Abstract

Sediaan parasetamol yang beredar di pasaran saat ini berbentuk tablet, tablet kunyah, sirup, suspensi dan emulsi. Ketersediaan formula obat parasetamol untuk anak di Indonesia masih terbatas. Tujuan dari penelitian ini untuk melakukan suatu pengembangan sediaan parasetamol yang acceptable atau mudah diterima oleh masyarakat dalam bentuk sediaan permen kenyal yaitu gummy candy.            Rancangan formula gummy candy parasetamol untuk di optimasi dengan metode Simplex Lattice Design (SLD) kemudian pembuatan 8 run gummy candy variasi konsentrasi gelatin dan karagenan. Pengujian mutu fisik masing-masing formula gummy candy meliputi pengujian organoleptik, kekenyalan, pH dan kadar air. Validasi metode analisis dengan parameter akurasi, presisi, spesifitas, linearitas dan LOD LOQ kemudian dilanjutkan dengan pengujian keseragaman kandungan sediaan gummy candy parasetamol dengan menggunakan spektrofotometer UV-Vis.            Hasil penelitian konsentrasi gelatin dan karagenan berpengaruh terhadap kekenyalan, pH dan kadar air sediaan gummy candy parasetamol. Gelatin berpengaruh dominan terhadap peningkatan kekenyalan, karagenan berpengaruh dominan terhadap peningkatan pH dan kadar air. Formula optimum didapatkan dengan proporsi gelatin 599,226 mg dan karagenan 400,774 mg.

Abstract

Dental caries is a dental infection with a high prevalence in Indonesia, one of the causes is Streptococcus mutans which demineralizes the tissues causing dissolution and dental caries. To reduce the number of microorganisms, people can utilize natural materials. One of the plants that have potential as an antibacterial is johar leaf and avocado leaf. The purpose of this study was to determine the antibacterial activity of the combination of ethanol extract of johar leaves (Cassia siamea Lamk) and avocado leaves (Persea americana Mill.) against Streptococcus mutans. This study uses the diffusion method. Extraction of johar leaves and avocado leaves was carried out by maceration method then the extracts were combined 1:1 with concentration series of 25%, 50%, 75% and 100%. Antibacterial testing uses the diffusion method to determine the inhibition. Streptococcus mutans bacteria were cultured at a density of 108 CFU/ml in MHA (Muller Hinton Agar) medium. Inhibitory power is known by measuring the diameter of the inhibition zone formed around the well. The average inhibition test results were obtained at concentrations of 25% (6 mm), 50% (6.1 mm), 75% (7.1 mm) and 100% (9.3 mm). The positive control of ciprofloxacin (30 mm) and the negative control did not show the diameter of the inhibition zone. So that the greater the concentration of the extract combination, the greater the diameter of the inhibition zone obtained to inhibit the growth of Streptococcus mutans and there is a significant difference in the diameter of the inhibition zone of the combination of extracts and ciprofloxacin positive control.

Abstract

Bacterial endophyte is an endosimbiont which lives within host plans without causing apparent disease on its host plans. Some endophytic bacteria found in plants have the capability to produce potential compounds as antibacterial. This study aimed to determine the antibacterial activity of the endophytic bacteria fermentation from the tubers of taro plants and determine the optimum time of fermentation which produce the highest antibacterial activity. This research was initially started by producing the suspension of the endophytic bacteria and test bacteria. The suspension that had been made then was used to characterize and identify the endophytic bacteria and test bacteria. Identification methods include colony identification, Gram staining, Spore staining and biochemical tests planted on MCH, SIM, LIA and Citrate mediums. The fermentation was then carried out to produce secondary metabolite compounds which were utilized to examine the antibacterial activity against Pseudomonas auriginosa. The antibacterial activity test was conducted by the disk diffusion assay. The results of the antibacterial activity assay showed that Bacillus siamenssis and Bacillis subtilis were both able to produce secondary metabolites that have potential as antibacterial against Pseudomonas earuginosa. The highest value was achieved by 72 hours fermentation time with a value of 8.7 by Bacillus siamensis. The endophytic bacteria Bacillus subtilis had the highest activity value at 48 hours fermentation time with an average of diameter of inhibition zone of 8.97.

Abstract

Reactive oxygen spesies merupakan radikal bebas yang apabila jumlahnya berlebih akan menyebabkan stres oksidatif. Reactive oxygen spesies dapat dihilangkan dengan pemberian antioksidan enzim, seperti enzim superoksida dismutase. Tanaman yang memiliki aktivitas superoksida dismutase salah satunya adalah temu hitam (Curcuma aeruginosa Roxb.). Penelitian ini bertujuan untuk mengetahui kadar protein total dan persen inhibisi ekstrak kasar enzim superoksida dismutase temu hitam dan konsentrasi amonium sulfat 25, 50, 75, 100% serta nilai persen inhibisi yang optimum dari konsentrasi amonium sulfat. Penelitian ini diawali dengan determinasi tanaman dan pengambilan sampel temu hitam. Ekstraksi dilakukan dengan cara penambahan dapar fosfat salin dan disentrifugasi. Pemurnian dilakukan dengan metode presipitasi amonium sulfat. Penetapan kadar protein total dengan menggunakan metode Lowry. Pengukuran aktivitas enzim dilakukan dengan menggunakan metode water soluble tetrazolium salt-1. Hasil penelitian menunjukkan bahwa kadar protein total ekstrak kasar dan presipitasi amonium sulfat 25, 50, 75, 100% secara berturut-turut nilainya sebesar 0,909; 0,639; 0,710; 0,752; 0,944 mg/ml. Kemudian nilai persen inhibisinya secara berturut-turut adalah 76,720; 23,810; 65,079; 70,370; dan 83,069% dengan konsentrasi yang optimum adalah 100%.

Abstract

Spirulina platensis, a blue-green algae abundant in tropical regions, is rich in minerals, vitamins, fibers, and pigments, with low nucleic acid content. It has unique chromoproteins called phycobiliproteins, notably phycocyanin, used in various applications. This study aims to optimize phycocyanin extraction using different solvents (distilled water and sodium phosphate buffer pH 6.7) and methods (freeze-thaw and sonication). Spirulina platensis biomass was extracted in both solvents, then some of them was freeze for 24 and 48 hours followed by thawing overnight. The other was sonicated for 2.5 minutes, 50 Hz then soaked for 1, 2, and 3 hours. All of the samples were centrifuged at 6000 rpm for 10 minutes and the absorbance was measured using a UV-Vis spectrophotometer at wavelengths of 280, 620, and 650 nm. with freeze-thawing for 48 hours yielded the highest phycocyanin concentration (0.55%), with a yield of 11.07 and purity of 0.21. Sonication improved phycocyanin concentration, yield, and purity significantly, yielding 1.108, 25.85, and 0.26, respectively.

Abstract

Sampai saat ini, Indonesia masih kesulitan menangani persoalan sampah. Oleh karena itu, perlu dilakukan perubahan paradigma yang memandang sampah sebagai sumber daya yang memiliki nilai ekonomis dan dapat dimanfaatkan. Kkhususnya limbah anorganik seperti kaleng dan plastik banyak dimanfaatkan untuk pembuatan kriya logam, campuran beton, bahan pembuatan MMC (Metal Matrix Composites) dan bahan dasar koagulan. Untuk mendukung warga Desa Doplang meningkatkan nilai jual komoditi bunga sedap malam, Teknologi Tepat Guna (TTG) alat destilasi/ penyulingan sederhana dari limbah anorganik dibuat. Metode yang digunakan adalah dengan pendekatan community development, empowerment dan community engagement. Setelah melakukan perijinan ke mitra, sosialisasi tentang potensi limbah anorganik dan pelatihan pembuatan alat destilasi sederhana dilakukan. Alat destilasi sederhana berhasil dibuat dengan memanfaatkan limbah organik. Dari kegiatan ini, warga Desa Doplang diharapkan mampu mengekstrak minyak atsiri bunga sedap malam menjadi produk turunannya.

Abstract

Changes in the social behavior of the Kuta Traditional Village community are becoming increasingly significant along with the impact of the ongoing modernization era. Kuta Traditional Village, a community rich in traditions and cultural values, is experiencing transformation in various aspects of their daily lives. This study explores how modernization affects the social behavior patterns of the Kuta Traditional Village community, from traditions to their social interactions. Through an in-depth qualitative approach, the results of this research describe the dynamics of social change occurring in this traditional village and seek an understanding of how they adapt to the ever-evolving modern era.

Abstract

Background: Antioxidants are compounds that inhibit cell damage caused by free radicals. Antioxidant enzymes include superoxide dismutase (SOD), glutathione, and catalase. SOD protects cells against free radicals implicated in various diseases. The guava plant (Psidium guajava L.) is known to possess SOD activity.Objective: This research was performed to determine the activity of the superoxide dismutase enzyme in guava leaf extract (Psidium guajava L.) with ammonium sulfate concentrations of 25, 50, and 75%.Methods: Guava leaves were extracted using phosphate buffer and centrifugation. The superoxide dismutase (SOD) enzyme was then purified using ammonium sulfate at concentrations of 25, 50, and 75%. The amount of protein was measured with the Lowry method, and the activity of the SOD enzyme was tested using the WST-1 assay to find out how much it was inhibited. The resulting data were statistically analyzed using one-way ANOVA.Results: Guava leaf extract exhibits SOD enzyme activity. The total protein content of the crude guava leaf extract and the extracts purified with 25, 50, and 75% ammonium sulfate were 9.683, 6.958, 8.842, and 11.269 mg/mL, respectively. The corresponding percent inhibition values were 69.652, 35.323, 61.69, and 79.104%. The 75% ammonium sulfate concentration yielded the highest percent inhibition.Conclusion: Guava leaf extract exhibits SOD enzyme activity, with the highest percent inhibition observed at an ammonium sulfate concentration of 75%.

Abstract

Endophytic bacteria are bacteria that carry out symbiosis with host plants without causing disease without the host plant. Some endophytic bacteria found in plants are able to produce potential compounds as antioxidants by producing the enzyme SOD (Super Oxide Dismutase). This research aims to determine the results of endophytic bacterial fermentation from the leaves of the breadfruit plant (Artocarpus altilis). This research began with the identification of endophytic bacteria, then continued with the extraction of the SOD enzyme by adding PBS and centrifuging. Purification of the SOD enzyme was carried out using the ammonium sulfate precipitation method. Determination of total protein content using the Lowry method. SOD activity was measured by measuring the percent inhibition value using the Superoxide Dismutase Activity Assay Kit WST-1. The results of the research showed that the isolated bacteria had morphological characteristics in sequence from colony shape, color, size, elevation, edges, cell shape and Gram type for AL 4 bacteria Filamentous, white, medium, flat, Rhizoid, coccus, Gram +, citrate -, Catalase -, for IL11 bacteria Irregular, white, large, flat, entire, bacilli, and Gram +, citrate +, Catalase +. The extraction results from the crude extract produced a sediment weight of 2,915 g for AL4 and 1,653 g for IL11. The total protein content of the crude extract and 30% ammonium sulfate precipitation for isolate AL4 was 1.74 mg/mL and IL11 1.79 mg/ml. Then the percent inhibition value for isolate AL4 with a concentration of 20% was 64.02% and a concentration of 80% was 71.43%.

Abstract

This article examines the potential for integrating digital technologies into the teaching of Catholic religious education. The study analyzes data from interviews with 20 Catholic religious teachers in secondary schools, which indicate that digital platforms can enhance teaching effectiveness and student engagement. However, challenges such as the digital divide and limited teacher training are also identified. This article provides practical guidance for overcoming these barriers and leveraging technology to support the mission of evangelization.
