Abstract

Background: Plasmodium falciparum Erythrocyte Membrane Protein 1 (PfEMP1) is an important protein responsible for the pathogenesis of severe malaria, including cerebral malaria. The protein is highly diverse. The CIDR1α-PfEMP1 binds endothelial protein receptor (EPCR) and may associated with the brain swelling in childhood malaria.Objective: To analyze the CIDR1α-PfEMP1 from Indonesian isolate and determine its association with cerebral malaria outcome.Methods: Fifteen blood samples of clinically mild to severe malaria-patient were collected for DNA extraction. Malaria diagnosis was conducted microscopically by Giemsa-stained thin blood smear. The CIDR1α domain was amplified by PCR using specific primer and PCR product was sequenced. The nucleotide sequences were analyzed by NCBI blast, DNASIS MAX 3 and translated into amino acid sequences using Expasy Translation Tool.Results: One out of fifteen samples was severe malaria case and infected with P.  falciparum, the rest were clinically mild to moderate malaria and infected with pure P. falciparum or mixed infection of P. falciparum and P. vivax. Amplification for CIDR1α domain resulted a single band of + 550 bp from a severe sample only. Sequencing of PCR product on both strands read 524 nucleotides and BLAST analysis confirmed as CIDR1α sequence. Multiple alignment showed 74-78% nucleotide sequence similarity with reference sequences, but amino acid sequences presented 23.5% homologous.Conclusion: An identified CIDR1α domain only from severe case implicating the potential association with the severe outcome including cerebral malaria, but the highly diverse of the domain needs further studies on the interaction with the pathological-causing receptor in the host.

Abstract

Konektivitas yang dibangun negara, baik dalam lingkup bilateral, regional, maupun global berkelindan satu sama lain. Misalnya, kerja sama subregional berbasis konektivitas lintas batas yang tengah dibangun dalam kerangka IMT-GT, BIMP-EAGA, dan TIA-GT akan mendorong dan menguatkan sebuah kerja sama regional, seperti ASEAN Community, sesuai dengan Master Plan on ASEAN Connectivity. Studi ini difokuskan pada tiga hal, yakni  pertama, capaian dan tantangan pembangunan konektivitas lintas batas (fisik, kelembagaan, dan antarmasyarakat) melalui kerja sama ekonomi subregional; kedua, strategi penguatan konektivitas antarwilayah pertumbuhan subregional untuk mendukung konektivitas ASEAN; ketiga, strategi pemanfaatan konektivitas ASEAN berbasis kerja sama ekonomi subregional bagi Indonesia. Dengan menggunakan metode penelitian kualitiatif yang bersifat eksploratif analitis, studi ini menemukan bahwa keterlibatan Indonesia dalam beberapa kerja sama ekonomi subregional membawa dampak positif bagi Indonesia, baik secara ekonomi, sosial budaya, maupun pertahanan keamanan. Namun, capaian dari beberapa kerja sama subregional ini lebih terlihat di kawasan Indonesia bagian barat dan tengah dibandingkan bagian timur. Tantangan ini yang harus dapat diselesaikan oleh pemerintah Indonesia untuk dapat mengoptimalkan pemanfaatan kerja sama subregional di semua kawasan di Indonesia dengan mendorong peran aktif pemerintah-pemerintah daerah untuk terlibat dalam kerja sama subregional yang melibatkan daerahnya. Kata kunci: konektivitas, lintas batas, subregional, IMT-GT, BIMP-EAGA, TIA-GT

Abstract

Aspartame is an artificial sweetener in foods and beverages. Continuous use of aspartame will result in oxidative stress in the liver. So we need antioxidants from outside the body such as periwinkle leaves.This study aims to determine the effect of periwinkle leaves (Catharanthus roseus L) on aspartame-induced liver histology. This study used 24 male mice divided into 4 groups, namely negative control (K-) without treatment; Positive control (K +) treatment of aspartame; D1: treatment of aspartame and periwinkle extract doses of 0.42 mg/kg. D2: aspartame and periwinkle  extract doses of 0.84 mg/kgbb. Administration of aspartame dose of 0.2 mg /kgbb by intraperitoneal  for 14 days followed by gavage administration of periwinkle leaf extract for 12 days. The liver was taken on the 27th day by surgery, then made slide histology with Hematoxylin-Eosin staining. The results show that the administration of Catharanthus roseus leaf extract at a dose of 0.84 mg/kgbw in aspartame-induced mice was effective in reducing hepatocyte damage, namely hydrophilic degeneration and necrosis.

Abstract

ABSTRACTDiazinon has a hepatotoxic effect since it is metabolized in the liver involving Kupffer cells (KCs) activation, increasing reactive oxygen species (ROS). The flavonoids contained in shallot (Allium cepa L.) skin act as antioxidants neutralizing oxidative stress. This study aims to determine the effect of shallot skin ethanol extract (SSEE) on liver MDA level and activated KCs histopathology. The total flavonoids level of SSEE was measured using the aluminum chloride colorimetric method and resulted in 228.1 mg QE/g. Rats were divided into normal, diazinon, and SSEE groups. Diazinon was administered at a dose of 40 mg/kg b.w. for 7 days, followed by SSEE at the dose of 600, 900, and 1,200 mg/kg b.w. for 7 days. Liver malondialdehyde (MDA) level was measured using the MDA-TBA method. The results revealed that diazinon increased liver MDA level (p <0.05), while SSEE at doses of 900 and 1,200 mg/kg b.w. decreased liver MDA level equal to normal (p>0.05). Activated KCs in the SSEE group at a dose of 1,200 mg/kg b.w. was impressively equal to the normal group. In conclusion, SSEE at dose of 1,200 mg/kg b.w. neutralize liver oxidative stress due to diazinon indicated by the decrease of liver MDA level and activated KCs equal to normal.Keywords: antioxidant, diazinon, Kupffer cell, liver, MDA, shallotCorrespondence: rosita.fk@unej.ac.id

Abstract

This research aimed to investigate the chlorine contents in rice circulation in Pasar Besar of which findings were supposed to be applicable as Biological learning resource. This research was officially conducted at March 29 – April 6 2016 in Putra Indonesia Malang Laboratory. This research was of descriptively quantitative design since the main aim of this research was to describe the chlorine contents in rice. The sampling technique engaging regarding this research was purposive sampling with two repetitions in total. The sample of rice that was going to be analyzed was originally from Pasar Besar comprised 19 types (considering the brands), they were: Piala, Mentari, Padi Wangi, Bintang Biru, Rosita Super, Kancil, Bintang Mas, Bulan Mas, Puteri,  Lombok, Maknyus, Apel, Manggis, Mangga, Cucak Rowo, Beras Padi, Raja Boga dan Monas. In addition, the methods accommodated in terms of testing the reaction included testing the color reaction test, testing the iodometric titration, and measuring the average percentage of chlorine contents in the whole sample.  Moreover, this research revealed that the 19 types sample of rice taken from Pasar Besar is 0 % or negative of chlorine contents. Consequently, they were safe to consume.

Abstract

        The purpose of this research is to analized the influence of organizational commitment to job satisfaction of public accountants with a role stress as moderating variabel. The samples of this research are 54 public accountants that worked at public accountants office which registered in Jakarta. The data were analized by multiple regression method with moderating variabel.        The result of this analysis of individual test show that role conflict is not moderated of relation by organizational commitment with job satisfaction. Also with role ambiguity which not moderated of relation by organizational commitment with job satis¬faction. The main practical implication of this research is that the public accountants organization must be attention to jobs expectation of their auditors. Because that is the factors that rising the organizational commitment and in the future it will be to give a high job satisfaction.Keywords: organizational commitment, role stress, role conflict, role ambiguity, job satisfaction.

Abstract

The niacin in energy drinks has metabolic product that cause oxidative stress and liver damage, while the liver damage can be prevented by hepatoprotective agents. Scoparone in Artemisia vulgaris L. can act as a hepatoprotector by its antioxidant effect. This study aimed to investigate the effectivity of Artemisia vulgaris L. extract as a hepatoprotector in wistar hepatocytes induced by niacin. This study used 25 male rats which were divided into 5 groups: normal, the negative control, the positive control, the treatment group I, and II. Treatment was conducted for 28 days. The samples were terminated and the hepatocyte were prepared for histological examination. Histological appearance was catagorized as mild, moderate, and severe damage with or without inflamatory cells activity. The data analysis by Kruskal Wallis showed significant difference (p<0,001). Further analysis by Mann Whitney revealed significantly difference (p<0,05) between normal group and all groups, negative control group and positive control group, and positive control group and treatment group I, but not significantly difference between negative control group and treatment group I, negative control group and treatment group II, positive control group and treatment group II, and between treatment groups. The study concluded that the effectivity of Artemisia vulgaris L. extract has not been proven as a hepatoprotector but further study is needed to draw a definite conclusion. Abstract

DMBA (7,12-Dimethylbenz(α)antracene) is one of Polycyclic Aromatic Hydrocarbon’s form that is abundant on air pollution, i.e. automobile exhaust, cigarette smoke, and wood stoves. DMBA is metabolized in liver result in DMBA-DE. DMBA-DE is a free radical compound that has hepatotoxic effect. Broccoli contains flavonoid that has antioxidant effect. It can stabilize DMBA-DE radical compound. This research was conducted to investigate the hepatoprotective effect of ethanolic extract of broccoli on the damage of rat’s liver cells induced by DMBA. This research used 30 white male rats that was divided into 6 groups: normal control (aquadest), positive control (DMBA) and the treatment groups(using brocolli ethanolic extract with dose : 250, 500, 1000, and 2000 mg/kgBB). The treatment was given for 7 days followed by single-dose DMBA 15 mg/kgBB induction in the 8th day. Histological appearance of liver cells were examined on the 12th day. Data were analyzed by using Kruskal Wallis test followed by Mann Whitney. The result showed that there was a significant difference among the groups with p<0.05. In conclusion, there were some differences of hepatoprotective effect of broccoli ethanolic extract on the histologically damage of rats liver cells induced by DMBA. Keywords: ethanolic extract of broccoli, DMBA, histopathology, hepatoprotective

Abstract

The number of cesarean section in Indonesia is 9,8% and approaching cesarean limit suggested by WHO which is 10%. The most common reason for cesarean section is prior cesarean. Cesarean section has higher maternal mortality risk compared to vaginal delivery. Vaginal delivery with prior cesarean has higher complication risk compared to cesarean section. Maternal complications and neonatal outcomes are important to decide delivery methods. Maternal complications that can be compared are postpartum infection, postpartum hemorrhage, and uterine rupture. Neonatal outcomes that can be compared are APGAR score and neonatal birth injury. This study aimed to compare maternal complications and neonatal outcomes between vaginal delivery and cesarean section in women with prior cesarean section. There was a significant difference in the incidence of postpartum infection (p=0.01) and neonatal birth injury (p=0.05). There was no significant difference in APGAR score and the incidence of postpartum hemorrhage. There was no uterine rupture observed. In conclusion, vaginal delivery is a viable option for pregnant woman with prior cesarean.

Abstract

Demensia merupakan penyakit pada lansia yang bermanifestasi terhadap kemunduran kemampuan intelektual. Di Indonesia, prevalensi demensia pada lansia berusia 65 tahun adalah 5 persen dari populasi lansia. Prevalensi meningkat menjadi 20 persen dari lansia yang berusia 85 tahun ke atas. Kategori lansia berusia 65 tahun ke atas di Indonesia pada tahun 2003, sebanyak 11.280.000. Jumlah ini diperkirakan akan meningkat menjadi 29 juta pada tahun 2013 atau 10 persen dari populasi. Ada beberapa faktor yang mempengaruhi penurunan memori salah satunya adalah aktivitas fisik . Aktivitas fisik secara teratur telah terbukti dapat mengurangi risiko demensia, termasuk penyakit Alzheimer sebesar 50 %. Penelitian ini menggunakan jenis penelitian analitik observasional dengan desain cross sectional. Variabel yang diamati adalah tingkat demensia pada lansia dan aktivitas fisik yang dilakukannya. Responden kemudian diukur menggunakan instrumen kuisioner MMSE (Mini Mental State Examination). Populasi dalam penelitian ini adalah semua lansia yang tinggal di UPT Pelayanan Sosial Lanjut Usia Jember sebanyak 43 responden. Hasil penelitian dengan uji Spearman Rho menunjukkan bahwa nilai signifikansi = 0,00 (p 0,05 > 0,000). Terdapat hubungan yang bermakna antara aktivitas fisik dengan kejadian demensia pada lansia di UPT Pelayanan Sosial Lanjut Usia Jember. Kata kunci: aktivitas fisik, demensia.
