Abstract

Background: Meatballs are a food that easily spoils if stored improperly. This spoilage is caused by microbial contamination. One way to reduce microbial contamination in meatballs is by using natural preservatives, such as cell-free supernatant from Lactobacillus plantarum. This cell-free supernatant contains antimicrobial metabolites that can effectively inhibit the growth of spoilage bacteria. Purpose: This study aims to determine the concentration of cell-free supernatant from Lactobacillus plantarum BAF514 as a biopreservative on the microbiological quality of beef meatballs stored at refrigerator temperature. Methods: A Completely Randomized Design (CRD) with 5 treatments and 4 replications was used in this study. The treatments consisted of: P1 = 100% cell-free supernatant, P2 = 90% cell-free supernatant + 10 ml sterile distilled water, P3 = 80% cell-free supernatant + 20 ml sterile distilled water, P4 = 70% cell-free supernatant + 30 ml sterile distilled water, and P5 = 60% cell-free supernatant + sterile distilled water. The variables observed were pH value, total microbes, Staphylococcus aureus, and Escherichia coli. Analysis of variance (ANOVA) was used in data analysis, Duncan's multiple range test was used if there was a significant effect. Results: The results showed that the use of several concentrations of 100% - 60% cell-free supernatant had a significant effect (P < 0.05) on pH and no significant effect (P > 0.05) on total microbes, Staphylococcus aureus, and Escherichia coli. The pH value of meatballs increased with the lower concentration of cell-free supernatant during storage. Conclusion: It was concluded that the use of cell-free supernatant concentration from Lactobacillus plantarum BAF514 on the microbiological quality of meatballs during refrigerator temperature storage did not increase the total microbes, Staphylococcus aureus and Escherichia coli but increased the pH value of the meatballs. The concentration of cell-free supernatant from Lactobacillus plantarum BAF514 up to 60% was still good for preserving beef meatballs stored at refrigerator temperature.

Abstract

Abstract. Sukmono T, Kaswari T, Utomo PEP, Wulandari T, Wibowo SE, Rolin F, Nurjirana. 2026. Revealing phylogenetic relationships of Cyprinidae based on DNA barcoding in Merangin Jambi Geopark, Indonesia. Biodiversitas 27 (3): d270317. https://doi.org/10.13057/biodiv/d270317. Cyprinidae, a dominant freshwater fish family, plays a crucial role in biodiversity and ecosystem functioning. The Merangin Jambi Geopark, under evaluation for the UNESCO Global Geopark Network, hosts a rich freshwater fish fauna, but morphological identification is challenging due to overlapping characters. This study employed DNA barcoding of the mitochondrial COI gene to identify species and investigate phylogenetic relationships of Cyprinidae. A total of 26 individuals representing 14 species across 10 genera were analyzed. The Cytochrome C Oxidase subunit I (COI) gene (~650 bp) was amplified and sequenced. Sequence analysis was performed using Basic Local Alignment Search Tool (BLAST), and a phylogenetic tree was constructed in MEGA 12. Sequence similarity ranged from 88.11% to 99.56%, with most species exceeding 96% identity, indicating strong intraspecific homogeneity and clear intergeneric divergence. Genetic distances ranged from 0.000 to 0.179, reflecting population stability in species such as Barbodes lateristriga and Rasbora spilotaenia, and high divergence in species like Luciosoma spilopleura and Epalzeorhynchos kalopterus. Phylogenetic reconstruction revealed two major clades, with samples clustering consistently with GenBank reference sequences, confirming accurate species identification and taxonomic placement. Based on IUCN status, most species were categorized as Least Concern, though several were Data Deficient or Not Evaluated, highlighting the need for local conservation attention. These findings demonstrate that DNA barcoding effectively complements morphological taxonomy and provides a robust framework for biodiversity monitoring. Furthermore, the study contributes to local conservation by informing habitat protection, species management, and sustainable freshwater resource utilization in the Merangin Jambi Geopark.

Abstract

This study aimed to determine the effect of administering red ginger extract (*Zingiber officinale* var. *rubrum*) and using a cool perch on heat stress indicators in broiler chickens. The study utilized 198 broiler chickens housed in 18 experimental pens, arranged in a Completely Randomized Design (CRD) consisting of three treatments and six replicates. The treatments were: P1 (control: no red ginger extract and no cool perch), P2 (red ginger extract without a cool perch), and P3 (red ginger extract with a cool perch). The variable observed was the heterophil/lymphocyte (H/L) ratio, an indicator of physiological stress in broilers. Data were analyzed using Analysis of Variance (ANOVA) followed by Duncan’s test via SAS software. The results showed that the treatments had a significant effect (P<0.05) on the broiler H/L ratio. The mean H/L ratio values for the treatments were 0.350 ± 0.080 for P1, 0.198 ± 0.071 for P2, and 0.206 ± 0.065 for P3. Duncan’s post-hoc test indicated that P1 differed significantly from P2 and P3, whereas there was no significant difference between P2 and P3. In conclusion, the administration of red ginger extract—either alone or in combination with a cool perch—is effective in lowering the heterophil/lymphocyte ratio, thereby reducing heat stress levels in broiler chickens.

Abstract

Jembrana disease is an infectious disease that causes significant economic losses in Bali cattle farming. Muara Papalik District in West Tanjung Jabung Regency has a large Bali cattle population; therefore, farmers' understanding of disease prevention measures is crucial for maintaining livestock productivity. This study aimed to identify farmer characteristics and analyze the factors influencing their perceptions regarding Jembrana disease prevention. Data were collected via a questionnaire survey and analyzed using descriptive statistics and multiple linear regression. The results showed that the majority of farmers had an elementary school education (38.2%), 2–5 years of farming experience (61.8%), and a herd size of 5–10 cattle (61.8%). Farmers' knowledge of Jembrana disease was low; 50% were unfamiliar with the disease, and 79.4% had never attended extension programs. Farmers' perceptions regarding Jembrana disease prevention were rated as reasonably good, with an average score of 60.16%. Regression analysis yielded an R² value of 0.657, revealing that farming experience (Sig = 0.011) and knowledge level (Sig = 0.001) significantly influenced farmers' perceptions. In conclusion, greater farming experience and higher levels of knowledge correlate with more positive perceptions regarding Jembrana disease prevention. Enhancing farmer capacity through continuous education and extension services is necessary to strengthen awareness and the implementation of preventive measures in the field.

Abstract

Abstract

Jejunal intussusception is an uncommon cause of intestinal obstruction in dogs and is associated with abnormal motility, enteritis, parasitic infection, and foreign body ingestion. The diagnosis, surgical management, and outcome of a 3-year-old Poodle (5 kg) are described. The dog presented with vomiting, anorexia, weakness, abdominal pain, and foreign body ingestion. Abdominal radiography and hematology supported the diagnosis of intestinal obstruction and inflammatory response. Exploratory laparotomy confirmed jejunojejunal intussusception with congestion and hemorrhage without necrosis or perforation. The segment was reduced and repositioned without bowel resection or anastomosis. The abdominal wall was closed in layers, followed by postoperative therapy and monitoring, with wound healing by day 7 without complications.
