Abstract

This research aims to evaluate the safety of ethanolic extract of Acalypha hispida (A. hispida) leaves with acute toxicity test using 15 female rats strain Sprague-Dawley. A single dose of different doses of extract (2, 4, 8, and 16 g/kg body weight) was administrated orally, and theobservation was conducted for 14 days. The results revealed that the ethanolic extract of A. hispida leaves was relatively harmless (LD50 16 g/kg BW), did not affect body weight, and did not show clinical signs of toxicity during the observation periods. The parameters of blood serumbiochemistry of all extract-treated groups (alanine aminotransferase, aspartate aminotransferase, creatinine, and urea) did not change significantly  compared to the control group. The histological observation of the liver showed a significant increase in eosinophilic cytoplasm and basophilic nuclei at all doses. However, the ethanolic extract of A. hispida leaves did not significantly affect glomerulus/Bowman’s capsule ratio, glomerular cell density, and the proportion of normal cell tubule. In conclusion, the ethanolic extract of A. hispida leaves was relatively harmless with LD5016 g/kg BW and seems to be safe in low doses (2 g/kg BW).

Abstract

Gentavar® merupakan produk antibiotika topikal yang dapat digunakan untuk mengurangi infeksi bakteri dalam penyembuhan luka terbuka Penelitian ini bertujuan untuk menguji efektifitas kandidat obat penyembuh luka pada hewan model tikus. Tujuan lain penelitian ini adalah untuk menguji secara klinis sediaan yang digunakan pada hewan anjing. Hewan coba tikus dibuat model luka terbuka dengan metode insisi. Luka sayat dibuat sepanjang 1 cm pada bagian punggung tikus. Kemudian liakukan pengolesan sediaan. Dilakukan observasi selama 14 hari. Parameter yang diamati berupa kondisi luka dan histopatologi. Penelitian pada anjing dilakukan dengan mengoleskan sediaan pada luka terbuka anjing. Pengamatan dilakukan selama 7 hari terhadap kondisi luka. Persembuhan lebih cepat ditemukan apda kelompok tikus yang dioleskan sedian dua kali dosis dibandingkan dengan kontrol positif. Perbaikan yang ditemukan terlihat dari proporsi isa luka, pengamatan kondisi luka dan histopatologi. Pada hewan anjing ditemukan persembuhan lebih cepat ditemukan pada kelompok yang diberikan sediaan salep uji dibandingkan dengan kelompok yang tidak diberikan sediaan. Sediaan Gentavar® memiliki khasiat dalam penyembuhan luka lebih efektif dibandingkan dengan kontrol positif. Efek maksimal ditemukan pada pemberian dua kali dosis sediaan.

Abstract

The purpose of the study was to examine the safety of combination of basil, turmeric and bean sprouts extract solution on the hematological parameters and blood biochemistry profile of rats. The combined ingredients used in the study were obtained from different places. A total of 15 rats were divided into 3 groups based on the treatment dose. Each group consisted of 5 rats. The rats in control group (K) did not given extract combination, while the rats in group KKT 1 % and KKT 5% received combination of basil, turmeric and bean sprouts extract at dose of 1% and 5%, respectively. Effectiveness and safety tests were carried out by evaluating the hematological and blood biochemical profiles of female rats. The data were analyzed using one-way analysis of variance (ANOVA). The results showed that the rats given combination of basil, turmeric and bean sprouts extract at a dose of 1% and, dose of 5% did not had significant different compared to control (P0.05), but tend to have positive effects in increasing  several blood components that play an important role in maintaining immunity during pregnancy. It can be concluded that the combination of basil, turmeric and bean sprouts at dose of 1% and 5% do not adversely affect hematology profile and blood biochemistry of rats. This indicates that the extract combination does not cause any toxicity effects on the rats.

Abstract

Rambut berfungsi untuk memberikan kehangatan, perlindungan, keindahan serta penunjang penampilan. Kerontokan rambut yang dapat mengakibatkan kebotakan merupakan salah satu problema yang paling dikhawatirkan setiap orang. Kebotakan biasanya diobati dengan mengkonsumsi obat tertentu namun keamanan dari obat-obatan tersebut belum tentu terjamin. Bahan alami menjadi salah satu alternatif yang dapat dipilih, diantaranya adalah sari buah apel (Malus domestica). Penelitian ini bertujuan menguji dan mempelajari efektivitas sari buah apel sebagai penumbuh rambut serta menganalisis frekuensi pengolesan terbaik dengan parameter pertumbuhan rambut dan reaksi iritasi pada tikus (Rattus norvegicus). Sebanyak 6 ekor tikus dibagi menjadi 4 perlakuan pada masing-masing tikus yaitu kontrol (tidak diberi olesan), 1 kali oles sehari, 2 kali oles sehari, dan 3 kali oles sehari. Pengukuran panjang rambut tikus dilakukan pada hari ke-6, 9, 12, dan 15 serta menganalisis skor eritema dan edema atau Primary Dermal Irritation Index (PDII). Hasil penelitian menunjukkan sari buah apel memiliki efek yang signifikan sebagai pemacu pertumbuhan rambut. Pemberian olesan sebanyak 3 kali sehari menjadi kelompok dengan frekuensi pemberian olesan terbaik karena memiliki perbedaan nyata terhadap kelompok kontrol. Tidak ada reaksi iritasi yang ditimbulkan dari sari buah apel saat dilakukan uji iritasi pada tikus.

Abstract

Arabica green coffee bean is the most widely produced coffee bean in Indonesia. Studies have shown that arabica green coffee beans are rich in polyphenols and antioxidants, stimulating immune cell proliferation. This study investigated the immunomodulatory activity of arabica green coffee beans (Coffea arabica). Thirty mice (Mus musculus) with an average body weight of 30 g were divided into five groups: negative control, positive control, and groups administered arabica green coffee bean powder extract at doses of 0.03, 0.06, and 0.18 mg/30 g BW. It was orally administered once daily for 14 days. On the 15th day, the mice were intraperitoneally injected with non-pathogenic Staphylococcus aureus. After an hour, mice were necropsied for peritoneal fluid collection. Peritoneal fluid was stained with a peripheral blood smear, and the number of macrophages and lymphocytes was observed under a microscope. Data were analyzed using one-way analysis of variance (ANOVA), followed by Tukey’s test. The group administered with 0.18 mg/30 g body weight (BW) dose showed the highest average of macrophage and lymphocytes compared to the negative control group and any other groups. Arabica green coffee bean powder extract was shown to have immunomodulatory activity, with the highest activity observed at a dosage of 0.18 mg/30 g BW.

Abstract

Latar Belakang: Ayam kampung merupakan salah satu kebutuhan protein hewani yang sering dicari masyarakat. Pemeliharaan ayam kampung dengan sistem dibiarkan lepas menjadi predisposisi terinfeksi oleh parasit gastrointestinal. Tujuan: Penelitian ini bertujuan mengkaji perubahan sel epitel dan hipertrofi sel goblet pada saluran pencernaan ayam kampung yang terinfeksi cacing cestoda Raillietina spp. secara alami. Metode: Penelitian ini menggunakan tujuh preparat arsip histopatologi usus halus ayam kampung yang terinfeksi cacing cestoda. Sampel usus halus ayam kampung diambil dari dua pasar yang berbeda, yaitu pasar Pluit di Jakarta Utara dan pasar Kebayoran Lama di Jakarta Selatan. Hasil: Hasil pengamatan histopatologis pada preparat usus ditemukan deskuamasi epitel vili dan proliferasi sel-sel kripta yang diakibatkan oleh infeksi cacing cestoda. Sel goblet yang mengalami hipertrofi paling banyak ditemukan pada sampel preparat yang diambil dari pasar Kebayoran Lama, Jakarta Selatan (p>0,05). Simpulan: Dapat disimpulkan bahwa deskuamasi vili dan jumlah hipertrofi sel goblet terjadi akibat infeksi cacing cestoda pada mukosa usus halus ayam kampung.

Abstract

Ilmu forensik veteriner belum mendapat perhatian yang memadai di Indonesia, sehingga perlu ditingkatkan pengembangannya. Penelitian ini bertujuan mempelajari ilmu forensik veteriner melalui histopatologi hati ayam broiler 48 jam pasca kematian. Ayam broiler sebanyak 75 ekor berumur 7 hari dieutanasia dan dibagi menjadi 25 kelompok berdasarkan jam nekropsi pasca-eutanasia (n=3). Kadaver ayam dinekropsi setiap 2 jam postmortem, organ hati dikoleksi untuk dibuat potongan histopatologi dan diwarnai dengan Hematoksilin-Eosin (HE). Histopatologi hati evaluasi dan hasil yang didapat dianalisis menggunakan software ImageJ versi 1.53a. Hubungan peubah histopatologi dengan waktu postmortem dianalisis dengan metode Pearsons. Hasil penelitian menunjukkan bahwa pembusukan dimulai pada jam ke-18 post mortem yang ditandai dengan jumlah inti sel hepatosit, jam ke-20 ditemukan bakteri pembusuk, dan jam ke-22 terjadi peningkatan jarak antar hepatosit. Berdasarkan nilai korelasi Pearson, jumlah inti sel hepatosit, jarak antar hepatosit dan keberadaan bakteri pembusuk mempunyai hubungan kuat hingga sangat kuat dengan waktu postmortem sehingga hasil ini dapat menjadi parameter penentu waktu kematian (Post Mortem Interval/PMI) pada hewan.

Abstract

The aim of this research was to determine the effectiveness of sugar dressing treatment from Lawang sugar cane on incision wound healing process of rabbit skin. Nine New Zealand white rabbits were divided into three treatment groups with three rabbits per group. In negative control group (KN), the wounds were only treated with distilled water, while in SD60 and SD100 groups, the wounds were treated with 60% and 100% sugar solution, respectively. The treatment was given once daily for 7 days. The results showed that the sugar dressing treatments have significant effect (P0.05) on the size of the wound area and increased the collagen density, but did not have significant effect on the remaining length of the wound.Based on the results of the study, it can be concluded that sugar dressing dressing treatment from Lawang sugar cane is effective in increasing collagen density in the wound healing process.

Abstract

Currently, the use of calf mesenchymal stem cell Wharton's jelly (MSCWJ) could be an alternative for pet animal therapy. The benefits of MSC included regulating cell signaling pathways, low immune system rejection in allogeneic administration, no risk of malignancy, stable storage, and easy transportation. Calf umbilical cord culture had not been reported on the development of WJ explants, especially the growth process of WJ explants from attach to confluent. The aim of the study was to determine the development of calf umbilical cord WJ explants. Factors examined included explant attachment, outgrowth appearance, cell morphology, and cell confluence. Explants were able to attach as much as 42% on day 8 of culture and 88% (day 12). Cell outgrowth was observed in 45% of explants (day 8) and 100% (day 12). The visible cell morphology was fibroblastic. Cell confluence was obtained on day 12, day 16, and day 20 at 3%, 83%, and 100%, respectively

Abstract

Probiotics are viable microorganisms that promote health benefits by balancing positive microbes in the gut, and controlling pathogenic microbes in the digestive tract. Probiotic from genus Lactobacillus is the most widely used. In addition to effectiveness testing, development and research on probiotics need to be supplemented with evidence from clinical and non-clinical data. This study was aimed to collect non-clinical data by examining the effectivity and toxicity of a probiotic candidate Lacticaseibacillus rhamnosus MG5511 from BIma horse milk (KRG) in the intestine and their effect on the intestinal mucosa. Animal study was conducted with administration of the specific strain in MRS broth in 107 to 1010 CFU/mL concentrations to mice. Parameters observed were clinical signs, daily body weight measurement, gross organ pathology, and growth of bacterial populations. Based on the results, the strain Lacticaseibacillus rhamnosus MG5511 was able to survive the gastrointestinal tract for 14 days and can be classified as relatively harmless based on the results of toxicity test.
