Abstract

Ikan tuna (Thunnus albacares) merupakan salah satu komoditas hasil laut yang banyak ditemukan di Pusat Pendaratan Ikan (PPI) Kota Gorontalo dan sering dikonsumsi masyarakat. Penanganan pasca tangkap yang kurang baik dapat memicu kontaminasi bakteri sehingga berdampak pada penurunan mutu ikan. Penelitian ini bertujuan untuk mendeteksi cemaran bakteri Escherichia coli dan Salmonella sp. pada ikan tuna jenis T. albacares di PPI Kota Gorontalo. Penelitian bersifat deskriptif dengan pengambilan sampel dari tiga titik berbeda, masing-masing satu sampel. Pengujian E. coli dilakukan dengan metode MPN tiga seri tabung, sedangkan Salmonella sp. diuji menggunakan metode kultur pada media selektif Salmonella Shigella Agar (SSA). Hasil penelitian menunjukkan bahwa seluruh sampel ikan tuna mengandung E. coli dengan nilai MPN berturut-turut 7,8 × 10³ APM/gr, 6,5 × 10³ APM/gr, dan 14,1 × 10³ APM/gr, sehingga tidak memenuhi syarat mutu SNI 2729:2013 yang mensyaratkan E. coli kurang dari 3 APM/gr untuk ikan segar. Sebaliknya, uji Salmonella sp. menunjukkan hasil negatif pada semua sampel, sehingga memenuhi baku mutu SNI 2729:2013 yaitu negatif per 25 gram (0/25 g).

Abstract

Differentiated learning is the core of the independent curriculum now.The aim of this research is to develop an instrument for measuring the cognitive learning outcomes of high school students based on differentiated learning on elasticity material and to obtainits characteristics. Differentiation, in this case, is differentiation of student learning styles. The instrument grid is arranged based on indicators of cognitive learning outcomes, which are then used to compile question items. The test instrument consists of two test sets, each of which has 9 items including two anchor itemsso that the total number of items is 16 items and has been validated by 6 experts. The validated instrument was tested on 252 respondents spread across high schools in the low, medium and high categories. Dichotomous data were analyzed using the Partial Credit Model (PCM). The trial results showed that all 16 items and the differentiated learning-based student cognitive learning outcomes test instrument were proven to be valid, fit the PCM model, and reliable, which means all items were in the good category. Thus, the test instrument developed meets the requirements for measuring the cognitive learning outcomes of high school students on elasticity material based on differentiated learning.

Abstract

Actinomycetes show promise as biological control agents, reducing fungicide use in agriculture due to their bioactive compounds and ecological resilience. This study focused on the initial screening and ecological bioprospecting of actinomycetes from the Hungayono karst ecosystem in Gorontalo by determining their antagonistic activity against the pathogenic fungi Fusarium oxysporum and Neocosmospora solani, as well as their tolerance to the fungicides thiram and benomyl. Potential isolates were identified using molecular methods. Samples were collected from three locations. Rhizosphere soil samples were obtained from plant species: Alocasia macrorrhizos, Ficus microcarpa, and Acrostichum aureum L. Antagonistic activity against root-pathogenic fungi and tolerance to fungicides were used as initial screening criteria to assess their potential as biological control agents. This study successfully isolated 9 actinomycete isolates. The results showed that only 33% of the isolates exhibited inhibitory activity, with isolate RzAH-07 showing a moderate inhibition zone (5.22 mm) against Fusarium oxysporum and weak inhibition (4.18 mm) against Neocosmospora solani, while also demonstrating tolerance to the fungicide benomyl up to 1000 ppm. RzAH-07 was selected as the top candidate due to its two key advantages. Phylogenetic analysis based on 16S rRNA gene sequences indicates that RzAH-07 is closely related to the genus Streptomyces, with a similarity percentage ranging from 98.35% to 99.14% based on GenBank data.

Abstract

Actinomycetes are soil microorganisms that play a crucial role in nutrient cycles and interactions, particularly in nutrient-limited environments such as karst ecosystems. This study aimed to evaluate the preliminary functional potential of actinomycetes isolated from the plant rhizosphere in the Hungayono karst ecosystem, Gorontalo, as phosphate solubilizers and producers of indole acetic acid (IAA). The study employed a quantitative descriptive approach with soil sampling at a depth of 10–15 cm using purposive sampling. Isolation was performed using the spread plate method on Starch Casein Agar medium, followed by qualitative and quantitative testing of phosphate solubilization and IAA production capabilities. Identification of selected isolates was conducted through 16S rRNA gene-based phylogenetic analysis. The results showed that 11 actinomycete isolates were obtained from 4 host plants, of which two isolates exhibited low-index qualitative phosphate solubilization activity. However, quantitative results indicated that some isolates that did not form clear zones still produced measurable amounts of soluble phosphate, suggesting differences in method sensitivity. The highest phosphate-solubilizing activity was exhibited by isolate RzPO-09. In addition, four isolates were capable of producing IAA at varying levels, with the highest production observed in isolate RzPH-07 at 0.896 ppm. Phylogenetic analysis showed that isolate RzPO-09 (PZ458677) is closely related to the genus Pseudonocardia, while RzPH-07 (PZ458434) is closely related to the genus Streptomyces. Overall, the isolates obtained show initial functional potential; however, further validation is required through optimization of culture conditions, genetic characterization, plant growth tests, and testing both in greenhouses and in the field to ensure their practical feasibility.
