Abstract

The aim of this study was to determine the estrus cycle and the length of estrus in Tragulus javanicuson the basis of its vaginal cytology. Vaginal swabs were collected daily at 7 am for two months. Smears ofthe swab were then prepared on glass slide and they were stained with Giemsa. Vaginal epithelial cells;parabasal, intermediet and superficial cells were counted and their percentages during proestrus, estrusand diestrus were determined. Diestrus was characterized by the absent of superficial cells in the vaginalepithel. Proestrus was characterized by the progressive increase in percentage of intermediet/superficialcells in vaginal epithel, whereas estrus was characterized by the presence of superficial/cornification cellsin most vaginal epithel. On the basis of its vaginal cytology, it was determined that the estrous period offemale Tragulus javanicus was 3 days (48-60 h) and the length was 11 days (10-14 day). The change in thecytology of vaginal epithelial cells appeared to a good marker to determine the estrus cycle phase of theTragulus javanicus

Abstract

Ovarian dynamics in the garut ewes had never been studied continuously by using ultrasonography.The aim of this study was to observe development of the follicles and corpus luteums in the estrous cyclein the garut ewes. Garut ewes (n=6) with body weight 30.00±4.05 kg which had normal estrous cycle wereused in this study. All ewes were synchronized by using CIDR-G implantation for 14 days. Ovulation of thedominant follicle, development of the follicle waves and corpus luteum were observed continuously everyday during the estrous cycle after CIDR-G removal. The number of small (2-3 mm in diameter), medium (4-5 mm in diameter) and large (>5 mm in diameter) follicles were aligned during the estrous cycle. Follicleand corpus luteum diameters were measured by using built in caliper in the ultrasound. The resultsshowed 1) the average length of estrous cycle was 19,2±0,8 days; 2) ovarian follicle growth occurred inthree waves during the estrous cycle; 3) the number of preovulatory follicles were 1-2 follicles; 4) theaverage maximum diameters of preovulatory follicle was 7.5±0.5 mm; 5) the average maximum diametersof corpus luteum was 7.3±0.4 mm. In conclusion, the estrous cycle in garut ewes was 18-20 days with 3follicular waves.

Abstract

Super-ovulation is conventionally performed by injection of FSH twice daily for four days. Thistreatment needs frequent attention by farm-personnel and relatively increases the possibility of failuresdue to mishandling and errors in administration of the treatment. This study was conducted to evaluatethe responses of superovulation trough a single injection of FSH into epidural space in ongole cattlecrossbreed. In Experiment 1, a combination of single dose injection of FSH was applied into epidural spaceplus intramuscular (epi+i.m group) compared to the group of intramuscular injection of FSH, which wastreated twice daily for four days (intramuscular group), using equal total dose of FSH 400 mg. Superovulationresponse of epi+im group (n=4) was not significantly different compared with intramusculargroup (n=4). In experiment 2, it compared two treatmentof FSH in different concentration(280 mg versus160 mg) in a single dose applied into epidural space. Data of Epi+im group from experiment 1 was used ascontrol. Group of 280 mg FSH (n=4) resulted total collection of ova/embryo and transferable embryos(9,00+2,65 and 3,33+2,52 respectively) was significantly different compared to the 160 mg group (n=4)(which were 2,00+1,26 and 0,00) P <0,05, although they werenot significantly different compared to thecontrol (9,33+5,68 and 3,67+3,21). In conclusion, injection of a single dose of FSH at 280 mg into epiduralspace result in a comparable transferable embryo which similar to the conventional method that appliedintramuscular injection of FSH twice daily for four days.

Abstract

A research was conducted to study the ovarian dynamics in kacang goats. Six non pregnant kacang goatswere used in this research. They were clinically healthy, 2 3 years old and showed normal estrous cycles. Theestrous cycles of the kacang goats were synchronized by using prostaglandin 0.5 mg/kg body weight duringluteal phase. The ovarian dynamics were observed daily by using transrectal ultrasonography. Ovarian dynamicswere characterized by follicular wave. The estrous cycle of kacang goats was 20.0±1.1 days. An ovulatoryfirst follicle wave was appeared on days 1-4, the second follicles wave was on days 3-7, the third folliclewave was on days 8-13, while the ovulatory follicular wave emerged on day 17-18. The dominant folliclesreached the maximum diameter in 3.1±0.6 days from wave emergence and the preovulatory dominantfollicle, the diameter was 6.5±0.5 mm. The corpus luteum of kacang goat was observed on 2.0±0.5 daysafter ovulation up to 2.8±0.8 days before the next ovulation. Follicular development consists of three orfour follicular waves and CL developments were static.

Abstract

This study was conducted to isolate and identify the caused bacteria of endometritis in Frisian Holstein (FH) and assessed the effectiveness of nanochitosan against bacterial endometritis in vitro. Sample was collected from six FH cows with subclinical endometritis was examined by ultrasonography, then was calculated the number of colonies, isolation and identification of bacteria. Nanochitosan was calculated the protein content and made into neutral pH with concentrations of 0,5%, 1% and 2%. The results showed that the highest total plate count in the endometrial fluid was founded at number 626 with 4,7 ± 0,6 Log cells/mL and the lowest was found at number 532 with 3,3 ± 0,8 Log cells/mL. Bacteria in the Endometritis fluid was founded Gram-negative bacteria were Escherichia coli, Pseudomonas aeruginosa, and Klebsiella sp. Furthermore, Gram-positive bacteria were founded Staphylococcus aureus, Bacillus sp. and S. pyogenes. The challenge test of nanochitosan 1% and 2% with E. coli showed insignificant results when compared with positive controls (P>0,05). Nanochitosan with all concentrations can reduced number colonies of P. aeruginosa and Bacillus sp. significantly compared with negative controls (P<0,05). The challenge test of nano chitosan 0,5% and 2% were able to reduced the number of S. aureus and Klebsiella sp. colonies but could not reduce of S. pyogenes. These findings showed treatment of nanochitosan as an antibacterial is able to reduce the number of E. coli, Klebsiella sp. and P. aeruginosa and Bacillus sp. and S. aureus

Abstract

Administration of pregnant mare serum gonadotropin (PMSG) hormone prior to mating increasesquality of lamb born on one and two litter sizes, but in three litter size the motality is higher. Administrationof traditional herbal medicine “jamu” consisting of ginger,green chiretta, cinnamon, Zingiberzerumbet, andpepper during gestation increases performance of lambs. This research was conducted to explore theeffectivity of PMSG injection prior to mating and jamu administration during gestation to increase lambsperformance. Eighteen priangan fat-tail ewes with weight around 20-25 kg were injected with PGF2á at adose of 10 mg/ewe twice with 11 days interval to synchronize estrous cycle. Injection of PMSG at a dose of200 IU/ewe was conducted at the second PGF2á injection. The experimental ewes were mated naturally anddivided into a randomized design with a 2 x 3 factorial arrangement with three replications. The firstfactor was dose of PMSG with two levels i.e., 0 and 200 IU/ewe. The second factor was dose of jamu withthree levels i.e., 0, 15, and 30 mL/ewe. Jamu was administered orally every week during gestation. Injectionof PMSG and administration of jamu decreased prenatal mortality, increased ratio of lamb per ewe, increased total lamb born with average birth weight and total birth weight were higher in jamu at a doseof 15 and 30 mL/ewe by 30.02 and 31.76%, repectively. During the first month postnatal, lambs born toewes injected by PMSG and administered jamu had higher number of lambs survive, average weight, totalweight around 3 times as compared to control. It was concluded that injection of PMSG prior to mating andadministration of jamu during pregnancy increase lamb birth weight and improve the quality of lamb.

Abstract

The aim of this study was to determine the optimal mating time in Etawa Crossbred Goat. This studyused 10 ewes, 2-3 years old, with normal estrous cycle. Initiation of estrous using Controlled Internal DrugRelease Implants (CIDR) intra vaginal and released after 12 days. The development of follicles in theovaries wasobserved using transrectal Ultrasonography (USG) every six hours until reach for 66 hoursafter CIDR removal from the vagina. Clinical signs of standing heat and vulva signs were also observedwhile under USG observation. The follicles were divided into three groups based on their diameter:small(<3 mm), medium (4-4.9 mm) and large (>5 mm) which were observed during theestrous phase. Ovulationwas predicted when large follicles (dominant follicles) havenot been seen anymore. The resulta showedthat the average follicles number in ovary after 36 hours CIDR removal were 0-1 (0.8±0.4 mm) small, 0-1(0.9±0.6 mm) medium and 1-2 (1.7±0.8 mm) large, respectively. The large follicles (7-8 mm) wereseen after36 hours CIDR removal or 24 hours after standing heat. Forty two hours after CIDR removal, the largefollicles werenot being seen anymore. It means, the ovulation were predicted in 36-42 hours after CIDRremoval or 24-30 hours after standing heat. The relationship between follicular development and clinical vulva signs (redness, swelling, mucus) revealed that the highest vulva signs (+++) wereseen in 36-42 hoursafter CIDR removal. It was concluded that the ovulation time occurred in 36-42 hours after CIDR removal,with the high intensity of vulva changes. Therefore the optimal mating time in PE 24-36 hours of standingheat or 36-42 hours after CIDR removal.

Abstract

This study was aimed to identify ovarian dynamics using ultrasonography (USG) in simmental crossbreed cows diagnosed prolonged oestrus treatment with human chorionic gonadotrophin (hCG). A total of four Simmental crossbreed cows diagnosed with prolonged oestrous were used in this study. The first oestrous observations in all four cows were a control group, where as the next oestrous cycle observation to the same four cows treated with 1500 IU hCG in jugular vein refer to treated group. The observation of ovarian dynamics was started from the first oestrous day until ovulation occurs in the second oestrus cycle using ultrasonography (USG) with a linear rectal probe. At the same time, follicle and corpus luteum (CL) images were recorded every day. Data were analyzed using paired data comparison test (pair sample t-test). The observation results ovarian dynamics on prolonged oestrus cows showed that the long of oestrous cycle was 22.5 ± 0.5 days, and there are three follicular wave patterns in oestrous cycle. The first wave of dominant follicle was firstly identified on day 3.3 ± 0.5, reached maximum diameter 12.0 ± 0.3 cm. The second and third dominant follicle appeared on day 10.8 ± 0.5 and 15.8 ± 0.5 cm reached maximum diameter 1.2 ± 0.3 cm dan 1.5 ± 0.3 cm. Therapy with hCG does not significantly affect the dominant follicle’s diameter during oestrous (p>0.05). The conclusion of this study showed that 1500 IU of hCG therapy does not affect the development of dominant follicle, however, it can shorten the duration of oestrous and ovulation time.
