Abstract

Pectinase is used in a variety of industries, including fruit juice production, textile processing, papermaking, biodegumming, coffee and tea manufacturing, medicines, feed, and nutraceuticals. The demand for pectinase enzymes grows year after year. Industrial applications require high-activity enzymes that can increase considerable product yields while also improving based on needs and byproduct use. Indonesia's industry continues to rely on imported pectinase enzymes. Indonesia, on the other hand, possesses megabiodiversity, particularly marine microorganisms, which have the potential to provide new enzymes with high activity for industrial applications. This work aims to undertake bioprospecting of marine actinomycetes producing pectinase enzymes that have the capacity to hydrolyze pectin polymer, both from commercial and biomass sources. A total of 20 marine actinomycetes isolates from sediment, seawater, and sponges were bioprospected, and one isolate was selected with high pectinase activity (BLH 1.20), which was then used to characterize pH, temperature, hydrolysis analysis on pectin polymers, and isolate identification using 16s rRNA analysis. The selected isolate (BLH 1.20) performed best in a sodium phosphate buffer with a pH of 6.0 and a temperature of 30°C, achieving an activity of 5.4 U/mL. The 16S rRNA analysis revealed that the isolate is from the genus Streptomyces and the species Streptomyces sampsonii. Abstract

The converting process of coffee cherries to green coffee beans was conducted through the fermentation process using lactic acid bacteria (LAB) and yeasts to degrade coffee's mucilage. This present study aims to isolate acid producing bacteria and yeasts from three type of samples obtained from Arabica Coffee Plantations in Central Java and East Java, Indonesia. A total of 31 bacteria and 15 yeasts were obtained from this study. From 31 bacteria, 6 strains were identified as Lactobacillus sp. isolated from all samples; 15 strains were identified as P. pentosaceus; 4 strains as L. pseudomesenteroides; 1 strain as K. quasipneumoniae; and 5 strains as A. tropicalis from exocarp of coffee fruit. While from 15 yeast, 8 strains were identified as C. tropicalis isolated from soil; 2 strains as P. kudriavzevii from Luwak feces, and 5 strains as C. fabianii from exocarp of coffee fruit. A total of 25 strains of lactic acid bacteria can release lactic acid bacteria on MRS agar supplemented with 0.5% CaCO3, but 1 strain of K. quasipneumoniae and 5 strains of A. tropicalis produced acid weakly. Four strains of C. tropicalis showed the ability to form thin clear zone formation contrariwise with four other strains. This study adduced that waste materials and soil from the plantation coffee’s area are potential sources to obtaining acid-producing bacteria which is utilized in fermentation process of coffee cherries.

Abstract

Tea is one of the most popular beverages in the world. Produced from Camellia sinensis leaves, tea has been studied to provide health benefits due to the content of important metabolites. This study aimed to investigate the catechin contents, antioxidant, and antibacterial activities of Indonesian tea varieties, namely green tea, black tea, and white tea. Tea infusion was prepared by extracting 1 g of each sample into 10 mL of distilled water and incubated at 80 °C for 60 min. The catechin and epigallocatechin gallate (EGCG) contents of tea extracts were determined using high-performance liquid chromatography (HPLC). Antioxidant activity was measured using the free radical method with 2,2-diphenyl-1-pycrylhidrazyl (DPPH), while antimicrobial activity was assesed using paper disc diffusion assay. The results indicated that green tea had the highest contents of catechin (646 ± 17.14 mg/L) and EGCG (997.8 ± 36.72 mg/L), and antioxidant activity with IC50 of 5.65 µg/mL. Furthermore, green tea and white tea extracts showed inhibitory activity against Gram-positive bacteria such as Micrococcus luteus, Bacillus subtilis, and Staphylococcus aureus whereas black tea had no activity against all bacterial strains tested. Generally, we concluded that white tea and green tea contributed to the higher content of catechins and exhibited strong antioxidant and antibacterial properties.

Abstract

Bioprospecting has developed to all biological taxa including procaryotic. Actinomycetes become interesting procaryotic because of the ability to produce important secondary metabolite for human life. Actinomycetes are known as the largest antibiotic producer that has a broad range habitat. Some research has been done to find new antibiotic from the various habitat of actinomycetes. One of the interesting habitats of actinomycetes which never been explored in Indonesia is lichens... Lichens as the symbiotic structure of alga and fungi areknown as the ecological niche of various kinds of microorganisms including actinomycetes. Cibinong Science Centre (CSC) and Cibodas Botanical Garden (CBG) have various species of trees as the habitat of lichens. These areas are known as one of the research locations to explore the biodiversity of Indonesia. The aims of this research is to study the diversity and antimicrobial potency of actinomycetes isolated from 10 lichen samples with various type of thallus; crustose, fructose and foliose. Lichen samples were grown on the bark of 9 trees species in CSC and CBG. Isolation process used three agar media; HV, YIM6 and YIM711 with cycloheximide and nalidixic acid. Molecular identification based on 16S rRNA gene sequence. Antimicrobial activity was tested to 65 isolates by agar diffusion method to Bacillus subtilis BTCC B.612, Escherichia coli BTCC B.614, Candida albicans BTCC Y.33, Staphylococcus aureus BTCC B.611, Micrococcus luteus BTCC B.552. Isolation process retrieved 125 isolates with the highest number grow on HV agar medium. Based on the sample, the highest number of actinomycetes were isolated from crustose lichen attached on the bark of Averrhoea carambola. A total 69 isolates were identified as the genera Actinoplanes, Amycolatopsis, Angustibacter, Kribbella, Micromonospora, Mycobacterium, and Streptomyces. The screening process showed 24 isolates have antimicrobial activity, with the highest inhibitory activity against Micrococcus luteus BTCC B.552.

Abstract

Lactic acid bacteria (LAB) are important for prevention of spoilage and pathogenic bacterial growth in foods due to their ability to generate antimicrobial substances. The objective of this study was to screen LAB for antimicrobial activity and to optimize culture medium for antimicrobial production using Response Surface Methodology (RSM) with Central Composite Design (CCD). Optimization of antimicrobial production of selected LAB was conducted with different combinations of glucose, NaCl, inoculum, and temperature. Our experimental results showed that from 129 LAB isolates, 55 showed significant inhibition against Bacillus subtilis, Escherichia coli, Micrococcus luteus, Staphylococcus aureus, Aspergillus niger, and Candida albicans. No isolates inhibited the growth of Aspergillus flavus. Lactobacillus plantarum LIPI13-2-LAB011 was selected for further study on culture medium optimization to inhibit the growth of C. albicans. From statistical analysis, the production of antimicrobial substances was significantly influenced by temperature, NaCl, and concentration of glucose. Furthermore, the optimum concentrations of glucose, concentration of inoculum, temperature, and NaCl were 1.63 %, 3.03%, 33.74°C, and 3.4%, respectively, with a maximum predicted inhibition index of 1.916, which increased 3.56-fold compared to that obtained in medium before optimization processes. The result was confirmed as when the optimum concentration of nutritions used, the inhibition index increased 3.12-fold.

Abstract

In this issue, 5 original research papers are presented. The fIrst paper explains the authors attempts to purify and characterize an extracellular enantio seJective lipase from Aspergillus niger, an important component for the manufacture of c1ural drugs such asibuprofen. The second papers describe the identification of a highly abundant lectin in Arabidopsis thaliana. with potential application for biopesticides against nematode. In the third paper, the authors explain an association of a polymorphism in the a-lactoglobulingene on milk yield and quality in local sheep. The fourth paper describes the authors efforts to construct a synthetic human errythropoietin (EPO) in an attempt to obtain high-yield production of the protein in yeast. And the fifth papers describes the newly develop a method to do micropropagation of buah merah (Pandanus conoideus Lam), which is a potentially important source for anticancer medication. The Editor
