Abstract

Miana plant (Coleus) is a plant with compound flowers. Miana leaves have various bioactive compounds widely used to treat diseases such as fever, cold, cough, headaches, asthma, constipation, and skin diseases. Based on phytochemical screening of Coleus atropurpureus Benth. Several bioactive compounds are produced, such as sterols, terpenoids, alkaloids, phenols, flavonoids, glycosides, lactones, catechols, and tannins. Miana leaf extract has been tested for antioxidants using the DPPH method, and the IC50 value obtained from the antioxidant test results is 3.80 ppm and is categorized as a powerful antioxidant. There were four formulations of miana leaf extract nanoemulsion. Based on the characterization on the Miana leaf extract nanoemulsion preparation, formulations with good results were obtained in F1 and F3 with precise results and high transmittance values. In testing the miana leaf extract nanoemulsion, an organoleptic test was carried out on each formulation. It was found that all formulations had a distinctive smell of miana leaves, and no phase separation occurred, with the clarity of F1 and F3 classified as straightforward but F2 and F4 not clear, and the resulting color increased from F1 -F4. Meanwhile, for the physicochemical test, an O/W type nanoemulsion was produced, physically stable, with a particle size of F1 153.9 nm and F3 795.6 nm, with a zeta potential of F1 of 16.4 and F3 of -28.266.

Abstract

One of the efforts to increase the growth of Liberica coffee of Tungkal composite seedlings is through organic fertilization as considered inorganic fertilizers replacement. The purpose of this study was to look for the best dose of chicken manure for the growth of Liberica coffee of Tungkal composite seedlings. This study used a randomized block design with 4 replications with one factor, namely chicken manure consisting of 7 treatment levels, namely 0, 50, 100, 150, 200, 250 and 300 g of chicken manure/polybag. The variables observed were increase in seedling height, increase in stem diameter, increase in number of leaves, leaf area, crown dry weight, and root dry weight. The results showed that the application of chicken manure was able to increase the increase in seedling height, increase in stem diameter, increase in the number of leaves, and crown dry weight of liberica coffee seedlings, but was not able to increase leaf area and root dry weight. The recommended dose of chicken manure to increase the growth of liberica coffee seedlings was 211 g of chicken manure/polybag.

Abstract

This research was conducted to determine the effect of the addition of CPO (Crude Palm Oil) lecithin as an Emulsifier on the physicochemical and sensory characteristics of dark chocolate, and to determine the amount of lecithin addition as an appropriate Emulsifier to produce chocolate with the best physico-chemical and sensory characteristics. This study used a completely randomized design (CRD) method with a treatment level of 0%. 0.1%, 0.2%, 0.3%, 0.4% with 4 repetitions to obtain 20 experimental units. Parameters observed included color degee, antioxidant activity, fat blooming, stability level, color sensory test, taste, aroma, texture and overall acceptability. The data obtained were analyzed using ANOVA levels of 1% and 5%. If there is a significant effect on the treatment, it will be continued with the Duncan's Multiple Range Test (DNMRT) at the 5% level. The best concentration of adding lecithin to dark chocolate is 0.3% which contains 66.76% antioxidant activity, color degee L*28.58, a* 13.67, b* 22.67, hue 58.75, good stability (melting properties), able to prevent fat blooming, has good color very chocolatey (4.20), very distinctive aroma of chocolate (4.44), bitter taste (2.68), smooth texture (3.36) and the overall acceptance of the panelists really liked it (8.16).

Abstract

The paper reports the in vivo and in vitro antidiabetic activity of bioactive compounds composed of ethyl acetate fractions from Sungkai (Peronema canescens Jack). The fractions are obtained from vacuum liquid chromatography (F1, F2 and F3). Phytochemicals screening, UV-Vis and FTIR spectrometry analysis is applied to detect secondary metabolite composed of the fractions. In addition, in vitro antidiabetic capability is measured inhibiting a-amylase activity, and in vivo evaluation is conducted using diabetic rat's model induced by alloxan. It was found that fraction collected as a white precipitate. The phytochemical analysis indicated positive for flavonoid. UV-Vis and FTIR spectra support the data. Moreover, capability of the fractions to inhibit a-glucosidase activity in the range 5-100 ppm does not show the activity. Conversely, the in vivo evaluation from fraction 2 at concentration 600 mg/Kg of body weight has antidiabetic in 50.16%. This result comparable to that of glibenclamide as a control. This finding lead to further evaluation for structural elucidation.

Abstract

Cancer is the process of forming new tissue that is abnormal and malignant. Efforts to discover anti-cancer drug compounds continue to be made to minimize the toxic effects of chemotherapy, so it is necessary to look for other alternatives to treat cancer. This research aims to determine the anti-cancer activity of putat plant bioactive compounds through identification of receptor targets and interaction studies using the molecular docking method against the VEGFR2 (Vascular endothelial growth factor receptor-2) receptor. The results showed that in the test of eight putat plant bioactive compounds the best results were obtained for the compound 3-oxo-N-(1,3-thiazol-2-yl) butanamide with a binding free energy value of -9.86 kcal-mol-1 with an inhibition constant value of 1.47 Micromolar. The best docking result has an inhibition constant value that is higher than the native ligand and has a binding free energy that is almost close to the binding free energy of the native ligand with an inhibition constant value of 42.38 Micromolar and a binding free energy value of 10.06 kcal-mol-1. Therefore, the results of the docking of the bioactive compound 3-oxo-N-(1,3-thiazol-2-yl) butanamide with the VEGFR-2 receptor are considered capable of being an alternative as a candidate for anti-cancer drugs.

Abstract

Sungkai (Peronema canescens Jack) is a medicinal plant in the Verbenaceae family that is known to have antidiabetic activity. The secondary metabolite content found in sungkai leaves is in the form of alkaloids, flavonoids, triterpenoids, steroids, phenolics, and saponins. This research aimed to identify the biological activity of compounds in sungkai plants as antidiabetic agents using α-glucosidase inhibitors in silico. Sungkai leaf extract with ethanol solvent was identified using a Liquid Chromatography Mass Spectrometer (LC-MS/MS). There are 15 compounds resulting from LC-MS/MS analysis which will then predict antidiabetic activity using molecular docking. Molecular docking was carried out using AutoDock Tools software and visualized with Discovery Studio. The highest scoring result obtained from the molecular docking test was the compound C13H23N6S with a free bond energy result of -7.31 and an inhibition constant value of 4.41 μM, which binds three hydrogen bonds in GLU 78, ALA 75, and GLU 198. Abstract

Sungkai (Peronema canescens Jack) merupakan tumbuhan obat yang banyak dimanfaatkan secara tradisional oleh masyarakat sebagai obat luka, diare, obat kumur dan malaria. Kegiatan pengabdian ini bertujuan untuk memberikan pemahaman khasiat daun sungkai secara saintifik melalui pelatihan pemanfaatan daun sungkai sebagai minuman herbal. Kegiatan pendampingan dimulai dari sosialisasi dan edukasi untuk peningkatan pemahaman serta pelatihan pengolahan daun sungkai menjadi teh berstandar dengan penambahan herbal lainnya guna meningkatkan keterampilan masyarakat dan diakhiri dengan survei untuk evaluasi kegiatan. Teh daun sungkai dibuat dalam empat variasi: variasi 1 (daun sungkai + gula aren + kayu manis), variasi 2 (daun sungkai + gula aren + lemon), variasi 3 (daun sungkai + kayu manis + madu) dan variasi 4 (daun sungkai + lemon + madu). Hasil observasi dan survei terhadap masyarakat terkait variasi teh sungkai, ditemukan bahwa masyarakat menyukai variasi teh sungkai mencapai 87 – 94% dengan tingkat preferensi 25% pada masing-masing varisi 1, 2, 3, dan 4. Kegiatan ini meningkatkan pengetahuan dan keterampilan masyarakat mencapai 95%. Sungkai (Peronema canescens Jack) is a medicinal plant traditionally used by the community as a medicine for caterpillars, diarrhea, mouthwash, and malaria. This activity aims to provide an understanding of the scientific efficacy of sungkai leaves through training in the use of sungkai leaves as herbal drinks. The mentoring activity begins with socialization and education to increase understanding and training in processing sungkai leaves into standardized tea with the addition of other herbs to improve community skills. It ends with a survey to evaluate the activity. Sungkai leaf tea is made in four variations: variation 1 (sungkai leaves + palm sugar + cinnamon), variation 2 (sungkai leaves + palm sugar + lemon), variation 3 (sungkai leaves + cinnamon + honey), and variation 4 (sungkai leaves + lemon + honey). The results of observations and surveys of the community regarding variations of sungkai tea found that the community liked the variation of sungkai tea, reaching 87 - 94% with a preference level of 25% for each variation 1, 2, 3, and 4. This activity increased community knowledge and skills by 95%.

Abstract

Secondary metabolite compounds contained in plants are bioactive substances related to the chemical content in plants, so that some plants can be used as medicinal materials. Putat (Planchonia valida) is a medicinal plant that has bioactive compounds that have potential as anticancer. Compound analysis of LC-MS test results has been carried out which shows several metabolite compounds contained in putat. The method used is analyzing compounds obtained from LC-MS results, screening compounds using pchembl and chemspider websites. And using the pChEMBL webserver to find out the bioactivity of these compounds. The purpose of this study is to determine and predict the potential bioactivity of the compounds contained therein as new drug candidates. The results obtained from this study are metabolite compounds in putat have the potential as anticancer based on the screening results carried out using the pChEMBL webserver by looking at the highest pIC50 value, obtained the highest pIC50 value of 8.18 with target proteins namely Serine/threonin mTOR and Cholinesterase which are target proteins that have anticancer activity.

Abstract

Coffee pulp is a by-product of coffee cherry processing. It has the potential to be used as a source of antioxidant activity. The dried coffee pulp is commonly referred to as cascara tea, which can be used as a functional drink. The coffee pulp contains caffeine, phenols, and chlorogenic acid that has antioxidant activity. Cascara tea can be fermented into kombucha as a functional drink that contains probiotics. In Jatimulyo Village, coffee cultivation and processing have been developed, but coffee skin waste is not used. Seeing the potential for processing coffee pulp waste into cascara and kombucha cascara tea products can provide added value (value added) to coffee pulp waste. The existence of added value to a product can increase the selling price in the market. The processing of coffee pulp waste into a product that is useful and has a sale value can be used as an effort to develop a creative economy. The results achieved from this activity are increasing the knowledge and skills of Jati Mulyo Village residents in utilizing coffee pulp waste into food products that are useful and have economic selling value.
