Abstract

Penelitian ini bertujuan untuk melakukan inventarisasi konsep dan praktik etnofarmasi Jamu Cekok pada pengobat tradisional di Kabupaten Ngawi. Kami menggunakan kombinasi metode kualitatif dan kuantitatif etnografi. Data kearifan lokal tentang konsep dan praktik etnofarmasi Jamu Cekok dikumpulkan melalui wawancara mendalam dan semi-terstruktur dengan informan lokal yang dipilih melalui purposive sampling. Hasil penelitian menunjukkan bahwa sebanyak 7 resep Jamu Cekok dengan menggunakan 11 spesies tumbuhan dimanfaatkan oleh masyarakat melalui berbagai pengolahan dan untuk berbagai macam perawatan medis. Informasi tersebut diwarisi dari generasi sebelumnya melalui penyuluhan dan pelatihan. Cara yang paling disukai memanfaatkan tanaman obat adalah dengan mengkonsumsi daunnya. Di antara semua tanaman obat yang teridentifikasi, kunyit (Curcuma longa L.) memiliki nilai SUV (1) tertinggi. Famili Zingiberaceae teridentifikasi memiliki nilai FUV tertinggi sebesar 0,71. Pengobat tradisional mempercayai konsep sehat sakit berdasarkan sebab personalistik dan naturalistik.

Abstract

This study evaluated phytochemical composition, and in vitro and in silico antibacterial activity of Elaeocarpus ganitrus extract. Elaeocarpus ganitrus leaves, seed and fruit powder were extracted with absolute ethanol. Then, the extract was identified phytochemical compounds qualitatively and evaluated the antibacterial activity through in vitro against Staphylococcus aureus and E. coli. Molecular docking was conducted to evaluate the antibacterial mechanism of Elaeocarpus ganitrus extract. Elaeocarpus ganitrus leaves, seeds, and fruits extract presented positive tannin, saponin, cardiac glycoside, quinone, steroids, terpenoids, and anthocyanins. In vitro analysis performed Elaeocarpus ganitrus leaves strong inhibited Staphylococcus aureus growth and medium inhibition against E. coli. structure activity relationship revealed 14 of 72 compounds have high antibacterial activities. molecular docking of 7 compounds showed inhibition activity of D-alanin ligase of Staphylococcus aureus. Those compounds blocked the activity of D-alanine ligase at inhibitor sites of enzyme, and might be disrupted the cell wall synthesis. In conclusion, Elaeocarpus ganitrus contained several phytochemical compounds and has antibacterial activity both in vitro and in silico investigation.

Abstract

Background: Genitri (Elaeocarpus ganitrus) is a local plant, growing well in tropical forests. Genitri seeds are used as jewelry. Utilization of genitri in the health sector is still lacking. Objectif: To analyze the potential of genitri leaves on the leukocyte profile and splenic histopathology of Wistar rats. Methods: Experimental true approach, using experimental male wistar rats aged 5-6 months, weighing 280-360 grams, totaling 24 rats. Experimental animals were placed in group cages at 27oC, 12 hours of light, fed and watered ad libitum. Acclimatization was carried out 7 days before treatment. Treatment P0 (negative control), P1 induced influenza virus (Vaxigrip trivalent) 0.5 ml (positive control), P2 induced influenza virus 0.5 ml and 1 gram of genitri leaves. Induction of virus from intramuscular vaccine. Induction was carried out on day 0 and day 14. Genitri leaves were administered using the oral gavage method on the 3rd day for 29 days. On the 33rd day, blood sampling was carried out through the heart and surgery and spleen retrieval. Total leukocyte count using a hemocytometer. Histopathologically examined using hematoxylin and eosin (HE) staining. Results: The total leukocyte count on the 19th day, P0 4263.82 cells/mm3, P1 6314.46 cells/mm3, P2 8443.72 cells/mm3. Total leukocytes on day 33, P0 4241.66 cells/mm3, P1 4983.33 cells/mm3, P2 7520.83 cells/mm3. Diameter of germinal center spleen on day 19, P0 231 nm, P1 186 nm, P2 202 nm. Germinal center diameter on day 33, P0 239 nm, P1 191 nm, P2 242 nm. Tissue structure of the P1 spleen was congested and haemorrhagic, the structure of the spleen P2 had leukocyte infiltration, the marginal zone and germinal center were thickened. Conclusion: genitri leaves can increase the number of leukocytes and the diameter of the splenic germinal center of Wistar rats .

Abstract

Osteoporosis adalah kondisi penurunan kepadatan mineral tulang dan berisiko patah tulang yang disebabkan oleh defisiensi estrogen. Fitoestrogen adalah senyawa pada tanaman yang memiliki kemampuan berikatan dengan reseptor estrogen untuk menggantikan fungsi estrogen. Daun M. crenata Presl. diketahui mengandung senyawa fitoestrogen. Tujuan penelitian ini adalah untuk membuktikan ekstrak etanol 96% daun M. crenata Presl. dapat meningkatkan jumlah sel osteoblas tulang trabekular femur dan vertebra pada mencit yang mengalami osteoporosis. Penelitian ini dilakukan dengan pemberian ekstrak etanol 96% daun M. crenata Presl. dosis 1,2; 2,4; 4,8; dan 9,6/20 g BB mencit/hari. Analisis dilakukan dengan cara pemeriksaan histomorfometri menggunakan pewarnaan HE, dan pengamatan sel osteoblas menggunakan mikroskop cahaya Olympus dengan software optilab. Data dianalisis menggunakan metode one way ANOVA. Hasil yang didapat yaitu kelompok PIV dengan dosis 9,6/20g BB mencit/hari dapat meningkatkan jumlah rerata sel osteoblas dengan nilai tertinggi yaitu 224,60 pada tulang trabekular femur dan 540,11 pada trabekular vertebra. Nilai perhitungan ED50 ekstrak etanol 96% daun M. crenata Presl. yaitu 1,908 mg pada tulang trabekular femur dan 2,878 mg pada tulang trabekular vertebra

Abstract

Latarbelakang: Antioksidan merupakan senyawa yang mampu berfungsi dalam menangkal radikal bebas serta meningkatkan sistem imun dan kekebalan tubuh. Selain itu, buah genitri memiliki efek farmakologis yang meliputi antiinflamasi, antidiabet, antimikroba.Tujuan: Untuk mengetahui nilai IC50 serta senyawa metabolit sekunder yang terkandung didalam buah genitri dengan pembanding asam askorbat.Metode:Menggunakan metode ekperimental laboratorium dengan pengujian antioksidan menggunakan metode DPPH. Hasil: Nilai IC50 dengan katagori sangat kuat  sebesar  40,75 mg/ml, sedangkan nilai IC50 asam askorbat berkatagori sangat kuat sebesar 1,96 mg/ml. Pada uji skrining fitokimia buah genitri mengandung senyawa alkaloid, flavonoid, dan tanin yang memiliki peranan penting dalam aktivitas antioksida. Analisis hasil uji one way anova yaitu nilai sig 0,048 0,05 yang menunjukan bahwa hasil tersebut terdapat perbedaan yang signifikansi antara aktivitas antioksidan buah genitri dengan asam askorbat. Kesimpulan: Dapat dijadikan sebagai penangkal radikal bebas dalam tubuh.

Abstract

The ethnopharmacological significance of Elaeocarpus longifolius (Jambu Wer), an indigenous medicinal plant of the Tengger Tribe, remains underexplored. This study evaluates the antibacterial and phytochemical properties of E. longifolius fruit extracts using ethanol, n-hexane, ethyl acetate, and chloroform as solvents. Antibacterial activity was assessed against Escherichia coli and Shigella dysenteriae, pathogens commonly associated with severe diarrhea, using the agar disk-diffusion method. Phytochemical screening was conducted using reagent-based tests and thin-layer chromatography (TLC). The ethanol extract exhibited the highest antibacterial activity, with inhibition zones exceeding 20 mm against both bacterial species. Phytochemical analysis revealed the presence of alkaloids, flavonoids, and tannins (compounds known for their pharmacological properties). These findings highlight the potential of E. longifolius ethanol extract as a promising natural therapeutic candidate for the development of Fitofarmaka targeting diarrheal diseases.

Abstract

Granule preparations are lumps of small particles that are uneven in shape and become single, larger particles. The process of making granule preparations uses a technique, namely granulation. In the granulation process, a binder is needed to unite the powder particles into a granule preparation. The binder material that is often used is polyvinyl pyrrolidone (PVP). The use of PVP is able to produce an optimal granule preparation that meets the requirements. This research reviews the use of PVP as a binding agent in making granule preparations. This research method uses a literature study method in the form of primary references in the form of national journals from the last 10 years searched in the Google Scholar, PubMed, and ScienceDirect databases. Search for this data source using the keywords Use of PVP as a binding agent in granule preparations. The results and conclusions of the research show that the use of PVP as a binding agent in granular preparations can produce preparations that meet the requirements.

Abstract

Black rice is a food crop with black pigments, which helps reduce the risk of various diseases and improve health. One way to improve food quality and avoid food contaminants is by irradiating. This study evaluated the phytochemical profile, antioxidant activity, and anticancer activity of irradiated black rice bran extract (IBRBE) in vitro and in silico. Black rice was irradiated with a Gamma cell 220 type irradiator at a 7.5 kGy/hour dose rate. Extraction of irradiated black rice bran was carried out using the maceration method. Phenolic and flavonoid components were quantified using the Folin–Ciocalteau and AlCl3 methods, respectively. Phytochemical compounds were identified by liquid chromatography-high-resolution mass spectrometry (LC HRMS). The antioxidant activity of IBRBE was carried out against 2,2-diphenyl-1picrylhydazyl (DPPH). The cytotoxic activity of IBRBE against WiDr cells (colorectal cancer) and Vero (nonhuman cell lines) used the MT test method. Prediction of the inhibitory mechanism of compounds in the extract against target proteins EGFR and GPX7 was carried out in silico. Total phenolics and flavonoids were 2.57 ± 0.28 mg GAE/g and 19.12 ± 0.18 μg QE/ml, respectively. Twenty-four types of active compounds were obtained in IBRBE. The results of antioxidant activity obtained an IC50 value of 1198.45 ± 92.86 μg/ml. IC50 in WiDr cells and Vero cells were obtained at 36.08 ± 11.71 μg/ml and 570.58 ± 130.25 μg/ml, respectively. In silico results, the compound 4 Dodecylbenzene sulfonic acid has the highest binding affinity to the EGFR protein, with a value of -5.9. Meanwhile, the Monoolein compound has the highest binding affinity to the GPX7 protein, with a value of -5.4.

Abstract

Gel merupakan salah satu sediaan topikal yang memiliki keuntungan seperti bewarna jernih, memberikan sensasi dingin, tidak berminyak, mudah menyebar di kulit sehingga dapat maksimal memberikan efek teraupetik. Gelling agent merupakan komponen penting dalam pembentukan gel. Kitosan merupakan polimer alam yang dapat membentuk gel pada larutan asam. Sedangkan karbopol 940 dapat menghasilkan gel bewarna jernih dan stabilitasnya baik selama penyimpanan. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh kombinasi karbopol 940 dan kitosan pada sediaan gel ekstrak daun kelor terhadap karakteristik fisik dan uji stabilitas selama 4 minggu. Pembuatan gel ekstrak daun kelor menggunakan ekstrak etanol daun kelor sebesar 5%. Kombinasi gelling agent yaitu karbopol 940 1% dan kitosan 0,5%, 1%, 1,5%. Uji karakteritik dan stabilitas meliputi organoleptis, homogenitas, pH, viskositas, daya sebar dan stabilitas selama 4 minggu. Hasil menunjukkan gel ekstrak daun kelor bewarna coklat tua, kental, bau khas daun kelor serta homogen dan tidak mengalami perubahan signifikan selama disimpan 28 hari. Nilai Uji pH sediaan gel ekstrak daun kelor tidak mengalami perubahan setelah disimpan selama 4 minggu (4,94-5,20) menjadi 5,09-5,65). Stabilitas viskositas sediaan gel ekstrak daun kelor mengalami penurunan dari minggu ke-1 2,964 cps-4420 cps menjadi 2,438 cps-3556 cps setelah minggu. Pada uji daya sebar terjadi peningkatan setelah disimpan selama 4 minggu (4,2 cm- 5,9 cm menjadi (5,1 cm – 6,9 cm). Berdasarkan hasil tersebut semua formula gel kitosan-karbopol 940-ekstrak daun kelor telah memenuhi persyaratan.
