Abstract

The leaves of Syzygium polyanthum is widely used in Indonesia as a spice. It has been reported to have significant antioxidant activity. However, the reports on compounds responsible for the activity are very scarce. The purpose of this research was to characterize the chemical profile of S. polyanthumfractions which showed the highest antioxidant activity. The dried leaves of S. polyanthum were fractionated using the different combination of non-polar to polar solvent, those are n-hexane, acetone, water. Fourteen fractions were collected and analyzed for their DPPH antioxidant activity. The chemical profile of selected fractions was characterized using UHPLC-HRMS to identify compounds that might associate with the antioxidant activity of S. polyanthum. The results showed that at the dose of 500 μg/ml, four fractions showed the highest antioxidant activity with the following order: F9 (acetone:water=80:20), F10 (acetone:water=60:40), F1 (hexane 100%) andF3 (hexane:aceton=80:20). UHPLC-HRMSanalysis successfully identified several compounds in F9 and F10. Gallic acid, pyrogallol, and myricetin were identified in F9.  Aconitic acid was identified in F10. All of the identified compounds were previously reported to have antioxidant activity

Abstract

One of the effective methods for mineral destruction is by using microwave digestion. However, some parameters in the microwave manual book are different with AOAC method such as the volume of acid, temperature, and the length of destruction time (Δt). Thus, a validation for mineral analysis method of Fe, Zn, and Cu by using microwave digestion method is required. In this study, carbohydrate-rich samples such as rice and canned pineapple were being used for the validation of mineral analysis method of Fe, Zn, and Cu using microwave digestion. The parameters of this research include LOD, LOQ, LDM, linearity, precision, accuracy, and reproducibility. The accuracy result for Fe, Zn, and Cu mineral was found in the range of 75-120% with good precision value (% RSD <% 2/3RSD Horwitz). Based on the results, accuracy for mineral Fe, Zn and Cu found in the range of 75-120% with good precision value (% RSD <% 2/3RSD Horwitz). The regression curves of the Fe, Zn, and Cu minerals gave the value of R2> 0.990. For LDM values in the rice samples consecutively for Fe, Zn, and Cu minerals were 0,06; 0,07; 0,04 mg/L, while for the canned pineapple were 0,07;0,10;0,01 mg/L, respectively. The results of a modified method for minerals Fe, Zn and Cu analysis in carbohydrate-rich samples was not significantly different from AOAC (2012). Thus the method was acceptable to be used in laboratory analysis.

Abstract

Perubahan gaya hidup telah menempatkan penyakit degeneratif sebagai penyebab kematian utama, baik di dunia maupun di Indonesia. Salah satu upaya pencegahan penyakit ini dapat dilakukan melalui pengaturan pola makan. Pembuatan beras analog selain bertujuan untuk mendukung program diversifikasi pangan, tetapi juga dapat dirancang dari bahan-bahan tertentu (seperti sorgum, bekatul, dan kedelai) agar memberikan manfaat fungsional khusus bagi kesehatan. Beras analog telah diteliti memiliki aktivitas hipokolesterolemik, nilai indeks glikemik yang rendah, serta menghambat proliferasi sel kanker kolon secara in vitro. Aktivitas tersebut selain berasal dari komponen fitokimia yang terdapat pada bahan-bahan tetapi juga dapat muncul akibat proses pengolahan, seperti terbentuknya pati resisten maupun terlepasnya komponen aktif dari matriks pangan. Hanya saja penambahan komponen fungsional terkadang memberikan dampak yang tidak diinginkan pada atribut sensori. Oleh karena itu, pengembangan beras analog fungsional dari segi sensori harus terus dilakukan supaya dapat diterima dengan baik oleh masyarakat.The changes of lifestyle nowadays bring degenerative diseases as the most leading cause of death, both in worldwide and Indonesia. It has been very well known that incident of the diseases could be prevented through food diet. Not only aimed to support the government’s diversification program, rice analogue development could be designed from certain materials (such as sorghum, rice bran, and soybean) to provide specific functional health benefit. Some reports showed that rice analogue has low glycemic index, as well as its activity in lowering cholesterol level and inhibiting the proliferation of in vitro colon cancer cell. The activity was expected to come from the phytochemical compounds naturally present in the ingredients and might also arise during the processing, such as the formation of resistant starches as well as the release of the active compounds from food matrix. However, the addition of functional ingredients sometime impacted on undesirable sensory attributes. Therefore, all sensory attributes of functional rice analogue should be improved to achieve the increment of public acceptance.

Abstract

Type 2 diabetes mellitus pathogenesis is characterized by high blood glucose which can be triggered by several factors such as oxidative stress, unhealthy diet, and poor lifestyle. The high content of bioactive components and low content of digestible carbohydrate in black soybeans tofu might contribute to the body antioxidant supply and control blood glucose levels. This study aimed to evaluate the effects of black soy tofu consumption toward plasma SGOT/SGPT and antioxidant capacity. The intervention of black soybean tofu was conducted for 28 days in 15 type-2 diabetic respondents. The results showed that black soybean tofu significantly (p ˂ 0.05) increased plasma antioxidant activity level of respondents, from 45.79 ± 3.31 percent to 53.05 ± 4.44 percent. SGOT / SGPT tests showed a decrement ((p ≥ 0.05), from 14.27 ± 3.81 / 21.07 ± 6.73 (U/L) to 12.73 ± 2.34 / 18.60 ± 4.29 (U/L). The processing of black soybeans into tofu caused a decrease in antioxidant activity, from 144.06 mg AEAC to 45.27 mg AEAC and the total anthocyanin, from 12.27 mg to 1.805 mg/100 g. The results suggested that black soybean tofu improved the health profile of the diabetic respondents by increasing plasma antioxidant capacity and decreasing plasma SGOT/SGPT level.

Abstract

The leaves of yacon (Smallanthussonchi folius [Poepp. &Endl.] H. Robinson) or in Indonesia known as “Daun insulin” (Insulin leaf), scientifically has various medicinal properties and one of them as antidiabetic. The aim of this research is to know α-glucosidase enzyme inhibitor compound in the ethanol 96% extract of yacon leaves. Fractionation of ethanol extract subjected to column chromatography (SiO2; dichloromethane-methanol-water = 5 : 5 : 1) gave 5 fractions (Fr -1 ~Fr – 5). The results of the -glucosidase enzyme inhibition activity test in vitro against the five fractions obtained the 3rd fraction as the most active of 87.84 %. Purification of the 3rd fraction with preparative thin layer chromatography (SiO2; n-BuOH-ethanol-water = 4 : 1 : 2.2) gave pure isolate as Fr 3-1. Identification of pure isolates Fr 3-1 was carried out by interpreted spectra of Ultraviolet, Fourier Transform-Infra Red, Mass Spectra (from LC-MS) and compared data on the chemical shift of the Nuclear Magnetic Resonance (proton and carbon) and determined as Nystose (GF3) compound. The results of the -glucosidase enzyme inhibition activity showed that Fr 3-1 has IC50 of 21.36 ppm

Abstract

Kecombrang (Etlingera elatior) is a well-known herb in Indonesia that is widely used in traditional medicine and as a flavour in local dishes. The aim of this study was to identify retention time of antibacterial components from kecombrang flower extract (Etlingera elatior) by HPLC-based metabolomics method. The antibacterial activity of ethanol extracts, chloroform, and water fractions was assessed by well-diffusion agar method against Staphylococcus aureus. Only chloroform fraction showed medium inhibition activity against Staphylococcus aureus. The HPLC chemical profiles of the fractions were then correlated to their antibacterial activity profile by means of Orthogonal Projection to Latent Sructure (OPLS) analysis. Peak area with retention time of 0.96 - 1.12 min present in chloroform fraction at UV 250 nm was found to significantly correlate to the antibacterial activity (Y related coefficient value 0.73).

Abstract

Destruction process using both dry and wet conditions serves as a significant step in mineral quantification. Wet destruction using microwave-assisted digestion refers to the standard method of AOAC (2012); in this work, we modified the destruction procedures provided in microwave digestion manual book, including temperature, reagent volume (HNO3 and H2O2), and length of destruction. The experiment works aimed (1) to validate the procedures in destruction process using microwave digestion; (2) to quantify Fe, Zn and Cu in various food matrices (canned peas, canned fish, full cream powdered milk) using a validated method. The method was evaluated according to linearity, accuracy, precision, absolute and relative LOD, LOQ, and intra-reproducibility. Measurement of Fe, Zn and Cu were considered to have a satisfying accuracy at a range of 80–115%, with a good precision value (% RSD < 2/3 CV Horwitz), while regression curves R2 > 0.995. The results showed that data collected from the modified method was not significantly different compared to those from the AOAC method. The currently developed method also fulfilled the acceptability requirements for laboratory analysis.

Abstract

Smallanthus sonchifolius [Poepp. & Endl.] H. Robinson (Asteraceae) also known as Yacon or insulin plant, is traditionally used for treating diabetes. Varying geographical origins and postharvest handling, however, seem to affect quantitative and qualitative metabolites in the leaves of Smallanthus sonchifolius [Poepp. & Endl.] H. Robinson (Yacon). The study was conducted to compare and differentiate metabolites profile/fingerprint of Yacon leaves which were grown and obtained from different locations in Pulau Jawa i.e. Lembang (Jawa Barat) and Wonosobo (Jawa Tengah). Three different solvents (95% ethanol, 50% ethanol and water) were used to synthesize Yacon leaves extracts, in order to determine the suitable solvent to produce discernable differentiation through FTIR and HPLC-based metabolomics. Principal Component Analysis (PCA) of FTIR data (4000–400 cm1 wavenumber) indicated that Yacon leaves extracted with ethanol at 95%, had a distinctive FTIR fingerprint profile when compared to others. However, the FTIR-based PCA could not differentiate the extracts based on their geographical origins, although PCA analysis of HPLC-data successfully differentiated the extracts based on their geographical origins. Furthermore, the prominent peak for the leaves extract from Lembang and Wonosobo as regards retention time, was observed at 21.59–25.10 min and 20.69–21.695 min respectively. Notably, R2Y and Q2 value obtained by cross-validation and permutation tests showed all multivariate models were statistically reliable. Overall, there is the need to conduct further research using a more sophisticated tool such as LC-MS, to identify which metabolites represented by the aforementioned FTIR and HPLC data.

Abstract

In Tanah Datar Regency, West Sumatra, Indonesia, the waste of pruning coffee leaves (Coffea canephora) is utilized as a traditional beverage called "Kawa Daun". For a consistent quality of Kawa Daun functional beverage, we evaluated the effect of different smoking times (0, 2, 4, and 6 h) on its in vitro DPPH antioxidant activity. Estimation of antioxidant components from the coffee leaf was conducted in silico using Peroxiredoxin V (PrxV) with 3MNG code as a receptor, and 33 phytochemicals were reported to be present in the coffee leaves as ligands. As a result, Kawa Daun, with a 2-h smoking time, had the highest antioxidant activity. Molecular docking between PrxV and the 33 compounds resulted in the ten most potential compounds based on the affinity energy. They were xanthone (-4.9 kcal/mol), uric acid (-4.8 kcal/mol), xanthosine (-4.8 kcal/mol), caffeine (-4.6 kcal/mol), 3-methylxanthine (-4.6 kcal/mol), 7-methylxanthosine (-4.6 kcal/mol), theobromine (-4.5 kcal/mol), theophylline (-4.5 kcal/mol), caffeic acid (-4.5 kcal/mol), and xanthine (-4.4 kcal/mol). These ten ligands had stronger interactions than the control ligand 1.2-dithiane-4.5-diol (-3.6 kcal/mol). This research showed the potential of Kawa Daun as a functional beverage with antioxidant activity. Further confirmation on the antioxidant potential of this beverage using an in vivo method is recommended.

Abstract

Beef price is relatively expensive, which makes this commodity vulnerable to be counterfeited. The development of rapid, cheap and robust analytical methods for meats authentication has therefore become increasingly important. In this study, colorimetric and digital image analysis methods were used to characterize and classify four types of meat (beef, buffalo, pork, wild boar) and two muscle types from each sample (Semitendinosus and Vastus lateralis). Multivariate data analysis (PCA and OPLS-DA) was used to observe classification pattern among species using different color parameters data obtained from meat chromameter and digital image measurement. The results showed that PCA and OPLS-DA successfully classified meat from different species and different muscle type based on color, both in chromameter and in image analysis. It was shown that pork had the highest lightness level, and was the most different among the four types of meat tested. Beef was predominated by yellowish color, while buffalo meat had the highest reddish color level. Semitendinosus and Vastus lateralis muscles had different color intensity where Vastus lateralis exhibited darker color intensity. This study showed that meat color analysis using chromameter and imaging techniques can be used as cheap and quick tools to discriminate meats form different species and different muscles type.
