Abstract

Goats are small ruminants that are reared by the rural community for financial income and nutrition. This study was aimed to determine the effect of vitamin C addition in tris–egg yolk extender on the lifespan of Sapera goat spermatozoa stored in 5o C. Semen was collected from 1.5 years old Sapera male goat. The ejaculates were diluted in Tris-egg yolk (T-EY) extender added with 0, 0.2, 0.3, and 0.4 g vitamin C/mL extender for T0, T1, T2, and T3 groups, respectively. The spermatozoa motility, viability, and morphological abnormality were assessed along with storage at 5° C. The result showed that spermatozoa motility was the highest (p <0.05) in T1 compared to other groups and qualified for AI use for up to 72 hours. In 24 hours storage, the spermatozoa viability was not significantly different (p >0.05) among the groups. The spermatozoa viability in the T1 group was the highest (p <0.05) among the groups along 48-96 hours of storage. The spermatozoa morphological abnormalities of the T1 group was the lowest (p <0.05) compared to other treatment groups in the range of 24-72 hour storage. In the control group (T0), the less than 5% spermatozoa morphological abnormalities (qualified for artificial insemination) were only in the 24 hours storage, while those of the T1 group were up to 72 hours. It could be concluded that the addition of 0.2 g vitamin C/100 mL T-EY extender maintained the quality of Sapera goat semen for 72 hours at 5°C.

Abstract

Sapera goat is a dairy goat resulting from a cross between a Saanen buck and an Ettawah cross doe. These small ruminants are reared by rural households for poverty reduction and undernutrition prevention. Breeding of Sapera goat through artificial insemination (AI) technique was expected to accelerate the increase in population. Unfortunately, the frozen semen of Sapera buck is not available yet. This study aims to determine the effect of the distance between semen straw and liquid nitrogen surface during pre-freezing process on the quality of frozen semen of Sapera goats. Semen was obtained from 1.5 years old Sapera buck. The ejaculates were diluted in tris-egg yolk extender to obtain a concentration of 120 x 106 spermatozoa/mL, then equilibrated at 5°C for one hour. The extended semen was packaged in 0.5 mL French straws (60 x 106 spermatozoa/straw). The filled straws were pre-frozen in the cold handling cabinet (Minitube, Germany) held at 10, 11, 12, 13, 14, and 15 cm above liquid nitrogen respectively for T1, T2, T3, T4, T5, and T6 groups, each with four replicates. After pre-freezing for eight minutes the straws were immediately plunged, and stored in liquid nitrogen (-196°C) for two days, followed by quality assessment. The best post-thawed motility (41.75 ± 2.06 %), viability (49.00 ± 0.82 %) and morphological abnormalities (4.75 ± 0.96 %) were obtained from pre-freezing stage with a distance of 10 cm. It could be concluded that in tris-egg yolk extender, post-thawed spermatozoa viability and motility of Sapera buck were qualified for AI when fresh semen was pre-frozen at 10 cm distance of straws from the surface of liquid nitrogen for eight minutes.

Abstract

Antibiotic Growth Promoters (AGP/AGPs) have been used to improve feed efficiency in poultry, but are banned in Indonesia due to food safety concerns. This study evaluated the potential of probiotics as an AGP alternative in ISA Brown male layer chickens during the finishing period, focusing on blood glucose and cholesterol levels. A total of 180 chickens were reared for 42 days. Treatment groups received basal feed supplemented with probiotics at 1–5 mL/kg, equivalent to 1.2×10⁹ to 6×10⁹ CFU/kg feed, containing Lactobacillus acidophilus, Lactobacillus plantarum, and Bifidobacterium spp. Blood cholesterol was analysed using an enzymatic colorimetric method at 500 nm, while glucose was measured using the glucose oxidase method at 505 nm. Results showed that probiotic treatments (P2, P3, P5) significantly reduced glucose levels compared to the control (P0) and AGP group (P1). A moderate decrease in cholesterol levels was also observed in probiotic groups. These effects may be linked to improved gut microbial balance and nutrient metabolism. The probiotic combination used in this study shows potential as a safer and more sustainable alternative to AGPs. Further studies are recommended to optimize dosage and assess long-term impacts on poultry health and productivity.

Abstract

Lead exposure exerts toxic effects on the female reproductive system, particularly on the ovaries and uterus, resulting in hypofunction and organ atrophy. Oxidative stress induced by lead accumulation may be attenuated by antioxidants such as vitamin E and zinc sulfate. This study aimed to evaluate the protective effects of vitamin E, zinc sulfate, and their combination on ovarian and uterus weights in albino rats (Rattus norvegicus) exposed to lead acetate. Twenty-five female rats were randomly divided into five groups (K, T0, T1, T2, and T3) and treated orally for 21 days. On day 22, the rats were euthanized, and the ovaries and uterus were collected and weighed. The average ovarian weights for K, T0, T1, T2, and T3 were 25.80 ± 2.49; 18.62 ± 4.36; 28.40 ± 5.67; 33.78 ± 6.09; and 36.84 ± 5.15 mg, respectively. The average uterus weights for K, T0, T1, T2, and T3 were 292.48 ± 36.81; 249.96 ± 25.97; 315.86 ± 26.56; 325.54 ± 31.53; and 375.00 ± 36.55 mg, respectively. In conclusion, vitamin E and zinc sulfate demonstrated protective effects against lead-induced reproductive toxicity, with the combined treatment showing the greatest improvement in ovarian and uterus weights.

Abstract

The intensification of the global poultry sector is currently challenged by the need to balance productivity with the mitigation of antimicrobial resistance (AMR) and environmental pollution. This study addresses the technological gap among smallholder farmers in Banyuwangi, Indonesia, by implementing a comprehensive technology transfer program focused on the functional substitution of antibiotic growth promoters (AGPs) with probiotics and the development of waste management cycles. Employing a Participatory Action Research (PAR) framework, the intervention integrated cognitive coaching, technical demonstrations, and longitudinal field applications to bridge the divide between biotechnological innovation and grassroots practice. Data were obtained from 27 broiler farmers through pre- and post-test evaluations and Focus Group Discussions (FGDs) to measure literacy shifts and technical adoption. Quantitative analysis revealed a significant transformation in biotechnology literacy, with understanding of probiotic mechanisms increasing from 37.0% (baseline) to 85.2% post-intervention. Furthermore, technical competence in functional feed formulation improved by 33.4% (p < 0.05), while the intention to adopt sustainable practices increased to 96.3%. These findings indicate that barriers to adoption among smallholders are primarily driven by limited access to applied science rather than cultural resistance. The study concludes that this hybrid extension model successfully establishes a viable micro-circular economy, offering a replicable strategy for upstream AMR mitigation in developing agropolitan regions.

Abstract

Antibiotics are commonly used to treat vibriosis in shrimp. However, their prolonged use at high doses may lead to the development of antibiotic resistance. As a preventive measure against antibiotic resistance, the use of natural antibacterial agents is required. Bajakah tampala root (Spatholobus littoralis Hassk) has been reported to possess antibacterial properties. This study employed six treatment groups: a positive control using a tetracycline antibiotic disc, a negative control using distilled water and CMC-Na, and four treatment groups (P1, P2, P3, and P4) treated with Bajakah tampala root extract (Spatholobus littoralis Hassk) at concentrations of 25%, 50%, 75%, and 100%, respectively. After treatment, the diameters of the inhibition zones were measured. The data obtained were analyzed using the Shapiro–Wilk normality test, followed by one-way analysis of variance (ANOVA). The results showed that treatment P4, with a 100% concentration of Bajakah tampala root extract (Spatholobus littoralis Hassk), produced an inhibition zone diameter comparable to that of the positive control.
