Abstract

Penelitian ini bertujuan untuk mengetahui peningkatan populasi dan laju pertumbuhan Daphnia sp. pada beberapa media yang menggunakan kotoran ayam, kotoran kuda, kotoran kambing, dan kombinasi kotoran tersebut serta mengetahui media kultur terbaik dalam menunjang peningkatan populasi Daphnia sp. Disamping itu penelitian ini juga bertujuan untuk mengetahui korelasi antara kepadatan fitoplankton media dengan kepadatan Daphnia sp. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) yang terdiri dari enam perlakuan yang diulang 3 kali, yaitu P1: kotoran ayam 100%, P2: kotoran kambing 100%, P3: kotoran kuda 100%, P4: kotoran ayam 50% + kotoran kambing 25% + kotoran kuda 25%, P5: kotoran kambing 50% + kotoran ayam 25% + kotoran kuda 25%, P6: kotoran kuda 50% + kotoran ayam 25% + kotoran kambing 25%. Parameter yang diamati adalah kepadatan populasi Daphnia sp., laju pertumbuhan populasi spesifik, pertumbuhan fitoplankton sebagai pakan Daphnia sp., uji proksimat (kotoran ayam, kambing, kuda, dan kombinasinya), dan kualitas air. Hasil penelitian menunjukkan bahwa kombinasi kotoran ternak (ayam, kambing, dan kuda) memberikan pengaruh yang nyata terhadap kepadatan populasi Daphnia sp. (p<0,05) serta berpengaruh nyata juga terhadap laju pertumbuhan populasi spesifik Daphnia sp. (p<0,05). Perlakuan P4 kombinasi kotoran ayam 50% + kotoran kambing 25% + kotoran kuda 25% menghasilkan kepadatan puncak populasi Daphnia sp. terbaik sebesar 1.840 individu/L dan laju pertumbuhan populasi spesifik sebesar 56,51%/hari.

Abstract

Mikroalga merupakan salah satu produsen primer yang diduga memiliki kandungan lipid tinggi untuk dimanfaatkan sebagai energi alternatif. Mikroalga yang memiliki kandungan lipid cukup tinggi diantaranya adalah Porphyridium sp. dan Isochrysis sp. Pemberian stress osmotik diduga dapat meningkatkan kadar total lipid mikroalga Porphyridium sp. dan Isochrysis sp. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian stress osmotik berupa perbedaan salinitas terhadap kadar total lipid pada mikroalga Porphyridium sp. dan Isochrysis sp. Penelitian ini telah dilaksanakan pada bulan Januari - Februari 2016 di Laboratorium Biologi Molekuler Jurusan Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Lampung. Penelitian ini menggunakan Rancangan Acak Lengkap dengan faktorial yang terdiri dari 2 jenis mikroalga serta 4 taraf salinitas 20 ppt, 25 ppt, 30 ppt, dan 35 ppt, serta 3 kali ulangan. Parameter yang diamati yaitu kepadatan populasi, laju pertumbuhan, dan kadar total lipid. Data dianalisis dengan Analysis of Variance (ANOVA) dan diuji lanjut dengan Uji Tukey HSD pada taraf 5 %. Hasil penelitian menunjukkan bahwa kepadatan populasi tertinggi terjadi pada perlakuan Isochrysis sp. pada salinitas 35 ppt. Mikroalga yang menyumbang lipid tertinggi pada tiap selnya adalah Porphyridium sp. pada salinitas 20 ppt sebesar 6x10-6 g/sel dan Isochrysis sp. pada salinitas 20 ppt sebesar 45x10-7 g/sel.

Abstract

Foraminifera bentik merupakan salah satu Protozoa yang dapat terawetkan dalam sedimen perairan. Organisme bersel tunggal ini telah banyak digunakan sebagai indikator lingkungan karena sangat sensitif terhadap perubahan lingkungan di sekitarnya. Penelitian ini bertujuan untuk mengetahui keanekaragaman foraminifera bentik dalam 20 sampel sedimen dasar perairan Teluk Balikpapan, Kalimantan Timur dalam kaitannya dengan beberapa parameter lingkungan air seperti kedalaman, kecerahan, temperatur, pH, turbiditas, salinitas, dan oksigen terlarut serta pembuatan peta sebaran spasialnya. Hasil penelitian menunjukkan bahwa terdapat 3 spesies foraminifera bentik yang mendominasi yaitu Asterorotalia trispinosa (1.553 spesimen) diikuti oleh Rotalia sp.1 dan Rotalia sp.2. Jumlah spesies terbanyak didapatkan pada titik TB-05 (33 spesies) yang terletak di bagian luar teluk Balikpapan. Indeks Keanekaragaman bervariasi dari 0 sampai 4. Besarnya nilai Korelasi Pearson untuk kedalaman, kecerahan, temperatur, pH, turbiditas, salinitas, dan oksigen terlarut berturut-turut yaitu 0,874; 0,962; 0,962; 0,966; 0,829; 0,966; dan 0,968. Nilai korelasi tersebut bervariasi dengan distribusi foraminifera bentik di daerah penelitian.

Abstract

Cagar Alam Laut Krakatau berada di daerah vulkanik Gunung Anak Krakatu. Aktivitas seismik yang diakibatkan oleh magma chamber Anak Krakatau menyebabkan goncangan pada dasar laut yang memungkinkan bergesernya substrat yang menjadi tempat terumbu karang tumbuh, Selain itu debu vulkanik akan mempengaruhi kalsifikasi dan pertumbuhan karang sehingga karang yang terbentuk akan rapuh dan rentan terhadap pengaruh lingkungan, seperti arus dan goncangan. Tujuan penelitian ini adalah untuk mengetahui kondisi terkini komunitas serta keanekaragaman terumbu karang di Cagar Alam Laut Krakatau. Penelitian dilakukan dari bulan Juli sampai Oktober 2012. Perairan yang menjadi stasiun penelitian adalah perairan Pulau Rakata. Metode manta tow digunakan pada saat survei pendahuluan dan metode Line Intercept Transect (LIT) digunakan dalam pengambilan data terumbu karang. Stasiun penelitian yang berda di perairan Pulau Rakata dibagi menjadi empat titik pengambilan sampel. pada kedalaman 5 meter dengan panjang transek 50 meter searah garis pantai. Hasil pengamatan menunjukkan bahwa dari 27 spesies dalam 7 famili terumbu karang yang ditemukan pada stasiun penelitian kondisinya bervariasi dari baik sekali hingga rusak dengan tingkat tutupan berkisar antara 90,88%, 56,54%,dan 38,32 %.

Abstract

Glyphosate herbicide widely used by farmers in Indonesia. As a result of exposure to herbicides, the production of free radicals or ROS exceeds the body's defense system called oxidative stress. Therefore, the activity of antioxidant enzymes in the body decline and thus can cause tissue damage and certain organ dysfunction. ROS can be suppressed by administration of antioxidants. One source of the antioxidant is taurine. Taurine has a protective effect against oxidation and free radical capture in a variety of cells and tissues against toxic oxidant component. This study aims to determine the effect of taurine in the liver of mice induced herbicide glyphosate on the activity of the enzyme superoxide dismutase (SOD), malondialdehida (MDA), changes in morphology and structure of liver histology. The research used completely randomized design (CRD). Mice were divided into three treatment groups such as, control group (K0), a group of glyphosate (K1) at a dose of 25 mg /kgBW/day orally for 25 days, and a group of taurine+glyphosate (K2) at a dose of taurine 7800 mg/kgBW/day + glyphosate dose 25 mg/kgBW/day orally for 25 days. Each treatment consisted of 8 mice as replication. Results of analysis with one ways ANOVA (P<0.05) followed by the Tukey test showed taurine dose of 7800 mg/kgBW/day can reduce liver cells of male mice that were treated by the herbicide glyphosate but it not be able to increase the levels of SOD and to decrease MDA levels.

Abstract

−Eggshells are one of the organic waste that is commonly found and comes from households. Based on observations in the field, people generally have not done waste management properly such as separating organic and inorganic waste. However, the utilization of household waste in the form of eggshells has not been widely done by the community. The solution to overcome this problem is to conduct training for the community to utilize waste in the form of eggshells to be used both for nurseries and further processing as organic fertilizer to increase plant growth. The desired expectation in the implementation of this service is an increase in skills and knowledge in making organic fertilizer from eggshells. There was an increase in knowledge and skills of the service participants with a pretest score of 55.88 and post-test to 90.00. There was an increase in value of 34.12. The percentage increase in value is 62.86%.

Abstract

Plastic waste will be degraded into microplastic which has a size of ≤ 5 mm. These microplastic particles change in size and color due to various natural processes that occur in the environment. The purpose of this study was to determine the shape, color and size characteristics of microplastics found around Pasaran Island. Sampling was carried out by taking microplastic samples in water and green mussels around Pasaran Island. Based on the research results, microplastic fragments, films, fibers, granules and foam were found. Microplastic colors consist of black, blue, green, yellow, red, clear, and white. Microplastic fragments, films and fibers were found in the <50µm to 1000 - 5000 µm size group. Granular microplastic types were found in the <50 µm to 100 - 500 µm size group and foam types were found in the <50 µm to 500 - 1000 µm size group.

Abstract

Hyperglycemia is an early symptom of diabetes caused by the pancreas not being able to produce enough insulin. This condition can disrupt blood glucose metabolism, thereby damaging organs in the body such as the pancreas. Hyperglycemia can be treated using traditional plant medicines. Songgo Langit leaves (Tridax procumbens L.) are reported to have secondary metabolite compounds that have potential as antidiabetic drugs. This study aims to determine the effect of administering Songgo Langit leaf extract on blood glucose levels and changes in the histopathological structure of the pancreas in hyperglycemic mice induced by alloxan. Using Complete Random Design, mice were divided into 5 groups of 5 mice each. Group K (-) was a negative control (not treated), group K (+) was a positive control (only induced by alloxan), group P1 was induced by alloxan and extract of 0.45 mg/g BW, group P2 was induced by alloxan and extract 0.9 mg /g BW, and group P3 was induced by alloxan and extract 1.8 mg/g BW for 14 days. Glucose level data were analyzed using ANOVA and post hoc test with LSD. Pancreatic histopathological structure data were analyzed using the Kruskal-Wallis test and the Mann-Whitney post hoc test. The results showed that the treatment of ethanol extract of Songgo Langit leaf at doses P1, P2, and P3 were significantly able to reduce blood glucose levels by 61.70%, 65.11%, and 74.79% and was able to repair damage to pancreatic histopathology of mice induced by alloxan.

Abstract

Taurine is a non-essential sulfur-containing amino acid with the chemical formula C2H7NO3S. These compounds play an important role in maintaining the smooth running of various processes in the body. Tetraselmis sp. is one of the microalgae known to have a taurine biosynthetic pathway via the serine/sulfate pathway. The purpose of this study was to determine the effect of culture salinity on taurine content produced by microalgae Tetraselmis sp. which was dissolved using 96% ethanol. The samples obtained were then tested using UV-Vis spectrophotometry using standard synthetic taurine as a comparison. Based on the research results, the maximum absorption was obtained at a wavelength of 630 nm, and the regression equation y = 0.001x + 0.033 was obtained and had a correlation coefficient of r = 1. Based on the analysis results showed that all samples contained taurine. Samples at a salinity of 25 ppt contained an average of 2.82 ml/100g of taurine, while a culture salinity of 35 ppt contained an average of 4.03 ml/100g of taurine. From these data it is known that the highest taurine level was found at a culture salinity of 35 ppt, while the lowest taurine level was at a culture salinity of 25 ppt. This indicates that the higher the culture salinity, the higher the taurine content produced by the microalgae Tetraselmis sp. The phenomenon of increased taurine due to salinity stress is thought to be due to an increase in free amino acids in microalgae cells to produce metabolites that are used to defend themselves due to salinity stress. The function of taurine in this microalgae is thought to be an organic osmolyte in the cells of the Tetraselmis sp. microalgae.

Abstract

Nile tilapia (Oreochromis niloticus) is a fish that is in demand because of its high economic value and thick meat. Tilapia is easy to breed, uncontrolled spawning can occur so that growth is inhibited. The growth of male tilapia is faster than females at the same age. To increase the growth rate, sex reversal is carried out towards males through immersion in steroid hormones. Bioactive substances contained in sea urchins are steroid compounds. The purpose of this study is to determine the effect of the length of immersion in sea urchin gonad extract (Diadema setosum) on the formation of male phenotypes of tilapia larvae and tilapia survival. The method used is a completely randomized design with 4 treatments and each treatment with 3 replicates at a dose of 4 mg/L with soaking times of 0, 12, 18, and 24 hours. Data are analyzed using SPSS 16 software with analysis of variance (One-way ANOVA). The results show that different lengths of immersion in sea urchin steroid extract at a dose of 4 mg/L affect the formation of male individuals. A soaking time of 18 hours is quite effective in directing the sex of fish to males by 66%. Immersion time also affects the survival of tilapia larvae.
