Abstract

Protein N-myristoylation is the covalent attachment of myrstate, via an amide bond, to the N-terminal glycine residue of a nascent polypeptide assisted by myristol-CoA protein: N-myristoltransferases (NMT). PROSITE motif describes 5 amino acid after glycine site that would give rise to myristoylation site. However, applied to whole database extract, this motif give too many false postive results. Therefore 2 new tools were developed for N-myristoylation prediction.Taking physical properties into consideration inceases prediction scores greatly. However, these algorithms still cannot predict myristoylated site correctly 100% of the time due to limited understanding of the real mechanims underlying N-myristoylation

Abstract

Pseudomonas aeruginosa is a bacterial pathogen that causes various kinds of infections. The use of antibiotics is usually done to treat the infection. However, Pseudomonas aeruginosa has various resistance abilities that can avoid the efficacy of antibiotics. The emergence of drug resistance events encourages the need for the discovery of new antibiotic drugs, one of which comes from fungals secondary metabolite. Soil fungi from mangrove ecosystems have not been studied much. Interestingly, these soil fungi have unique adaptive responses. This response also affects the secondary metabolites it contains. Knowing this information, mangrove soil samples were cultured on fungi media which were then tested against Pseudomonas aeruginosa through antagonist test and microdilution test. Three fungal cultures with yeast characteristics were obtained and each of the secondary metabolite ethyl acetate extracts of these three fungi had antibacterial activity. The antibacterial activity was indicated by the % inhibition parameter. The terpenoid content in fungal cultures may be responsible for the antibacterial activity against Pseudomonas aeruginosa. This research is an initial indohoki77 juga dikenal karena layanan pelanggannya yang responsif dan profesional. Tim dukungan pelanggan tersedia 24/7 untuk membantu para pemain dengan segala pertanyaan atau masalah yang mereka hadapi screening of the search for new antibiotic candidates, so further research study such as isolation and species determination needs to be done.

Abstract

The emergence of antibiotic resistance becomes a barrier in controlling infectious diseases, so it is necessary to find new alternative antibiotics. This study aimed to explore the potential of estuarine soil fungals secondary metabolites isolated from Katialada Village Gorontalo Location One and test their antibacterial activity against Staphylococcus aureus. The antibacterial parameter observed was the percent inhibition of ethyl acetate extracts against Staphylococcus aureus. The estuarine soil samples were cultured in PDA media producing five single fungal isolates differentiated based on color morphology, texture, and shape coded IS2-BTG-4.1.1, IS2-BTG-4.1.2, IS1-BTG-4.2, IS2-BTG4.3.1, and IS2-BTG-4.3.2. Those five samples were fermented using PDB media and the fermented products were extracted with ethyl acetate. The extracts were screened for its compound using a TLC plate and antibacterial testing with a single concentration of 100 g/mL in the microdilution method. The screening results showed that the five ethyl acetate extracts contained terpenoid compounds and code IS2-BTG-4.3.2 also had alkaloids. Based on the antibacterial test results, the five ethyl acetate extracts had growth inhibitory activity on the Staphylococcus aureus, with the percentage showed by IS2-BTG-4.1.1 50,2% ± 14,8%, IS2-BTG-4.1.2 35,6% ± 12,9%, IS1-BTG-4.2 13,0% ± 7,3%, IS2-BTG-4.3.1 13,6% ± 6,0%, and IS2-BTG-4.3.2 by 12,4% ± 9,2%.

Abstract

Indonesia was ranked 61 out of 164 countries (6.32 out of 10) based on the Basel AML Index in 2024. The Basel AML Index score reflects a country's vulnerability to money laundering and terrorist financing. Although not among the countries with the highest risk globally, this score indicates that Indonesia still has work to do to improve the effectiveness of its AML/CFT/CPF framework. Moreover, the newly emerging money laundering schemes, one of which involves untraceable assets like cryptocurrency, make it more difficult to carry out legal proceedings. The novelty in this study is the discovery of assets disguised in money laundering, which is expected to ease the Financial Transaction Reporting and Analysis Center (Pusat Pelaporan dan Analisis Transaksi Keuangan-PPATK) to track hard-to-trace assets. This study was conducted using a juridical-normative method. The result showed various methods in money laundering activities, namely transferring and/or investing funds obtained from illicit activities, corruption, bribery, fraud, irregularities through the banking sector, capital markets, and other means such as deposits, stocks, bonds, and various financial instruments. The various modus operandi carried out by corrupt individuals in money laundering in various sectors require more attention from PPATK.
