Abstract

This experimental research was performed to observe the influence of an agricultural liquid organic fertilizer called Bio Ferti on the growth and biomass of Spirulina platensis, aiming at replacing inorganic fertilizer with the liquid organic one. The cultivation of the microalgae was conducted over seven days at Nogotirto Algae Park. The liquid organic fertilizer, namely Bio Ferti, was obtained from the Faculty of Biology, Universitas Gadjah Mada, and prepared to have doses of 2, 4, 6, 8, and 10 mL. For comparison, an inorganic fertilizer with the same doses was also prepared. The variables to be observed were cell density, dry cell weight, and growth kinetics. The culture medium conditions observed were temperature, pH, and salinity (the optimum salinity was 20 ppt). The growth kinetic analysis was performed mathematically using numerical simulations using the Contois and the Haldane models. This research’s results showed that Bio Ferti affected the growth rate of Spirulina platensis. With a dose of 2 mL, it became the optimum medium which produced the highest density and dry weight of 1.78x106 cells/mL and 160 mg/mL, respectively. Meanwhile, the inorganic fertilizer with a dose of 10 mL produced the highest density and dry weight of 2,13x105 and 80 mg/mL, respectively. The temperature ranged from 28 to 31°C, while the pH ranged from 8.01 to 9.02 for each medium. The suitable model to describe the growth kinetics of Spirulina platensis was the Haldane model.

Abstract

Endophytic fungi have been reported to promote plant growth and resistance to biotic and abiotic stresses. This study aimed to determine the effect of endophytic fungi application isolated from the sengon (Falcataria moluccana) seeds on the growth of sengon. The seed-borne endophytic fungi were applied when the session was three weeks old by sprinkling each suspension of endophytic fungi (103 to 106 spores/ml) on the soil around the plant roots. The experimental parameters evaluated were height, diameter, and total wet and dry weight. This study showed that endophytic fungi treatment has no significant effect on the stem diameter, total wet weight, and water content; but significantly affected the height and total dry weight. Cladophialophora sp. and Ascotricha sp. fungi increased the height growth of sengon, while Aspergillus sp. fungi increased the total dry weight against the control. Ochroconis sp. treatment was not significantly different from the control. The shoot-root ratio values range from 1.13-1.43, indicating that the seeds already have roots supporting plant growth. The Dickson quality index range from 0.65-1.03 suggested that the seedlings have good biomass distribution. Abstract

Community forests play significant role in supplying national demand of woods, sustaining local ecosystem and increasing benefit for local community. Tree plantings for both community forests and rehabilitation of degraded forests and lands, however often neglect utilization of good quality seeds, which resulted in low quality of forest plantation. In order to support the procurement of good quality seeds, it is necessary to raise awareness of local community, seeds providers and forest extension workers on the production of good quality seeds through tree selection program. This paper reported the activity to build the capacity of local community, seeds providers and forest extension workers on plus trees selection, as sources of good quality seeds production. The training activity took place two times at Ngancar Village, Kediri Regency, East Java. The participants in each training received lectures in a classroom and hand-in practical work on tree selection techniques in the field. The topics given are considered as important by participants, as evident from the increasing number of participants in the second time of training. Analysis of pre- and post-test conducted showed that the participants knowledge and skill increased by 27,9 % after training, regardless of their ages and educational backgrounds.

Abstract

Penanaman sengon yang luas di Kediri, Jawa Timur menyebabkan meluasnya serangan hama, salah satunya adalah serangan hama penggerek batang boktor. Petani belum menemukan cara untuk mencegah dan menanggulangi serangan hama yang efektif. Hal tersebut dapat disebabkan karena kurangnya pengetahuan petani akan jenis-jenis hama dan cara pengendalian hama tanaman sengon. Tujuan dilakukannya sosialisasi yaitu untuk menambah wawasan masyarakat sehingga mereka dapat memilih teknik pengendalian hama sengon yang tepat. Kegiatan sosialisasi dilaksanakan pada tanggal 14-15 Juni 2022. Kegiatan dilaksanakan di Desa Ngancar, Kabupaten Kediri, Jawa Timur. Peserta sosialisasi dan pelatihan berjumlah 20 orang, yang terdiri atas perwakilan Perum Perhutani selaku mitra program pengabdian, anggota Lembaga Masyarakat Desa Hutan (LMDH) binaan Resort Pemangkuan Hutan (RPH) Pandantoyo, serta perangkat Desa Ngancar. Hasil yang diperoleh yaitu 50 % peserta sosialisasi belum pernah mendapatkan penyuluhan pengenai hama tanaman sengon. Terdapat kenaikan persentase nilai pre-test ke post-test sebesar 12 %. Kegiatan sosialisasi mengenai hama tanaman sengon dapat menambah pengetahuan masyarakat.

Highlight Research One of the content of microalgae that is beneficial for health is polysaccharides Polysaccharides of Indonesian microalgae can be promoted as anti-malarial Polysaccharides from Glagah, Spirulina and East Java microalgae inhibited the growth of plasmodium in vitro and had IC50 values of 3.18 µg/mL, 5.43µg/mL and 9.87 µg/mL, respectively   Abstract Malaria is an infectious disease caused by protozoan parasites of the genus Plasmodium that categorized as deadliest diseases in the world. Artemisinin and its derivatives are still recommended drugs for malaria therapy, however, there have been indications that Plasmodium parasites are resistant to this drug. Therefore, a study on polysaccharides from microalgae may be a potential as bioactive compound for anti-malaria. The aim of this study was to determine the effectiveness of the mixed microalgae polysaccharides as anti-malarial in vitro. Polysaccharides were extracted from three microalgae Spirulina sp., mixed microalgae Glagah and mixed microalgae East Java using the alkaline extraction method. The anti-malarial activity test refers to the concentration of polysaccharides used in calculating the IC50 value by probit analysis.  The concentration of polysaccharides of the three microalgae used were 0; 0.01; 0.01, 1, 10 and 100 µg/mL. The results showed that the IC50 values of polysaccharides of Glagah, Spirulina sp. and East Java microalgae were 3.18 µg/mL, 5.43µg/mL, and 9.87 µg/mL, respectively. In Conclusion, polysaccharides of Indonesian mixed microalgae can be promoted as anti-malarial.

Abstract

Hutan rakyat adalah hutan tanaman yang tumbuh di atas lahan milik rakyat atau masyarakat. Sengon (Falcataria moluccana) dan balsa (Ochroma pyramidale) merupakan jenis multi fungsi dan cepat tumbuh yang banyak dibudidayakan di Indonesia, khususnya di Pulau Jawa. Serangan penyakit hutan merupakan ancaman yang berpotensi mengganggu kegiatan budidaya kedua jenis ini, serta dapat merugikan secara ekologi, ekonomi, dan sosial. Pengenalan penyakit hutan merupakan kegiatan preventif yang sangat penting dalam pengelolaan penyakit hutan itu sendiri. Tujuan pengabdian pada masyarakat dalam “Program Dosen IPB Pulang Kampung” ini adalah sebagai upaya capacity building masyarakat sekitar hutan melalui penyuluhan, pelatihan, dan pendampingan terkait pengenalan (identifikasi) dan pengelolaan penyakit pada hutan tanaman yang ditujukan bagi para masyarakat petani hutan dan/atau pihak (stakeholder) terkait di Desa Ngancar, Kabupaten Kediri, Jawa Timur. Secara umum, program ini membantu meningkatkan pengetahuan dan pengalaman peserta dalam mengidentifikasi penyakit hutan tanaman, khususnya untuk jenis sengon dan balsa. Hal tersebut diindikasikan dari hasil penilaian dan evaluasi terhadap kuisioner (pre- dan post-test) para peserta pelatihan yang menunjukkan peningkatan, masing-masing, yaitu 7% (pelatihan Gelombang I) dan 9% (pelatihan Gelombang II). Community forests are forest plantations that grow on land owned by the people or community. Sengon (Falcataria moluccana) and balsa (Ochroma pyramidale) are multi-functional and fast-growing species that are widely cultivated in Indonesia, especially on the island of Java. Forest disease outbreaks are a potential threat that can disrupt the cultivation of these two species, and can cause ecological, economic, and social harm. The introduction of forest disease is a preventive action that is very important in managing of forest diseases. The aim of the community service in the “Program Dosen IPB Pulang Kampung” is to build the capacity of communities around the forest through counselling, training, and assistance related to the identification and management of diseases in forest plantations, targeting forest farmers and/or stakeholders in Ngancar Village, Kediri Regency, East Java. In general, this program helps to increase the knowledge and experience of participants in identifying forest diseases, specifically for sengon and balsa species. This is indicated by the results of the assessment and evaluation of questionnaires (pre- and post-tests) of the training participants, which showed an increase of 7% (Batch I) and 9% (Batch II).

Abstract

Perguruan tinggi sebagai tempat insan akademik berkiprah saat ini mengalami berbagai tantangan dan ancaman yang dapat mengganggu fungsinya sebagai pusat pengembangan ilmu pengetahuan dan teknologi (iptek), serta menjadi penjaga keberlanjutan peradaban dan moralitas insan akademik agar tidak terdistorsi dan tererosi. Kemajuan teknologi (khususnya teknologi informasi), dekadensi moral global, tekanan ekonomi, orientasi praktis dan fokus pada pencapaian jangka pendek, perubahan paradigma pendidikan serta iklim ketidakpastian, menjadikan nilai-nilai tradisional perguruan tinggi terancam tergeser menjadi mesin penghasil tenaga kerja semata. Perguruan tinggi, yang seharusnya berperan sebagai benteng terakhir penjaga nilai-nilai akademik, serta sebagai agen perubahan (agent of change) yang berorientasi pada nilai kebaikan dan penjaga nilai moral (guardian of value), perlu jati diri insan akademik yang unggul, sehingga peran dan fungsi perguruan tinggi dapat tetap dipertahankan. Untuk memberikan pertimbangan dalam upaya menjaga tersebut, Policy Brief ini disusun melalui serangkaian proses diskusi anggota tim PAH Jati Diri Insan Akademik dan proses FGD yang melibatkan narasumber internal dan eksternal IPB. Policy Brief ini diakhiri dengan rekomendasi yang dapat diusulkan kepada penyelenggara pendidikan tinggi Indonesia agar fungsi perguruan tinggi dan insan akademik dapat tetap terjaga

Abstract

Rencana rehabilitasi tanaman hutan membutuhkan benih dan bibit dalam jumlah besar. Pemenuhan kebutuhan bibit dapat diperoleh melalui sertifikasi benih tanaman hutan. Penggunaan benih bersertifikasi juga dapat meningkatkan produktivitas tanaman sehingga hasil panen memiliki nilai ekonomi yang lebih tinggi. Salah satu sumber benih dan bibit tanaman hutan bersertifikat yang diperlukan masyarakat adalah sengon. Pelatihan seleksi pohon plus melalui kegiatan pengabdian masyarakat dilakukan untuk memberikan informasi dan membantu petani dan masyarakat desa hutan (LMDH) binaan UPTD Perbenihan Jawa Tengah dalam memilih pohon induk yang unggul. Kegiatan pelatihan dilakukan dengan metode ceramah, diskusi interaktif, dan praktikum langsung di lapang. Evaluasi keberhasilan pelatihan dilihat dari peningkatan nilai post-test peserta pelatihan. Hasil post-test peserta mengalami peningkatan sebesar 48%. Berdasarkan tingkat pendidikan, peningkatan tertinggi terdapat pada peserta dengan tingkat pendidikan S1 yaitu sebesar 40,4 poin. Berdasarkan rentang usia, peningkatan nilai tertinggi berada pada rentang usia 25-35 tahun dan 36-46 tahun. Pelatihan seleksi pohon plus dapat dikatakan berhasil meningkatkan pengetahuan peserta pelatihan. Semua pihak berharap kegiatan serupa dapat diadakan kembali.

Abstract

Gyrinops versteegii and Aquilaria malaccensis are two important species of the Aquilarieae tribe. The main problem of this tribe is the challenge of species identification that is strongly dependent on the presence of flowers and fruit, which are not always available. The availability of whole genome information is expected to address the problems of species identification. This research aims to construct and compare the chloroplast and mitochondrial genomes of G. versteegii and A. malaccensis from short-read data using the NOVOplasty and GetOrganelle assembler. The chloroplast genome assembly revealed a full-length quadripartite circular structure with sizes of 174.814 bp (G. versteegii) and 174.821–174.822 bp (A. malaccensis), with highly conserved gene and organization. Meanwhile, the mitochondrial genome is multipartite with a size of 400.012 bp (G. versteegii) and 400.000 bp (A. malaccensis), with highly variable genes and organization due to the presence of gene cluster repeats. The LSC/IR/SCC region borders and phylogenetic analysis in chloroplasts indicate variations between the genomes of these two species. The investigation of nucleotide diversity in the chloroplast genome revealed that the trnL-rpl32 region had the highest nucleotide diversity (Pi = 0.03). This information will be useful in the future for a variety of downstream analyses.

Abstract

Sengon (Falcataria falcata (L.) Greuter and R. Rankin) plantations in Indonesia are threatened by attacks from Boktor stem borers and gall rust disease. Controlling pests and diseases is difficult therefore, planting resistant trees obtained from tree selection programs is necessary. Currently, genomic breeding often incorporates GWAS, which uses thousands of SNP markers to identify markers with significant associations with the traits studied. This study aimed to bypass such expensive studies by identifying and developing SNP markers from sequences of putative resistance genes to Boktor stem borer and gall rust disease, identified from sengon transcriptomic data analysis. A total of 496,194 putative SNP sites were identified from transcriptomic sequences using the SAMtools and BFCtools programs, of which 119 SNP sites were associated with resistance genes. Of the 101 non-synonymous SNPs selected, only 12 were located in the conserved domain of each gene and were used for primer design. Of the 13 primers designed, only 10 were successfully amplified. Validation of 10 developed SNP markers on 100 sengon accessions using the HRM method confirmed a significant association between SNP markers and resistance traits, with a -log 10 (P-value) between 10.49 and 16.63. A few SNPs markers developed from putative resistance gene sequences are associated with resistance traits in sengon. Therefore, the SNP markers could be applied in selection programs for sengon trees resistant to Boktor stem borers and gall rust disease.
