Abstract

Patchouli oil (Pogostemon calbin Benth) has economic benefits which are widely used as raw material for the pharmaceutical, perfume and food industries. Although widely used in various fields, patchouli oil or essential oils are generally young to evaporate, are susceptible to high temperatures and humidity, so we need a way / solution to overcome this, one way is encapsulation. Encapsulation is a technique to protect the core material which was originally liquid from becoming a solid / powder form and can protect the core material from losing its taste and aroma. Coating is a material that can mix with the core material, is not reactive to the core material and can form a layer around the core material. Chitosan is a good coating for essential oil encapsulation. This study aims to determine the particle size and microcapsule characteristics of patchouli oil (P.calbin Benth) using chitosan coating. The microcapsules were made using a freeze dryer with the freeze drying method. The dried samples were analyzed for particle size, morphological shape and content using SEM (Scanning Electron Microscopy). The results showed that the samples with a coating material composition of 1:2 were completely dry freeze drying. The average particle size <3.5 µm with a magnification of 10,000x with round morphology and various shapes. Analysis of the chemical content in the sample showed the similarity of the main compounds in patchouli essential oil, namely α-guaiene, azulene, α/β/ γ-patchoulene, seychellene, and α-himachalene and there was the addition of carbon (C) 76.07% and oxygen. (O) 16.46% originating from coating material

Abstract

The abundance of lignocellulosic waste makes it a potential source for advanced biomaterials through various transformation processes. Lignocellulosic biomass transformation to advanced biomaterials involves enzymes from extracellular metabolites of microorganisms capable of hydrolyzing lignocellulose. This research was to molecularly screen fungi found in symbiotes of lichen endemic to trees growing in North Minahasa (North Sulawesi) with lignocellulolytic transformation enzymes. Molecular screening was conducted from identified fungi isolates based on partial genetical analysis on the locus of Internal Transcribed Spacer (ITS) of the fungi’s ribosomal DNA. Fungi isolates screening identified Trichoderma koningiopsis (isolate HZA8 and isolate HZA6), Penicillium sumatraense (strain CBS 127365 and strain CBS 130380), Trichoderma hamatum (isolate PAN12-45 and isolate PAN12-05), Aspergillus aculeatus (strain A1.9 18S),  Aspergillus aculeatus (isolate XSD-74), Trichoderma reesei (strain S2606 and isolate 5A14).  Molecular identification and BLAST homology of potentially lignocellulolytic fungi isolates rDNA indicated that isolate KB2 had close relationship with Trichoderma reesei at 100% degree of closeness and an index of cellulolytic activity of 1.19. While isolate KB3 appeared closely related to Aspergillus aculeatus at 99.83% degree of closeness and an index of cellulolytic activity of 1.57. Therefore, the potential of developing bioprocess industries in general and in particular is most probable.

Abstract

The application of the conversion results from Eco-Enzyme into dishwashing soap, the fermentation process lasts for one months, where the resulting liquid is dark brown in color and has a very strong sweet and sour fermented aroma. There is a lack of public knowledge about the very profitable benefits of Eco-Enzyme, such as as a multi-purpose cleaner as a multi-functional liquid which can be converted, one of the ways, into dishwashing soap. This research aims to determine the application of the results of converting Eco-Enzyme into dishwashing soap. This research used experimental methods and a research period of 3 months. This Eco-Enzyme is made with a ratio of organic ingredients: sugar: water 3:1:10. The Eco-Enzyme harvest is converted into dishwashing soap by adding lerak and Methyl Ester Sulfonate (MES). Eco-Enzyme conversion soap was then applied to several treatment groups: plastic plates, cans and glass. The results of the application show that the dishes washed with Eco-Enzyme using lerak have not completely removed dirt, oil and odors in each treatment, whereas Eco-Enzyme using Methyl Ester Sulfonate (MES) when compared with Sunlight brand liquid soap sold on the market shows that there are similarities both in terms of cleanliness, tapestry and smell. The research conclusion is that the application of the Eco-Enzyme conversion results has significant potential to be developed into an active ingredient for environmentally friendly dishwashing soap.

Abstract

Giant swamp taro (GST/Cyrtosperma merkusii), a local North Sulawesi coastal plant, is underutilized but has high po-tential for cultivation as a paludiculture plant on rewetted peatland restorations. GST has high starch content and is currently being explored for potential use. This study explored the potential of using the biomass pulp yielded as resi-dues of the starch extraction process. Endolichenic fungi (Trichoderma reesei and Aspergillus aculeatus) isolated from local lichens, were inoculated on the GST biomass in a solid state fermentation (SSF). Findings of this study indicate that both isolates were effective in degrading the lignocellulosic biomass of GST over an incubation period of 28 days, focusing on cellulose as the targeted polysaccharide. Although T. reesei requires a more adjusted carbon source than A. aculeatus to effectively process the biomass, the effectiveness of the bioprocess after adjustment matched that of A. aculeatus. When hemicellulose is targeted, other strategies must be used to prevent destruction of hemicellulose during the process. This phenomenon should be explored in further studies. These findings indicate the potential of industrial-izing cellulose-based products from GST using local North Sulawesi fungal species abundantly occurring in local eco-systems.
