Abstract

Jati Belanda Leaves (G. ulmifolia Lamk.) has beenused traditionally to reduce obesity 1). G. ulmifoliaLamk. leaves extract contained alkaloids, tannins, saponins, flavonoids, terpenoids, glycosides, andsteroids 2). Phytochemical screening resultsconducted by Iswantini et al. showed that theflavonoids and tannins were detected high levels inthis extract 3). Flavonoids contained in the leaves ofG. ulmifolia Lamk. were catechin, kaempferolglycosides, procyanidin B-2, procyanidin B-5,procyanidin C-14).Obesity can occur when the body of excess adiposetissue (fat), particularly white adipose. Excess whiteadipose tissue (WAT) is considered as the maincause of obesity 5). The growth of adipose tissueoccurs due to the formation of new adipocytes andincrease in size of adipocytes, called adipogenesis.Adipogenesis involves the multiplication of cells(proliferation) and cell maturation (differentiation). The treatment which can affect the size and numberof adipocytes and signals expression are involved inthe inhibition or stimulation of specific adipokineshave been used to prove the bioactivity associatedwith antiobesity 6).Natural products that have a specific target in thispathway have the potential to prevent and treatobesity. Some polyphenols, including flavonoidshave anti-obesity effects and also specific effects onadipocytes 7). The presence of the flavonoids i.e.catechin, kaempferol, tilirosida, and prosianidin in G.ulmifolia Lamk leaves. may affect adipogenesis. This research will be conducted extraction andfractionation leaves of G. ulmifolia Lamk. Extracts,non-polar fractions and semi-polar fractions testedtheir effect on the proliferation and differentiationof Wistar rat preadiposit.

Abstract

The sponge Aaptos suberitoides can produce high secondary metabolite with some farmocological activities as antimicrobial, antiviral, antiinflamatory, and anticancer agents. The purpose of this research is to find out correlations between activities of the etanol extracts of marine sponges Aaptos suberitoides on the cancer growth of subcutaneous mice (Mus musculus) injected by Benzo(a)piren. For the purpose the mice were divided into six groups, i.e. I, II, III, IV, V, and VI. Each group was treated with carcinogenic inductions by intravenously injecting the Benzo(a)piren concentration of 0.3g/0.2 ml oleum olivarum. After the cancer was appeared at the fifteenth day, the mice were treated by the anticancer extracts of marine sponges Aaptos suberitoides with concentrations of 500 mg/kg BB (Group IV), 1000 mg/kg BB (Group IV), and 1500 mg/kg BB (Group IV). The treatments were orally done each day for two weeks. At the twentieth week, subcutaneous cancer tissues were taken to make histological preparates using a parafin method. Result of the histological observation indicates that cancer in the mice was fibrosarcoma characterized by the thickening dermis layers, necrosis, mitosis, and nuclear polymorphsm. Necrosis, mitosis, and nuclear polymorphism occurred in Groups II, IV, V, and VI, and did not in Groups I and II. Presentation of necrosis was 20-60%, mitosis was in 3-4 cells, and nuclear polymorphism was 100%. Result of the statistical analyses by using the Kruskal-Wallis method and the Pair Comparison test indicates that the anticancer extracts of marine sponges with the concentrations of 500 mg/kg BB, 1000 mg/kg BB, and 1500 mg/kg BB had no activity inducing mice skin cancer.

Abstract

Background: Diabetes Mellitus (DM) is a complex chronic disease characterized by increased blood glucose. The incidence of this disease is rising, especially type 2 diabetes which is caused by insulin resistance in the body. SUR1-Pancreatic KATP Channel is a receptor as an antidiabetic target because its inhibition process can increase insulin production so that it can reduce blood glucose in people with type 2 diabetes. Objective: This study aims to identify the in-silico activity of the SUR1-Pancreatic KATP Channel macromolecules. Methods: Identification of macromolecular binding sites using Protein Plus software, then carried out molecular docking using AutoDock software, where the formed molecular interactions are further identified using the BIOVIA Discovery Studio software. Results: After determining the macromolecular binding site, the RMSD value was 1.253, allowing for further molecular docking. Molecular docking showed that the Ligands of mangostin (α, β, γ-mangostin) and sinensetin derivatives had a good affinity, namely α-mangostin -6,31 kcal/mol; β-mangostin -5.78 kcal/mol; γ-mangostin -6.17 kcal/mol and sinensetin -4.75 kcal/mol. Conclusion: The affinity sequence in the docking process for the SUR1 KATP channel macromolecules is α-mangostin > γ-mangostin > β-mangostin > sinensetin. The highest affinity for the docking process on the macromolecule SUR1 KATP channel was α-mangostin with a value of ΔG -6.31 kcal/mol Ki 23.65 μM.

Abstract

Enzymes are proteins that control and carry out metabolic activities in the body, either directly or indirectly. Enzymes have various advantages over traditional pharmaceuticals in terms of therapeutic efficacy and toxicity. They have great specificity and affinity for their target, minimizing toxicity. Use the terms "Serratiopeptidase enzyme" and "inflammation" to search publications in Google Scholar, Sciencedirect, SAGE journal, and Pubmed databases. According to ten articles, the serratiopeptidase enzyme has been shown to have an anti-inflammatory effect by reducing inflammatory cytokines and adhesion molecules, thereby regulating the movement of inflammatory cells to the site of inflammation, which in clinical trials can reduce swelling of edema and metabolism of scar tissue in the body.
