Abstract

This study was conducted in response to low student learning outcomes on the topic of the Social Impact of Informatics, which was attributed to the dominance of lecture-based teaching methods that limited student independence and engagement. The research aimed to evaluate the effectiveness of the Self-Directed Learning (SDL) model in enhancing the learning outcomes of seventh-grade students at SMP Negeri 5 Samarinda. Using a pre-experimental one-group pretest-posttest design, the study involved 24 students. Data were collected through pretest and posttest assessments, observation sheets, and student response questionnaires. The results showed a notable improvement in learning outcomes, with the average pretest score of 70 increasing to 89.17 in the posttest. N-Gain analysis produced a score of 0.63, indicating a moderately effective learning gain. In addition, student responses to the SDL model were highly positive, with an average questionnaire score of 81.29%, demonstrating strong acceptance and enthusiasm for independent learning approaches.

Abstract

Several approaches have been designed to control malaria, a disease with high morbidity and mortality, but they face some hurdles. Antimalarial resistance is one of the major challenges for malaria elimination, so the detection of antimalarial resistance is essential. Several molecular markers for antimalarial resistance have been identified, including Plasmodium falciparum multidrug resistance 1 (pfmdr1) gene. This study determined the optimization of molecular techniques to identify the pfmdr1 gene as an antimalarial resistance predictor in Indonesia. The study included patients diagnosed with uncomplicated or severe malaria originating from the health district of Kerom Regency, Papua Province, and Dr. Soebandi Hospital, Jember, East Java Province. Blood samples were collected in the Whatmann filer paper after informed consent. DNA was isolated from dried blood filter paper, and nested PCR was performed using a specific primer, the pfmdr1-A and pfmdr1-B genes. The PCR cycle was optimized based on previous studies. The pfmdr1-A has a similar setting to the earlier study, but the pfmdr1-B had a different optimum setting from the previous study with the annealing temperature of 57oC for nested-1 and 62oC for nested-2. This PCR setting could be used for further examination. The positive results of the amplification indicated the potential for antimalarial resistance in the parasite population. A study on the gene copy number and polymorphism is essential to determine the definitive conclusion on antimalarial resistance.

Abstract

Malaria masih menjadi masalah kesehatan dunia terutama di negara-negara berkembang. Data World Malaria Report 2020, kasus malaria di dunia tahun 2019 mencapai 229 juta kasus dengan total kematian mencapai 409.000 jiwa. Kematian tertinggi terjadi pada pasien usia balita yang mencapai 272.000 jiwa.  Angka kematian akibat malaria berat di dunia sangat tinggi sehingga perlu berbagai pendekatan penanganan termasuk vaksin malaria. Domain Cysteine-rich Interdomain Regions 1α-Plasmodium falciparum Erythrocyte Membrane Protein 1 (CIDR1α-PfEMP1) merupakan kandidat vaksin malaria karena berperan penting dalam pathogenesis malaria. Pengembangan vaksin malaria berbasis CIDR1α-PfEMP1 dilakukan melalui teknologi rekombinan DNA dimana tahapan yang sangat penting adalah transformasi. Tujuan penelitian ini untuk mengoptimasi kondisi transformasi gen CIDR1α-PfEMP1 pada Escherichia coli BL21(DE3) menggunakan metode heat shock guna mendapatkan efisiensi transformasi tertinggi. Variable yang dianalisis adalah durasi heat shock selama 30, 50, dan 70 detik dan volume plating 100 µL dan 200 µL. Hasil penelitian menunjukkan efisiensi transformasi tertinggi sebesar 1,9 x 102 CFU/µg dicapai pada kondisi heat shock selama 50 detik dan volume plating 200 µL. Ini adalah kondisi optimum transfomasi gen CIDR1α-PfEMP1 yang dapat digunakan untuk penelitian lanjutan guna mengembangkan vaksin malaria.

Abstract

Animated cartoons are media that contain a collection of images that are processed in such a way as to move and are equipped with audio so that they are memorable and store learning messages. The purpose of this study was to determine the effect of counseling using animated cartoon media on dental and oral health knowledge. Based on the results of the initial data examination conducted on 15 students, the results obtained were 2 students with good knowledge, 2 students with moderate knowledge, and 11 students with less knowledge. This research method uses a Quasi Experiment One Group Pre test-Post test design. The population in this study were all fifth-grade students of SDN 2 Lamcot Aceh Besar totaling 30 respondents. The sampling technique used was the Total Sampling technique. This research was conducted on 12-18 May 2022. Data collection using a questionnaire. This study used the SPSS Paired Sample T-test and analyzed univariately and bivariate. Based on the results of the study, there were differences in the mean before and after the intervention with a difference in the mean value of 40.1. The results of statistical tests obtained a significance value of p = 0.000 (p = <0.05). The results of this analysis indicate that there is a significant difference between before and after counseling using animated cartoon media. It can be concluded that in this study there was an effect of counseling using animated cartoon media on dental and oral health knowledge of fifth-grade students at SDN 2 Lamcot Aceh Besar. It is recommended to teachers and health workers at the local health center use animated cartoon media in increasing students' dental and oral health knowledge.

Abstract

Penggunaan diazinon dalam membasmi hama tanaman akan meninggalkan residu pada tanaman sehingga jika dikonsumsi memberikan dampak buruk. Metabolisme diazinon di hati meningkatkan jumlah radikal bebas dan menurunkan aktivitas enzim antioksidan yang akan mengakibatkan kerusakan sel hati. Kulit bawang merah (Allium cepa L.) mengandung kuersetin yang dapat memperbaiki kerusakan jaringan akibat stres oksidatif. Penelitian ini bertujuan mengetahui hubungan antara dosis infusa kulit bawang merah (IKBM) dan tingkat kerusakan hati tikus yang diinduksi diazinon dan menentukan dosis efektif maksimum IKBM. Tikus dibagi menjadi kelompok normal, diazinon, dan IKBM. Diazinon diberikan dengan dosis 40 mg/kgBB selama 7 hari dilanjutkan dengan IKBM dengan dosis 125, 250, 500, 1.000, dan 2.000 mg/kgBB selama 7 hari. Pengamatan mikroskopik dilakukan terhadap 100 sel hati dengan pewarnaan hematoxylin-eosin (HE) berdasarkan skor Manja Roenigk. Tingkat kerusakan hati antara dua kelompok dianalisis menggunakan uji Mann-Whitney, sedangkan korelasi antara dosis IKBM dan tingkat kerusakan hati, dan dosis efektif maksimum IKBM ditentukan dengan uji regresi. Rerata skor histopatologi hati tertinggi didapatkan pada kelompok diazinon dan terendah pada kelompok normal (p<0,05). Uji regresi menunjukkan terdapat hubungan eksponensial antara dosis IKBM dan tingkat kerusakan hati (p<0,05), yaitu semakin tinggi dosis IKBM, semakin rendah skor histopatologi hati dan dosis efektif maksimum IKBM adalah 1.347,28 mg/kgBB. Sebagai kesimpulan, semakin tinggi dosis IKBM, semakin rendah tingkat kerusakan hati tikus yang diinduksi diazinon. Pemberian IKBM melebihi dosis 1.347,28 mg/kgBB akan meningkatkan kerusakan hati. Abstract

Transmigration is the program which cannot be separated with the success story of palm oil development in Indonesia. Transmigrants becomes the engine to build the palm oil smallholder. The involvement of transmigrant in the PIR-Trans program, primary credit for member Scheme (KKPA), and revitalization scheme has been accelerating the palm oil plantation development in Indonesia. However, the “success” of developing palm oil plantation in Indonesia through scheme mentioned above was not without flaws. Unclear of land status or overlapping claim become the biggest problems faced nowadays. This paper aims to observe the development transmigration program and palm oil development in Indonesia. By analyzing policies of both transmigration programs and palm oil development from different political administration, this paper shows that, first, transmigration policy went from narrative of security, economic growth to border development and security. Second, under those different policy narratives, transmigration policy has been accommodated to the development of palm oil in Indonesia. Transfer of central authority to local government in executing transmigration movement and settlement has been addition to local government operational permission authority for palm oil development.

Abstract

Background: Cigarette smoking damages the alveoli through oxidative stress. Shallot peels containing flavonoids, especially quercetin, potentially serve as a daily antioxidant supplement to impede lung tissue damage induced by cigarette smoke. However, the maximum effective dose is yet to be determined. Objective: This research was designed to establish the maximum effective dose of shallot peel infusion (SPI) to prevent oxidative stress and histopathological lung damage induced by cigarette smoke. Methods: This experimental laboratory was a posttest-only control group design. A total of 24 male Rattus norvegicus Wistar strain, were allocated into 6 groups: a control group and 5 SPI-treated groups. All rats were exposed to 2 cigarettes/day and were treated for 28 days with aquabidest and different doses of SPI (0 mg/kgBW, 125 mg/kgBW, 250 mg/kgBW, 500 mg/kgBW, 1,000 mg/kgBW, and 2,000 mg/kgBW). The level of oxidative stress in serum was measured malondialdehyde (MDA) level with ELISA, and histopathological lung damage was estimated using the lung histopathological damage scoring method assessing inflammatory cells, alveolus lumen and inter-alveoli junction.Results: The quadratic regression analysis revealed the maximum effective dose of SPI to prevent oxidative stress and lung damage were 1,435 mg/kgBW and 1,206 mg/kgBW, respectively. In the histopathological examination of the lungs, the administration of SPI up to a dose of 1206 mg/kg BW prevents the inflammatory process caused by cigarette smoke, which is indicated by the number of inflammatory cells, the thickness of the alveolar septum, and the increasingly normal shape of the alveolar lumen.Conclusion: SPI doses of less than 1,206 mg/kgBW are safe and effective daily antioxidant supplements in rats exposed to cigarette smoke and have the potential to be further studied for application in humans.

Abstract

Shallot (Allium cepa L.) contains bioactive compounds with antioxidant effects. In addition to the tuber, the shallot’s peel is a rich source of flavonoids with demonstrated capacity to mitigate oxidative stress. Prior studies have evaluated the antioxidant efficacy of shallot peel in ameliorating organ damage and have established its maximum effective dose. The subsequent step required for its potential therapeutic application is toxicity test. This study aimed to analyze the liver histopathological changes in the acute toxicity test of shallot peel extract (SPE) in rats based on OECD TG 420. This research was an experimental with a post-test-only control group design. Five female rats were used as the control group administered dimethyl sulfoxide (DMSO), while 5 female rats were used in the treatment group administered SPE at 5,000 mg/kg body weight (BW) (one of them had been used for a preliminary test with the same dosage). On day 15, a necropsy was conducted, followed by histopathological observation of the hematoxylin-eosin (HE)-stained liver histopathological slide. The damage to hepatocytes was evaluated using Manja Roenigk criteria. The average histopathological score per liver cell of the control group was 1.226 ± 0.0065 and the treatment group was 1.235±0.0079. The Mann-Whitney test showed that the liver histopathological score of the treatment group was not different from that of the control group (p>0.05). It can be concluded that SPE 5,000 mg/kg does not show acute toxic effects in rats, with LD₅₀ estimated at >5,000 mg/kg according to OECD standards.

Abstract

Legal assistance for applications is a crucial form of legal aid provided to the public in submitting various types of applications to the courts or related agencies. This research is motivated by the urgent need to understand the effectiveness and implementation of such legal aid services, particularly for underprivileged communities who face barriers in accessing the justice system. This article aims to discuss and describe in detail the application processing process carried out by the Legal Aid Post (Posbakum) when operating and collaborating with the Legundi Legal Aid Institute (LBH Legundi), specifically in the context of name change applications. This research uses normative legal methods with a conceptual and statutory approach to analyze the applicable procedures. The results show that Posbakum plays a central role in drafting name applications for communities in need through a phased assistance process that includes initial consultation, verification of document requirements, technical assistance in scanning and nazegel, drafting application letters, and guidance on case registration. This assistance model has proven effective in overcoming procedural obstacles and increasing access to justice for underprivileged communities, while simultaneously transforming the community from a passive to an active role in fighting for their civil rights.

Abstract

This study aims to analyze the service quality of the SIAKAD website at Universitas Dinamika Bangsa Jambi using the WebQual 4.0 method. The issues raised include the impact of access speed, the SIAKAD system on user expectations, the completeness and accuracy of data, and an overall analysis of the SIAKAD service quality. The study uses three main variables: Information Quality, Interaction Quality, and Usability Quality, with 311 respondents categorized by gender, age, and academic program cohort. The results show that Information Quality and Interaction Quality have a significant impact on user satisfaction, while Usability Quality does not have a significant effect. An R-Square value of 85% indicates that the service quality of SIAKAD at Universitas Dinamika Bangsa Jambi is good and positively influences user satisfaction. This study concludes that users are satisfied with the quality of the SIAKAD website services, except in terms of Usability.
