Abstract

Pengaruh dan penyebaran Ramayana sebagai sebuah konsep cerita yang bersumber dari sastra Ramayana tidak dipungkiri telah menyebar di Indonesia semenjak Hindu dikenalkan. Ada berbagai sumber yang dapat dijadikan bukti hidupnya cerita Ramayana di dalam kehidupan masyarakat Hindu khususnya. Di Bali sendiri cerita ramayana tidak saja menjadi pergulatan pengamat budaya khususnya sastra-sastra yang seringkali secara eksis digemakan lewat bentuk seni budaya. Kecak merupakan salah satu bentuk karya seni pertunjukan klasik yang mengambil inspirasi dari cerita Ramayana. Sangat beralasan ketertarikan karena menggugah pandangan masyarakat tentang nilai-nilai yang terkandung di dalamnya menjadikan bentuk bentuk garapan semakin menarik. Kecak yang seringkali diasosiasikan dengan kera tampaknya memberikan sentuhan harmoni musikal yang sangat tepat dengan latar budaya Ramayana. Ketokohan yang mengagumkan diperlihatkan oleh Hanoman atau kera berbulu putih yang senantiasa hadir demi menjaga dharma atau kebenaran dianggap menjadi inspirasi di dalam kehidupan masyarakat Hindu di Bali Inspirasi tersebut tampaknya menjadi alasan kuat bagi seorang peneliti India untuk memperlihatkan kepada dunia tentang pengaruh wiracerita yang dianggap berhasil menginspirasi masyarakat dunia.

Abstract

Merefleksikan unsur-unsur kesenian, khususnya seni pertunjukan, dalam ritual keagamaan, seni tari wali atau tari sakral merupakan salah satu unsur yang menunjang rangkaian segala upacara keagamaan yang ada di Bali. Artikel ini membahas salah satu tari sakral atau wali bernama Tari Baris Kekupu yang berada di Banjar Lebah Desa Adat Sumerta Kaja, Denpasar. Berdasarkan hasil penelitian, tarian ini merupakan bagian integral dari kehidupan masyarakat setempat yang selalu terikat dengan peristiwa ritual atau upacara Mamukur. Bentuk Tari Baris Kekupu, ditarikan oleh penari anak-anak perempuan karena didalam Tari Baris Kekupu ini terdapat unsur-unsur Tari Legong, Idenya terinspirasi dari hiasan kupu-kupu pada damar kurung yang dipasang saat upacara Mamukur. Selain itu, secara fungsional tarian ini dapat dipahami memiliki fungsi religius dan fungsi pelestarian budaya.

Abstract

The contamination of waste oil and grease to the aquatic environment from industrial food processing, restaurants and kitchens or by accidental oil spills is increasingly being found. Biodegradation using a consortium of bacteria can be an effective way to overcome this. The aim of this research was to isolate and identify bacterial strains from waste oil and grease that are capable of biodegradation and the potential of degrading bacteria to degrade oil and grease in vitro. Samples were taken from waste oils and fats. Enrichment techniques were carried out for all samples followed by isolation of bacterial strains to determine which strains could degrade oil and fat in vitro. Identification of bacteria using the identification kit BBL Crystal System. The total bacteria count was carried out using the dilution method. The results showed that there were 3 strains of oil and fat degrading bacteria, namely Bacillus licheneformis, Bacillus coagulant and Psedomonas diminuta. The consortium of the three types of bacteria was able to degrade waste oil and fat with a total of 624x105 CFU / ml to 816x105 CFU / ml bacteria with a control without the addition of a bacterial consortium which was 56x101 CFU / ml. The total bacteria in various treatment wastes with a consortium of bacteria based on molasses in the invitro test showed the ability to live and develop from a fairly high bacterial consortium, namely 292x104 CFU / ml to 904x104 CFU / ml. The bacteria found had the ability to degrade working oils and fats. synergistically. The benefit of this research is that bacteria that have been found can be used as a starter for processing waste fats and oils in environments contaminated with domestic waste fats and oils.

Abstract

Tanaman cabai rawit (Capsicum frutescens L.) merupakan salah satu tanaman hortikultura yang memiliki nilai gizi dan nilai ekonomi tinggi. Peningkatan produksi cabai dapat dilakukan dengan upaya perbaikan kesuburan tanah melalui pemupukan yang optimal dan ramah lingkungan. Air cucian beras dapat dimanfaatkan sebagai POC yang dikombinasikan dengan molase dan mikroba pelarut fosfat (MPF). Tujuan penelitian untuk mengeksplorasi pemanfaatan formulasi POC terhadap pertumbuhan tanaman cabai rawit dan menentukan perbandingan yang tepat antara air cucian beras dan molase dengan penambahan konsorsium MPF dalam memformulasi POC. Penelitian dilakukan dengan menggunakan Rancangan Acak Lengkap (RAL) satu faktor dengan 4 ulangan. Perlakuan ada 7 yaitu Y1 = Kontrol I (tanpa perlakuan); Y2 = Kontrol II (POC komersial); Y3 = 5% air cucian beras + 5% molase + 10 mL konsorsium MPF; Y4 = 10% air cucian beras + 10% molase + 10 mL konsorsium MPF; Y5 = 15% air cucian beras + 15% molase + 10 mL konsorsium MPF; Y6 = 20% air cucian beras + 20% molase + 10 mL konsorsium MPF; dan Y7 = 25% air cucian beras + 25% molase + 10 mL konsorsium MPF. Hasil penelitian menunjukkan bahwa pemberian perlakuan Y7 memberikan respon yang baik terhadap pertumbuhan tanaman cabai rawit dibandingkan dengan pemberian perlakuan formulasi POC lainnya.

Abstract

Escherichia coli O157:H7 and Staphylocccus epidermidis are bacteria that can cause disease in humans. Alternative control efforts are very important because the use of antibiotics can cause adverse side effects. This study aims to obtain extracts of white galangal (Alpinia galanga L.) in inhibiting the growth of E. coli O157:H7 and S. epidermidis. In this test, white galangal extract was macerated with three solvents at different polarity levels (96% ethanol, n-hexane and ethyl acetate). The ability of white galangal extract in inhibiting bacteria was tested using the Kirby Bauer method (paper disc). The concentration of galangal extract tested was 0%; 5% ; 10% ; 15%; 20% (w/v), control (ciprofloxacin) and MIC (Minimum Inhibition Concentration). To determine the toxicity of the extract, the LC50 test was carried out. Meanwhile, to determine the class of compounds contained in the galangal extract, phytochemical tests were carried out using the phytochemical screening method. The results showed that white galangal extract with 96% ethanol as solvent and n-hexane was able to inhibit the test bacteria. Inhibition against E. coli O157:H7 was most effective in n-hexane solvent with an extract concentration of 20% (inhibition zone diameter 20.2 mm). while for S.epidermidis was n-hexane solvent with a concentration of 20% (inhibition zone diameter 23.5 mm). LC50 against E. coli O157:H7 with 96% ethanol and n-hexane as solvents 11.2% and 10.4%, respectively. Meanwhile, the LC50 of S. epidermidis with 96% ethanol and n-hexane solvents were 9.9% and 9.3%, respectively. Qualitatively, white galangal plant extract contains a class of flavonoid compounds, saponins, terpenoids, and tannins.

Abstract

This study aims to find out the potency of garlic extract (Allium sativum L.) to the growth of Colletotrichum acutatum fungus which cause rot of red chili fruit post-harvest. The treatments were the administration of garlic extract in-vitro and in-vivo. In the in-vitro test, the inhibition of garlic extract was measured using well diffusion method. The in-vivo test was conducted by measuring the diameter area of C. acutatum on red chilies after treatments. The concentration of garlic extract given was 15%(b/v), 20%(b/v), 25%(b/v), 30%(b/v), and 35%(b/v) with positive control of synthetic fungicides and negative control of ethanol solvents 95% and 4 times replications. The average diameter result on the 25%(b/v) concentration of bland power test had the most effective bland zone diameter of 20.1±0.08. Statistically different was real (P?0.05) with negative controls. While in vivo tests the average area of infection area at a concentration of 35%(b/v) had an average diameter area of infection area which was best in inhibiting the growth of C. acutatum fungus.

Abstract

Seaweeds like other marine organisms develop a strong association with prokaryotic communities, especially bacteria. Many of these associated bacteria play an important role in synthesizing secondary metabolites which are beneficial for their host, especially against predation or infection. In Indonesia, Eucheuma cottonii is one of the seaweed species that has been cultivated widely.  However, only limited study has been reported on bacterial isolates and their antibacterial activity from E. cottonii.  Bacterial isolates from E. cottonii were cultivated in six agar media namely Zobell Marine Agar, Nutrient Agar, Plate Count Agar, ISP-1, ISP-2, and Starch-M protein agar. Cultivation of bacterial isolates yielded 23 isolates with nine bacterial isolates were classified as Gram-positive bacteria and 14 isolates were grouped among Gram-negative bacteria. All the bacterial isolates were screened for their antibacterial activity against six bacterial indicator strains namely Staphylococcus aureus ATCC 25923, Streptococcus mutans FNCC 0405, Escherichia coli ATCC 25922, and Klebsiella pneumoniae ATCC 700603 using perpendicular streak and agar block method. Six out of 23 bacterial isolates displayed antibacterial activity against at least one of the bacterial indicator strains. The bacterial isolate ISP1RL4 showed the highest antibacterial activity with average inhibition of  > 20 mm against all bacterial indicators. Overall, our result indicated the potential of bacterial isolates associated with E. cottonii as an antibacterial producer.

Abstract

Ikan kerapu adalah komoditas perikanan yang bernilai ekonomis tinggi pada pasar domestik dan internasional. Akan tetapi, dalam budidaya ikan kerapu masih sering ditemukan kasus penyakit vibrosis yang berdampak terhadap kerugian ekonomi akibat kematian yang ditimbulkan. Penyakit ini disebabkan salah satunya oleh bakteri Vibrio parahaemolyticus yang faktor virulensinya diregulasi oleh mekanisme kuorum sensing (QS). Pencegahan penyakit bakteri selama ini sering dilakukan dengan menggunakan antibiotik yang berisiko munculnya bakteri resisten antibiotik. Salah satu upaya untuk menghindari hal tersebut adalah dengan menghambat sistem QS menggunakan bakteri pendegradasi molekul sinyal N-acyl homoserine lactone (AHL). Pada penelitian ini dilakukan isolasi kandidat bakteri pendegradasi AHL pada media menggunakan AHL komersial sebagai sumber N tunggal dan juga melakukan isolasi bakteri V. parahaemolyticus pada media Thiosulfate-Citrate-BileSalts-Sucrose Agar dari saluran pencernaan ikan kerapu. Pengaruh penambahan isolat kandidat pendegradasi AHL terhadap tiga faktor virulensi V. parahaemolyticus yaitu motilitas, produksi kaseinase dan hemolisa diuji dengan mengkultur V. parahaemolyticus dengan dan tanpa penambahan isolat pendegradasi AHL. Hasil penelitian menunjukkan bahwa 2 isolat kandidat pendegradasi AHL yaitu SNA02 dan SNA03 mampu menurunkan ketiga faktor virulensi yang diuji tersebut. Hal ini mengindikasikan bahwa isolat SNA02 dan SNA03 berpotensi sebagai kandidat bakteri untuk mencegah penyakit vibriosis. Hasil ini diharapkan dapat membantu pembudidaya ikan kerapu dalam mengendalikan patogenitas bakteri V. parahaemolyticus.

Abstract

ABSTRACT Kombucha is a beverage made from tea as its basic ingredient and fermented by active starter of microbial consortia. The flavor of this fermented beverage is determined by many factors, such as supplementation of the main raw ingredient with various fruits, sugar, and types of tea used as its main raw ingredient. The main objective of this research was to elucidate the effect of fruit supplementation (addition of Fuji apple and strawberry) on the organoleptic quality of the products, following incubation periods of 4, 8, and 12 days. A triplicate complete randomized factorial design (types of fruits combined with incubation period) was applied in the experiment. Organoleptic tests (include taste, aroma, and color of the products) were conducted by applying quisionere method involving 7 panelis. The results showed that types of fruits and incubation period significantly affected (p<0.05) the organoleptic quality of the kombucha products. Overall, combination of stroberry and incubation period of 4 days gave the best results (p<0.05) when compared to control (commercially sold kombucha) and other treatments, indicating that such product has potential to be widely commercialized. Keywords: Alcohol, fermented beverage, fermentation, kombucha, organic acids ABSTRAK Kombucha merupakan minuman hasil fermentasi yang terbuat dari bahan dasar teh setelah diinokulasi dengan mikroba starter. Cita rasa kombucha sangat ditentukan oleh penambahan suplemen, seperti gula, buah-buahan, dan jenis teh yang dipakai. Tujuan utama penelitian ini untuk mengelusidasi pengaruh penambahan jenis buah (buah apel dan buah strobery) dan lama waktu fermentasi (4, 8, dan 12 hari) terhadap kualitas organoleptik produk kombucha yang dihasilkan. Penelitian ini menggunakan rancangan acak lengkap factorial (jenis buah yang dikombinasi dengan lama fermentasi) dengan 3 kali ulangan untuk setiap kombinasi perlakuan. Uji organoleptik (rasa, warna, dan aroma) dilakukan dengan metoda kuesioner yang melibatkan 7 orang panelis. Hasil penelitian menunjukkan bahwa jenis buah dan lama fermentasi berpengaruh nyata terhadap tingkat kesukaan panelis pada produk kombucha. Secara keseluruhan, perlakuan kombinasi penambahan buah stroberi dan waktu fermentasi selama 4 hari memberikan hasil terbaik, dan berbeda nyata dengan kontrol maupun perlakuan lainnya. Abstract

iThis researchhaimed to determineethe potency off Trichoderma asperellum TKD ffiltrate in inhibiting Aspergillus parasiticus contamination in peanut seeds. The treatment given to A. parasiticus was done in vitro and in vivo. In vitro there were three types methods, dual culture and the inhibition test of the T.asperellum TKD ifiltrate using the diffusion well and the colony method with concentrations of 40% (v/v), 50%(v/v), 60%(v/v),70%(v/v) and 80%(v/v). While in vivo, it was done on peanut seeds added T.asperellumiTKD filtrate with some concentrations similar to the treatment given in vitro. The parameters measured in vitro test were determined through PIRG (Percentage Inhibition of Radial Growth) and diameter inhibition zone, in vivo test parameter was the A. parasiticus colony populations. The results showed T.asperellum TKD filtrate significantly (P?0,05) was able to inhibit A. parasiticus growth iin vitro and iin vivo. The percentageeof T.asperellum TKD bland power against A.parasiticus in vitro with the highest dual culture method was 60.65%. In the method of colony and diffusion well, the concentration of 80% was the highest concentration in inhibiting A. parasiticus, which were 83.00±1.60% and 15.09±0.12 mm. T.asperellum TKD filtrate with a concentration of 80% added to peanut seeds had the highest inhibition on the number of population of A. parasiticus colonies with a percentage decrease of 88.3%.
