Abstract

Plectranthus amboinicus (Lour.) is a perennial plant belonging to the family of Lamiaceae which have been widely used in traditional herbal medicine. In the present study, the extracts of the leaves of P. amboinicus that was extracted by different solvents were evaluated for their antioxidant and antibacterial properties. The fresh leaves of the plant were extracted using subsequent methanol, n-hexane and dichloromethane. The antioxidant activity was determined by DPPH free radical scavenging assay and the antimicrobial activity was tested against Escherichia coli and Staphylococcus aureus by agar well diffusion assay. The results of antioxidant activity test showed that dichloromethane extract of the leaves had the highest antioxidant activity among all with  IC50 value of 50.2 μg/mL Methanol and n-hexane extracts were still active in DPPH with IC50 value of 71.3 μg/mL and 205.9 μg/mL, respectively. Furthermore, the dichloromethane and methanol ektracts have shown a promising antibacterial activity in E. coli. The findings indicated that dichloromethane and methanol extracts of the leaves of P. amboinicus were potential further study as herbal medicine as they showed high antioxidant and antibacterial activities.

Abstract

Flavonols a derivatives of 2'-hydroxyacetophenon and 3,4,5-dimethoxy benzaldehid has been synthesized under basic condition (KOH). The structure of the compound was characterized based on the interpretation of spectroscopic data, including UV, FTIR, NMR and HRMS. Anticancer activity was evaluated using the MTS assay against MCF-7 cells, which showed that the flavonol 2'-hydroxycalone derivative was potentially active as an anticancer substance with IC50 values <1000 µg / mL.

Abstract

Three pyridine chalcones including (Z)-1-(4-bromophenyl)-3-(pyridin-2-yl)prop-2-en-1-one (1), (Z)-1-(4-bromophenyl)-3- (pyridin- 3-yl)prop-2-en-1-one (2) and (Z)-1-(4-bromophenyl)-3-(pyridin-4-yl)prop-2-en-1-one (3) were synthesized by aldol condensation reactions from pyridinecarbaldehyde with 4-bromoacetophenone. In antibacterial assay, compound 3 exhibited strong activity against Staphylococcus aureus, Bacillus subtilis and Escherichia coli with the inhibition zone of 19.9; 19.5, and 17.5 mm, respectively.

Abstract

The use of plants as traditional medicine has long been known. One of them isBingkek (Entada phaseoloides merr). Seedl of this plant has been being used in folk medicine for stomach ache, hernias and restore the condition of women after childbirth, so as well as food. The study aimed to determine the antioxidant bioactivity of various fractions of seeds Bingkek and to identifity potential antioxidant compounds from this plant. The process started by macerating 500 g of seeds kernel by methanol. After 72 hours, the extract filtered and concentrated by rotary evaporator. Then, 80 g methanol exstract is fractioned by hexane, dicloromethane and athyl acetate. The result gained are 0.8 g hexane, dicloromethane 0.4 g, ethyl acetate 0.7 g and residual extract 23 g. the test result of each fraction antioxsidant bioactivity wit DPPH, showed 909,35 μg/mL hexane extract, 507,74 μg/mL dicloromethane, 712,61 μg/mL ethyl acetate and 77,60 μg/mL residual extract. The best value closest to vitamin C is the residual extract. Then, the phyrochemical profile showed that in residual extract contains phenolic compounds, triterpenoids, coumarin and saponins. It can be concluded that the residual extract has potential as good antioxidant. Further more it is necessary to carry out the study of of the potential compounds of the antioxidant.

Abstract

Tumbuhan paku kaki tupai (Davalia denticulata) merupakan salah satu tumbuhan paku epifit yang tumbuh pada kelapa sawit dan digunakan untuk mengurangi kandungan asam urat.. Beberapa spesies dari genus ini dilaporkan telah digunakan sebagai obat tradisional untuk mengobati penyakit infeksi dan juga kanker di Taiwan.  Tetapi, pada spesies ini kandungan metabolit sekunder dan tingkat toksisitasnya belum di laporkan. Penelitian ini bertujuan untuk menguji tingkat toksisitas dari beberapa ekstrak dari tumbuhan paku kaki tupai. Tumbuhan dikering-anginkan kemudian diekstraksi dengan metode maserasi dengan pelarut metanol, etil asetat dan n-heksana. Masing-masing ekstrak dilakukan pengujian toksisitas dengan menggunakan metode Brinne Shrimp Lethality Test (BSLT). Pada penelitian ini didapatkan empat ekstrak dari pelarut yang berbeda-beda. Masing-masing ekstrak dilakukan pengujian BSLT. Hasil dari pengujian ini di dapat bahwa ekstrak etil asetat memiliki toksisitas yang tinggi dengan LC50 sebesar 119,8 ppm diikuti oleh ekstrak methanol dengan LC50 sebesar 215,5 ppm. Sedangkan ekstrak heksan memiliki nilai LC50 > 1000 ppm. Berdasarkan laporan yang ada, jika hasil BSLT menunjukkan bahwa ekstrak tumbuhan bersifat toksik maka dapat dikembangkan ke penelitian lebih lanjut untuk mengisolasi senyawa sitotoksik tumbuhan sebagai usaha pengembangan obat alternatif anti kanker. Pengujian toksisitas dari tanaman ini didapatkan bahwa ekstrak etil asetat dan methanol memiliki tingkat toksistas yang tinggi. Oleh karena itu diperlukan pengujian isolasi senyawa metabolit sekunder dari ekstrak tersebut dan dilakukan pengujian toksisitasnya sehingga diharapkan memiliki senyawa yang bersifat antikanker.

Abstract

The objective of this study was to analyze the total phenolics, total flavonoids, andantioxidant activities of Amaranthus spinosus L leaves. During cultivation, these plantswere treated with 2 treatments, biocontrol Effective Microorganism 5 (EM5) garlic andcontrol (water). The A. spinosus L leaves were dried using freeze dryer and extracted bydissolving n-hexane, dicloromethane (DCM) and methanol. The results showed thatA.spinosus L leaves treated with biocontrol had the highest total phenolic, totalflavonoid and antioxidant activity content compare to the control. The total phenolic,total flavonoid, 2,2 diphenyl-1-picrylhydrazyl (DPPH) and Nitric Oxide (NO) radicalscavenging were the highest on DCM extract with each value of 74,656 mg Gallic AcidEquivalent (GAE)/g of dry weight (DW), 22,337 mg Catechin Equivalent (CE)/g DW,0,373 mg/mL and 0,025 mg/mL. n-Hexane extract had the highest antioxidant activityon Ferric Thiocyanate ( FTC) test (27,615 %), meanwhile methanol extract indicated thehighest antioxidant activity on FRAP test (1,022 mmol Fe +2 /g DW). From this study,it can be concluded that A. spinosus L leaves with biocontrol EffectiveMicroorganism 5 (EM5) garlic can increase the phenolic, flavonoids and antioxidantactivitiy contents of A. spinosus L plant.

Abstract

"> Enzymatic transglycosylation of pinocembrin was successfully carried out by cellulase from Trichoderma asperellum LBKURCC1. However, the reproducibility of the reaction is low, making detection by reverse phase HPLC (High Performance Liquid Chromatography) difficult when the enzyme activity decreases. It is believed that the initial flavonoid concentration used in the transglycosylation reaction influences the detection pattern of transglycosylation product by HPLC, when the enzyme activity is low. In this research, transglycosylation of pinocembrin was done by a concentrated extract of T. asperellum LBKURCC1 cellulase and using CMC (Carboxymethyl Cellulose) as glycosyl donor. The reaction was carried out for 30 hours at 40oC and 170 rpm shaking. Results of HPLC analysis showed that cellulase with activity of (4.3±0.2) U/mL could only obtain 1.7% conversion of pinocembrin to its glycosylated product when the substrate concentration was 6 mg/mL. At these conditions, a barely detectable HPLC peak was observed at retention time of 8.4 minutes. This peak increased noticeably when the initial concentration of pinocembrin was decreased to 0.6 mg/mL. Percent conversion of pinocembrin to its transglycosylation product was 4.3% when the initial substrate concentration was 0.6 mg/mL and the enzyme activity was (4.3±0.2) U/mL.

Abstract

Pyrazolines have been reported to possess  various  biological activities such as antimicrobial,  antidepressant,  antidiabetic,  anti-inflammatory,  antipyretic,  anticancer, and  anticonvulsant  activity. In this research, a halogenated pirazoline 5-(4-bromophenyl)-3-(naphthalene-1-yl)-4,5-dihydro-1H-pyrazole was synthesized  by cyclization  reaction between chalchone with hydrazine  hydrate  using acetate acid as catalyst under microwave irradiation. The compound showed a good yield, and then was characterized by  UV,  IR, 1H-NMR, and 13C-NMR spectroscopy. The antibacterial activity of pyrazolines was determined using the agar diffusion method  against  Gram-negative bacteria:  Escherichia  coli  and  Salmonella  typhi  and  Gram-positive bacteria: Bacillus  subtilis  and  Staphylococcus  aureus. This compound  showed  inhibitory  activity against those four bacteria. Test results of antibacterial activity indicated that pyrazoline analogues were potential as antibiotics.

Abstract

Pyrazolines have been reported to possess many of interesting biological activities such as antimicrobial,  antidepressant,  antitumor, antioxidant, antiinflammatory, anticancer and  anticonvulsant  activity .  In this research, a  compound  of 1-phenyl-3-(2-naphthyl)-5-(3-chlorophenyl)-2-pyrazoline  was synthesized by cyclization of  (E)-3-(3-chlorophenyl)-1-naphthalene-2-yl-prop-2-en-1-on and phenylhydrazine  using glacial acetic acid  as  a  catalyst under  microwave  irradiation. The compound showed a good yield and  it  was characterized by UV, IR, 1 H-NMR  and 13 C-NMR  spectroscopy. The antibacterial activity of this compound was determined using agar diffusion method against Gram positive bacteria  (Bacillus subtilis  and  Staphylococcus aureus) and Gram negative bacteria  (Escherichia coli  and  Salmonella typhi). The compound showed very poor activity against Salmonella typhi.

Abstract

Cerbera odollam  Gaertn is a plant  found  in Indonesia which is used as traditional medicine. Isolation and toxicity assay on fruit flesh of this plant has  been  done. The methanol macerating method was applied to isolate the secondary metabolites from  part of this plant. The separation was carried out by vacuum liquid chromatography (VLC), gel chromatography and KLT preparative. Characterization of the fractions  was established using UV-Vis and FTIR spectroscopy. Toxicity assay was conducted by Brine Shrimp Lethality Test  (BSLT) method. The LC50  of  7th to 11th fractions of the methanol extract  were 107,86 ; 5,19; 136,42 ; 41,33 and 720,60 ppm, respectively. The 5th and 6th fractions were not toxic, the 11th fraction was toxic whereas other fractions were very toxic and potential to be used as an anticancer.
