Abstract

Roselle (Hibiscus sabdariffa L.) has been known as traditional medicinal plants. It needed to evaluate the safety of roselle extract on long-term oral administration. The research aimed to determine the savety of ethanolic extract of roselle calyx. Subchronic toxicity study of ethanolic extract of roselle calyx had been carried out on 60 Sprague Dawleys (SD) rats for 28 days. The roselle extract was administered orally every day, with doses of 50,100 and 200 mg/kgBW. There are 2 satellite groups, kept for another 14 days after the treatment in order to detect a delayed occurrence of toxic effect. Satellite group was given extract 200 mg/kgBW for 28 days and followed aquadest treatment for 14 days. At the end of experiment the blood was collected for meansuring SGPT, SGOT and ALP activity. The result showed that treatment of roselle calyx extract had no significant changes in SGPT SGOT, ALP activity. The delayed effect was not also observed. The ethanolic extract of roselle calyx didnt show toxic effects on the liver on subchronic administration.

Abstract

This study aims to determine the antibacterial activity and TLC-bioautography profile of the active fractions of Muntingia calabura L. leaves against Staphylococcus aureus. Muntingia calabura L. leaves were macerated with ethanol 96% then fractionated with n-hexane, ethyl acetate, and methanol solvent, respectively. The antibacterial activity was tested by the Kirby-Bauer method to determine the most active fraction and the lowest concentration that inhibited the growth of Staphylococcus aureus. TLC-bioautography was tested using chloroform: ethyl acetate (2:8) as the mobile phase and silica gel F254 as the stationary phase. Antibacterial activity test of n-hexane and ethyl acetate fractions at a concentration of 10% w/v showed activities with inhibition zone diameter of 0.330.288 and 9.665.77 mm, respectively. At the same time, the methanol fraction showed no activity. The lowest concentration of ethyl acetate fraction which still showed the inhibition zone was 0.312% w/v. The TLC-bioautography profile showed active spots with an Rf value of 0.82 and had an inhibitory zone diameter of 4.0130.864 mm. It can be concluded that the ethyl acetate fraction was the most active fraction that inhibited the growth of Staphylococcus aureus and had one active spot on the bioautography test.

Abstract

Free radicals were reactive species caused oxidation of lipids membrane and DNA. Superoxide dismutase (SOD) is one of the primary antioxidants to inhibit free radicals. The purpose of this study was to determine the effect of ethanol extract of roselle calyx on SOD enzyme activity of the Sprague Dawley (SD) rats induced by 7.12-dimethylbenzen[a]anthracene (DMBA). The animals age of four weeks divided in to five groups, groups I were normal group. Group II is the negative control group only induced by DMBA 75 mg/kgBW; and group III ,IV , and V were treatment groups that treated by ethanolic extract of roselle calyx at dose of 10, 50 and 100 mg/kgBW/day for 7 days. After treatment with roselle, animals were induced by DMBA 75 mg/kgBW. On day 8 after DMBA induction, animals were fasted for 16 hours and blood was collected to measured SOD activity. Data were analyzed using One Way Analysis Of Variance (ANOVA) and LSD (P <0.05). The result showed the significancies on increasing of SOD activity at 10 mg/kgBW (50.87±1.98), 50 mg/kgBW (69.98±3.58) and 100 mg/kgBW (73.01±6.95) compared with DMBA (43.74±1.95) (p<0,05).

Abstract

The administration of high-fat diets can increase the body's lipid level and damage the organs. Purple sweet potato leaf (Ipomoea batatas L.) was reported as an antioxidant against free radicals. This study aimed to observe the sweet potato leaf extract's activity on decreasing lipid profile and hepatoprotective effect in high-fat diet fed rats. The treatment animals were divided into five groups, namely normal control, high-fat diet (HFD) control, the treatment group of purple sweet potato leaf extract (SPLE) doses 100 mg/kg BW, 200 mg/kg BW and 400 mg/kg BW which fed with high-fat diet for 14 days and SPLE for 28 days. After treatment was completed, the blood was collected for the detection of cholesterol, triglyceride, serum glutamic oxaloacetic transaminase (SGOT), and serum glutamic pyruvate transaminase (SGPT). After that, the animals were sacrificed, and a liver histopathology observation was conducted using Haematoxylien and Eosin staining. The result showed a significant decrease in cholesterol and triglyceride levels (p≤0.05) compared to the negative group in all treated groups. The SGOT and SGPT enzymes in all of treatment groups were also found to decrease compared with HFD control. The result was confirmed by the histopathological observations. The finding suggested the potency of SPLE for antihyperlipidaemic and hepatoprotective agent.

Abstract

Radikal bebas bersifat reaktif yang menyebabkan penyakit degeneratif sehingga tubuh memerlukan antioksidan eksogen. Salah satu contohnya adalah buah Jamblang (Syzygium cumini (L)) yang memiliki aktivitas antioksidan tinggi karena antosianin alaminya.. Antosianin bersifat lebih stabil dalam suasana asam sehingga cocok jika ekstrak buah jamblang difortifikasi dalam sediaan yoghurt. Yoghurt merupakan produk fermentasi susu yang menggunakan bakteri asam laktat yaitu: Streptococcus thermophilus, Lactobacillus bulgaricus, Lactobacillus acidophilus. Tujuan penelitian ini untuk mengetahui kadar antosianin ekstrak buah jamblang pada formulasi yoghurt dengan konsentrasi 0%, 5%, 7,5% dan 10% serta syarat mutu SNI. Pengujian meliputi uji sifat fisika, kimia dan mikrobiologi yaitu sensoris, viskositas, kadar asam, kadar lemak, dan jumlah starter bakteri, serta uji stabilitas yoghurt dengan penyimpanan pada suhu 4°C dan uji total antosianin pada hari ke-1, ke-7, ke-14, 21 dan hari ke-28. Hasil penelitian menunjukkan bahwa kadar antosianin total pada formula yoghurt terfortifikasi ekstrak buah jamblang pada konsentrasi 10% (60.87±2.24 mg/L) menghasilkan stabilitas antosianin lebih baik, sesuai dengan persyaratan SNI.

Abstract

Permasalahan kulit tidak hanya merugikan kesehatan fisik namun dapat juga menimbulkan masalah psikologi, terutama jika penyakit kulit ini terjadi di wajah. Pemeriksaan yang dilakukan oleh dokter saat ini hanya mengandalkan penglihatan objek mata dan riwayat penyakit pasien, sehingga memungkinkan terjadinya kesalahan pada analisis dan diagnosa penyakit. Penelitian ini dilakukan untuk menghasilkan sistem klasifikasi penyakit kulit wajah menggunakan metode convolutional neural network (CNN). Data yang digunakan pada penelitian ini sebanyak 500 sampel data citra dengan 5 Jenis penyakit kulit wajah yaitu jerawat, eksim, milia, herpes, dan melasma. Pada penelitian ini proses klasifikasi menggunakan metode CNN. Arsitektur model yang digunakan adalah VGG16. Dalam pemodelan VGG16 terdapat dua lapisan yaitu feature extraction dan classification. Di dalam feature extraction terdapat convolution layer menggunakan 3 layer dengan kedalaman filter yang berbeda pada setiap layernya. Fungsi aktivasi ReLu dan avarage pooling digunakan untuk mengurangi feature map. Lapisan classification terdapat fully connected layer dan softmax digunakan untuk klasifikasi. Metode CNN yang digunakan pada penelitian ini memperoleh hasil yang sangat baik, dimana didapat hasil proses pelatihan sebesar 98% dan hasil validasi 88% dengan data pealtihan sebanyak 325 citra dan data validasi sebanyak 125. Hasil akurasi yang didapat pada pengujian terhadap data baru sebesar 90% dengan data uji sebanyak 50 citra. Dengan demikian, dapat dikatakan hasil yang diperoleh pada pengujian penelitian ini sangat baik.

Abstract

Parsea americana Mill. is a natural resource that has been studied for its antibacterial properties. The pulp, peel, and seed of Parsea americana Mill. have potential as an antibacterial agent. This study aimed to determine the inhibitory activity and phenolic content of Parsea americana Mill. peels extract and fraction against Staphylococcus aureus ATCC 25923. Parsea americana Mill. was macerated with 96% ethanol and then fractionated with n-hexane, ethyl acetate, and methanol solvent. Determination of the fraction that has the greatest antibacterial activity against S. aureus was carried out using the Kirby Bauer method.  The total phenolic content in the extract and fraction was calculated as gallic acid equivalent (GAE) using the Folin-Ciocalteu method spectrometrically. Antibacterial activity test of the 96% ethanol extract, ethyl acetate fractions, and methanol fractions at a concentration of 10% w/v showed activity with a measurable inhibition zone. On the other hand, the n-hexane fraction showed no inhibition zone. The highest inhibition zone was the ethyl acetate fraction with approximately 8.33 ± 0.58 mm. The ethyl acetate fraction of Parsea americana Mill. resulted in 536.26 ± 14.29 mg GAE/g fraction. The conclusion was that the ethyl acetate fraction had the highest total phenolic content and was the most active fraction in inhibiting the growth of Staphylococcus aureus.

Abstract

Tujuan dari penelitian ini adalah untuk mengetahui gambaran keterampilan motorik halus anak usia 5-6 tahun. Harapannya, hasil penelitian ini bisa dijadikan dasar dalam memberikan intervensi yang sesuai kepada peserta didik. Penelitian ini melibatkan 21 subjek pada sebuah Taman Kanak-Kanak. Identifikasi dilakukan dengan metode observasi selama 3 hari dan wawancara kepada guru, sehingga dapat dilakukan triangulasi sumber. Identifikasi keterampilan motorik dalam penelitian ini berfokus pada kegiatan berikut: menganyam, mewarnai gambar sederhana, menggambar kolase, menggunting, meniru lipatan kertas sederhana, melukis dengan jari, dan garis meniru. Kemudian, hasilnya dianalisis dengan menggunakan statistik deskriptif dengan 4 kategorisasi. Berdasarkan hasil analisis, ditemukan bahwa dari 21 anak dapat dikategorikan sebagai berikut: rata-rata 67,5% anak berada pada kategori Belum Berkembang (BB), rata-rata 28,5% anak masuk dalam kategori Mulai Berkembang (MB), kategori 4% Berkembang Sesuai Harapan (BSH), dan 0% anak berada dalam kategori Berkembang Sangat Baik (BSB). Dengan demikian, lebih dari 75% peserta didik di kelompok ini memerlukan intervensi berupa stimulasi motorik halus dengan strategi dan media yang menarik.

Abstract

Perception is an individual response about something that can be influenced by knowledge. Good individual knowledge of alcohol will affect their perception of alcohol-containing product.  The MUI fatwa permits the use of alcohol in cosmetics. The purpose of this study was to observe the perception of alcohol content in cosmetics. The observation was carried out using a questionnaire technique with a nonprobability sampling technique with accidental sampling. The questionnaire was conducted by Pearson correlation test, validity test, reliability test, normality test, chi-square test, and description. In this study, using 98 respondents and get an average value of knowledge about cosmetics 6,73 with SD value of 0,49; knowledge of alcohol 4,54 with SD 1,17 and behavior with SD 1,46. There is a relationship between age with the level of cosmetic knowledge with p-value <0.05. As many as 46.94% stated, the halal label's urgency on cosmetics is very important and 39.80% is important, where the religion of the individual influences the urgency level of cosmetics halal. The urgency of halal labels on cosmetics is high in line with the behavior when buying cosmetics. The 81.63% of respondents stated that halal labels on cosmetic products affect the interest in buying cosmetics. As many as 61.24% of respondents have the perception that halal cosmetics do not contain alcohol. The Pearson halal label correlation test confirms this obtained a p-value value of 0.024 that the halal label influences respondents' perceptions. Individuals were having a perception that cosmetics with halal labels do not contain alcohol, which is influenced by halal labels' knowledge.

Abstract

Javanese women often consume kunir asam to relieve dysmenorrhea symptom. It is a traditional herbal medicine which is conventionally prepared from curcuma domestica. One of the active compound of curcuma domestica is curcumin. The purpose of this research is to determine the amount of curcumin available in kunir asam selling in daily market of Kota Gede. Curcumin level in daily batches consistency was also evaluated. Curcumin analysis was conducted by qualitative and quantitative tests by TLC densitometry method. A total of 50.0 ml kunir asam was extracted with 25.0 ml of chloroform, and then the mixture was dried by evaporation. The dried residue of extract was then reconstituted in 5.0 ml of 96% ethanol. 5 μl of sample supernatant and standard solutions spotted on the plates of silica gel 60 F254 that has been activated previously. The organic solvent mixture comprise of chloroform: ethanol: glacial acetic acid (94:5:1) was used as the mobile phase for elution, and the spots were then analyzed by using densitometry at wavelength of 425 nm. Curcumin levels was calculated by inputting the size of spot area data into the standard curve linear regression equation of curcumin concentration versus the size of spot area. The data analysis was performed by normality test Kolmogorov-Smirnov test, Levene test of homogeneity followed by Post Hoc / Test LSD (Least Significant Difference) or Mann Whitney. TLC analysis conducted on four samples of “kunir asam” displayed a disparities on the curcumin level between the three days of sampling in a row. Statistical analysis of the curcumin content in traditional herbal medicine of “kunir asam” samples showed that the levels were not consistent in the three days of the examination. Curcumin levels in all four samples were also shows a significant difference in variations.
