Abstract

Selain sebagai biopestisida ekstrak etanol 70 % biji bengkuang dilaporkan mempunyai aktivitas antibakteri terhadap bakteri gram positif Staphylococcus aureus dan bakteri gram negatif Escherichia coli dan Candida alibican. namun efeknya terhadap Staphylococcus epidermidis dan Pseudomonas aeruginosa, belum dketahui. Penelitian ini ditujukan untuk menentukan aktivitas antimikroorganisme ekstrak etanol 70 % biji bengkuang lokal  terhadap  Staphylococcus epidermidis, Pseudomonas aeruginosa dan  Candida Albican menggunakan teknik difusi agar dan metoda dilusi.  Aktivitas antimikroorganisme ekstrak diteliti pada konsentrasi 10% 12% 14%, 16% dan sebagai kontrol negatif, kontrol positif antibakteri dan kontrol positif antifungal masing masing digunakan akuadestilata.  ertromisin 10 ppm, ketokonazol 10 ppm. Data yang diperoleh menunjukkan bahwa ekstrak efektif sebagai antibakteri terhadap Staphylococcus epidermidis dan Pseudomonas aeruginosa serta sebagai antifungi terhadap Candida albican, pada konsentrasi bunuh minimum 10 mg/ml. Kata kunci: Ketokonazol, Eritromisin Pseudomonas biji bengkuang Candida albikan

Abstract

Betle and ginger juice extracts have been reported to have antibacterial activity against E. coliand M. gallisepticum. However, fractionation analysis showed the have differences in their antibacterialpotency which appeared to be associated with the nature of the solvent polarity. This study wasconducted to obtain information about antibacterial activity of Piper betles and gingers extract againstE. coli and M. gallisepticum. Three types of betle, P. betle, P. betle var nigra, P. crocatum, wereexamined for their antibacterial activity using disc method and three types of ginger, Zingiber officinale,Z. officinale Linn var rubrum, and Z. majus rumph which were extracted by soxhletation using ethanol30%, 60% and 90% as well as distestillation for three and five hours. Piper betle and P. crocatumconsistently have antibacterial effect against E. coli whereas Z. officinale consistently has antibacterialeffect against M. gallisepticum, either extracted by distillation or soxhletation. Piper betle is potentialyield of distillate and extract that has the highest antibacterial activity against E. coli, and M.gallisepticum with inhibitory zone were 9.76 mm and 22 mm respectively.

Abstract

The aim of the study was to analyze the potency of Centella asiatica extract to induce proliferation andneurogenesis process of newborn rats cortex cerebri.   Research has been conducted on in vitro culture ofthree days old rat (Sprague Dawley) cerebrum cells in DMEM (Dulbecco’s Modiûed Eagle’s Medium)containing 10% NEAA (Non Essential Amino Acid), 1 mM NaHCO3, 10% NBCS (Newborn Calf Serum)and 50 µg/mL gentamycin (mDMEM), with and without Centella asiatica (CA) leaf extracts. The experimentwas set in five groups of treatment consisted of positive control (mDMEM+30 µg/mL asiaticoside (AC)),negative control (mDMEM), and mDMEM with three concentration of CA extract i.e. 100 ppm, 200 ppmand 400 ppm. Culture was done in 5% CO2 incubator at 37oC for six days. The parameters observed werecells proliferation based on Population Doubling Time (PDT), neuron and glia composition, and the lengthof axon and dendrite. Cells concentration were counted using Newbauer hemocytometer.  Neuron and gliacells were determined based on morphology after Hematoxylin-Eosin staining, and the length of axon anddendrite were measured using eyepiece micrometer. Data were analyzed using ANOVA and Duncan test.The results showed that Centella asiatica extract at concentration 100 ppm could induce neurogenesisand increased the axon length growth.  However, at concentration 200 and 400 ppm, CA extract  inhibitedthe neuronal cells proliferation and the axonal growth (P<0,05). In conclusion, induction of Centella asiaticaextracts at concentration of 100 ppm on the cortex cell cerebrum cells culture increase the axon lengthgrowth and tends to induce neurogenesis; however at higher concentration CA extract was neurotoxic.

Abstract

Coxiella burnetii belongs to rikettsia group living obligate intracellularly and as the agent of zoonosisQ fever. Enrofloxacine is an antibiotic in quinolon group used to treat infection of C. burnetii in chicken,goat, calve, pig, dog, cat,  and horse. From ruminant practical experience, enrofloxacine if combined withBioATP  can enhance the enrofloxacine activity. Research for the effecivity of enrofloxacine and BioATP totreat C. burnetii has never been carried out. The research was conducted to explore effect of enrofloxacinewith supplementation BioATP against C. burnetii. Enrofloxacine pharmacokinetic study was carried outby using simental beef as an experimental animals. The effectivity of BioATP supplementation onenrofloxacine activity to treat C. burnetii was tested by using Vero cell tissue culture. The results showedthat combination of enrofloxacine and BioATP increased kinetic profile of enrofloxacine in term of onset,duration, pharmacology intensity, and bioavailaibility. Enrofloxacine had activity to treat C. burnetii withvalue of minimal inhibitory concentration (MIC) at 1-2 ppm and value of minimal bactericidal concentrationat 4 ppm. Supplementation of BioATP improved the effectivity of enrofloxacine in treating C. burnetii.

Abstract

Mimusops elengi L. , Averrhoa carambola L. and Curcuma xanthorrihiza Roxb singly were reported to have hypothensif effect. Mechanism underlying decrease of blood pressure was suggested through modification of cardiac activity. The Effect of Mimusops elengi L., combined with Averrhoa carambola L. and Curcuma xanthorrihiza Roxb extract on cardiac activity were studied using ECG on 12 male cats that were grouped to be a control group administered aquadestilata, and treatment group administered extract of 21 mg and 82 mg/2 kg bw orally respectively. The extracts were given 3 h prior to ECG. The results showed a decrease in P wave, QRS complex and the speed of the heart rate after administration of combination of extract . However QT and PR intervals were increased. This showed that the extract can weaken a contraction of the Atria and ventricles, prolonging the onset of the occurrence of atrial contraction towards ventricular contraction, extending the onset of contraction and relaxation of the ventricles and cause a decrease in heart rate. Based the cardiogram, It was concluded that their combination is valuable for treatment of aritmia.

Abstract

Diabetes is marked by hyperglicemia with severe symptoms included polydipsi, polyuri, polyphagia,weight loss, and decreased of insulin sensitivity index specially at type 2 diabetes mellitus. The objective of this research was to analyze the characteristics of diabetes mellitus in rat (Rattus novergicus) which induced by dexamethason. A total of 24 rats were divided into four groups which divided as follow: (1) aquabidest subcutaneously (K0), (2) subcutaneous dexamethasone 1 mg/kgBW daily (K1), (3) subcutaneous dexamethasone 2.5 mg/kgBW daily (K2) and subcutaneous dexamethasone 5 mg/kgBW daily (K3). The data of fasting blood glucose, fasting blood insulin, body weight and feed intake, urine volume and water intake collected at day 0, 5, 10 and 15 (five days after the last treatment). The result showed that there was a significant elevation of glucose level followed by significant decreased of insulin sensitivity index and the body weight of rats in group wich induced by dexamethasone but there was no change in feed intake, water intake, volume of urine and fasting blood insulin before and after the treatment. There was insignificant different of total beta pancreatic islet between treatment and control grup. Induction of dexamethasone in mice causes reversible hyperglycemia and has not met the characterisics of either type 1 or 2 diabetes mellitus models. The profile of dexamethasone induced diabetes model has their own spesific type.

Abstract

Permasalahan utama dalam kegiatan usaha budidaya perikanan antara lain adalah ketersediaan benih ikan yang tepat waktu, tepat jumlah dan berkualitas. Ketersediaan benih ikan sangat tergantung pada jumlah telur yang dapat dibuahi dan ditetaskan. Semakin tinggi presentase pembuahan dan penetasan maka akan semakin tinggi pula jumlah larva yang dapat dihasilkan. Penelitian ini menggunakan ikan lele sebagai hewan coba dengan tujuan untuk mengetahui pengaruh suplementasi kurkumin dan tiroksin pada induk ikan lele terhadap kualitas dan performa telur yang dihasilkan. Suplematasi kurkumin dan hormon tiroksin melalui pakan selama 12 minggu pemeliharaan. Parameter yang dianalisis berupa konsentrasi vitelogenin telur, diameter telur, koefisien keragaman diameter telur, konsentrasi lipid dalam telur (triglserida, kolesterol dan HDL), derajat pembuahan dan derajat penetasan. Hasil penelitian menunjukkan bahwa kelompok perlakuan yang diberikan kombinasi penambahan kurkumin dan hormon tiroksin memiliki nilai konsentrasi vitelogenin (8.17+2.74 µg/mL) (p<0,05), diameter telur (1.43+0.00 mm) (p<0,05), dan konsentrasi trigliserida (4.89+0.53 mg/g) (p<0,05) tertinggi diantara semua perlakuan, sementara itu kelompok perlakuan yang baik hanya diberikan penambahan kurkumin, maupun kombinasi penambahan kurkumin dan tiroksin cenderung memiliki nilai derajat pembuahan dan penetasan yang lebih tinggi dibandingkan dengan kelompok perlakuan yang tidak diberikan penambahan kurkumin.  The main problem in aquaculture is the availability of fish seedlings on time, in the right quantity and quality. The availability of fish seeds is determined by the number of eggs that can be fertilized and hatched. The higher the percentage of fertilization and hatching, the higher the number of larvae that can be produced. This study uses catfish as experimental animals because catfish are one of the most consumed cultivation commodities. The aim of this study is to determine the effect of curcumin and thyroxine supplementation on catfish brooders on the quality and performance of eggs produced. Catfish used in this experiment were given the addition of curcumin and thyroxine hormone through the feed for 12 weeks of rearing. Parameters analyzed were egg vitellogenin concentration, egg diameter, coefficient of diversity in egg diameter, lipid concentration in eggs (triglycerides, cholesterol, and HDL), degree of fertilization and degree of hatching. The results showed that the group given the combination of the supplementation curcumin and the thyroxine hormone had the values of vitellogenin concentration (8.17+2.74 µg/mL) (p <0.05), egg diameter (1.43+0.00 mm) (p <0.05), and triglyceride concentration (4.89+0.53 mg/g) (p <0.05) the highest among all treatments, meanwhile the group is only given the addition of curcumin, and the combination of supplementation of curcumin and thyroxine tends to have higher values of fertilization and hatching compared to the group which is not given the addition of curcumin.

Abstract

This experiment was designed to study the effect of pegagan (Centella asiatica) and turmeric (Curcuma longa) extracts on the activities of glutathione peroxidase (GSH- Px) in liver of white Spraque Dawley rat aged 2 months old with average weight of 200 grams. Pegagan was extracted using infuse method, and turmeric was extracted using dekokta method. The activity of GSH-Px was measured using Flohe and Gunzler’s methods. Extract pegagan and turmeric were assumed to have both preventive and curative treatments. For the preventive treat- ment, extract pegagan and turmeric at concentrations of 22.05 mg/ml : 184.1 mg/ml could increased the activity of GSH-PX enzyme, i.e. 232.60 ± 21.40 mU / mgprotein. This was approximately 79% than the levels of GSH- Px enzyme in normal liver without paracetamol which was 190.78 ± 9.28 mU/mgprotein. For the curative treatment, extract pegagan and tumeric at concentration of 22.05 mg/ml : 184.1 mg/ml could improved GSH- Px enzyme activity (i.e. 239.01 ± 47.40 mU/ mgprotein). This figure was about 92% higher than the levels of GSH-Px enzyme in the liver normal without paracetamol which was 190.78 ± 9.28 mU/mg protein. Pegagan and turmeric extracts with a high concentration could increased the activity of GSH-Px enzyme, thus potential as curative treatment.

Abstract

Nyeri adalah perasaan sensorik atau emosional berupa perasaan tidak nyaman yang berhubungan dengan kerusakan dari suatu jaringan. Obat analgesik adalah obat yang memiliki aktivitas mengurangi rasa nyeri tanpa menghilangkan kesadaran. Penelitian ini bertujuan untuk membandingkan efek analgetik ekstrak etanol 96% dan ekstrak air, menentukan dosis ekstrak daun ungu yang paling efektif dan menentukan jangka waktu efek analgesik ekstrak obat pada mencit putih jantan (Mus musculus L.). Hewan coba  digunakan sebanyak 20 ekor mencit putih jantan yang dibagi menjadi 5 kelompok perlakuan mulai P1 sampai P5. Hasil penelitian menunjukkan ekstrak daun ungu dengan pelarut etanol 96% mempunyai efektivitas analgesik setara dengan kontrol positif. Sediaan paling paling efektif sebagai analgesik adalah Ekstrak Etanol Daun ungu 12 mg/20 g BB dengan nilai rata-rata lama mencit menahan rasa nyeri selama 16,08 detik yang diberikan pada kelompok P3, sedangkan kontrol positif Asetosal 1,82 mg/20 g BB memberikan waktu  rata-rata 17,47 detik  untuk menahan rasa sakit pada mencit.

Abstract

Kurkumin yang terdapat dalam rimpang temulawak, selain dapat menginduksi terjadinya proliferasi sel progenitor pada otak tikus dewasa juga dapat menghambat kerja enzim tirosinkinaseyang berperan penting dalam mengatur pertumbuhan dan diferensiasi sel. Meskipun demikianrespon sel saraf terhadap ekstrak temulawak pada masa pertumbuhan perlu kajian lebih lanjut. Efekekstrak temulawak terhadap proliferasi dan diferensiasi sel otak besar atau serebrum pada masapertumbuhan anak diteliti pada sel otak anak tikus Spague Dawley berumur tiga hari yangditumbuhkan dalam media DMEM (Dulbeccos Modified Eagles Medium). Perlakuan dikelompokkan dalam kelompok kontrol positif (mDMEM+30 g/mL asiaticoside (AC), kontrolnegatif (mDMEM), kelompok yang memperoleh ekstrak temulawak (CZ) 100 ppm (mDMEM+100ppm CZ), CZ 200 ppm (mDMEM+200 ppm CZ), dan CZ 400 ppm (mDMEM+400 ppm CZ). Kultur diinkubasi pada suhu 37 o C dalam inkubator CO 5 % selama 6 hari. Parameter yang diamatiadalah population doubling time, komposisi sel saraf dan sel glia, panjang akson dan dendrite yangdiukur masing masing menggunakan hemositometer, pewarnaan Hematoxyilin Eosin (HE) danmikrometer. Hasil penelitian menunjukkan ekstrak temulawak pada konsentrasi 100 ppmmemperlambat prolfperasi, pada konsentrasi 400 ppm meningkatkan diferensiasi sel yangditunjukkan dengan meningkatnya ratio sel glia terhadap sel saraf dan mempengaruhi panjangakson dan dendrite.Kata kunci : Curcuma xanthorrhiza Roxb., neuron, sel glia, dendrite
