Abstract

This study aimed to determine the effect of a combination of extracts of Noni leaves (Morinda citrifolia L) and fruit kandis (Garcinia cowa Roxb) on body weight of male rats induced by high- fat calories food Test animals were used 25 rats male Sprague Dawley strain age of 2 months with body weight 200 g. White mice were divided into 5 groups of 4 rats each. The treatment committed agains an obesity rats. The treatment group consisted of rats doses of 1 (8,75mg/kgBB noni leaves ekstract and sour fruit kandis 7mg/kgBB), dose II (4,75 mg/kgBB noni leaves ekstract and sour fruit kandis 3,5mg/kgBB), dose III (17,5 mg/kgBB noni leaves ekstract and sour fruit kandis 14mg/kgBB), the positive control group were given Orlistat and negative group were only given food to eat and drink. The observed parameters include weight loss, abdominal width, body length, skin fold, body mass indeks. The results showed that noni leaf extract and fruit kandis can be used as a weight loss with the most effective dose is 17,5 mg /kg BB noni leaf + 14 mg/kg BB fruit kandis with the most effective treatment is 14 days.Key words : Noni Leaf and fruit kandis, Feeding a High-Fat Calories, Obesity Parameters, Rats male Sprague Dawley.

Abstract

Turmeric contains curcumin which has antibacterial and anti-inflammatory activities. In addition, turmeric contain also carbohydrates that has potency as a prebiotic agent. Research on turmeric extract that was obtained by maceration in variety of solvent, temperatures and time have been carried out to get the most potent prebiotic extract of turmeric to stimulate L. plantarum growth. Maceration with water at 28oC for 1 hour resulted in extract which can enhance the growth of L. plantarum approximately is higher than controls at a concentration of 1 ppm. L. plantarum growth rate increases with increasing concentration of extract. Sugars content of oligosaccharide in the extract were stakiosa, sucrose and lactose of 3014, 164, and 82 ppm respectively. It is concluded that oligosaccharide in turmeric extract is potential as prebiotic agent to increase L. plantarum growth.

Abstract

Background: Mucus is pre-epithelial gastric layer that may prevent damages due to direct contact between aspirin and gastric epithelial cells. The integrity of gastric mucosa and mucous cellular reaction may serve as primary and secondary prevention of extended aspirin-induced gastric mucosal damage. The aim of this study was to prove the function of mucus as defensive factor in rats. Method: The study was conducted in twenty white rats of the Sprague-Dawley strain at Department of Pathology and Clinical Reproduction, Bogor Agricultural University, between January and December 2008. The rat in the treatment group were given 400 mg aspirin diluted in aqua bidest through intra- gatric canules; while the control group received aqua bidest only once daily for 3 days. Necropsies, macroscopic and microscopic observation were performed by counting the number of Alcian blue- periodic acid Schiff-stained mucous cells at fundus/corpus and antrum/pylorus regions. Data analysis was performed using ANOVA and Duncan test. Results: The number of mucous cells with positive lesions in the treatment group was significantly different from the control group at both regions. There was no significant difference of negative lesions between treatment and control group at both regions. At antrum/pylorus region, there was no difference of negative lesions between treatment and control groups; however, both groups demonstrated significant difference of positive lesions in treatment group. Conclusion: In primary prevention for gastric mucosal lesions, there is no increasing number of mucous cells in normal mucosa. Increasing number of mucous cells is a secondary prevention against extended aspirin-induced gastric mucosal damage. Keywords: NSAIDs/ASA, mucus cells, gastric mucosal lesion, rat

Abstract

Background: Nonsteroidal anti-inflammatory drugs (NSAIDs) gastropathy is a common complication, which has characteristic symptoms of dyspepsia syndrome. Mostly, it includes epigastric discomfort with bloating and nausea. The aim of this study was to provide evidences that clinical symptoms of dyspepsia are related to macroscopic changes of gastric form in rats, which are expected to be applied in human. Method: The study was conducted in 20 white rats (Rattus norvegicus, Sprague-Dawley strain) at the Department of Pathology and Clinical Reproduction, Bogor Agricultural University between January and December 2008. The rats were divided to treatment group and control group and each group consisted of 10 rats. Acetyl salicylic acid (ASA/aspirin) was administered at 400 mg dose, diluted in distilled water and was given to the treatment group using gastric cannula, once daily for three days period; while the control group had received aquabidest only. Subsequently, necropsies were conducted for both groups, followed by macroscopic observation and measurement of sagittal and transversal diameter. Gastric incisions along the minor curvature were performed in both groups to recognize any macroscopic changes of gastric mucosa. ANOVA test was utilized for data analysis, which was followed by Duncan test when the results were significant. Results: Gastric diameters in treatment group with positive lesion were significantly different from the control group and the treatment group with negative lesion on anthrum/pylorus region, with p 0.05. Conclusion: Prominent gastric dilatation at anthrum/pylorus region found in the treatment group may become the initial cause and signs of dyspepsia in human. Abstract

Pacific whiteleg shrimp (Litopenaeus vannamei) is a very important commodity, which accounts for almost 70% of the total world shrimp production. However, this production is still threatened by disease attacks, one of which is caused by white spot disease (WSD). Herbs are widely used as immunostimulants as an effort to prevent diseases. Black cumin (Nigella sativa) and stone breaker (Phyllanthus niruri) have long been known as excellent immunostimulants in human. This study aimed to evaluate the potential of N. sativa and its combination with P. niruri as an immunostimulant in L. vannamei for preventing WSD. Tested shrimp (±3.0 g) were treated by N. sativa (N), P. niruri (P) and the combination of both (1N:1P, 2/3N:1/3P, 3/4N:1/4P, 1/3N:2/3, 3/5N:2/5P, 1/4N: 3/4P, 2/5N:3/5P) through feed for 28 days. They were then infected by white spot syndrome virus (WSSV) and observed for 7 days. The parameters observed included immune response and production performance.  N. sativa was able to reduce the mortality of test shrimp infected with WSSV, with an RPS value of 71%. N. sativa can be combined with P. niruri in the right composition (1/2N:1/2P and 2/3N:1/3P), while the other combinations are antagonistic. The administration of the best treatments in this study, relatively did not affect the value of total hemocyte count, phenoloxidase activity, respiratory burst activity, hemolymph clotting time and production performance of tested shrimp.

Abstract

Penelitian ini bertujuan mengetahui pengaruh penyuntikan pregnant mare serum gonadotropin (PMSG) terhadap peningkatan respons berahi kambing kacang. Sebanyak 20 ekor kambing kacang betina dewasa kelamin dibagi secara acak ke dalam 5 perlakuan dosis penyuntikan PMSG, yaitu 0,0 (K1); 1,0 (K2); 7,5 (K3); 15,0 (K4); dan 20,0 (K5) IU/kg bobot badan dan diulang 4 kali. Hasil penelitian menunjukkan bahwa pada K3, K4, dan K5 terjadi peningkatan jumlah folikel dominan, total diameter folikel dominan, total volume folikel dominan, dan konsentrasi estrogen dalam serum darah (P0,05), serta menunjukkan gejala berahi yang lebih jelas dibandingkan K1. Konsentrasi estrogen dalam serum darah memiliki korelasi positif dengan jumlah folikel dominan, total diameter folikel dominan, total volume folikel dominan, onset berahi, durasi berahi, perubahan vulva menjadi merah, kebengkakan vulva, peningkatan respons betina terhadap pejantan, dan pergerakan ekor pada saat berahiatau fleging berturut-turut sebesar 53,10; 82,60; 48,80; 53,0; 45,50; 39.40; 32,60; 32,60; dan 80,20% dengan pola kuadratik. Dari hasil penelitian disimpulkan bahwa penyuntikan PMSG pada dosis 7,5; 15,0; dan 20,0 IU/kg bobot badan mampu meningkatkan konsentrasi estrogen dan respons berahi kambing kacang.

Abstract

The accession of mulberry (Morus sp.) with salinity tolerance unavailable in Indonesia is needed to overcome the problem of marginal land, especially in the saline areas of the country. Therefore, this research aims to determine the accession of salinity tolerance of mulberry from 6 origins in Indonesia based on growth and physiological parameters. The method used was a factorial randomized block design with the first treatment of stem cutting-derived mulberry accession being M1, M2, M3, M4, M5, M6, and M7, while the second was the concentration of NaCl at 0 mM, 40 mM, 60 mM, and 80 mM. The results showed that M2 accession from Bogor was categorized as tolerance to high salt stress, and M1 was classified as sensitive accession. Increasing salt concentration causes a decrease in growth parameters. It also decreased physiological parameters such as leaf and media relative water content while increasing dissolved sugars. Genotypic scores indicate salinity tolerance and the potential number of leaves, while Na+ content and leaf necrosis percentage showed sodium toxicity in the leaf. Therefore, genotypic score, Na+/K+ ratio, and leaf necrosis percentage are the best parameters to select and identify mulberry accession with high salt stress tolerance.

Abstract

Lactobacillus casei merupakan salah satu bakteri saluran pencernaan yang memberi dampak positif terhadap kesehatan tubuh inang. Pertumbuhan bakteri ini dipengaruhi oleh beragam jenis karbohidrat terutama karbohidrat yang resisten terhadap enzim pencernaan.Adanya kandungan serat pangan dalam labu siam, memungkinkan ekstrak air labu siam (Sechium edule) dapat digunakan sebagai nutrisi untuk bakteri probiotik seperti Lactobacillus casei. Potensi ekstrak buah dan daun labu siam sebagai prebiotik dikaji terhadap pertumbuhan bakteri Lactobacillus casei menggunakan metoda dilusi untuk penentuan KTM dan menggunakan metoda difusi kertas cakram yang diresapi ekstrak buah dan daun labu siam pada konsentrasi 10%, 15%, 20%, 25% untuk menentukan LDT. Hasil pengujian menunjukkan konsentrasi tumbuh minimum ialah konsentrasi 10%. Rata-rata LDT buah labu siam pada konsentrasi tersebut sebesar 2,40 mm sedangkan pada daun labu siam yaitu 1,99 mm. sehingga dapat disimpulkan bahwa ekstrak buah dan daun labu siam berpotensi sebagai prebiotik.

Abstract

The aim of this study is to compare the potential of ethanolic and water extracted of teki grasstuber on   white  male  rat strain Sprague  Dawley  at  two  months of age and ± 200 g of body weight. The rats were divided among four groups. Each group consists of six rats. The  groupswere ethanolic and water extracted  of  teki  grass tuber, positive control group (Paracetamol) and a negative control group (distilled water). Analgesic potency was tested by inducing pain stimuli previously to the animals, in the form of  heat stimuli  by putting it ona hot plate with  the temperature of 55°C. The responses of rats observed were licking and moving or lifting of their legs. Observations  were made for 1 minute, before  and  after  administration  of  pain stimulus at 15, 30, 45  and 60 minute. The results showed that ethanol and  water  extract  at an effective dose of 49 mg / 200 g of BW, gave relatively the same effect as an analgesic or   positive control group. Keywords : analgesic, ethanol and water extract of teki grass tuber, pain
