Abstract

This study was aimed to develop the model to predict the maturity and ripeness of durian based on its physical and chemical characteristics using neural network. The physicochemical and acoustic characteristics measurement was fed into the model as the inputs, which provided the levels of maturity and ripeness as the output of the model. The results suggested that the physico-chemical properties and the acoustic charcteristic decreased with the increase of both maturity and ripeness level of durian. The total solid soluble, the water content and the total sugar increased according to the fruits maturity. The total acids increased in the beginning of durian maturing process and then decreased when the maturity and ripeness level reached the mature-over ripened stage. Data training were done by model of neural network: model 4 output, with various node in the hidden layer 4, 6, 8 and 10 nodes. The results recommended that the best model to be applied was model 4 output with 4 nodes in the hidden layer and iteration 1000 and 5000 with the model accuration 87.5 % - 100%.

Abstract

Carnation mottle virus (CarMV) merupakan virus penting pada tanaman anyelir di Indonesia maupun di dunia.  Deteksi virus yang mudah dan cepat, diperlukan untuk memantau sumber induk anyelir bebas virus. Tujuan penelitian adalah mengevaluasi tiga metode preparasi RNA total yang mudah dan cepat dari tanaman anyelir sebagai templat one step RT-PCR.  Sumber RNA total adalah dari daun dan batang anyelir terinfeksi CarMV. Metode yang dievaluasi yaitu SDT, SEM, dan kit komersial sebagai pembanding. Optimasi dilakukan terhadap konsentrasi akhir primer (0.4 – 1.0 µM) dan MgCl2 (1.5 dan 2.0 mM).  Metode SDT dan SEM berhasil mendapat RNA total dari tanaman anyelir baik dari sampel daun maupun batang.  Keberhasilan yang didapat dengan metode SDT dan SEM sebanding dengan kit komersial.  One step RT-PCR RNA total yang digabungkan dengan metode SDT dan SEM menghasilkan intensitas DNA yang sebanding dengan kit komersial.  RNA total dari daun sebagai sumber templat one step RT-PCR terbaik dibandingkan batang. Preparasi RNA total dengan metode SDT dan SEM adalah metode cepat, mudah, dan murah dalam menyediakan templat one step RT-PCR. Konsentrasi primer 0.4 µM dan MgCl2 2 mM merupakan konsentrasi optimum dan menghasilkan hasil amplifikasi terbaikKeywordsCarmovirus; Simple extraction method (SEM); Simple direct tube (SDT); One step RT-PCRAbstrackCarnation mottle virus (CarMV) is an important virus on carnation plants in the world, including in Indonesia. A rapid and easy virus detection is necessary to monitor the source of virus free carnation mother plant. The aim of the research was to evaluate three methods of rapid and easy total RNA preparation from carnation plants as template of one step RT-PCR. The total RNA source is from the CarMV infected leaf and stem of carnations. The evaluated method namely simple direct method (SDT), simple extraction method (SEM), and commercial kit as comparison. Optimization was performed to a final concentration of premiere (0.4 – 1.0 µM) and MgCl2 (1.5 and 2.0 mM). SDT and SEM method successfully obtained a total RNA from both leaves and stems samples. The obtained success by the SDT and SEM methods were comparable with these of commercial kit. One step RT-PCR of total RNA combined with SDT and SEM methods produced DNA intensity comparable with commercial kits. Total RNA from leaves known to be the best source of one step RT-PCR template compared to these from stem. Total RNA preparation by SDT and SEM method is a method of quick, easy, and inexpensive to provide a template one step RT-PCR. Premiere and MgCl2 concentration of 0.4 μM and 2 mM, respectively were optimum concentration and produced best amplification result.

A database system has been developed to prodict land suitability and cultivation information of national valuable fruits. The database system covers commodity of fruits database, agro-ecological database, and salaccia and mango information database. "A computer program for determining the best of land suitability was designed based on fruits database and agroecological database.

Abstract

Orange (Citrus grandis L.) is one of horticulture commodity which is consummed because of its delicious taste and plenty vitamins and minerals that is required by the humm body. The respiration rate of the minimally processed of orange at 10, 15 and 27.5 OC were 8.10, 13.24 and 28.212 ml/kg. h, re.jp,pectivily. The respiration rate of whole (unpeeled) orange at the same storage temperature were 10.15, 19.13 and 42.28 ml/kg. h, respectivily.

Abstract

Carnation has a highly economic demand of cut flower in Indonesia.  Field observations in West Java Indonesia was conducted in order to find the typical mottle symptoms that was a suspect caused by a virus disease. Identification of the virus was respectively conducted by performing ELISA test with four anti sera and characterizations held by bioassay, observing of virion particles, detecting of nucleic acid by RT-PCR and nucleotide sequencing. Total of 403 samples were collected from plants with or no virus-like symptoms. Among those all tested, 83% were found to be infected by Carnation mottle virus (CarMV), but negatively against Carnation ringspot virus (CRSV), Carnation laten virus (CLV), and Carnation vein mottle virus (CVMV) antisera. By mechanical inoculation, the virus was able to infect systemically Cenopodium quinoa and locally infect on others. However on Phalaenopsis sp and Gomprena globosa, there was symptompless found. The isometric CarMV particles size was approximately 30 nm. RT-PCR using specific primers of CP gene of CarMV successfully amplified a DNA sized 1000 bp.  CarMV West Java Indonesian (Idn-WJ) isolates possessed the highest nucleotide and amino acid homology with CarMV from Spain and was in the same cluster with CarMV from China, Taiwan and Israel.

Abstract

Protoplast fusion is a useful technique for citrus genetic improvement. Establishment of protoplast technology could produce triploid seedles citrus and new citrus genetic combination. The aim of this research was to establish protoplast isolation, protoplast fusion between pummelo ‘Nambangan’ (Citrus maxima) and keprok ‘Garut’ (C. reticulata), and its protoplast culture. Protoplasts were isolated from leaves of in vitro seedlings of pummelo ‘Nambangan’ and embryogenic callus of keprok ‘Garut’ with two different compositions of enzymes. The results showed that optimum enzymes compositions for cells mesophyl wall degradation were consisted of 0.5 % Onozuka cellulase RS10, 0.5 % Macerozyme R10 and 0.1 % Pectolyase Y23. Optimum enzymes composition for callus embryogenic cells wall degradation were 0.5 % Onozuka cellulase RS10 added with 0.5 % Macerozyme R10. Protoplast fusion was conducted with PEG-mediated. Protoplast mixture were incubated in 40 % PEG for 8-10 min and after that could be observed 40.6 % of unfused protoplasts, 4.6 % of homofused mesophyl protoplasts, 28.2 % of homofused callus protoplasts, 7.8 % of heterofused and 9.6 % of multifused protoplasts from mesophyl and callus. Cells divided at the first week and began to form cell colonies afterwards in liquid culture.

Abstract

Indonesia has number of accessions and cultivars of pummelo which are prospective to be developed. Pummelo contains higher antioxidants thus beneficial for health. This research aimed to get information of physical and chemical quality differences, antioxidant capacity, and explain the relationship between fruit quality and antioxidant capacity of selected seeded pummelo cultivars. Fruit was harvested in Banyuwangi and Magetan while fruit quality assesment was conducted in Laboratory of Agronomy and Horticulture Depart-ment, IPB. The results showed that physical qualities of fruit (weight loss, peel softness and peel color) and chemical qualities (total soluble solids and total titratable acidity) were changed during storage. Adas Nambangan and Banyu-wangi cultivars have better physical and chemi-cal qualities than other cultivars during storage until 10 weeks after harvest because of good visual appearance, the lowest decreased in weight loss and the good ratio of TSS:TTA. Seeded pummelo cultivars with dark red to reddish white fruit pulp had significant higher total phenolic, carotenoid, anthocyanin and anti-oxidant capacity than white fruit pulp. There were negative correlations between antioxidant capacity with colored pulp and total phenolic content. Banyuwangi had the highest antioxidant capacity in the pulp, followed by Bali Merah, Adas Nambangan, Pamelo Magetan, Srinyonya, Bali Putih cultivars.

Abstract

Jeruk pamelo mengandung senyawa antioksidan, antihiperlipidemik, antidiabetik, protein, kalsium dan senyawa bermanfaat lainnya. Indonesia memiliki dua musim, yakni musim penghujan dan musim kemarau. Musim dan ketersediaan air mempengaruhi ketersediaan hara tanaman dan kualitas buah yang dihasilkan. Tujuan dari penelitian ini adalah mengkaji perbedaan kualitas buah pamelo yang dipanen pada dua musim, yakni musim penghujan dan musim kemarau. Penelitian ini dilaksanakan pada bulan Oktober 2018 sampai September 2019 menggunakan rancangan acak lengkap (RAL) 1 faktor yaitu musim. Bahan penelitian menggunakan kultivar Adas Duku yang buahnya dipanen pada musim penghujan dan kemarau. Pengamatan lingkungan dilakukan pada suhu, relative humidity (RH) dan curah hujan. Peubah yang diamati meliputi diameter buah panen secara membujur dan melintang (cm), bobot buah panen (g), tebal kulit buah (cm), jumlah biji (biji), bagian dapat dimakan (BDD) (%), padatan terlarut total (PTT) dalam satuan ºBrix, asam tertitrasi total (ATT) dalam satuan %, rasio PTT/ATT, Vitamin C mg/100 mL dan IC50 dalam %. Buah panen yang diperoleh berukuran lebih besar pada musim penghujan dibandingkan musim kemarau namun buah yang dipanen kemarau memiliki PTT, ATT dan kapasitas antioksidan yang lebih tinggi daripada buah yang dipanen musim penghujan.
