Morpho-anatomical characterizationand DNA barcoding analysisof Pluchea indica (L.) Less.. Biodiversitas 23: 4272-4282. Pluchea indica (L.) Less. is a member of the Asteraceae family and is popular as a medicinal plant. It is very important to authenticate plants using various markers to confirm the labeling of plants and to reveal the bioactive compounds for pharmaceutical research. This study aims to characterize the morpho-anatomical and DNA barcoding aspects of P. indica. Three samples obtained from Taman Husada Graha Famili, Surabaya, Indonesia were used in this study. Morphological studies were carried out descriptively, while the paraffin method was used for anatomical observations. DNA barcoding analysis was performed by amplifying and aligning the rbcLand matKgenes. Morphologically, P. indicahas organs like Asteraceae plants in general, with specific characteristics in leaves and flowers. P. indica has an anatomical organ similar to that of Asteraceae plants globally, only that there are specific histological features on its leaves, including anomocytic stomata, glandular cells, and multicellular glandular trichomes. Alignment and reconstruction of the phylogenetic tree showed that the sample plants were closely related to P. indica in the GenBank database. Based on morpho-anatomical characters and DNA barcodes, the plant under study was confirmed as P. indica L. (Less).

Biodiversitas 23: 4097-4108. Secondary metabolites in plants have various benefits, therefore their distribution in plant body parts is important to study. The identification process of plants should be conducted using several characters, including morphological and molecular data. This study aimed to identify and confirm Cosmos caudatus Kunth using morpho-anatomy characters and DNA barcodes. The plants used were three samples taken from the Husada Graha Famili Park, Surabaya. The morpho-anatomical investigation was carried out descriptively, while the DNA barcoding study was processed by amplifying and sequencing the rbcL and matK genes. Morphological studies indicate that the roots and stems of C. caudatus are similar to the organs of dicots shrubs in general. The leaves have a pinnate type with two different colors on each side of the leaf. Compound flowers are cup-type with two types of flowers, namely disc flowers and ray flowers. Based on anatomical observation, all vegetative organs have the same tissue as dicotyledonous plants. The anatomical characteristic of C. caudatus lies in its trichomes due to its anthocyanin content and stomata which are anomocytic and anisocytic types. DNA barcoding results showed that the three sample plants had identical similarities with several Cosmos genera contained in the GenBank database with percentage identities values ??above 98%, query covers 96-100%, and e-values ??of 0.

Antibacterial and biosurfactant activity of endophytic bacteria isolated from mangrove plant in Lamongan, Indonesia. Biodiversitas 25: 3035-3042. This study aims to determine the antibacterial and biosurfactant activity of 61 endophytic bacteria from mangrove plants in Kutang Lamongan Beach. Antibacterial screening of supernatant of endophytic bacteria was performed using the disc diffusion method against Escherichia coli ATCC 25922 and Staphylococcus aureus ATCC 25923. The most potent isolate was identified using 16s rRNA. The chemical compounds of the most potent isolate were identified using GC-MS analysis. The data were analyzed descriptively. Biosurfactant activity was also performed on the most potential isolate. All the isolates showed inhibitory zones against E. coli and S. aureus. Isolate LMG-2 produced the highest antimicrobial activity, averaging 9.57±0.4 mm against E. coli and 8.87±0.36 mm against S. aureus. The biosurfactant activity of LMG-2 isolate produced a surface tension of 39.09+0.49 mN/m and emulsification activity of 65.03+0.03% against kerosene, 47.63+0.10% against crude oil and 46.53+0.27 to diesel. The isolate LMG-2 had 98,41% similarity with the Bacillus amyloliquefaciens strain NBRC 15535 with a query cover of 100%. The biosurfactant extract contains 9-octadecenoic acid methyl ester, (E)-, 9,12-octadecadienoic acid (Z, Z) methyl ester, and 9,12,15-octadecadienoic acid methyl ester. These compounds, known as Fatty Acid Methyl Esters (FAMEs), were integral components of biosurfactants, surface-active agents produced by microorganisms.

Diversity of Vachellia nilotica rhizosphere bacteria in the savanna of Baluran National Park, Indonesia and their potential for hydrolytic enzyme production. Biodiversitas 26: 1754-1767. Vachellia nilotica, a leguminous plant introduced to Baluran National Park as a firebreak, is now causing environmental problems due to its invasiveness, which hinders the growth of native plants. Despite significant ecological changes, there is no data regarding the impact of this plant invasion on soil microorganisms. This study aimed to assess the bacterial diversity in the rhizosphere of V. nilotica and investigate the bacteria's ability to produce hydrolytic enzymes. The bacterial community was identified using Next-Generation Sequencing (NGS), and a conventional isolation technique. A total of 99,520 Operational Taxonomic Units (OTUs) were obtained, encompassing a rich diversity of 4,631 species. These species belonged to four phyla (i) Pseudomonadota; (ii) Actinomycetota; (iii) Bacillota; and (iv) Acidobacteriota. Additionally, 26 isolates were obtained through conventional isolation, and all of them can produce hydrolytic enzymes, including protease, lipase, amylase and cellulase. A total of 9 isolates showed highest proficiency in producing hydrolytic enzymes, of which 8 belonged to the Bacillota phylum and 1 isolate showed similar characteristic with Acidobacteriota phylum. The isolates SAV 2.NA.1.5.6 and SAV 2.NA.2.4.3 exhibited multiple enzyme activities and were classified as Bacillota. The findings indicate that the rhizosphere of V. nilotica contains a high diversity of bacteria, including those capable of producing hydrolytic enzymes. This information can serve as a foundation for managing V. nilotica and promoting sustainable development by leveraging the potential of rhizosphere bacteria in this plant. To our knowledge, no prior research has been conducted on this topic in the savanna area of BNP.

Microbial community profiling of volcanic-affected rice soils near Mount Semeru, Lumajang, Indonesia using Next-Generation Sequencing for biofertilizer development. Biodiversitas 26: 4424-4434. This study represents the first attempt to apply Next-Generation Sequencing (NGS) based on the 16S rRNA gene to analyze the microbial community structure in paddy soil affected by the eruption of Mount Semeru in Lumajang. This study aims to profile the microbial communities of volcanic-affected rice soils near Mount Semeru. Sequencing was conducted using ONT, with Guppy and NanoFilt for bioinformatic processing. Soil chemical analysis revealed a moisture content of 1.22%, organic carbon at 0.96%, and nitrogen at 0.1%. Available phosphorus was measured at 11 ppm, with a Cation Exchange Capacity (CEC) of 6.22 cmol(+)kg?¹. The microbial community was highly diverse, encompassing 100,046 observed species, with a Shannon-Wiener diversity index of 5.528 and estimated species richness values of 2,337.51 (Chao1) and 1,242.00 (ACE). This diversity spanned 44 phyla, 109 classes, and more than 5,000 species. The dominant phylum was Pseudomonadota (46.35% of total OTUs in the sample), with major genera including Lysobacter (5.90%) and Gaiella (4.44%). Opportunistic pathogens were also detected, underscoring the need to strengthen health and safety measures in post-eruption agricultural areas. These findings provide critical insights into the resilience of microbial communities and highlight their potential application in the development of biofertilizers and environmental biotechnology in Indonesia, offering hope for the future of agriculture in volcanic-affected areas.
