Abstract

Betta dennisyongi, locally known as Jielabu fish, is an endemic ornamental species originating from southwest Nanggroe Aceh Darussalam, Indonesia. The fish currently faces increasing exploitation and habitat degradation which raise conservation concerns and emphasize the need for domestication efforts. This study aimed to determine an appropriate stocking density for the early culture of B. dennisyongi. The research was conducted from October to December 2023 using a completely randomized design with four stocking densities (1, 2, 3, and 4 fish L⁻¹) with three replications. Juvenile fish collected from the wild were acclimated and reared for 60 days in 10-L aquaria. Biological parameters measured included survival, specific growth rate, length gain, weight gain, feed conversion ratio, and feed efficiency, while water quality was monitored periodically. Stocking density significantly affected survival rate and growth (p < 0.05) of B. dennisyongi, with the lowest density (1 fish L⁻¹) producing the most favorable performance, while densities of 3–4 fish L⁻¹ resulted in reduced outcomes. Feed conversion ratio and feed efficiency did not differ significantly among treatments (p > 0.05). All measured water quality parameters remained within acceptable ranges for Betta fish culture. Overall, the study indicates that lower stocking densities support better biological responses, and that 1 fish L⁻¹ is the most suitable for cultivation, while 2 fish L⁻¹ may serve as an acceptable alternative for practical application. These findings provide a foundation for developing effective domestication protocols and support conservation-oriented aquaculture of this vulnerable endemic species.

Abstract

Whiteleg shrimp (Litopenaeus vannamei) dominates global aquaculture production due to its adaptability to intensive systems. However, intensive systems often experience excess accumulation of nitrogenous waste and total organic matter (TOM), which can destabilize microbial communities and affect water quality. While protozoa are known as bioindicators, few studies have explored how their functional composition interacts with nitrogen cycling and production performance in shrimp ponds. This study investigated the relationships between nitrogenous compounds, TOM, bacterial and protozoan abundance in two intensive shrimp ponds (HP: high protozoan abundance and LP: low protozoan abundance). Water quality parameters, including Total Ammonia Nitrogen (TAN), nitrite, nitrate, TOM, and phosphate, were monitored weekly alongside microbial assessments of total bacterial count (TBC), total Vibrio count (TVC), and protozoa abundance. Protozoa were identified microscopically, while shrimp performance was measured by growth, feed conversion ratio (FCR), survival, and productivity. TOM emerged as the primary ecological driver, significantly correlating with Vibrio abundance (r = 0.585, p < 0.05). Although the high-protozoa pond featured greater bacterial biomass and more bacterivorous taxa (e.g., Ciliata, Vorticella), it had lower shrimp productivity. Conversely, the low-protozoa pond dominated by detritivores (Euplotes, Strombidionopsis) achieved superior growth, FCR, and final biomass, despite higher TOM and nitrite levels. These findings suggest that protozoan functional composition, rather than total abundance, critically influences nutrient cycling, microbial stability, and production outcomes. Managing TOM and fostering beneficial microbial loops are essential strategies for sustainable shrimp farming.

Abstract

Litopenaeus vannamei is a key species in global aquaculture; however, its susceptibility to vibriosis presents a persistent challenge. The aim of this study was to investigate the effects of dietary supplementation with alginate and Spirulina hot water extract (SWE) on the immune response and gene expression associated with pathogen resistance. Shrimp fed diets containing 3 g·kg⁻¹ of alginate and 5 mg·kg⁻¹ of SWE exhibited marked improvements in immune parameters, including total hemocyte count, phagocytic activity, phagocytic index, phenoloxidase activity, and superoxide dismutase levels, as measured on days 3 and 7 (P< 0.05). Following challenge with Vibrio parahaemolyticus, V. vulnificus, and V. harveyi, treated shrimp demonstrated a 90% survival rate compared to 0% in the control group. Gene expression analysis revealed a 2-fold and 1.5-fold upregulation of prophenoloxidase (proPO) and lipopolysaccharide β-glucan binding protein (LGBP), respectively, indicating the activation of the proPO cascade and pathogen recognition pathways. These results provide direct evidence that alginate and SWE act as immunostimulants by modulating hemocyte function and immune-related gene expression. The findings of the present study suggest that oral supplementation with these natural bioactive compounds can serve as a functional dietary strategy to enhance disease resilience in shrimp and promote more sustainable aquaculture systems, in seven days period.
