Abstract

Cellobiose dehydrogenase (CDH, EC 1.1.99.18) is an enzyme classified under the oxidoreductase group. CDH has been widely utilized in various fields, including healthcare such as in wound dressing and in the energy sector for bioenergy production. To support these industrial applications, it is necessary to investigate the characteristics of CDH to enhance process efficiency. Therefore, this study aims to characterize CDH produced by the fungus Trametes versicolor strain TRAMS. The research involved constructing a CDH production curve followed by enzyme activity assays, protein content determination, isoelectric point (pI) analysis, and kinetic studies using Michaelis-Menten and Lineweaver-Burk plots. The results showed that the enzyme exhibited an activity of 3.441 U/mL on the 8th day of production. The enzyme's specific activity reached 61.57 U/mg at the 80% ammonium sulfate fraction, with an isoelectric point at pH 4. Enzyme kinetics based on the Michaelis-Menten plot revealed a Vmax of 6.7 µM/min and a Km of 3.35 µM, while the Lineweaver-Burk analysis indicated a Vmax of 9.625 µM/min and a Km of 0.23 µM.

Abstract

Title: Small Steps-Big Impact: Walking Tour as Key to Sustainable Tourism in Kampung Peneleh Surabaya A way to preserve historical areas through tourism is by organizing walking tours. Recently, there has been a surge in walking tours offering experiences in spatial and architectural history for both domestic and international tourists. One example is the locally organized walking tours. However, studies focusing on tourist experiences during these walking tours, directly involving tourists as participants, are still limited. Identifying observation points along the walking tour route is crucial to determine which sites elicit positive or negative responses from participants. This article aims to explore tourists' spatial and architectural experiences and assess the contribution of walking tours to sustainable tourism. The research focuses on a case study in the historical village of Peneleh, Surabaya, where walking tours are conducted by a tourism awareness group (pokdarwis). The study employs a qualitative approach, utilizing observation and photo surveys, with six tourists serving as participants who joined other tourists on a walking tour package. The results show 154 photos representing four participant responses: liked photos (50.6%), disliked (10.4%), helpful during the tour (19.5%), and areas needing improvement (19.5%). Participants captured liked objects while also noting areas that require improvement. Additionally, walking tours offer broader access to various tourist sites that are not always open to the public. Those initiated by Pokdarwis can be sustainable tourism activities, continually innovating tour themes and offering new experiences for tourists.

Abstract

The decline in morality and spirituality among adolescents has become a significant challenge in modern education. Junior high school (SMP) is a crucial phase for character formation, particularly in instilling religious values. This study aims to describe and analyze the development of students' religious character at SMP Birrul Walidain Muhammadiyah Plupuh through the lens of Plato’s philosophy of idealism. Using a qualitative case study approach, this research provides a rich, in-depth analysis supported by detailed observations, in-depth interviews, and documentation. The findings indicate that religious character development is conducted through daily religious habituation, teacher role modeling, and the integration of Islamic values into the learning process. Activities such as congregational prayers, daily Qur'an recitation, weekly Islamic studies, and teachers’ religious examples demonstrate the school’s efforts to guide students toward Plato’s ideals of truth, goodness, and beauty. This study confirms that religious character education goes beyond cognitive learning and serves as a spiritual and moral journey toward the ideal self.

Abstract

This study aims to analyze the educational financing system at SMP Birrul Walidain Muhammadiyah Plupuh and its impact on improving the quality of education. The research employs a qualitative approach using a case study design. Data collection techniques include in-depth interviews, observations, and document analysis. The findings reveal that the school's educational financing is sourced from the school committee, parental contributions, and grants from the foundation. Fund management is carried out transparently and accountably, with appropriate allocation toward operational needs and quality improvement efforts. Effective financial management significantly contributes to enhanced teaching quality, improved school facilities, and the professional development of educators. This study concludes that sound financial management is a key factor in enhancing the quality of education in private schools.

Abstract

This study aims to determine the effectiveness of using Kahoot, a game-based learning platform, in improving student learning outcomes in Social Studies (IPS) at SMP Birrul Walidain Muhammadiyah Plupuh. The research employed a quasi-experimental method with a Nonequivalent Control Group Design. The subjects were 28 seventh-grade students, divided into two groups: VII A as the experimental group (using Kahoot) and VII B as the control group (using conventional teaching methods), each consisting of 14 students. Data were collected through pretest and posttest assessments. The results showed that the average posttest scores of the experimental group were significantly higher than those of the control group. The independent t-test confirmed a significant difference in learning outcomes between the two groups. It can be concluded that the use of Kahoot as a game-based learning medium is effective in enhancing students’ understanding and engagement in Social Studies learning.

Abstract

Anemia is a condition of not having enough number of red blood cells or hemoglobin to carry oxygen to supply the body's physiological needs. The occurrence of anemia in adolescent girls can be influenced by various factors. This study aimed to determine the factors that influence the incidence of anemia in adolescent girls at Senior High School in Samarinda city. The design of this research was analytical observational with the cross sectional approach. Primary data was obtained using the interview with Knowledge, Attitudes, and Practice (KAP) Questionnaire and SAMANTA Questionnaire, and hemoglobin levels were examined using haemometer digital. The sampling technique used was purposive sampling with 98 respondents represented by a 10th grade student in Senior High School 6 Palaran District. The bivariate analysis used was the chi-square test. The results of the analysis between anemia status and the level of knowledge about anemia, nutritional status, menstrual patterns, parental income, and maternal education sequentially obtained showed p-values of 0.645; 0.279; 0.654; 0.551; 0.776. Based on the results of the study, it is concluded that there is no significant relationship between the factors and the incidence of anemia in adolescent girls at Senior High School 6 Palaran District.

Abstract

Poteran Island is among the largest islands located within the Sumenep Regency, characterized by lowland agricultural areas with a dry E5 climate, allowing only one planting cycle of secondary crops per year. However, during the rainy season, the region tends to experience an abundance of wood-decaying fungi. This study aims to identify and evaluate the potential of wood-decaying fungi from Poteran Island as a source of MnP using morphological approaches. Screening of MnP-producing fungal isolates was carried out on L-MSM medium supplemented with methylene blue, and the clear zones were observed. The isolates were then identified macroscopically on petri dishes and microscopically using Lactophenol cotton blue, with reference to the book Pictorial Atlas of Soil and Seed Fungi: Morphologies of Cultured Fungi and Key to Species. Based on the morphological examination of 20 fungal isolates, 11 isolates displayed white mycelium, 1 isolate had gray mycelium, 2 isolates showed greenish-white mycelium, and 2 isolates had yellowish-white mycelium. Furthermore, 14 isolates exhibited circular colony morphology, 3 isolates were semi-circular, and 3 isolates were filamentous (filiform) in shape. MnP-producing fungal isolates were screened using L-MSM medium supplemented with methylene blue. Only one isolate, DS 18, demonstrated MnP production potential, indicated by the decolorization of methylene blue on medium. Macroscopic and microscopic analyses confirmed that isolate DS 18 is Trametes versicolor, as it exhibited similar characteristics, including a cottony texture, white color, smooth colony edges, non-septate hyphae, elliptical spores, and hyphae with clamp connections.

Abstract

Shipbuilding industry is an industry group which has high risk. For that reason, the managementshould include risk assessment. The production process of new buildings in theshipbuilding industry is grouped into three major parts, namely the work of design, materialprocurement and production processes. Each stage of the production process will bring therisk and will accumulate on the overall risk. If the risk is not anticipated, the possibility ofdelays in the production process will be even greater. Risk assessment performed on eachproduction process by using a probabilistic approach to the principle of multiplication in thetheory of opportunity. Risk analysis performed on the construction of fast patrol boats was inthe construction number 268, 269 and 270 on the PT. PAL Indonesia. From the analysis, itwas obtained the greatest probability of occurrence of sequential delay which is in the processof the material, the production, and design group. The potential for loss is due to the riskof unanticipated costs due to factors affected by delays in production processes. Performancefactors are still required for further study.
