Abstract

Daluga (Cyrtosperma merkusii) starch processing yields waste with 70% of its total weight consist of organic biomass materials that can be utilized as advanced composite biomass materials, a potential base for reinforcing materials. Processing the Daluga waste (debris) requires stages of pretreatment and activations to eliminate inhibiting agents within the hydrolisis process (fermentation process). The purpose of this research is to understand the effects of biological pretreatments using the Effective Microorganism (EM4) bacterial consortium towards the chemical properties of Daluga debris. The chemical compositions of debris Daluga before and after each treatment were determined by NDF and Van Soest method for observing the cellulose, hemicellulose, lignin, fiber, ash content, crude protein, silicate and crude fat. These parameters were analysed following the methods outlined in the Indonesian National Standard (SNI 01-2891-1992) and while protein and carbohydrate content was measured with the Dumas combustion method. Results indicated that lignin concentration after delignification process decreased considerably to 1.80 ± 0.07% although lignin content of these Daluga debris initially wasn’t so high to begin with 4.11 ± 0.02%. Herewith, hemicellulose also decreased giving an appearance for the increase and exposure of cellulose. Thus, providing a clear path to further processing the cellulose to advance cellulosic materials such as cellulose nano crystals (CNC).

Isolation and identification of thermophilic amylolytic bacteria from Likupang Marine Hydrothermal, North Sulawesi, Indonesia. Biodiversitas 22: 3326-3332. The aims of the research were to isolate and identify the amylase-producing thermophilic bacteria from Likupang Marine Hydrothermal, North Sulawesi, Indonesia. The bacteria were characterized based on the colony and cell morphology and subsequently screened for their amylase activities. The bacterial isolates were identified based on 16S rRNA gene sequences. There were 12 thermophilic bacteria isolates from Likupang Marine Hydrothermal that were able to produce amylase. Two selected isolates (L3 and L9) had an amylolytic index value in the range of 3.04-3.52 at 55oC. The colonies of L3 and L9 are circular, and they are Gram positive, rod-shaped, and motile bacteria. Based on the 16S rRNA gene sequences and phylogenetic tree analysis showed that L3 was closely related to Bacillus caldotenax with 93% similarity while L9 had 99% similarity to B. caldotenax. Hence, we identified L3 isolate as Bacillus sp. strain L3 and L9 isolate as B. caldotenax strain L9. Both of these bacteria were grouped as Geobacillus thermoleovorans.

Abstract

Penelitian ini mengkaji inovasi mesin pengaduk berspatula yang dirancang khusus untuk meningkatkan efisiensi produksi selai pala di Industri Kecil Menengah (IKM) Sari Fruit Barik, Desa Beong, Sitaro. Tujuan utama penelitian adalah mengoptimalkan proses pengadukan yang selama ini dilakukan secara manual. Hasil uji coba menunjukkan bahwa penggunaan mesin pengaduk berspatula dapat mempercepat waktu pengadukan hingga 40% dibandingkan dengan metode manual, serta menghasilkan kualitas selai pala yang lebih konsisten dan baik. Efisiensi waktu yang dicapai mencapai 37,5%, dengan kapasitas produksi meningkat dari 20 kilogram per jam menjadi 30 kilogram per jam. Penelitian ini juga menyoroti pentingnya penerapan teknologi untuk mendukung pengembangan usaha secara berkelanjutan dan meningkatkan produktivitas IKM Sari Fruit. Melalui inovasi ini, diharapkan dapat memberikan kontribusi signifikan terhadap peningkatan ekonomi lokal serta menjadi model bagi IKM lain dalam menghadapi tantangan serupa.
