Abstract

The research aimed to investigate the effect of dehydroepiandrosterone (DHEA) on lipid peroxidation activity and nitric oxide concentration in the aorta of male Wistar rats due to exposure to cigarette smoke. This study was a laboratory experiment using 21 rats that were divided into 3 groups: KN-14 (negative control that is given smoke exposure within 14 days), DHEA-1 (15 mg/kgBW + smoke exposure for 14 days), and DHEA-2 (30 mg/kgBW + smoke exposure for 14 days). The exposure was carried out in 25-45 minutes with 4 cigarettes/day. Surgery was performed in the 15th days to remove the aorta to examine malondialdehyde (MDA) as a biomarker of lipid peroxidation and nitric oxide (NO) concentration using Enzyme-linked Immunosorbent Assay (ELISA). The results showed that a significant increase in MDA concentrations was found in the group administered with DHEA, particularly at a dose of 30 mg/kgBW (p<0.05). Meanwhile, the administration of DHEA at doses of 15 and 30 mg/kgBW did not have a significant effect (p>0,05) on aortic NO concentrations compared to the group KN-14. Therefore, it can be concluded that the administration of DHEA could increase lipid peroxidation activity, as indicated by a significant increase in aortic MDA concentrations, without affecting aortic NO concentrations in rats exposed to cigarette smoke for 14 days.Keyword: Dehydroepiandrosterone (DHEA), MDA, NO, lipid peroxidation, exposure of cigarettes smoke

Abstract

This study investigated the effect of dehydroepiandrosterone on the lungs of male Wistar rats exposed to cigarette smoke. A total of 21 male Wistar rats were divided into 3 groups, namely the first group (negative control exposed to cigarette smoke for 14 days), the second group (given DHEA at a dose of 15 mg/kgBW and exposure to cigarette smoke), the third group (given DHEA at a dose of 30 mg/kgBW and exposure to cigarette smoke). DHEA was administered orally through a cannula 30 minutes before exposure to cigarette smoke with an exposure duration of about 25-40 minutes, equivalent to 4 cigarettes/day. After 14 days of experimentation, the rats were euthanized. Right lungs were harvested for malondialdehyde level measurement using the thiobarbituric acid reactive substance method measured by UV-Visible spectrophotometer at a wavelength of 532 nm. The left lungs were also harvested for histopathological examination. This study showed that pulmonary malondialdehyde levels in second and third group tended to be higher than in first group. Meanwhile, histopathological examination showed that the percentage of lung tissue damage in second and third group tended to be lower than in first group. Thus, it can be concluded that DHEA at doses of 15 and 30 mg/kgBW tended to act as a pro-oxidant but might potentially improve lung histology due to cigarette smoke exposure.

Abstract

Hati adalah organ metabolisme di mana racun diserap dari saluran usus dan diangkut ke hati, menyebabkan berbagai jenis kerusakan pada hati. Kulit telur mengandung kalsium karbonat, yang mengurangi parameter oksidatif dan kerusakan jaringan hati. Kebaruan penelitian ini karena peneliti menganalisis pengaruh pemberian suspensi cangkang telur terhadap kerusakan hati pada tikus wistar. Penelitian ini bertujuan untuk mengevaluasi pengaruh suspensi kulit telur terhadap kerusakan hati yang diinduksi parasetamol pada tikus Wistar jantan. Metode penelitian menggunakan 25 tikus Wistar jantan (Rattus norvegicus) dengan berat 150-200 g dibagi menjadi 5 kelompok (n = 5). Selama satu bulan, tikus diberi 750 mg/kg BB parasetamol secara oral sekali sehari, menyebabkan kerusakan hati. Setelah 1 bulan, mereka dirawat dengan suspensi kulit telur (6,13, 10,0, dan 26,0 mg/kg BB) secara oral selama 7 hari. Setelah perawatan selesai, darah tikus ditarik melalui mata untuk pengukuran ALT dan AST. Nekropsi dilakukan untuk pemeriksaan histopatologis hati. Hasil penelitian menunjukkan tiga dosis suspensi kulit telur secara signifikan mengurangi kadar ALT dan AST pada tikus yang diinduksi parasetamol. Histopatologi hati pada tikus yang diinduksi parasetamol mengungkapkan hingga 50% nekrosis dan sel-sel inflamasi. Pada P2 dan P3, persentase kerusakan hati menurun masing-masing sebesar 25%. Kesimpulan menunjukkan bahwa suspensi kulit telur memiliki efek hepatoprotektif pada kerusakan hati yang diinduksi parasetamol, ditandai dengan penurunan kadar ALT dan AST dan peningkatan histologi hati dengan dosis 10,0 dan 26,0 mg/kg BB.Kata Kunci : Kerusakan hati; Cangkang telur; Alanin transaminase; Aspartat transaminase; Histopatologi Hati. Abstract The liver is a metabolic organ in which toxins are absorbed from the intestinal tract and transported to the liver, causing various types of damage to the liver. The eggshell contains calcium carbonate, reducing oxidative parameters and liver tissue damage. The novelty of this study is because researchers analyzed the effect of eggshell suspension administration on liver damage in wistar rats. This study aimed to evaluate the effect of eggshell suspension on paracetamol-induced liver damage in male Wistar rats. The study method used 25 male Wistar rats (Rattus norvegicus) weighing 150-200 g divided into 5 groups (n = 5). For one month, rats were given 750 mg/kg of BB paracetamol orally once a day, causing liver damage. After 1 month, they were treated with eggshell suspensions (6.13, 10.0, and 26.0 mg/kg BB) orally for 7 days. After the treatment was completed, the rat's blood was drawn through the eyes for measurements of ALT and AST. Necropsy is performed for histopathological examination of the liver. The results showed three doses of eggshell suspension significantly reduced ALT and AST levels in paracetamol-induced rats. Hepaticology of the liver in paracetamol-induced mice revealed up to 50% necrosis and inflammatory cells. In P2 and P3, the percentage of liver damage decreased by 25%, respectively. The conclusions showed that the eggshell suspension had a hepatoprotective effect on paracetamol-induced liver damage, characterized by a decrease in ALT and AST levels and an increase in liver histology at doses of 10.0 and 26.0 mg/kg BB.Keywords : Liver damage; Eggshell; Alanine transaminases; Aspartate transaminases; Hepatic Histopathology.

Abstract

Kelompok darah ABO dan jenis kelamin yang berbeda dapat memberikan hasil yang berbeda dari waktu perdarahan dan waktu pembekuan. Kebaruan penelitian ini karena meneliti tentang perbandingan bleeding time dan clotting time pada wanita terhadap golongan darah ABO. Penelitian ini bertujuan untuk membandingkan waktu perdarahan dan waktu pembekuan pada wanita berdasarkan golongan darah ABO. Penelitian ini merupakan penelitian deskriptif dengan desain cross sectional dan sampel total 115 subjek. Pengukuran waktu perdarahan menggunakan metode duke, waktu pembekuan menggunakan metode slide dan golongan darah menggunakan metode aglutinasi slide. Penelitian ini menggunakan uji komparatif numerik dengan one way ANOVA, dilanjutkan dengan analisis post hoc untuk membandingkan waktu perdarahan dan waktu pembekuan antara golongan darah ABO. Hasil penelitian ini menunjukkan waktu perdarahan pada golongan darah B yang secara signifikan lebih lama dibandingkan golongan darah lainnya (p=0,001). Sementara itu, waktu pembekuan pada golongan darah B secara signifikan lebih lama dibandingkan golongan darah lainnya (p= 0,001). Kesimpulan bahwa wanita dengan golongan darah B memiliki waktu perdarahan dan waktu pembekuan yang jauh lebih lama daripada golongan darah lainnya.Kata Kunci: Waktu pendarahan; Waktu pembekuan; Golongan darah ABO. AbstractDifferent ABO blood groups and sexes can give different results from bleeding and clotting times. This study's novelty is that it examines the comparison of bleeding time and clotting time in women to the ABO blood type. This study aimed to compare bleeding and clotting time in women based on ABO blood type. This study is a descriptive study with a cross-sectional design and a total sample of 115 subjects. Measure bleeding time using the duke method, clotting time using the slide method, and blood type using the slide agglutination method. This study used a comparative numerical test with a one-way ANOVA, followed by post hoc analysis to compare bleeding and clotting time between ABO blood types. This study showed that the bleeding time in blood type B was significantly longer than in other blood types (p=0.001). Meanwhile, the clotting time in blood type B is considerably longer than in different blood types (p= 0.001). In conclusion, women with blood type B have much longer bleeding and clotting times than other blood types.Keywords: Bleeding time; Freezing time; ABO Blood type.

Abstract

AbstrakFibrosis paru adalah gangguan paru-paru yang menunjukkan pembentukan jaringan paru yang melibatkan infiltrasi sel inflamasi, proliferasi fibroblas, spesies oksigen reaktif (ROS) dan akumulasi matriks ekstraseluler yang berlebihan di parenkim paru-paru. Kebaruan dalam penelitian ini karena meneliti tentang efek pemberian suspensi cangkang telur terhadap fibrosis paru tikus putih yang diinduksi bleomisin. Penelitian ini bertujuan untuk membuktikan efek anti-fibrotik dan anti-inflamasi dari suspensi kulit telur pada model fibrosis paru tikus. Penelitian percobaan dengan rancangan post-test only control group menggunakan wirar jantan Rattus norvegicus dengan berat 200-250 g sebanyak 25 ekor tikus dibagi menjadi 5 kelompok (n = 5). Pada penelitian ini dilakukan pemeriksaan histopatologi paru-paru yang dianalisis menggunakan metode Ashcroft yang dimodifikasi dan menghitung Neutrophil Lymphocyte Ratio (NLR). Hasil penelitian menunjukkan bahwa gambaran histopatologis paru-paru pada K1 tidak menemukan fibrosis sedangkan pada kelompok K2 fibrosis paru berkembang derajat 6 hingga 7 dengan massa fibrotik besar ≥50% bidang visual. Pengurangan luas fibrosis pada kelompok P1 dan P2 menjadi derajat 4 sampai 5. Sedangkan, pada kelompok P3 derajat fibrosis adalah 2 sampai 3. Hasil ini menunjukkan bahwa area fibrosis paru menurun dengan meningkatnya dosis suspensi kulit telur. Nilai NLR untuk semua kelompok dinyatakan dalam batas normal (4). Kesimpulan bahwa suspensi kulit telur memiliki efek anti-fibrotik dan anti-inflamasi pada model fibrosis paru-paru tikus yang disebabkan oleh bleomycin.Kata Kunci: Bleomisin; Fibrosis; Histologi; NLR. AbstractPulmonary fibrosis is a lung disorder that indicates the formation of lung tissue involving infiltration of inflammatory cells, the proliferation of fibroblasts, reactive oxygen species (ROS), and excessive accumulation of the extracellular matrix in the lung parenchyma. The novelty of this study is that it examines the effect of eggshell suspension administration on bleomycin-induced white rat pulmonary fibrosis. This study aims to prove eggshell suspension's anti-fibrotic and anti-inflammatory effects in models of rat pulmonary fibrosis. In this study, a histopathological examination of the lungs was carried out, which was analyzed using the modified Ashcroft method and calculated the Neutrophil Lymphocyte Ratio (NLR). The experimental research with a post-test-only control group design using Rattus norvegicus male wire weighing 200-250 g, as many as 25 mice divided into five groups (n = 5). The results showed that the histopathological picture of the lungs in K1 did not find fibrosis.Reduction of fibrosis area in groups P1 and P2 to degrees 4 to 5. In contrast, in the K2 group, pulmonary fibrosis developed degrees 6 to 7 with a sizeable fibrotic mass of ≥50% visual field. Meanwhile, in the P3 group, the degree of fibrosis is 2 to 3. These results show that the area of pulmonary fibrosis decreases with increasing doses of eggshell suspension. NLR values for all groups are expressed within normal limits (4). The conclusion is that eggshell suspension has anti-fibrotic and anti-inflammatory effects on a model of rat lung fibrosis caused by bleomycin.Keywords: bleomycin; Fibrosis; Histology; NLR.

Abstract

Objective: This study aims to assess the effectiveness of giving Bagea Sago date cakes in increasing the weight of children aged 6-12 months. Method: The design of the study used queasy experiments with a pretest and post-test approach with a control group. The study sample was 40 babies aged 6-12 months. The samples were divided into two groups, namely the intervention group (20 children), who were given date Sago Bagea cakes and the control group (20 children) who were given Sago Bagea cakes. The research begins with weighing weight before the intervention is carried out. Purposive sampling according to predetermined criteria. The instrument used was in the form of a questionnaire used to assess the BB of the body of children aged 6-12 months as well as observation sheets to control the administration and ascertain whether respondents consumed the date Sago Bagea cake given. Data analysis using t paired test and t independent test. Results: In the mean±SD intervention group, the body weight before treatment was 7.6±0.995 kg, and after the intervention, there was a weight gain with a mean±SD of 8.36±0. 811 kg. For the control group, the mean±SD body weight before treatment was 7.36±1,140 kg, while at the time± of post±SD weight gain was 7. 48±1. 141 kg. The results of the bivariate analysis showed that there was a difference in body weight in the group that received the intervention in giving date Sago Bagea cake with a value of p = 0.015< 0.05. Conclusion: Bagea Sago date cake can be used as an alternative to complementary breast milk (Makanan Pend besides breast milk) in children aged 6-12 months.

Abstract

This modern era has changed the pattern of how humans do their daily activities in 24 hours. The pattern change includes time-shifting, most human activities now are performed at night. The consequences of the changes in human activities pattern and the increase of artificial lighting use in the night were assumed unhealthy, but how the mechanism worked was not identified yet. This was an experimental study conducted on male Wistar rats aged 12-16 weeks which were divided into 4 categories: 1 intervention group exposed for 15 days, 1 intervention group exposed for 30 days, and 2 control groups for each time exposure, light intensity 1200lux. At the end of exposures, the blood samples were taken from rats that had been anesthetized before by using ether. Then blood samples were taken to analyze their cortisol levels using the Enzyme-linked immunosorbent assay (ELISA) method and the hematology was examined through autoanalyzer Abaxis. The data were analyzed using an independent T-test and Mann-Whitney test and considered significant if p-value<0,05. Overall, the cortisol and hematology levels indicated, especially Red Blood Cell (RBC), Mean Corpuscular Hemoglobin Concentration (MCHC), and neutrophil specifically for the group which had been exposed by LED lighting for 30 days.

Abstract

Latar Belakang: Anemia merupakan masalah kesehatan di seluruh dunia terutama di negara berkembang. Remaja merupakan kelompok yang rentan terhadap anemia karena dalam masa pertumbuhan dan perkembangan. Pemerintah melakukan upaya suplementasi tablet tambah darah selain itu pemanfaatan pangan lokal yang kaya akan zat besi. Tujuan: Penelitian ini bertujuan untuk mengetahui efektivitas biskuit beras merah dan kedelai terhadap kadar hemoglobin remaja putri anemia. Metode: Jenis penelitian quasi experiment (nonequivalent control group design). Penelitian ini dilakukan selama 30 hari dengan sampel sebanyak 36 sampel. Sampel dibagi menjadi 4 kelompok yaitu Kelompok Beras Merah (6 orang), Kedelai (6 orang), Beras Merah dan Kedelai (6 orang) dan Kontrol (18 orang). Instrumen yang digunakan Hematologi Analyzer. Analisis yang digunakan One Way Anova dan Post Hoc Test. Hasil: Hasil analisis menunjukkan bahwa rata-rata kadar hemoglobin kelompok Beras Merah, Kedelai, Beras Merah dan Kedelai, Kontrol sebelum intervensi adalah 11,40 g/dl, 10,85 g/dl, 10,68 g/dl, 11,34 g/dl dengan nilai p=0,037. Setelah dilakukan intervensi didapatkan rata-rata kadar hemoglobin  kelompok Beras Merah, Kedelai, Beras Merah dan Kedelai, Kontrol yaitu 11,90 g/dl, 12,51 g/dl, 11,95 g/dl, 11,72 g/dl dengan nilai p=0,001. Rata-rata selisih kadar hemoglobin adalah 0,5 g/dl, 1,66 g/dl, 1,27 g/dl, 0,38 g/dl dengan nilai p=0,001. Dapat disimpulkan, pemberian biskuit beras merah (Oryza Nivara) dan Kedelai (Glycine Max L) efektif meningkatkan kadar hemoglobin remaja putri.

Abstract

Tujuan, mengetahui kemampuan e-modul dalam meningkatkan keterampilan mahasiswa DIII kebidanan dengan menggunakan metode belajar mandiri dalam lingkup laboratorium. Metode, Quasi Eksperimen dengan rancangan pre-test post-test non-equivalent control group. Populasi seluruh mahasiswa DIII kebidanan tingkat III. Teknik penarikan sampel menggunakan teknik total sampling. Sampel 102 mahasiswa DIII kebidanan tingkat III, kelompok intervensi yang diberikan modul elektronik sebanyak 51 orang, kelompok kontrol yang diberikan modul cetak sebanyak 51 orang, pre-test dan post-test menggunakan daftar tilik. Post-test dilakukan sehari setelah pre-test. Analisis data menggunakan uji McNemar dan Chi-Square. Hasil, dengan menggunakan uji McNemar didapatkan, nilai p=0.000 pada pre-test dan post-test pada kelompok kontrol dan nilai p=0.000 pada pre-test dan post-test kelompok intervensi, yang artinya secara statistik pada kedua kelompok terdapat perbedaan peningkatan keterampilan yang bermakna setelah diberikan perlakuan. Kemudian dengan menggunakan uji Chi-Square pada post-test antar dua kelompok, didapatkan nilai p=0.014 < ?=0.05, yang artinya secara statistik ada perbedaan peningkatan keterampilan yang bermakna antara modul elektronik dan modul cetak dimana modul elektronik lebih mampu dalam meningkatkan keterampilan mahasiswa, sedangkan berdasarkan persentase peningkatan keterampilan, kelompok modul elektronik 37,3% lebih tinggi dari pada kelompok modul cetak. Kesimpulan, terdapat peningkatan yang signifikan pada keterampilan mahasiswa DIII kebidanan yang di berikan modul elektronik sehingga disimpulkan bahwa e-modul alat kontrasepsi bawah kulit yang diberikan mampu untuk meningkatkan keterampilan mahasiswa DIII kebidanan.

Abstract

Pemeriksaan hematologi rutin dapat digunakan sebagai penanda inflamasi untuk mengidentifikasi penyakit kardiovaskular, termasuk penyakit jantung koroner (PJK). Penelitian ini bertujuan untuk menganalisis gambaran profil hematologi rutin pasien penyakit jantung koroner yang menjalani angiografi koroner serta hubungannya dengan stenosis lesi arteri coronaria. Penelitian analitik observasional ini dirancang dengan desain cross-sectional. Sampel dipilih dengan teknik purposive sampling dan diperoleh 15 subjek penelitian. Tingkat stenosis ditentukan sesuai panduan PERKI 2017. Data disajikan dalam mean±SD, median (IQR), dan persentase n (%). Data parametrik diuji dengan uji t independen atau one-way ANOVA, data nonparametrik diuji dengan Mann-Whitney test atau Kruskal Wallis test, sedangkan data kategorik diuji dengan Fisher’s exact test. Hasil penelitian menunjukkan kadar nilai WBC, neutrofil, limfosit, basofil, RBC, hemoglobin, MCV, MCH, MCHC, PLT, RDW-CV, dan MPV berada dalam kategori normal. Dengan rerata monocyte/lymphocyte ratio 0,27 ±0,06 yang cenderung meningkat, tidak ditemukan adanya perbedaan nilai MLR antara kedua kelompok berdasarkan signifikasi stenosis arteri coronaria (p>0,05). Oleh karena itu, dapat disimpulkan bahwa tidak terdapat hubungan signifikan antara parameter hematologi dengan stenosis arteri coronary pada pasien PJK.
