Abstract

The aim of this study was to identify the antifungal activity of coumarin isolated from Ageratum conyzoides L. leaves and to observe its influence on Candida albicans cells by scanning electron microscope (SEM) and transmission electron microscope (TEM). Antifungal activity testing with the disk diffusion method showed coumarin was active toward pathogenic fungus Candida albicans with an MIC value of coumarin of 125 g mL-1. The results show that this compound damaged the cell by pores formation on the cell wall. Death of cells occurred due to leakage and necrosis of cytoplasmic content.

Abstract

Quercetin (3,3'4',5,7-pentahydroxyflavone) has been reported to show anti-inflammatory activity. However, its low oral bioavailability limits the application of quercetin in therapy. Ester derivatives of quercetin have been reported to have higher bioavailability than quercetin. This research aimed to study the anti-inflammatory activities and gastric ulcer-inducing properties of tetraacetylquercetinas well as tetrapivaloylquercetin. Synthesis of tetra-acyl derivatives of quercetin was conducted using acetic anhydride or pivaloyl chloride in the presence of pyridine and the structure was confirmed by 1H-NMR and 13C-NMR spectroscopy as well as elemental analysis. At a dose of 20 mg/kg bw, oral administration of quercetin only showed 20% inhibition activity on carragenan induced rat paw edema, while tetraacetyl and tetrapivaloyl derivatives at equimolar dose showed 11-33% and 5-15% inhibition activity respectively. Contrary to the gastric ulcer healing-promoting action of quercetin, tetraacetylquercetin caused mild gastric ulcers. However, no gastric ulcer was observed after administration of tetrapivaloylquercetin. It was concluded that acylation enhances the anti-inflammatory activity of quercetin but causes mild gastric ulcers in the case of tetraacetylation.

Abstract

Natural ingredients can have extraordinary potential as alternative medicines due to their accessibility and cost-effectiveness. Application of these ingredients should consider solubility and permeability, which determine the success of pharmaceutical characteristics formulation and biological activity indication. In this context, physical manipulation, specifically particle size reduction, is an effective strategy to address these issues. Previous research has explored active compounds in the stilbenoid group, found in the outer skin, hard shell, and endosperm of melinjo (Gnetum gnemon L.) seeds, functioning as antioxidant. Based on the potential as antioxidant, stilbenoid compounds, including resveratrol, contained in melinjo seed hardshell have shown significant pharmacological effects. Therefore, this research aimed to investigate the potential of melinjo seed hardshell extract as a natural antioxidant alternative by modifying the particle size through a grinding process to obtain nanoparticles. The analysis was carried out using ball milling to enhance the solubility of melinjo seed hardshell extract by increasing the saturated solubility and surface area of the particles. The results showed that the total phenol content and the antioxidant power increased significantly (p < 0.05) after ball milling. Melinjo seed hardshell nanoextract is reported herein as a promising source of natural antioxidant from local Indonesian plants.
