Abstract

Heliconia merupakan tanaman pisang hias yang banyak dibudidayakan di daerah tropis. Keberadaan Heliconia di Indonesia memiliki kemelimpahan tinggi yang menjadikan tanaman tersebut berpotensi untuk bersaing di pasar bunga Internasional. Heliconia merupakan kelompok tumbuhan belum banyak dipelajari karakter molekulernya, sehingga perlu diperhatikan karena memiliki keterkaitan dengan pentingnya upaya konservasi dan pemuliaan plasma nutfahnya. DNA barcoding dapat digunakan sebagai yang efektif dalam mendeterminansi spesies. Metode yang dilakukan merupakan studi in silico menggunakan database Genbank (NCBI). Sekuen DNA yang dikoleksi dari database pada NCBI yaitu terdiri dari region inti sel dan plastida, yakni rbcL, matK, ITS, psbA dan trnL. Analisis data penyejajaran sekuen, identifikasi sekuen yang berpotensi sebagai barcode dan konstruksi pohon filogenetik dilakukan dengan menggunakan MEGA X. Analisis bioinformatik pohon filogenetik Heliconia menunjukkan bahwa hasil algoritma UPGMA Tree menggunakan gen matK dan rbcL sekuen parsial sangat konservatif dan gagal dalam diskriminasi spesies, sedangkan region ITS memiliki tingkat variasi genetik tinggi dan kemampuan diskriminasi yang baik. Region psbA dan trnL dianggap sebagai gen pelengkap untuk identifikasi tumbuhan khususnya Heliconia, karena kurangnya ketersediaan aksesi online untuk spesies-spesies tumbuhan.

Abstract

The Bogor Botanical Garden (KRB) is a significant conservation site in Indonesia, serving as a sanctuary for a diverse range of plant species, including the genus Averrhoa, which holds both economic and ecological value. Plant morphology studies are essential for species classification and biodiversity conservation. This study aims to identify the morphological characteristics of leaves from three Averrhoa species (Averrhoa bilimbi, Averrhoa carambola, and Averrhoa dolichocarpa) as a tool for field identification. Conducted at the Morphology, Anatomy, and Cytology Laboratory of the Bogor Botanical Garden, the research utilized visual observation and digital microscopy to analyze key leaf traits. Principal Component Analysis (PCA) and cluster analysis revealed significant differences in adaxial leaf color and lamina shape, although certain characteristics, such as abaxial color and leaf margin type, were similar across species. These findings demonstrate that leaf architecture is an effective means of identifying Averrhoa species and contribute to taxonomy and biodiversity conservation efforts. Further research is recommended to expand sampling and explore genetic correlations.

Abstract

Alocasia suhirmaniana Yuzammi & A. Hay belongs to the Araceae family and has the potential for ornamental use. Nevertheless, this plant exhibits a limited leaf count and pliable leaf stems, necessitating the application of breeding techniques for its improvement. One important strategy empeloyed to improve the quality of ornamental plants is the breeding, which involves the use of gamma ray irradiation techniques. The objective of this study is to determine the developmental patterns and appropriate dosage levels of LD20 and LD50 Gamma ray irradiation in A. suhirmaniana callus. The development of callus, the quantity of shoots, and the establishment of rooted explants exhibit a stochastic pattern, with a decrease observed at a dosage of 5 Gy and an increase observed at 10 Gy. The LD20 and LD50 values for Gamma ray irradiation in A. suhirmaniana callus fall within the dose range of 7.14 Gy and 15 Gy, respectively. This dosage range is recommendedto achieve a greater diversity in genotypes and phenotypes. By successfully finding the optimal dosage of gamma ray irradiation, plant selection can be improved through enhancing genotype and phenotype characteristics.

Abstract

   Bogor Botanic Gardens is one of the outstanding green areas in Bogor City. Thousands of plant species are growing and providing suitable habitats and feed for various pollinators, such as butterflies, bees, and wasps. A well-designed pollination garden will benefit the pollinators and also create an educational facility for the community. This study aimed to select and characterize species of flowering plants that effectively attract pollinators, particularly Lepidoptera and Hymenoptera, in the pollination garden candidate area. The observa-tion had been carried out on 12 ornamental plant species: Pennisetum purpureum, Cosmos sp., Pachystachys lutea, Cuphea hyssopifolia, Orthosiphon aristatus, Lantana camara, Citrus aurantifolia, Buddleja davidii, Ixora javanica, Nerium oleander, Hibiscus rosa-sinensis, and Aglaia odorata. The method for observing pollinators was a standard walking method. Pollinator photos, its number, its visit time, and the plants it visited were recorded. A total of 23 species of Lepidoptera (20 species from 4 families and 3 species were unidentified) and 9 species of Hymenoptera (4 families) were found in the area with the best obser-vation time at 08.30-09.30 (GMT+7). The most visited plants by pollinators were Cuphea hyssopifolia, Lantana camara and Orthosiphon aristatus. Lepidoptera and Hymenoptera preferred blue-purple and bright-orange-yellow flowers. In addition, the flower characters that might influence pollinator visits are also described. The result would be an essential recommendation for plant selection, landscape design, and construction of the pollination garden in Bogor Botanic Gardens.

Abstract

Studies of leaf architecture and morphometry in Syzygium spp. are investigated. Seven species of Syzygium are taken for the study with additional Psidium guineense Sw. used as comparison. The seven species include Syzygium urceolatum subsp. palembanicum (Miq.) P.S.Ashton, Syzygium tinombalum Sunarti, Syzygium sexangulatum (Miq.) Amshoff, Syzygium hirtum (Korth.) Merr. & L.M. Perry, Syzygium boerlagei (Merr.) Govaerts, Syzygium racemosum (Blume) DC., and Syzygium claviflorum (Roxb.) Wall ex Steud. A comprehensive description of leaf architecture for the studied taxa was provided including leaf type, laminar shape, leaf attachment, laminar length, laminar width, petiole, leaf margin, apex shape, base shape, leaf color, and laminar symmetry. The pattern showed primary vein framework, major secondary spacing, major secondary angle, major secondary vein framework, and tertiary veins. Taxonomically significant leaf features of examined taxa showed great diversity in leaf attachment opposite, primary vein framework pinnate, and major secondary angle smoothly decreasing towards base.ABSTRAK: Kajian arsitektur daun dan morfometri Syzygium spp. diamati dan dilaporkan. Tujuh spesies Syzygium digunakan dan satu spesies Psidium guineense Sw. sebagai pembanding. Jenis Syzygium yang diamati terdiri dari Syzygium urceolatum subsp. palembanicum (Miq.) P.S.Ashton, Syzygium tinombalum Sunarti, Syzygium sexangulatum (Miq.) Amshoff, Syzygium hirtum (Korth.) Merr. & L.M. Perry, Syzygium boerlagei (Merr. Govaerts), Syzygium racemosum (Bl) DC., dan Syzygium claviflorum (Roxb.) Wall ex Steud. Deskripsi arsitektur daun yang diamati mencakup ciri tipe daun, bentuk helaian, letak daun, panjang helaian daun, lebar helaian daun, tangkai daun, tepi daun, ujung daun, pangkal daun, warna permukaan daun, dan simetri helaian daun. Pola venasi menunjukkan primary vein framework, major secondary spacing, major secondary angle, major secondary vein framework, dan tertiary veins. Ciri dengan nilai signifikan ditemukan pada letak daun berhadapan, primary vein framework menyirip, dan sudut sekunder utama yang secara halus berkurang menuju dasar.

Abstract

Bogor Botanic Gardens is an ex-situ plant conservation area in Indonesia. Since BBG is 103 years old, many collections are 100 years old or older. These antique collections may sustain damage, such as broken or collapsing, endangering visitors and employees. As a result, monitoring tree health at BBG is a critical task. According to the tree health monitoring data, 73 of 244 trees were further checked using the PiCUS Sonic Tomograph. Trees from the Fabaceae (31%) and Myrtaceae (10%) families were the most frequently checked. Walnuts trees from the Burseraceae family had the most specimens (47,94%). The PST effectively provides an immediate picture of the stem condition by calculating solid and decaying wood percentage values.

Abstract

Cassine is a genus of plants belonging to the family Celastraceae. Species of Cassine are among those preserved through ex-situ conservation efforts at the Bogor Botanical Gardens. This study aims to provide a comprehensive overview of the anatomical characteristics of leaf stomata in four distinct Cassine species. Stomatal and epidermal observations were conducted using the replica method. The findings reveal that all Cassine species exhibit anisocytic stomata, characterized by a central guard cell surrounded by three neighboring cells of unequal size. Furthermore, stomata were confined to the abaxial (lower) surface of the leaf, with none present on the adaxial (upper) side. This distribution indicates that gas exchange in Cassine leaves primarily occurs on the lower surface, reflecting a significant physiological adaptation. These observations offer valuable insights into the structural and functional traits of Cassine species conserved at the Bogor Botanical Gardens. Karakteristik Anatomi Daun Genus Cassine (Celastraceae) ABSTRAK: Cassine adalah genus tanaman yang termasuk dalam keluarga Celastraceae. Tanaman Cassine merupakan salah satu spesies yang dilestarikan dalam konservasi ex-situ di Kebun Raya Bogor. Penelitian ini berfokus untuk memberikan gambaran yang komprehensif mengenai karakteristik anatomi stomata daun pada empat spesies Cassine yang berbeda. Pengamatan stomata dan epidermis diperoleh dengan menggunakan metode replika. Temuan penelitian mengungkapkan bahwa semua spesies Cassine menunjukkan stomata anisositik, yang ditunjukkan oleh sel penjaga pusat yang dikelilingi oleh tiga sel yang berdekatan dengan berbagai ukuran. Selain itu, stomata terbatas pada permukaan abaksial (bawah) daun, tanpa ada yang ada di sisi adaksial (atas). Hal ini menunjukkan bahwa pertukaran gas pada daun Cassine terutama terjadi di bagian bawah, yang mencerminkan adaptasi fisiologis yang penting. Pengamatan ini memberikan wawasan yang berharga tentang sifat struktural dan fungsional dari spesies Cassine yang disimpan di Kebun Raya Bogor.

Abstract

Aglaonema pictum (Roxb.) Kunth merupakan tanaman hias populer di dunia karena memiliki variasi corak warna daun. Studi tentang keragaman genetik A. pictum masih jarang dilakukan, sehingga upaya ini perlu dilakukan. Salah satu penanda molekuler yang dapat digunakan untuk mengetahui keragaman genetik yaitu Simple Sequence Repeats (SSR). Tujuan penelitian ini adalah mengidentifikasi kondisi optimum pada primer SSR untuk studi keragaman genetik A. pictum. Metode yang digunakan adalah pengumpulan sampel, ekstraksi DNA menggunakan cetyltrimethylammonium bromide (CTAB), amplifikasi DNA menggunakan primer SSR (OSR13, RM10, RM6470, OSR14, MRG2172_RT2172, MRG2153_RGP2153, RM249, RM100, MRG2881_RT2881, dan RM7240) dengan menggunakan Polymerase Chain Reaction (PCR) pada kisaran suhu 52-54℃, 55-59℃, 47.2-50.9℃, dan 47.6-50.4℃, elektroforesis dan dokumentasi gel agarosa, serta analisis data. Hasil penelitian menunjukkan bahwa dua dari sembilan primer SSR (RM249 dan RM7240) dapat diamplifikasi dengan baik pada suhu 49,3 ˚C dan 52,6 ˚C. Kedua suhu tersebut mengindikasikan suhu optimum yang dapat mengamplifikasi DNA sampel A. pictum. Sementara itu, primer OSR13, RM10, RM6470, OSR14, MRG2172_RT2172, MRG2153_RGP2153, RM100, MRG2881_RT2881 tidak berhasil teramplifikasi dengan baik dan spesifik pada rentang suhu annealing yang digunakan dalam penelitian ini.

Abstract

The presence of undocumented alien flora threatens Indonesia’s flora richness and diversity due to unpredictable invasion risks. Hydrocotyle leucocephala is an alien flora that has been reported as invasive in Sri Lanka and Nigeria and has distributed to Indonesia. Therefore, mapping the distribution and describe the morphological of Hydrocotyle leucocephala is necessary to detect the initial introduction or further spread of this species. The data collection was carried out by exploring the Bogor Botanical Gardens. Environmental factors are measured and documented where Hydrocotyle leucocephala occurs. For observation, Hydrocotyle leucocephala was taken and their morphology described. As a results, Hydrocotyle leucocephala was spread over vak V.L, V.G, V.M, IX.D, VIII.B and parking lot near the conservation building in the Bogor Botanical Gardens. It is also known that Hydrocotyle leucocephala is a creeping herbaceous plant with glabrous stems, kidney-shaped leaves, white flowers without petals and green fruit with persistent styles.
