Abstract

Pisang merupakan pangan lokal yang mengandung karbohidrat, lemak, dan protein yang tinggi dalam mencukupi kebutuhan nutrisi berpotensi memperbaiki metabolism tubuh. Penelitian ini bertujuan untuk mengidentifikasi gambaran histologi hati dan penambahan berat badan pada tikus model malnutrisi setelah pemberian tepung pisang. Penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan 6 perlakuan dan 4 ulangan, menggunakan tikus jantan (strain wistar), umur 7 minggu, rerata berat badan 120 g sejumlah 24 ekor. Kelompok perlakuan diantaranya; K- (pakan standar), K+ (pakan nasi aking), M+P0 (pakan standar), M+P1 (pakan 100% tepung pisang), M+P2 (pakan 75% tepung pisang +25% pakan standar), M+P3 (pakan 50% tepung pisang + 50% standar). Kelompok perlakuan K+ sampai perlakuan M+P3 dibuat malnutrisi dengan diberi nasi aking. Kelompok K- diberi pakan standar selama 11 minggu. Pengukuran berat badan dilakukan setiap minggu untuk masing- masing perlakuan. Setelah 11 minggu, tikus dikorbankan. Organ hati dikoleksi kemudian dibuat mikroanatomi dengan pewarnaan Hematoxylin-Eosin. Parameter kerusakan hati dideteksi melalui jumlah sel nekrosis, degenerasi hidropik, dan apoptosis pada 100 sel yang diamati. Hasil penelitian pemberian tepung pisang pada perlakuan M+P3 dapat menurunkan jumlah kerusakan sel nekrosis (p<0,05). Kombinasi pakan 50% tepung pisang dan 50% pakan standar dapat mempercepat regenerasi, menurunkan jumlah kerusakan sel, dan menghasilkan peningkatan berat badan secara signifikan (p<0,05).

Abstract

Fish is a food that rots easily due to contamination with microorganisms. When consumed, food that has been contaminated with pathogenic bacteria can endanger human health. Tilapia fish (Oreochromis mossambicus) is vertebrate animal that is often cultivated, especially in the Kintamani area. This research was conducted to determine total bacterial contamination, Coliform, E.coli, and Salmonella sp. on tilapia fish cultivated in floating net cages in the Lake Batur area, Kintamani, providing information on the safety and suitability of consuming tilapia fish by food safety standards, as well as providing an overview of differences in bacterial contamination in each location village. The samples were tested using the pour plate and streak plate plating method. Coliform and E. coli bacterial contamination tests were carried out using the Most Probable Number (MPN) method, examining Salmonella sp. carried out on selective Salmonella Shigella Agar (SSA) media. The ALT test results obtained the highest average in tilapia fish samples from Kedisan village, namely 5.9 x 106 CFU/g, the highest average total Coliform was in fish samples from Abang and Batur Tengah villages with values ​​> 1.1 x 104 MPN /g, the highest average E. coli contamination was shown by tilapia fish samples from Abang village, namely >4.64 x 103 MPN/g, and one sample from Batur Tengah village showed positive for Salmonella sp bacteria. Based on these results, all tilapia fish samples exceeded the threshold for ALT, Coliform and E. coli contamination and one sample exceeded the normal limit for Salmonella sp contamination. on food based on BPOM and SNI standards.

Abstract

Chicken meat is a source of animal protein which is easily damaged due to its high water content. Chicken meat that is bought and sold could potentially become a place for the growth of bacteria that cause foodborne diseases.This study aims to determine total bacterial contamination, Coliform, Escherichia coli, and Salmonella sp. on broiler chicken meat in Badung Regency and determine the suitability of broiler chicken meat samples following food safety standards. Samples were taken using a purposive sampling method in six sub-districts in Badung Regency, namely Petang, Abiansemal, Mengwi, North Kuta, Kuta and South Kuta sub-districts at traditional markets that sell broiler chicken meat. The samples were tested using the pour plate and streak platting method. Parameters tested include Total Plate Number (ALT), Coliform, Escherichia coli, and Salmonella sp. on broiler chicken meat samples. Coliform and Escherichia coli bacteria were detected using Petrifilm ready-to-use media, while Salmonella sp. tested on selective Salmonella-Shigella Agar (SSA) media and catalase test and Gram staining. The research result showed that the contamination rates in broiler chicken breast meat were: Total Plate Number (ALT) 105 CFU/g to 107 CFU/g; Coliform and Escherichia coli 101 CFU/g to 104 CFU/g; Salmonella sp. positive in all samples, so that the samples tested did not meet the safety quality requirements for chicken meat according to SNI 3924: 2009.
