Abstract

Thalassemia is a genetic disorder caused by point mutations on the globin gene that decrease the corpuscular index in thalassemia carriers. Three to five percent of Indonesians are thalassemia carriers, with β-thalassemia being the most common type. This research aimed to identify the relationship between the number of mutated regions on the β-globin gene and the decrease in corpuscular index among β-thalassemia carriers. Data was collected from 2012 to 2013 in Yogyakarta. Hematological analysis was performed by assessing the corpuscular index, including mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), and mean corpuscular hemoglobin concentration (MCHC) at Prodia Laboratory. Molecular analysis was conducted using the polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) method at the Laboratory of Genetics and Laboratory of Falitma, Biology Faculty, University of Gadjah Mada. Out of a total of 96 individuals screened, there were 9 suspected β-thalassemia carriers with 1 mutated region of the β-globin gene, showing an average MCV of 63.1 fl, MCH of 19.76 pg, and MCHC of 32.34 g/dl. Seven suspected β-thalassemia carriers with 2 mutated regions showed an average MCV of 61.16 fl, MCH of 19.74 pg, and MCHC of 32.3 g/dl. One suspected β-thalassemia carrier with 3 mutated regions showed an average MCV of 64.2 fl, MCH of 19.5 pg, and MCHC of 30.4 g/dl. The number of mutated regions in the β-globin gene was not the main factor affecting the decrease in corpuscular index among β-thalassemia carriers.

Abstract

Abstract The stingless bee or stinglessbee has recently been promoted as a very promising farm animal. However, there is still very little literature that can be used in the cultivation of these bees. . This study aims to determine the characteristics of beebread collected by 7 species of stinglessbee. This research was conducted in February – May 2022, the beebread samples were taken directly from bee hives at PT Suhita Lebah Indonesia, Bandarlampung city and from bee cultivators in Harapan Jaya village, Wayratai district, Pesawaran district. Samples were analyzed by acetolysis method and observed using a microscope and grouped according to the provisions of Wingenroth (2001). From the observations made, it is known that the species Tetrigona apikalis, Tetrigona vidua, and Tetragonula reepeni have the characteristics of beebread monoflora with pollen dominated by Cocos nucifera plants. Beebreads Tetragonula testaceitarsis and Tetragonula levicep have biflora characteristics, and Heterotrigona itama and Geniotrigona thoracica beebreads have multiflora characteristics. Of the seven species of bees observed, they collected pollen from the C. nucifera plant and had a high percentage. The significant amount of C. nucifera pollen was due to the Stinglessbee's preference for foraging on flowers with high sugar concentrations available throughout the year. Keywords : Beebread, Geniotrigona, Heterotrigona, Stinglessbee, Tetragonula, Tetrigona.

Abstract

Kopi menjadi salah satu komoditas andalan Indonesia.

Abstract

Stingless bees are an abundant natural resource in various areas of the world. The utilisation of stingless bees is associated with their honey production. Therefore, stingless bees are widely cultivated in the community. The increasing number of cultivators also occurs in Lampung Tengah Regency. However, this increase in the number of cultivators has not been followed by research on the diversity of stingless bees, including based on their morphology. This research was implemented from May to August 2024. Explorative fieldwork was performed in Sido Mulyo, Punggur, Lampung Tengah. According to the findings of the exploration, samples were taken from seven different colonies. The samples were then observed on 11 morphological characters including (1) eyes, (2) forewings, (3) body colour, (4) thorax, (5) abdomen, (6) hammuli, (7) mesonotum, (8) mesoscutetellum, (9) propodeum, (10) hair on posterior hind tibia, and (11) elliptical disc on basitarsus. The observed samples are closest to the characteristics of species in the genus Heterotrigona (samples SK3, SK5, SK6, and SM1) and Tetragonula (samples SK1, SK2, and SK4) on the basis of morphological characters.

Abstract

Robusta coffee (Coffea canephora) genetic diversity is very important for breeding programmes aimed at increasing yield and adaptability. This diversity can be assessed through various morphological characters, especially leaf phenotype characters, which serve as indicators of genetic variation. This research was conducted from May to August 2024. Data collection was carried out by purposive sampling using the exploration and characterisation method of 30 coffee plants in the Traditional Coffee Plantation area, Bogorejo Village, Gedong Tataan District, Pesawaran Regency, Lampung Province. Qualitative character data collection was carried out modifying IPGRI descriptors. Based on the data obtained, there were no significant differences in qualitative characteristics and quantitative characteristics of leaves on 30 coffee plants. This indicates a low genetic diversity in the plantation.
