Abstract

Background: Cigarette smoking damages the alveoli through oxidative stress. Shallot peels containing flavonoids, especially quercetin, potentially serve as a daily antioxidant supplement to impede lung tissue damage induced by cigarette smoke. However, the maximum effective dose is yet to be determined. Objective: This research was designed to establish the maximum effective dose of shallot peel infusion (SPI) to prevent oxidative stress and histopathological lung damage induced by cigarette smoke. Methods: This experimental laboratory was a posttest-only control group design. A total of 24 male Rattus norvegicus Wistar strain, were allocated into 6 groups: a control group and 5 SPI-treated groups. All rats were exposed to 2 cigarettes/day and were treated for 28 days with aquabidest and different doses of SPI (0 mg/kgBW, 125 mg/kgBW, 250 mg/kgBW, 500 mg/kgBW, 1,000 mg/kgBW, and 2,000 mg/kgBW). The level of oxidative stress in serum was measured malondialdehyde (MDA) level with ELISA, and histopathological lung damage was estimated using the lung histopathological damage scoring method assessing inflammatory cells, alveolus lumen and inter-alveoli junction.Results: The quadratic regression analysis revealed the maximum effective dose of SPI to prevent oxidative stress and lung damage were 1,435 mg/kgBW and 1,206 mg/kgBW, respectively. In the histopathological examination of the lungs, the administration of SPI up to a dose of 1206 mg/kg BW prevents the inflammatory process caused by cigarette smoke, which is indicated by the number of inflammatory cells, the thickness of the alveolar septum, and the increasingly normal shape of the alveolar lumen.Conclusion: SPI doses of less than 1,206 mg/kgBW are safe and effective daily antioxidant supplements in rats exposed to cigarette smoke and have the potential to be further studied for application in humans.

Abstract

Shallot (Allium cepa L.) contains bioactive compounds with antioxidant effects. In addition to the tuber, the shallot’s peel is a rich source of flavonoids with demonstrated capacity to mitigate oxidative stress. Prior studies have evaluated the antioxidant efficacy of shallot peel in ameliorating organ damage and have established its maximum effective dose. The subsequent step required for its potential therapeutic application is toxicity test. This study aimed to analyze the liver histopathological changes in the acute toxicity test of shallot peel extract (SPE) in rats based on OECD TG 420. This research was an experimental with a post-test-only control group design. Five female rats were used as the control group administered dimethyl sulfoxide (DMSO), while 5 female rats were used in the treatment group administered SPE at 5,000 mg/kg body weight (BW) (one of them had been used for a preliminary test with the same dosage). On day 15, a necropsy was conducted, followed by histopathological observation of the hematoxylin-eosin (HE)-stained liver histopathological slide. The damage to hepatocytes was evaluated using Manja Roenigk criteria. The average histopathological score per liver cell of the control group was 1.226 ± 0.0065 and the treatment group was 1.235±0.0079. The Mann-Whitney test showed that the liver histopathological score of the treatment group was not different from that of the control group (p>0.05). It can be concluded that SPE 5,000 mg/kg does not show acute toxic effects in rats, with LD₅₀ estimated at >5,000 mg/kg according to OECD standards.

Abstract

Legal assistance for applications is a crucial form of legal aid provided to the public in submitting various types of applications to the courts or related agencies. This research is motivated by the urgent need to understand the effectiveness and implementation of such legal aid services, particularly for underprivileged communities who face barriers in accessing the justice system. This article aims to discuss and describe in detail the application processing process carried out by the Legal Aid Post (Posbakum) when operating and collaborating with the Legundi Legal Aid Institute (LBH Legundi), specifically in the context of name change applications. This research uses normative legal methods with a conceptual and statutory approach to analyze the applicable procedures. The results show that Posbakum plays a central role in drafting name applications for communities in need through a phased assistance process that includes initial consultation, verification of document requirements, technical assistance in scanning and nazegel, drafting application letters, and guidance on case registration. This assistance model has proven effective in overcoming procedural obstacles and increasing access to justice for underprivileged communities, while simultaneously transforming the community from a passive to an active role in fighting for their civil rights.

Abstract

This study aims to analyze the service quality of the SIAKAD website at Universitas Dinamika Bangsa Jambi using the WebQual 4.0 method. The issues raised include the impact of access speed, the SIAKAD system on user expectations, the completeness and accuracy of data, and an overall analysis of the SIAKAD service quality. The study uses three main variables: Information Quality, Interaction Quality, and Usability Quality, with 311 respondents categorized by gender, age, and academic program cohort. The results show that Information Quality and Interaction Quality have a significant impact on user satisfaction, while Usability Quality does not have a significant effect. An R-Square value of 85% indicates that the service quality of SIAKAD at Universitas Dinamika Bangsa Jambi is good and positively influences user satisfaction. This study concludes that users are satisfied with the quality of the SIAKAD website services, except in terms of Usability.

Abstract

Betawi’s taffy made of sticky rice was one of the best economic commodities for sub-district Cibitung’s citizens. Furthermore, this product usually gave success in a yearly exhibition for district Bekasi. For a long time, demand a trust the costumers were longer completed through the serving of nutrient food and the hygiene concept. That became a challenge for the owner and the Sukajaya village’s leader to decrease the resellers. Afterward, the breakthrough way guided selling the product into the food and the gift store in the scale of the home industry, which attached the label below monitoring the Service of Healthy (P-IRT). Achieve that, the initial step has been done that supported the guiding in the Sukajaya village and LPPM Unsika. The next activity for the site work evaluation also has been executed. It is such as providing a base for raw material until product and cover for the roof in around production line, giving the name for each section place, and pin the license certificate. Neat and clean aspects gave an esthetic point. Furthermore, it positively affected the occupation program of Sukaya village in the district Bekasi level.

Abstract

Introduction: Shallots (Allium cepa L.) are known to contain quercetin, which is helpful as an antioxidant, but its accumulation in the body can be toxic. Objective: This study aims to determine the effect of administering red onion skin extract on histopathology and weight of the kidney, body weight, and clinical symptoms, as well as determining the LD50. Material & Methods: This research was conducted according to OECD guideline No. 420 Fixed Dose Procedures. Rats were allocated to a control group that was given 3% DMSO and a treatment group that was given red onion peel extract at a dose of 5000 mg/kgBW. Kidney histopathology slides with Hematoxylin-eosin staining were observed by two observers in five fields of view using Fiji ImageJ software and assessed based on the Kocoglu scoring. Kidney histopathology data was analyzed using the Mann-Whitney test, while kidney weight and body weight gain used the Independent Sample T-test. Results: The treatment group experienced significantly higher body weight gain than the control group. On the other hand, observations of the histopathological appearance and weight of the kidney did not show any significant differences. Observation of clinical symptoms showed normal results with no deaths. Conclusion: Administration of 5000 mg/kgBW red onion skin extract in the acute toxicity test had no effect on the histopathology and weight of kidney rats, but had a significant effect on body weight gain. The LD50 of red onion skin extract was found to be >5000 mg/kgBW. Keywords: Red onion skin extract, acute toxicity test, LD50, kidney histopathology.

Abstract

Malaria is a major infectious disease worldwide, and vaccination is essential for disease control. The Plasmodium falciparum Erythrocyte Membrane Protein-1 (PfEMP1) is a potential malaria vaccine candidate due to its involvement in pathogenesis. Injection of DBL2β-PfEMP1 recombinant protein in animal models induces IgG and CD4+ production and inhibits binding with host endothelial receptors. This study aimed to analyze the spleen immune response by measuring the white pulp diameter. The experimental study used Wistar rats (Rattus norvegicus) that were divided into four groups, a control group and three treatment groups, which were injected with 100, 150, and 200 μg of DBL2β-PfEMP1 protein. Injection was done thrice with three-week intervals (days 0, 21, and 42). On day 56, rats were euthanized, and spleens were prepared for histology examination. The white pulp diameter increased along with increasing the dose of protein. The ANOVA test showed a significant difference between groups (p=0.001). The posthoc Bonferroni test showed a significant difference between the control and the 150 and 200 μg groups and the 100 μg and 200 μg groups. In conclusion, the DBL2β-PfEMP1 recombinant protein injection increased the spleen white pulp diameter in Wistar rats, and the 200 μg dose resulted in the highest increase.

Abstract

Abstract: A malaria vaccine based on Plasmodium falciparum antigen is essential in controlling malaria morbidity and mortality. The Duffy-binding-like domain 2β of Plasmodium falciparum erythrocyte membrane protein 1 (DBL2β-PfEMP1) is a malaria subunit vaccine candidate expected to prevent severe malaria. Meanwhile, subunit vaccines often elicit a weak immune response. This study aimed to analyze the CD4 response induced by several doses of DBL2β-PfEMP1 recombinant protein injection. This was a post-test-only true experimental study using 30 male Wistar rats (Rattus norvegicus) aged 2-3 months, weighing 150-250 g. The rats were randomly assigned into two control groups: NaCl 0.9% and adjuvant groups, and four treatment groups: DBL2β-PfEMP1 0.75 mg/kgBW, 0.75 mg/kgBW + adjuvant, 1.5 mg/kgBW + adjuvant, and 2.25 mg/kgBW + adjuvant groups. Injection was performed subcutaneously twice on days 0 and 42, and the blood serum was collected on days 14 and 56 for CD4 measurement using an enzyme-linked immunosorbent assay (ELISA) method. The results showed that CD4 concentration tends to decrease with an increasing dose of the DBL2β-PfEMP1 protein, particularly in post-injection II. Analysis of the differences between groups using the Kruskal-Wallis test showed no significant differences with p=0.59 in post-injection I and p=0.68 in post-injection II, and the Wilcoxon signed rank test to examine the differences across injections within each group showed no significant differences (p>0.05) in all groups. The results indicated no statistically significant differences in CD4 concentration post DBL2β-PfEMP1 recombinant protein injection in Wistar rats. Further studies examining other induced immune responses should be conducted to emphasize the DBL2β-PfEMP1 recombinant protein as a malaria vaccine candidate. Abstrak: Vaksin malaria berbasis antigen Plasmodium falciparum penting untuk mengendalikan morbiditas dan mortalitas akibat malaria. Duffy-binding-like domain 2β dari Plasmodium falciparum Erythrocyte Membrane Protein 1 (DBL2β-PfEMP1) merupakan salah satu kandidat vaksin malaria subunit yang diharapkan mampu mencegah malaria berat. Namun, umumnya vaksin subunit menginduksi respon imun yang lemah. Penelitian ini bertujuan untuk menganalisis respon CD4 setelah injeksi beberapa dosis protein rekombinan DBL2β-PfEMP1. Sebanyak 30 tikus Wistar jantan berusia 2-3 bulan dengan berat 150-250 g dibagi ke dalam dua kelompok kontrol: NaCl dan adjuvan, dan empat kelompok perlakuan: DBL2β-PfEMP1 0,75 mg/kgBB, 0,75 mg/kgBB + adjuvan, 1,5 mg/kgBB + adjuvan, and 2,25 mg/kgBB + adjuvan. Injeksi subkutan dilakukan dua kali pada hari ke-0 dan 42. Serum dikoleksi pada hari ke-14 dan 56 untuk pemeriksaan CD4 menggunakan metode ELISA. Hasil pengukuran menunjukkan bahwa konsentrasi CD4 cenderung menurun seiring dengan peningkatan dosis protein rekombinan, terutama setelah injeksi II. Analisis perbedaan tiap kelompok menggunakan Kruskal-Wallis test menunjukkan tidak ada perbedaan yang signifikan dengan p=0,59 setelah injeksi I dan p=0,68 setelah injeksi II. Dan analisis menggunakan Wilcoxon signed rank test untuk menilai perbedaan tiap kelompok antar injeksi menunjukkan semua kelompok tidak ada perbedaan bermakna (p>0,05).  Hasil ini mengindikasikan bahwa injeksi protein rekombinan DBL2β-PfEMP1 menginduksi respon CD4 yang tidak signifikan. Penelitian lanjutan dengan menilai respon imun lain diperlukan untuk menekankan potensi protein ini sebagai kandidat vaksin malaria.
