Abstract

Porang is one of the plant commodities with high economic value due to its high glucomannan content in the tubers. Currently, the global demand for porang tubers continues to rise, but domestic production has not yet met this demand due to the limited supply of superior porang seeds. Mutation breeding with Ethyl Methane Sulfonate (EMS) can be one of the alternative methods for developing superior porang to enhance tuber production. This study aims to determine the response of porang bulbil explants to EMS mutagen treatment in vitro. The research used a Randomized Complete Block Design consisting of six EMS concentrations (0, 0.02, 0.04, 0.06, 0.08, and 0.1%). The research stages included pre-culture of bulbil explants on MS + BAP 3 mg.L-1 + NAA 0.1 mg.L-1 for two weeks in dark conditions and EMS mutagen treatment on bulbil cultures for four weeks. The results showed that adding EMS mutagen to the medium for four weeks caused the explants browning, inhibited shoots forming, and decreased the fresh weight and growth index. The higher the EMS concentration in the medium, the increased percentage of browning explants and reduced explant growth and shoot formation. The EMS concentration (≥0.06%) inhibited explant growth and shoot formation. While EMS concentration (0.02-0.04%) increased explant growth, shoot formation was not significantly different from control, even though there was an increase. Up to four weeks of culture, there was no explant death, so LD50 is not yet known. The LD50 EMS can also be calculated by explant browning and shoot proliferation. Based on explant browning and shoot proliferation, the LD50 EMS in porang bulbil explants was 0.07% and 0.09%. The EMS concentration and length of culture age need to be increased to obtain an LD50 survival rate. Keywords: Amorphophallus muelleri Blume, bulbil, EMS, In Vitro mutagenesis.

Abstract

Bacterial stalk rot (BSR) disease is caused by Dickeya zeae, where infection on maize could lead to an enormous yield loss. Although curative action to control BSR infection can be done using bactericides, preventing the establishment of infection is still the best approach in minimizing potential yield loss. Among the different methods in preventing BSR infection, the use of resistant maize hybrid is considered the best approach. In this study, Genome Wide Associated Study (GWAS) was employed to find SNP markers associated with BSR resistance in maize. Six hundred twenty four lines were divided into two observation groups and further phenotyped for BSR resistance at 5, 10, and 15 days after inoculation (DAI) with BSR. GWAS was performed in a time-series manner using MLM (Mixed Linear Model) controlling for population structure and kinship. We found one SNP marker in chromosome 2 displaying significant association to BSR resistance spanning the entire observation periods. We also found SNP markers with significant association to BSR spanning two consecutive observation days located in chromosome 2 and 5. These results would hopefully contribute to the development of hybrid maize with better resistance against BSR.

Abstract

Global warming that occurs due to rapid population growth and increasing of energy consumption makes energy supply decreases. Energy source from non-renewable energy (fossil fuel) cause increasing greenhouse gas emission level and scarcity of natural resources. This paper reports that agricultural residues especially areca palm wastes had been widely used as an alternative energy source to reducing fossil fuel consumption. About 4.780.000 tones areca palm produced per year around the world and the shell was burned in open air or just thrown in the environment. This work aimed to investigate energy potential from areca palm shell using direct combustion and pyrolysis method also do analysis about this technology from environmental point of view and that contribution for sustainable uses.

Abstract

The escalating demand for Edelweiss flowers as souvenirs due to tourism development threatens their availability and economic value, impacting the indigenous Tengger community, which relies on these flowers for irreplaceable traditional offerings. The Hulun Hyang farmer group was established in 2017 under the guidance of the national park to conserve Edelweiss flowers, and subsequently obtained a cultivation permit from the Ministry of Environment and Forestry. However, the sustainable management of Edelweiss Park necessitates bolstering the group's institutional capacity as a critical agent in its development. This research examines the endogenous growth and active participation of the Hulun Hyang farmer group to strengthen their institutional capacity, enabling the establishment of deeply rooted and locally responsive mechanisms. Community development initiatives aim to foster economically and socially independent communities by enhancing agroecological management capacity by employing a strategic group development approach. Findings indicate that the institutional capacity of farmer groups is directly influenced by member dynamism and participation and indirectly impacted by leadership, member capacities, the role of extension workers, external support, and farmer characteristics. Consequently, efforts to augment the group's institutional capacity focus on promoting member dynamism and active engagement in group activities.

Abstract

Porang (Amorphophallus muelleri Blume) is a tuberous plant that contains glucomannan as an alternative in reducing obesity. It is widely traded, but it is cultivated through conventional methods and takes a long time. In vitro mutagenesis technique with gamma irradiation is expected to increase the production of superior porang. This study aims to determine the effect of gamma irradiation on explant growth, shoot multiplication, and plantlet regeneration on in vitro culture. The doses of gamma irradiation, i.e. 0, 2, 4, 6, 8, and 10 gamma rays (Gy). Explants bulbil were precultured for 14 days on Murashige and Skoog (MS) medium fortified with 6-benzyl amino purine (BAP) 3 mg.L-1 before irradiation. The irradiated explants were then cultured on media containing MS supplemented with α-naphthalene acetic acid (NAA) 0.1 mg.L-1 and BAP 3 mg.L-1 for 4 and 8 weeks of culture. Plantlet regeneration media was MS supplemented with NAA 0.1 mg.L-1 for 4 weeks of culture. The results showed that a dose of 10 Gy gamma irradiation was able to inhibit explant growth (0.06 g), shoot multiplication (0%), and plantlet height (0.38 cm), while gamma irradiation at a dose of 6 Gy increased explant growth (0.16 g), shoot multiplication (40%), and plantlet height (5.60 cm). The lethal dose 50 (LD50) of gamma irradiation for explant survival rate is 8.79 Gy. Abstract

Sebagai komoditas dengan fungsi yang beragam dan seluruh bagian tanamannya dapat dimanfaatkan secara total, maka talas hitam sangat berpotensi untuk dikembangkan lebih lanjut sebagai bahan pangan alternatif. Oleh sebab itu penelitian terhadap spesies talas (Colocasia esculenta L.) yakni talas hitam (Alocasia plumbea nigra), kemudian dilakukan di wilayah Malang Raya sebagai salah satu sentra penghasil talas di Indonesia. Secara khusus penelitian ini bertujuan untuk mengetahui sebaran, keanekaragaman dan jarak genetik serta dominansi talas hitam pada keanekaragaman spesies (Colocasia esculenta L.) di Malang Raya berdasarkan karakter morfologi. Hipotesis yang diajukan yakni, terdapat sebaran tanaman talas hitam yang luas, keanekargaman genetik rendah dengan jarak genetik rapat, adanya dominansi talas hitam pada keanekaragaman spesies (Colocasia esculenta L.) di Malang Raya berdasarkan karakter morfologi. Abstract

Bunga telang (Clitoria ternatea L.) atau yang dikenal sebagai Butterfly Pea merupakan bunga asal Indonesia, tergolong dalam keluarga Fabaceae atau polong-polongan. Hasil dari bunga maupun polong tanaman masih bervariasi dan secara kuantitatif masih rendah, karenanya diperlukan perbaikkan hasil produktivitas bunga telang agar memiliki kualitas yang unggul dengan dilakukannya peningkatan keragaman pada bunga telang. Salah satu upaya yang dapat dilakukan yaitu dengan pemberian kolkisin, sebagai pemicu terjadinya tanaman poliploidi. Penelitian ini bertujuan untuk mengetahui keragaman dari setiap perlakuan genotipe bunga telang, membandingkan pengaruh karakter antara genotipe bunga telang yang diberi perlakuan kolkisin dengan perlakuan tanpa kolkisin serta menghasilkan tanaman poliploidi. Penelitian dilaksanakan di Rumah Kaca Fakultas Pertanian Universitas Brawijaya Malang pada bulan September-Januari 2022. Penelitian dilakukan menggunakan Rancangan Acak Kelompok (RAK), dengan 8 perlakuan yaitu 4 genotipe bunga telang A1 (Cte-56362-02), A2 (Cte-68483-01), B1 (Cte-69281-02), B2 (Cte-55891-03) serta 4 genotipe yang diberi perlakuan kolkhisin 700 ppm P1 (CT0-56362-02), P2 (CT0-68483-01), P3 (CT0-69281-02) dan P4 (CT0-55891-03). perlakuan diulang sebanyak 5 kali. Karakter pada seluruh perlakuan genotipe yang diberi kolkisin dan tanpa kolkisin menunjukkan perbedaan yang nyata pada setiap variabel pengamatan pada karakter kualitatif dan terdapat perbedaan pada karakter kuantitatif. Sedangkan pada uji t dengan membadingkan setiap genotipe perlakuan kolkisin dengan perlakuan tanpa kolkisin menunjukkan hasil perbedaan yang signifikan pada beberapa variabel pengamatan diantaranya panjang daun majemuk, luas daun, diameter batang, diameter polen, jumlah stomata, jumlah klorofil dan lainnya. Peningkatan organ tanaman tersebut menjadi salah satu indikator terjadinya penggandaan kromosom pada tanaman poliploid.

Abstract

Cabai merah (Capsicum annum L) merupakan salah satu tanaman yang memiliki banyak manfaat untuk kebutuhan industri ataupun rumah tangga. Rerata hasil produktivitas cabai pada tahun 2015-2019 berturut-turut 8.65, 8.47, 8.46, 8.77,  9.10 ton  (Direktorat Jendral Hortikultura, 2019). Banyaknya petani dalam usaha budidaya cabai masih belum mencukupi kebutuhan cabai di Indonesia. Upaya dalam perakitan varietas unggul dengan berdaya hasil tinggi mampu meningkatkan produktivitas cabai besar di Indonesia. Uji daya hasil merupakan tahap akhir pada program pemuliaan tanaman untuk mendapatkan varietas yang diinginkan. Penelitian ini bertujuan untuk mengetahui daya hasil dari calon varietas cabai merah (Capsicum annuum L.) dengan varietas pembanding Bewe dan Pilar. Penelitian ini menggunakan rancangan acak kelompok (RAK) dengan 4 calon varietas cabai merah dan 2 varietas pembanding dengan 4 ulangan. Perameter yang diamati meliputi karakter kualitatif dan kuantitatif. Hasil penelitian menunjukan bahwa keempat calon varietas berpengaruh nyata terhadap hampir keseluruhan karakter kuantitatif terkecuali pada karakter tinggi tanaman. CV-49 memiliki hasil unggul pada jumlah buah, jumlah bunga,umur berbunga, umur panen, bobot per tanaman, bobo per plot dan hasil per Ha yang sama tinggi dengan pembanding Bewe yang mencapai 27,75 ton/Ha. Berdasarkan pengamatan karakter bentuk buah, didapatkan hasil Narrowly Triangular untuk keseluruhan calon varietas yang diamati dan pola daun Dropping pada keseluruhan calon varietas yang diuji.

Abstract

Ciplukan (Physalis angulata L.) merupakan tanaman tropis yang berkembang di Indonesia. Ciplukan dapat dikonsumsi dalam keadaan segar maupun diolah dalam bentuk buah kering. Untuk meningkatkan nilai buah ciplukan segar dan kering dilakukan analisis keragaman karakter buah melalui program pemuliaan tanaman. Keragaman diperlukan karena digunakan sebagai dasar pemilihan aksesi yang berpotensi untuk dikembangkan atau sebagai bahan baku genetik untuk perbaikan karakter buah segar dan buah kering. Tujuan penelitian ini adalah mengetahui keragaman karakter buah segar dan buah yang dikeringkan pada aksesi ciplukan, dan memilih aksesi ciplukan yang potensial dikembangkan sebagai buah segar dan buah kering. Penelitian dilakukan di greenhouse lahan percobaan Fakultas Pertanian, Universitas Brawijaya, Kelurahan Jatimulyo, Kecamatan Lowokwaru, Kota Malang, Jawa Timur dari bulan September 2021 hingga November 2021. Penelitian dilakukan dengan rancangan acak kelompok terhadap 19 aksesi ciplukan dengan 3 ulangan. Hasil penelitian menunjukkan pada tanaman ciplukan terdapat keragaman rendah, sedang, dan tinggi. Karakter buah ciplukan yang mempunyai keragaman rendah terdapat pada jumlah buah per tanaman dan umur panen. Karakter panjang buah, diameter buah, panjang kelopak, bobot buah tanpa kelopak, dan berat buah kering konstan memiliki keragaman sedang. Karakter bobot buah dengan kelopak dan kadar kemanisan buah mempunyai keragaman tinggi.
