkasinya (perendaman kecambah, perendaman akar bibit, dan penyiraman bibit). Variabel yang diamati adalah keparahan penyakit, pertumbuhan tanaman (tinggi tanaman dan jumlah daun), hasil (panjang dan berat tongkol per tanaman), total fenol dan kandungan asam salisilat. Perlakuan isolat rizobakteri CgBd yang diaplikasikan melalui perendaman kecambah merupakan perlakuan terbaik, karena memberikan pengaruh terhadap pertumbuhan tanaman yaitu tinggi tanaman tertinggi, jumlah daun terbanyak, dan produksi yaitu panjang dan berat tongkol yang tertinggi; dan keparahan penyakit terendah. Rendahnya keparahan penyakit karena tanaman jagung memiliki ketahanan yang meningkat terhadap penyakit layu stewart. Indikator peningkatan ketahanan jagung terhadap penyakit layu stewart adalah peningkatan kandungan total fenol dan asam salisilat. Penelitian menyimpulkan bahwa rizobakteri isolat CgBd dan TLKC mampu berperan sebagai agens penginduksi ketahanan sistemik dan sekaligus sebagai pemacu pertumbuhan tanaman; dan cara aplikasi rizobakteri yang terbaik adalah melalui perendaman kecambah jagung pada suspensi rizobakteri. Kata kunci: jagung, Pantoea, rizobakteri, perendaman kecambah, keamanan lingkunganABSTRACTStewart wilt disease has caused major problems for corn producing countries. Disease caused by Pantoea stewartii subsp. stewartii was first reported its existence on corn plants in Bali, in 2017. The incidence of new diseases on essential commodities such as corn, it is very urgent to look for its control solutions. Devise strategies for disease control, it should refer to sustainable agriculture by considering the balance and the environmental security, the health of farmers and consumers. An alternative control which is the purpose of this research is the utilization of rhizobakteria as inducers of plant resistance to stewart wilt disease. Greenhouse studies was designed using a randomized block design (RBD) with factorial pattern was conducted to test the potential rhizobakteria as inducer agents of plant resistance. The study was used two factors, namely: rizobakteri isolates (TLKC isolate, CgBd isolate and without rizobakteri isolates i.e. using sterile water as a control) and the way applications (soaking of sprouts, soaking of seedling root and watering the seeds). The variables measured were disease severity, plant growth (plant height and number of leaves), productions (length and weight of cobs per plant), phenols total and salicylic acid contents. Treatment of CgBd isolate applied trough sprouts soaking was the best treatment. It was giving the effects on plant growth, namely: the highest plant height and leaves number; production that were the highest of length and weight of cobs per plant; and the lowest disease severity. The low level of disease severity because the corn plants had increased resistance to stewart wilt disease. The indicator of increased corn resistance to stewart wilt disease was an increase in total content of phenols and salicylic acid. The study concluded that CgBd and TLKC rizobakteri isolates able to act as inducer agent of systemic resistance as well as plant growth promoters; and the best way of rizobakteri isolates application was through sprouts soaking on rizobakteri suspension.Keywords: corn, Pantoea, rhizobakteria, sprouts soaking, environmental securityCitation: Temaja, I G. Abstract

Use of chemical pesticides has been shown to have many negative side effects, such as insecticide resistance and resurgence, an outbreak of secondary pests and diseases, the disappearance of parasitoid and predator species as well as residual effects on food crops and on the environment. Over the past 60 years, both the number of agricultural toxins in the environment and incidence rates of toxin-related diseases has increased dramatically. The most effective way to combat this problem is through the use of natural predators. One of the best examples of this is the use of host-specific bacteriophages to control bacterial diseases. The mechanism of infection is a very interesting one. To break through the bacterial cell wall the bacteriophages must produce a range of lytic enzymes. This review will examine and discuss studies of these site-specific cell wall lytic enzymes and their roles in the infection of bacteriophages.

Abstract

Sycanus aurantiacus Ishikawa & Okajima, found in Bali, was first described in 2007 as a new harpactorine species based on morphological and biological characteristics; however, its genome has not yet been sequenced. In this study, we examine the mitochondrial cytochrome oxidase subunit I (MtCOI) nucleotide sequence of S. aurantiacus in order to determine whether it represents a new harpactorine species. A sample from Pancasari, Bali, Indonesia was collected at the same location S. aurantiacus was first discovered in 2007. The selected mtCOI gene (650 bp) was successfully amplified using mtCOI primer pairs LCO1490 and HCO2198, and the resulting MtCOI sequence of the S. aurantiacus sample was compared with those from other hapactorine species recorded in GenBank. This comparison revealed low genetic similarity between S. aurantiacus and most other harpactorine species worldwide, except for the Genus Sycanus (JQ888697) from USA whose mtCOI shares approximately 91% similarity with the Pancasari sample. Phylogenetic analysis indicated a close genetic relationship between Sycanus from Bali and the Genus Sycanus (JQ888697) from the USA. The mtCOI sequence of S. aurantiacus had not been recorded previously, and our comparison with existing Sycanus sequences provides support to the understanding that S. aurantiacus is indeed its own species.

Abstract

Purpose: By adopting an agriculture system that is in harmony with nature through an organic farming system approach, it is expected to produce commodities that are healthy and safe for consumption and have a higher economic value so that the two missions can be achieved in the future. Research methods: Research on identifying the potential and development of organic farming systems towards Bali organic islands had been carried out in all regencies and cities in Bali Province on May - December 2019. The research was conducted through a field survey and the results were agreed through FGD with stakeholders. Findings: Development of organic farming systems potentially carried out in almost all districts in Bali except in Klungkung Regency and Denpasar City. Implications: The innovation result of this research is the online, integrated, compact, lightweight, and easy to operate organic database system for Bali Province.
