Abstract

Abstract

Plutella xylostella Linnaeus (Lepidoptera: Plutellidaae) is an important pest on Brassicaceae plants, especially cabbage. The combination of entomopathogenic fungi and botanical pesticides has been widely developed to increase effectiveness effectiveness against P. xylostella . This research aimed to determine the potential of Metarhizium anisopliae and Aglaia odorata Leaf Extract (Ao-LE) as a combined control strategy for P. xylostella. The parameters observed in this research were colony growth, viability of conidia of M. anisopliae, antifeedant activity, mortality, and lethal time 50 (LT50) of P. xylostella . The findings revealed that Ao-LE addition maintained colony growth comparable to that of the control treatment, while significantly enhancing the conidial viability of M. anisopliae. Notably, 0.80% and 1.00% Ao-LE in the M. anisopliae suspension resulted in maximum antifeeding activity. Moreover, incorporating 0.20-1.00% Ao-LE demonstrably improved P. xylostella mortality compared with M. anisopliae alone. The combination of M. anisopliae 107 conidia/mL and 1.00% Ao-LE exhibited the lowest LT50 value of 2.16 days, highlighting the synergistic effect of the combined treatment. In conclusion, this study provides compelling evidence for the compatibility and synergistic potential of M. anisopliae and Ao-LE, presenting a promising strategy for the sustainable management of P. xylostella infestations.

Abstract

This study aimed to explore the potential of phyllosphere bacterial isolates as biological control agents against Xanthomonas oryzae pv. oryzae (Xoo). We isolated a strain exhibiting typical morphological and biochemical characteristics of Xoo, such as a yellowish, spherical colony, Gram-negative, and starch hydrolyzing ability. This isolate was shown to cause necrotic lesions on rice leaves, corroborating its pathogenic nature. Subsequently, we isolated ten diverse phyllosphere bacteria from rice plants. These isolates were characterized based on various morphological and biochemical attributes including colony shape, margin appearance, color, growth time, and the ability to fluoresce under UV light when cultivated on King's B medium. Crucially, none of these isolates induced a hypersensitive response in tobacco leaves, an initial indicator of their potential as safe biological control agents. In an antagonistic assay against Xoo, all the phyllosphere isolates demonstrated varying levels of inhibition, suggesting their potential role in biological control. Two isolates, PGM1 and PGM4, exhibited the highest antagonistic effects against Xoo. These findings provide preliminary evidence for the potential use of these phyllosphere isolates in managing bacterial leaf blight in rice, warranting further research to confirm their efficacy and safety under field conditions.

Abstract

This study evaluates the effectiveness of Actinomycetes in encapsulating soybean seeds to control damping-off disease caused by Sclerotium rolfsii. With national soybean demand reaching 22 million tons per year and domestic production only 1 million tons, increasing production is essential. One major constraint is the attack of plant pathogens such as S. rolfsii, which can cause significant losses. Actinomycetes were isolated from the rhizosphere of weeds and tested for their antagonistic activity in vitro. Seed encapsulation using various carriers (kaolin, talc, and zeolite) was conducted to determine the most effective formula. In vivo tests showed that the Actinomycetes isolate PM2 exhibited the best antagonistic activity, with the zeolite formulation reducing pre-emergence damping-off disease incidence to 12.50% and post-emergence damping-off to 8.25%, compared to the positive control. These results indicate that the use of Actinomycetes in soybean seed encapsulation is an effective and sustainable method for controlling damping-off disease. Zeolite as a carrier proved to provide a supportive environment for antagonistic microbes, maintaining moisture and supplying necessary nutrients. This research reveals the great potential of Actinomycetes as a biocontrol agent in plant disease management strategies, which can significantly enhance soybean production and support national food security.

Abstract

Deleterious rhizobacteria (DRB) is one of the rhizobacteria that can be used as a bioherbicide and can stimulate plant growth. The objectives that can be obtained from this study are to determine the potential of DRB on the growth of weeds Echinochloa crusgalli and Echinochloa colona on rice. Variables observed in this study were identification of secondary metabolites, measurement of weed sprouts, percentage effectiveness of bacterial isolates against weed seeds, normal sprouts, abnormal sprouts, fresh seeds that did not grow and dead seeds. The aim of this study is to explore biological agents that can be used to control the main weeds of rice crops. This candidate could be a recommendation for environmentally friendly controls. Exploration of the DRB group was obtained by collecting soil samples around the roots of rice crops. Then isolate it, identify as morphologically and physiologically, and test it in vitro studies. The results showed that DRB had the potential to control weed growth and could stimulate the growth of rice. The application of PFA and PFB isolates had a significant effect on inhibiting the length of weed roots and buds, the average percentage of control effectiveness > 80-90%. Novelty in this study could be found as candidate groups of DRB bacteria with specific locations to control the weed Echinochloa sp. on rice crops. For society, this research could be an alternative to reduce dependence on synthetic herbicides . DRB has the potential to be formulated into a bioherbicide  to control weeds.

Abstract

Free-living nematodes often serve as the bioindicators of environmental quality. These nematodes are highly sensitive to even minute changes in the environment. Studies related to the presence of free-living nematodes have flourished, but to date, only a few studies have investigated the morphological and molecular characterization of free-living nematodes from agroecosystems in Indonesia. This study aimed to characterize the free-living nematode Ironus spp. from the rhizosphere of Arabica coffee plants in Bondowoso Regency, Indonesia. Nematodes were extracted from the soil using the white-head tray method and then preserved in a DESS solution to maintain their morphology and DNA. The morphometric analysis of Ironus species obtained in this study showed that the nematode species is identical to Ironus dentifurcatus. The small subunit (SSU) region of rDNA of the nematode was successfully amplified using primers (SSU F07 and SSU R 81). The results of phylogenetic analysis and sequence homology showed that the sequence in this study showed up to 95% homology to Ironus spp. In the database. This is the first study documenting I. dentifurcatus in Indonesia that integrates morphological and molecular characterization.

Abstract

Organic rice is cultivated without synthetic materials, offering high economic value and health benefits. Supportive biophysical components are essential for optimal growth and improving organic rice productivity. This research aims to identify the biophysical components that contribute to the land's carrying capacity for organic rice growth. The research was conducted in Rowosari Village, Jember Regency, East Java. The study analyzed on-farm activities, including producing organic fertilizers and pesticides, land mapping, agroclimate, and assessing biological components. The results show that biophysical factors significantly influence organic rice growth. Indicators of the success of organic fertilizer produced are N of 0.15%, P of 0.04%, K of 0.22%, and fertilizer pH of 4.98. The qualitative analysis of organic pesticides revealed the presence of secondary metabolites like tannin, saponin, flavonoid, alkaloid, and terpenoid with the code (+++). A high biological diversity was observed, with a Shannon-H index exceeding 1. Additionally, the concentration of organic fertilizers and pesticides significantly increased the number of rice plant saplings during the vegetative phase. The study concludes that the biophysical components of the land provide strong support for the growth of organic rice, demonstrating their vital role in the sustainability and productivity of organic farming systems.
