Abstract

Arabica green coffee bean is the most widely produced coffee bean in Indonesia. Studies have shown that arabica green coffee beans are rich in polyphenols and antioxidants, stimulating immune cell proliferation. This study investigated the immunomodulatory activity of arabica green coffee beans (Coffea arabica). Thirty mice (Mus musculus) with an average body weight of 30 g were divided into five groups: negative control, positive control, and groups administered arabica green coffee bean powder extract at doses of 0.03, 0.06, and 0.18 mg/30 g BW. It was orally administered once daily for 14 days. On the 15th day, the mice were intraperitoneally injected with non-pathogenic Staphylococcus aureus. After an hour, mice were necropsied for peritoneal fluid collection. Peritoneal fluid was stained with a peripheral blood smear, and the number of macrophages and lymphocytes was observed under a microscope. Data were analyzed using one-way analysis of variance (ANOVA), followed by Tukey’s test. The group administered with 0.18 mg/30 g body weight (BW) dose showed the highest average of macrophage and lymphocytes compared to the negative control group and any other groups. Arabica green coffee bean powder extract was shown to have immunomodulatory activity, with the highest activity observed at a dosage of 0.18 mg/30 g BW.

Abstract

Latar Belakang: Ayam kampung merupakan salah satu kebutuhan protein hewani yang sering dicari masyarakat. Pemeliharaan ayam kampung dengan sistem dibiarkan lepas menjadi predisposisi terinfeksi oleh parasit gastrointestinal. Tujuan: Penelitian ini bertujuan mengkaji perubahan sel epitel dan hipertrofi sel goblet pada saluran pencernaan ayam kampung yang terinfeksi cacing cestoda Raillietina spp. secara alami. Metode: Penelitian ini menggunakan tujuh preparat arsip histopatologi usus halus ayam kampung yang terinfeksi cacing cestoda. Sampel usus halus ayam kampung diambil dari dua pasar yang berbeda, yaitu pasar Pluit di Jakarta Utara dan pasar Kebayoran Lama di Jakarta Selatan. Hasil: Hasil pengamatan histopatologis pada preparat usus ditemukan deskuamasi epitel vili dan proliferasi sel-sel kripta yang diakibatkan oleh infeksi cacing cestoda. Sel goblet yang mengalami hipertrofi paling banyak ditemukan pada sampel preparat yang diambil dari pasar Kebayoran Lama, Jakarta Selatan (p>0,05). Simpulan: Dapat disimpulkan bahwa deskuamasi vili dan jumlah hipertrofi sel goblet terjadi akibat infeksi cacing cestoda pada mukosa usus halus ayam kampung.

Abstract

The aim of this research was to determine the effectiveness of sugar dressing treatment from Lawang sugar cane on incision wound healing process of rabbit skin. Nine New Zealand white rabbits were divided into three treatment groups with three rabbits per group. In negative control group (KN), the wounds were only treated with distilled water, while in SD60 and SD100 groups, the wounds were treated with 60% and 100% sugar solution, respectively. The treatment was given once daily for 7 days. The results showed that the sugar dressing treatments have significant effect (P0.05) on the size of the wound area and increased the collagen density, but did not have significant effect on the remaining length of the wound.Based on the results of the study, it can be concluded that sugar dressing dressing treatment from Lawang sugar cane is effective in increasing collagen density in the wound healing process.

Abstract

Saat ini penggunaan sel punca mesenkimal Wharton’s jelly (SPMWJ) sapi dapat menjadi alternatif bagi terapi pet animals. Manfaat SPM antara lain meregulasi jalur persinyalan sel, rendahnya penolakan sistim imun pada pemberian alogenik, tidak adanya resiko keganasan, stabil penyimpanannya, dan mudah transportasinya. Kultur tali pusat sapi belum dilaporkan perkembangan eksplan WJ terutama proses pertumbuhan eksplan WJ mulai dari attach hingga sel konfluen. Tujuan penelitian adalah menentukan perkembangan eksplan WJ tali pusat sapi. Faktor yang dilihat diantaranya attachment eksplan, munculnya outgrowth, morfologi sel, dan konfluensi sel. Eksplan mampu attach sebanyak 42% pada hari ke-8 kultur dan 88% (hari ke-12). Outgrowth sel teramati pada 45% eksplan (hari ke-8) dan 100% (hari ke-12). Morfologi sel yang terlihat adalah fibroblastik. Konfluensi sel didapat pada hari ke-12, hari ke-16, dan hari ke-20 yaitu 3%, 83%, dan 100%.

Abstract

Probiotik merupakan mikroorganisme hidup yang dapat memberikan efek kesehatan dengan menyeimbangkan mikroba positif usus, serta mengendalikan mikroba patogen pada saluran pencernaan. Probiotik genus Lactobacillus merupakan jenis probiotik yang paling banyak digunakan. Selain uji efektivitas, pengembangan dan penelitian mengenai probiotik perlu dilengkapi dengan bukti dari data klinis dan nonklinis. Penelitian ini bertujuan mengoleksi data nonklinis dengan menguji efektivitas dan toksisitas kandidat probiotik Lacticaseibacillus rhamnosus MG 5511 dari susu kuda BIma yang selanjutnya disingkat KRG dalam usus, serta efeknya terhadap mukosa usus. Penelitian di hewan dilakukan dengan memasukkan galur spesifik tersebut dalam kaldu MRS dengan konsentrasi 107 sampai 1010 CFU/mL ke mencit. Parameter yang diamati berupa pengamatan tanda toksik dan klinis, pengamatan berat badan secara harian, gross patologi organ, dan koleksi sampel isi usus untuk mengamati jumlah populasi bakteri yang tumbuh. Berdasarkan hasil penelitian, galur murni Lacticaseibacillus rhamnosus MG5511 mampu sintas sampai ke saluran pencernaan sampai 14 hari dan termasuk dalam kategori relatif tidak membahayakan berdasarkan hasil uji toksisitas.

Abstract

An in vivo test study to prove its effectiveness as an antihyperuricemia requires an animal model with a hyperuricemic condition. Foods that contain high purine bases have the potential to increase uric acid levels in the blood, including chicken liver and melinjo seeds. This study aims to determine the effectiveness of the combination of chicken liver juice and melinjo seed powder in producing hyperuricemia conditions with the most optimal onset and duration in rats in vivo. Six groups of white Sprague Dawley rats (n=5) consisted of a normal group, a positive control group, a chicken liver juice group, a melinjo seed powder group and two groups that were given a combination of chicken liver juice and melinjo seed powder with different compositions. The treatment was given every day for 15 days orally. The parameters observed were uric acid levels which were measured every 3 days and, body weight. The results showed that the combination of chicken liver juice and melinjo powder with a half dose of a single dose each had a better onset and duration. The conclusion of this study is that the combination of chicken liver juice and melinjo seed powder is potent as an inducer of hyperuricemia in rats.

Abstract

The objective of this research was to analyze the effect of olein and super olein palm oil on blood lipid profile (total cholesterol, LDL cholesterol, HDL cholesterol, and triglyceride) and fatty liver condition of Sprague Dawley rats. The design used was pre and post test with control group design. The intervention run for 8 weeks using 20 rats which were divided into four treatments i.e: (1) standard-low fat feed and water (negative control/KN), (2) standard-low fat feed and dried egg yolk (positive control/KP), (3)standard-low fat feed and olein palm oil (B), and (4) standard-low fat feed and super olein palm oil (A). Results showed that blood lipid profile of rats at baseline were not different among treatment groups, while at midline super olein group has total cholesterol, triglyceride, and indirect LDL cholesterol lower than olein group. At endline, HDL and indirect LDL cholesterol of super olein group were different compared to olein group, while total cholesterol and triglyceride were not different between both groups. Super olein group had HDL cholesterol higher and indirect LDL cholesterol lower than olein group. Based on histopathology, lipid accumulation in the liver of Sprague Dawley rats in super olein group were lower than in olein group. The data showed that percentage of lipid droplet in super olein group (A) was 22.01%, while in olein group (B) was 40.43%.Keywords: blood lipid profile, fatty liver, palm oil, rats

Abstract

The purpose of this study was to determine the effectiveness of stingless bee honey in healing incision wounds on rat skin based on collagen formation. A total of 30 Sprague-Dawley rats were divided into 3 observation days (n= 10 rats). The incisions performed on all rats but given different treatments. In negative control group, the incision wound was not treated, while in positive control, the incision was treated with bioplacentom. Rats in treatment 1 and treatment 2, the incision wound were treated with 75% and 100% stingless bee honey treatment was given once a day for 7 days. The results showed that the 75% and 100% stingless bee honey groups showed a significant effect on reducing the wound length and increasing the collagen density (P0.05). As conclusion, stingless bee honey is effective in increasing collagen density in the wound healing process of rat.

Abstract

Cardiovascular disease (CVD) is still the leading cause of death according to WHO in 2021. CVD generally results in myocardial infarction (MI), which occurred after tissue ischemia. Echocardiography is most likely a diagnostic tool without invasive effects. This study aimed to compare the blood flow in pig research animals treated with ligases of the circumflex arteries. Measurements were made in the mitral valve (MV), tricuspid valve (TV), semilunar pulmonary valve (SLP), and semilunar aortic valve (AO) of velocity during systole (Vs), velocity during diastole (Vd), time average peak velocity (TAMAX), and blood flow pressure at three-time points: before ligase treatment, after acute infarction (60 min), and 8 weeks after ligase treatment. All parameters in MV group increased at 60 min and decreased at 8th weeks. On TV, TAMAX and pTAMAX decreased at 60 min and week 8. In SLP, VS, pVS, and Vd decreased at 60 min and increased at 8th week. The pVd and pTAMAX parameters decreased at 60 min and 8th week, while TAMAX increased at 60 min and decreased at 8th week. In AO, the TAMAX and pTAMAX parameters increased at 60 min and 8 weeks; the other parameters were similar to MV and TV patterns.

Abstract

Ilmu forensik veteriner belum mendapat perhatian yang memadai di Indonesia, sehingga perlu ditingkatkan pengembangannya. Penelitian ini bertujuan mempelajari ilmu forensik veteriner melalui histopatologi hati ayam broiler 48 jam pasca kematian. Ayam broiler sebanyak 75 ekor berumur 7 hari dieutanasia dan dibagi menjadi 25 kelompok berdasarkan jam nekropsi pasca-eutanasia (n=3). Kadaver ayam dinekropsi setiap 2 jam postmortem, organ hati dikoleksi untuk dibuat potongan histopatologi dan diwarnai dengan Hematoksilin-Eosin (HE). Histopatologi hati evaluasi dan hasil yang didapat dianalisis menggunakan software ImageJ versi 1.53a. Hubungan peubah histopatologi dengan waktu postmortem dianalisis dengan metode Pearsons. Hasil penelitian menunjukkan bahwa pembusukan dimulai pada jam ke-18 post mortem yang ditandai dengan jumlah inti sel hepatosit, jam ke-20 ditemukan bakteri pembusuk, dan jam ke-22 terjadi peningkatan jarak antar hepatosit. Berdasarkan nilai korelasi Pearson, jumlah inti sel hepatosit, jarak antar hepatosit dan keberadaan bakteri pembusuk mempunyai hubungan kuat hingga sangat kuat dengan waktu postmortem sehingga hasil ini dapat menjadi parameter penentu waktu kematian (Post Mortem Interval/PMI) pada hewan.
