Abstract

Abstract

Introduction: Cervical cancer remains one of the leading causes of morbidity and mortality worldwide, including in Indonesia, involving metabolic reprogramming such as the Warburg effect in tumorigenesis. The proteins IGF2BP2 and MYCBP are known to play important roles in promoting cancer cell proliferation through the regulation of cellular metabolism. Objective: This study aimed to investigate the anticancer potential of Indole-3-Carbinol against cervical cancer by targeting IGF2BP2 and MYCBP using an in silico approach. Method: The study employed molecular docking analysis targeting IGF2BP2 and MYCBP proteins, using 5-fluorouracil and doxorubicin as comparator compounds. Result: The results showed that Indole-3-Carbinol exhibited binding energy (∆G) values of -3.20 kcal/mol for IGF2BP2 and -5.75 kcal/mol for MYCBP, with better binding affinity than 5-fluorouracil but lower than doxorubicin. Interaction analysis revealed that Indole-3-Carbinol interacted with both IGF2BP2 and MYCBP proteins, with IGF2BP2 showing a higher number of interacting amino acid residues. Further studies are required to confirm its preferential molecular target. Furthermore, Indole-3-Carbinol fulfilled Lipinski’s criteria, demonstrated high gastrointestinal absorption, the ability to penetrate the blood-brain barrier, and a favorable toxicity profile without carcinogenic effects. Conclusion: Indole-3-Carbinol shows preliminary in silico potential as a cervical anticancer candidate targeting the IGF2BP2 pathway and modulating the Warburg effect, with the advantage of being a relatively safe natural compound.

Abstract

Abstract:Background. Type 2 diabetes mellitus (T2DM) is a metabolic disorder characterized by chronic hyperglycemia due to insulin resistance and pancreatic β-cell dysfunction. Chronic hyperglycemia triggers oxidative stress and increases inflammation, thereby contributing to liver damage. The potential of coffee cherry ethanol extract as an antioxidant and anti-inflammatory agent could serve as a therapeutic option for improving liver damage. Objective. This study aims to evaluate the effects of coffee cherry ethanol extract on liver histopathology in a T2DM mouse model. Methods. This study used 28 male Wistar rats, divided into 7 groups with 4 rats in each group, administered coffee cherry ethanol extract at doses of 100 mg/kg body weight and 200 mg/kg body weight/day/oral. Results. The results showed that administration of coffee cherry ethanol extract improved hepatocyte structure, reduced inflammatory infiltration, and decreased fat accumulation in liver tissue compared to the untreated DMT2 group (p0.05). Conclusion. Coffee cherry ethanol extract has potential as an adjunct therapy to slow the progression of DMT2 complications in the liver. Further studies are needed to confirm its efficacy, mechanism of action, and long-term effects in humans. Keywords: ethanol extract, liver histopathology, oxidative stress, type 2 diabetes mellitus.

Abstract

Complementary feeding (MPASI) refers to food or beverages given to infants after six months of age to meet nutritional needs and support growth and development. Mothers’ knowledge of complementary feeding is an important factor in food selection and feeding practices that may influence the nutritional status of toddlers. This study aims to determine the relationship between mothers’ knowledge of complementary feeding and the nutritional status of toddlers at the Toddler Integrated Health Post (Posyandu) of Sambirejo Public Health Center, Binjai District, Langkat Regency. This study employed a descriptive design with a cross-sectional approach and was conducted from June to December 2024. The sample consisted of 45 mothers with toddlers selected using total sampling. Data on mothers’ knowledge were collected through questionnaires, while toddlers’ nutritional status was assessed through anthropometric measurements. Data were analyzed using univariate and bivariate analyses with the Spearman correlation test. The results showed that 24 mothers (53.3%) had good knowledge, 14 (31.1%) had moderate knowledge, and 7 (15.6%) had poor knowledge. Regarding nutritional status, 23 toddlers (51.1%) had good nutritional status, 14 (31.1%) were undernourished, and 8 (17.8%) were severely undernourished. A significant relationship was found between mothers’ knowledge and toddlers’ nutritional status (p<0.001; r=0.523). Better maternal knowledge was associated with better nutritional status. Complementary feeding education through Posyandu is needed to optimize toddlers’ nutritional status.

Abstract

Decreased levels of brain-derived neurotrophic factor (BDNF) are closely related to neurodegenerative diseases such as Alzheimer's. This causes a decrease in cognitive function, memory and behavior due to a decrease in the BDNF protein which has the main function of protecting nerve growth, plasticity, nerve resilience, as well as being an important shield against nerve degeneration. Objective: of this research is to analyze the interaction between the oleic acid and alpha tocopherol ligands with the BDNF protein by molecular docking, as well as to analyze the ADMETox compounds of oleic acid and alpha tocopherol. Methods: This research is descriptive analytical. Molecular docking uses the Autodock tools, LigandScout, Discovery Studio, and Marvin Sketch applications. ADMETox analysis uses the SwissADME, ADMETlab 2.0, and ProTox-II sites. Results: the binding energy of oleic acid compounds to BDNF, TrkB, and CREB is -3.13 kcal/mol, -6.13 kcal/mol, and -3.70 kcal/mol. The binding energy results for alpha tocopherol compounds on BDNF, TrkB, and CREB are -5.67 kcal/mol, -8.04 kcal/mol, and -7.04 kcal/mol. ADMETox found that alpha tocopherol and oleic acid compounds fulfill Lipinski's rule of five and do not inhibit cytochrome P450 enzymes. Conclusion: Oleic acid and alpha tocopherol bind to BDNF, TrkB and CREB proteins and are safe for oral consumption.
