Abstract

Spermatozoa in fresh semen of Sapudi ram has a limited life span. The storage of semen in cold temperatures (5 °C) is intended to prolong the spermatozoa's life. However, storage in cold temperatures can lead to increased production of reactive oxygen species (ROS). This condition reduces the quality of spermatozoa. The purpose of this study was to determine the effect of alphatocopherol supplementation in skim milk-egg yolk extender on viability, motility, and plasma membrane integrity of Sapudi ram spermatozoa. Fresh semen derived from Sapudi ram was divided into four treatment groups. Control treatment (P0): semen was added in the extender of skim milkegg yolk without alpha-tocopherol. Three other treatments: P1, P2, and P3 semen were added in skim milk-egg yolk extender with the supplementation of 0.25, 0.5, and 1 gram alpha-tocopherol/ 100 mL extender, respectively. The results showed that the viability, motility, and integrity of the spermatozoa plasma membrane decreased gradually according to the storage length. Supplementation of skim milk-egg yolk extender with 0.5 gram of alpha-tocopherol/100 mL (P2) was able to maintain spermatozoa quality longer (p <0.05) than the control group. It can be concluded that alpha-tocopherol with a concentration of 0.5 g/100 mL of skim milk-egg yolk extender effectively maintains the quality of Sapudi ram spermatozoa in storage at 5 ° C.

Abstract

This case study aims to determine the effect of a combination of prostaglandin F2α (PGF2α) with Gonadotropin (PG-600), and human chorionic gonadotropin (hCG) on the estrus and pregnancy rates in Holstein Friesian (HF) cows with persistent corpus luteum (PCL). A total of 15 Holstein Friesian (HF) dairy cows aged 3-5 years with body condition score (BCS) 5-7 (scale 1-9) diagnosed with PCL, were injected with 7.5 mg PGF2α intra-vulvar submucosa. Cows were then divided into three treatment groups. Group P0 was not given the combination treatment, while P1 group was injected with a  combination of 300 IU PG-600 and 150 IU hCG, meanwhile P2 group was was injected with a combination of 300 IU PG-600 and 300 IU hCG intra-muscularly. The PGF2α and PG-600 injections were carried out on the same day PCL was diagnosed. Meanwhile, the injection of hCG was carried out at the same time as the AI. The Pregnancy examination was performed 90 days after AI through rectal palpation. The results showed that all treated cows were estrus  observed with the fastest onset of estrus was the P2 group, that was significantly different (p <0.05) compared to the P0 and P1 groups. All treated cows were diagnosed as pregnant. It can be concluded that the administration of PGF2α or its combination with gonadotropins in HF cows with PCL could result in 100% estrus and pregnancy rates.

Abstract

This study was conducted to determine the effect of crude protein tyrosine kinase (PTK) isolates supplementation into skim milk-egg yolk based diluent to maintain the quality of Merino ram spermatozoa.  Four ejaculates of two Merino rams were divided into two groups for the control group (P0): Merino ram semen was diluted in skimmed milk-egg yolk based diluent, and the treatment group (P1): Merino ram semen was diluted in skim milk-egg yolk based diluent contained crude PTK 1,597 mg/ml diluent. All diluted semen was equilibrated for 2 hours at 5 °C and filled into 0.25 mL French straws. The filled straws were placed on steel racks (Cooltop, Minitube) held in liquid nitrogen vapour for 10 minutes at –140 °C, immersed immediately in liquid nitrogen at –196 °C, and stored for 48 hours for  later assessment. Post-thawed semen samples were evaluated for spermatozoa motility, viability, and morphological abnormality. The results showed that the spermatozoa motility of fresh semen of Merino ram was 82.5 ± 2.89, which was qualified for freezing. The post-thawing spermatozoa motility, viability, and morphological abnormalities of Merino ram in the P1 group were 34.11 ± 3.26%, 38.00 ± 3.00%, and 12.89 ± 4.54%, respectively. It were higher (p <0.05) than the control group of 24.44 ± 2.9%, 26.67 ± 3.32%, and 21.11 ± 3.02%. It was concluded that the addition of crude PTK isolates of 1.597 mg/ml skim milk-egg yolk diluent improved the quality of post-thawed spermatozoa of Merino ram.

Abstract

This research was conducted in the teaching farm of the Faculty of Veterinary Medicine, Universitas Airlangga. The present study aimed to determine the effect of age, temperature, and humidity on bull semen quality which was processed into frozen semen. Three age groups of bulls (young 3-4 years, middle age 5-6 years, and old 7-8 years) were collected for the temperature and humidity of semen examination. Fresh semen was collected using an artificial vagina, followed by macroscopic and microscopic examination. In addition, post-thawing motility was also evaluated. The results showed that there was no effect of temperature and humidity during semen collection process. Fresh semen quality was affected by age. Moreover, the post-thawing motility of fresh semen of spermatozoa was lower than those of the fresh semen; however, it still qualifies for artificial insemination. It could be concluded that the semen of bulls at the Teaching Farm of Universitas Airlangga has good quality, and is suitable for Artificial Insemination.

Abstract

Bali cattle, developed in Indonesia, are renowned for their high parasite resistance, adaptability, and reproductive health. This study aimed to evaluate the impact of age on the kinematic parameters of sperm in Bali bulls post-thawing. Semen samples were obtained by artificial vagina methods from three Bali bulls aged between 11 and 13 years, whose age was not indicated, and one bull of 9 years, which was used as a control. The experiment was conducted in December 2023 at the Singosari National Artificial Insemination Center in Malang, East Java, for sperm parameters, including viability, motility, and morphology. Computer-assisted sperm analysis was performed. The results showed a decline in sperm quality from fresh to frozen semen. Despite this, the semen from Bali bulls met the Indonesian National Standard, demonstrating the bulls' superior reproductive capabilities. Bulls aged 11–13 years exhibited variations in sperm quality; however, these differences may reflect individual variability rather than a consistent age-related trend. Semen from bulls aged over 10 years remain suitable for use, though regular testing is essential. Further research is needed to understand these variations and ensure optimal semen quality across different ages.
