Abstract

ABSTRACT‘Kristal’ guava fruit can easily be deteriorated during storage. The objective of this experiment was to study the effect of the waxing on the shelflife and quality of the ‘Kristal’ guava fruit. Experiment was conducted at the Postharvest Laboratory of Departemen of Agronomy and Horticulture, Faculty of Agriculture, Bogor Agricultural University from February to May 2016. The experiment was designed using completely randomized design (CRD) with single factor consisted of 5 treatments, i.e., 0% of beewax (control), 2% of beewax, 4% of beewax, 6% of beewax and 8% of beeswax. The results showed that beewax coating suppressed weight loss and softness of fruit compared to the control fruit during storage period. Beewax coating did not significantly affect chemical quality such as soluble solids content, titratable acidity and vitamin C. The beeswax coating with 2% and 4% were able to extend shelflife of ‘Kristal’ guava fruit 9 days longer as compared to the control. Keywords: ‘Krístal’, performance, quality, waxing, weight loss ABSTRAK Buah jambu ‘Kristal’ dapat dengan mudah rusak selama penyimpanan. Tujuan percobaan ini yaitu mempelajari pengaruh pelilinan terhadap daya simpan dan kualitas buah jambu ‘Kristal’. Percobaan dilakukan di Laboratorium Pascapanen, Departemen Agronomi dan Hortikultura, Fakultas Pertanian, Institut Pertanian Bogor dari bulan Februari sampai Mei 2016. Percobaan dirancang menggunakan rancangan acak lengkap (RAL) dengan faktor tunggal terdiri atas 5 perlakuan, yaitu 0% dari lilin lebah (kontrol), 2% dari lilin lebah, 4% lilin lebah, 6% lilin lebah dan 8% lilin lebah. Hasil penelitian menunjukkan bahwa pelapisan lilin lebah menekan susut bobot dan kelunakan buah dibandingkan dengan kontrol selama masa penyimpanan. Pelapisan lilin lebah tidak secara nyata mempengaruhi kualitas kimia buah seperti kandungan padatan terlarut, keasaman titrasi dan vitamin C. Pelapisan lilin lebah dengan konsentrasi 2% dan 4% mampu memperpanjang masa simpan buah jambu ‘Kristal’ 9 hari lebih lama dibandingkan dengan kontrol. Abstract

ABSTRACT Arrangement of Guava Flowering (Psidium guajava L.) ‘Krystal’ through the Application of Different Strangulation Times. The seasonal production of guava (Psidium guajava) requires flowering manipulation technique such strangulation in order to be available throughout the years. This study aimed to explain the relationship between strangulation time, the period of flowering and harvesting of guava cv 'Krystal'. The experiment was conducted from February to December 2017, in Cikabayan experimental garden of IPB University, Bogor, Indonesia. The experiment used a completely randomized design with single factor, i.e strangulation times that consisted of control (no strangulation), strangulation in March, in April and in May. The results showed that strangulation treatment was able to accelerate the emergence of flowers and increase the number of generative shoots, the number of flowers per tree and the number of fruits harvested. The increase of flowering response on strangulated trees was supported by a higher leaf C/N compared to control, which is related to the low leaf N content in strangulation treatments. The strangulation treatment could accelerate the time of flower emergence six days earlier than control.  Keywords: guava, C/N, flowering induction, ringing, seasonal production ABSTRAK Produksi buah jambu biji (Psidium guajava) yang bersifat musiman memerlukan teknik pengaturan pembungaan agar dapat tersedia sepanjang tahun. Penelitian ini bertujuan untuk menjelaskan keterkaitan antara waktu strangulasi dengan pola pembungaan dan panen buah jambu biji ‘Kristal’. Percobaan dilaksanakan mulai Februari sampai Desember 2017, di Kebun Percobaan Cikabayan IPB Bogor. Rancangan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan faktor tunggal yaitu waktu strangulasi yang terdiri dari 4 (empat) taraf, yakni tanpa strangulasi, strangulasi bulan Maret, strangulasi bulan April dan strangulasi bulan Mei. Hasil penelitian menunjukkan bahwa perlakuan strangulasi mampu mempercepat munculnya bunga dan meningkatkan jumlah tunas generatif, jumlah bunga per pohon, jumlah bakal buah per pohon dan jumlah buah yang dipanen. Peningkatan respon pembungaan akibat strangulasi didukung oleh C/N daun yang lebih tinggi dibandingkan tanpa stangulasi, yang berhubungan dengan rendahnya kandungan nitrogen daun pada perlakuan strangulasi. Perlakuan strangulasi mampu mempercepat waktu muncul bunga 6 hari lebih cepat dibandingkan tanpa strangulasi.Kata kunci: jambu biji, C/N, induksi pembungaan, pencekikan batang, produksi musiman

Abstract

The objectives of the study are : (1) to design  land  suitability database for cultivation of the national valuable fruits, (2) to design the activation information of  fruits production, including land preparation until primary postharvest handling, and (3) to design the decision support system for valuable fruits on people’s orchads to produce fresh fruits. The study was conducted in Yogyakarta Special Region, from November 1999 until March 2000.  Two kinds of data used in this study were: (a) primary data, from the direct discussion with the experts, and direct observation in the fruits production center, and (b) secondary data were collected from library studies and Agriculture Agency Offices in each regency or municipality of the Yogyakarta Special Region.  The data in the salaccias production center was established from farmers which owned orchad more than one hectare. The results of  the study were : (1) land suitability database for national valuable fruits cultivation based on relational data, (2) computerize information system, namely Salaccia (Pondoh cultivar) and Mango (Arumanis, Manalagi, Golek, and Gedong cultivar) included : land preparation, planting, fertilizing, cultivating, pest and disease control, pruning, irrigating, harvesting, cleaning of fruits, sorting of fruits, grading of fruits, ripening (specially for mango), and storage of fruits, and (3) the system has been done to determine land suitability for fruits in Yogyakarta Special Region, and the result showed that 62.39 % was suitable between output system designed and the fruits produced by the farmer, namely manggistas, pineapples, mangoes, rambutans, durians, salaccias, and lawzons.

Abstract

This study was aimed to develop the model to predict the maturity and ripeness of durian based on its physical and chemical characteristics using neural network. The physicochemical and acoustic characteristics measurement was fed into the model as the inputs, which provided the levels of maturity and ripeness as the output of the model. The results suggested that the physico-chemical properties and the acoustic charcteristic decreased with the increase of both maturity and ripeness level of durian. The total solid soluble, the water content and the total sugar increased according to the fruits maturity. The total acids increased in the beginning of durian maturing process and then decreased when the maturity and ripeness level reached the mature-over ripened stage. Data training were done by model of neural network: model 4 output, with various node in the hidden layer 4, 6, 8 and 10 nodes. The results recommended that the best model to be applied was model 4 output with 4 nodes in the hidden layer and iteration 1000 and 5000 with the model accuration 87.5 % - 100%.

Abstract

Carnation mottle virus (CarMV) merupakan virus penting pada tanaman anyelir di Indonesia maupun di dunia.  Deteksi virus yang mudah dan cepat, diperlukan untuk memantau sumber induk anyelir bebas virus. Tujuan penelitian adalah mengevaluasi tiga metode preparasi RNA total yang mudah dan cepat dari tanaman anyelir sebagai templat one step RT-PCR.  Sumber RNA total adalah dari daun dan batang anyelir terinfeksi CarMV. Metode yang dievaluasi yaitu SDT, SEM, dan kit komersial sebagai pembanding. Optimasi dilakukan terhadap konsentrasi akhir primer (0.4 – 1.0 µM) dan MgCl2 (1.5 dan 2.0 mM).  Metode SDT dan SEM berhasil mendapat RNA total dari tanaman anyelir baik dari sampel daun maupun batang.  Keberhasilan yang didapat dengan metode SDT dan SEM sebanding dengan kit komersial.  One step RT-PCR RNA total yang digabungkan dengan metode SDT dan SEM menghasilkan intensitas DNA yang sebanding dengan kit komersial.  RNA total dari daun sebagai sumber templat one step RT-PCR terbaik dibandingkan batang. Preparasi RNA total dengan metode SDT dan SEM adalah metode cepat, mudah, dan murah dalam menyediakan templat one step RT-PCR. Konsentrasi primer 0.4 µM dan MgCl2 2 mM merupakan konsentrasi optimum dan menghasilkan hasil amplifikasi terbaikKeywordsCarmovirus; Simple extraction method (SEM); Simple direct tube (SDT); One step RT-PCRAbstrackCarnation mottle virus (CarMV) is an important virus on carnation plants in the world, including in Indonesia. A rapid and easy virus detection is necessary to monitor the source of virus free carnation mother plant. The aim of the research was to evaluate three methods of rapid and easy total RNA preparation from carnation plants as template of one step RT-PCR. The total RNA source is from the CarMV infected leaf and stem of carnations. The evaluated method namely simple direct method (SDT), simple extraction method (SEM), and commercial kit as comparison. Optimization was performed to a final concentration of premiere (0.4 – 1.0 µM) and MgCl2 (1.5 and 2.0 mM). SDT and SEM method successfully obtained a total RNA from both leaves and stems samples. The obtained success by the SDT and SEM methods were comparable with these of commercial kit. One step RT-PCR of total RNA combined with SDT and SEM methods produced DNA intensity comparable with commercial kits. Total RNA from leaves known to be the best source of one step RT-PCR template compared to these from stem. Total RNA preparation by SDT and SEM method is a method of quick, easy, and inexpensive to provide a template one step RT-PCR. Premiere and MgCl2 concentration of 0.4 μM and 2 mM, respectively were optimum concentration and produced best amplification result.

Abstract

Orange (Citrus grandis L.) is one of horticulture commodity which is consummed because of its delicious taste and plenty vitamins and minerals that is required by the humm body. The respiration rate of the minimally processed of orange at 10, 15 and 27.5 OC were 8.10, 13.24 and 28.212 ml/kg. h, re.jp,pectivily. The respiration rate of whole (unpeeled) orange at the same storage temperature were 10.15, 19.13 and 42.28 ml/kg. h, respectivily.

Abstract

Carnation has a highly economic demand of cut flower in Indonesia.  Field observations in West Java Indonesia was conducted in order to find the typical mottle symptoms that was a suspect caused by a virus disease. Identification of the virus was respectively conducted by performing ELISA test with four anti sera and characterizations held by bioassay, observing of virion particles, detecting of nucleic acid by RT-PCR and nucleotide sequencing. Total of 403 samples were collected from plants with or no virus-like symptoms. Among those all tested, 83% were found to be infected by Carnation mottle virus (CarMV), but negatively against Carnation ringspot virus (CRSV), Carnation laten virus (CLV), and Carnation vein mottle virus (CVMV) antisera. By mechanical inoculation, the virus was able to infect systemically Cenopodium quinoa and locally infect on others. However on Phalaenopsis sp and Gomprena globosa, there was symptompless found. The isometric CarMV particles size was approximately 30 nm. RT-PCR using specific primers of CP gene of CarMV successfully amplified a DNA sized 1000 bp.  CarMV West Java Indonesian (Idn-WJ) isolates possessed the highest nucleotide and amino acid homology with CarMV from Spain and was in the same cluster with CarMV from China, Taiwan and Israel.
