Abstract

Learning in the 21st century must be ICT-based. Professional elementary school teachers in the 21st century are teachers who can facilitate student learning. PGSD Study Program, STKIP Kusuma Negara through MK KKL held a national webinar with the theme Creative and Innovative Elementary School Teachers through ICT Implementation. This webinar was held online via the zoom meeting application and was attended by 287 participants, consisting of elementary school teachers, educational practitioners, and students (STKIP Kusuma Negara and the general public). The aim of this training is to equip elementary school teachers to be creative and innovative through the implementation of ICT. The stages of implementing the national webinar go through three stages, namely planning, implementation and evaluation. The national webinar material is Creative and Innovative Teachers Through Digital-Based Implementation and Innovative I-Teaching Teaching and Digital Tools. Schools are places to facilitate teachers in implementing ICT in the learning process to make learning creative and innovative. However, in reality in the field only students are able to use ICT tools. So that ICT-based learning does not become two-way. As a result, teachers are increasingly distant from ICT. Teachers must be able to implement ICT to create e-teaching and e-learning based learning.

Abstract

Diazinon is a pesticide widely used by farmers to control pests. Exposure to the low doses of diazinon can occur continuously through a polluted environment and diazinon residues in agricultural products. It has a nephrotoxic effect through oxidative stress mechanism. Flavonoid as antioxidant can significantly neutralize oxidative stress. Shallot peel is a source of flavonoids. This study was designed to determine the antioxidant effect of shallot skin infusion (SPI) on kidney oxidative stress in diazinon-induced Wistar rats by measuring the flavonoid level of SPI and kidney malondialdehyde (MDA) level. Rats were divided into normal, diazinon, and SPI groups. Diazinon was administered at dose of 40 mg/kgBW for 7 days followed by SPI at doses of 500, 1,000, and 2,000 mg/kgBW for 7 days. The flavonoid level of SPI was measured using spectrophotometry method and the kidney MDA level was measured using ELISA method. The flavonoid level of SPI was 96.8 mg QE/L. The results showed that the normal group had the lowest kidney MDA level of 2.585 mM/mL, the diazinon group had the highest level of 2.708 M/mL, while the SPI group at dose of of 500, 1,000, and 2,000 mg/kgBW had renal MDA levels of 2.642 mM/mL, 2.644 mM/mL, and 2.593 mM/mL. Nevertheless, the result of statistical analysis showed that the kidney MDA levels seven days after diazinon administration was not significantly different from the normal group so that SPI administration did not affect the kidney MDA level in diazinon-induced Wistar rats. Abstract

Kata kunci: dampak lingkungan, kelapa sawit, LCA, pemanasan global, pembibitan ABSTRACT As the initial stage in the oil palm (Elaeis guineensis Jacq.) plant life cycle, oil palm seedling cultivation becomes a crucial aspect that requires in-depth evaluation. Understanding the environmental impact at the seedling phase is critical to designing more sustainable management strategies, reducing ecosystem damage, and achieving a balance between production and conservation. This study aims to present an environmental impact analysis of oil palm seedling cultivation at the Oil Palm Nursery Unit (UPKS) of Politeknik Negeri Lampung using the life cycle assessment (LCA) approach. This study focuses on a gate-to-gate life cycle assessment conducted from October to December 2023. Data were collected through field visits and interviews with UPKS managers. The LCA scope covers from seed acceptance to ready-to-distribute seedlings. Environmental impact modeling used openLCA version 2.0.3 with the CML-IA baseline method and supporting database Agribalyse v3.0.1. Results indicate that oil palm seedling nursery generally has an insignificant environmental impact. The most substantial contributions come from secondary impacts, mainly marine aquatic ecotoxicity at 190.52122 kg 1,4-DB eq. and abiotic depletion (fossil fuels) at 5.65733 MJ. Primary impacts are potential global warming (GWP100a) at 0.27033 kg CO2 eq., primarily from polybag use, and potential human toxicity at 0.08227 kg 1,4-DB eq. Efforts to mitigate environmental impacts can be achieved by using reusable pot trays or organic material-based polybags as alternatives, coupled with a combination of inorganic and organic fertilizers to reduce the dosage of inorganic fertilizers. Additionally, curtailing evapotranspiration rates from plants and growing media can be accomplished by employing organic mulch and growing media enriched with organic matter. Keywords:  environmental impact, global warming, LCA, nursery, oil palm

Abstract

Introduction: Diabetes mellitus (DM) comorbidity in drug-resistant (DR) pulmonary tuberculosis (TB) patients can be associated with the treatment outcome. In DR-TB patients with DM, the immune system is impaired, which will decrease the success of treatment. Sputum conversion time is an indicator used to predict the treatment outcome. However, there is still no further study related to the association between DM comorbidity and the sputum conversion time in DR-TB patients, especially in Jember. This study aimed to determine the association between DM and sputum conversion time in DR-TB patients at Dr. Soebandi Regional General Hospital, Jember. Methods: This was an analytic observational study with a cross-sectional design. A total of 122 samples of DR-TB patients were taken using the purposive sampling method in 2018-2023 at Dr. Soebandi Regional General Hospital, Jember. The data were analyzed using the Chi-square and logistic regression statistical test. Results: Chi-square analysis showed that DM (p = 0.015; OR = 2.604; 95% CI 1.195-5.674) and age (p = 0.021; OR = 0.377; 95% CI 0.162-0.878) were associated with sputum conversion time. Logistic regression showed that DM was the most associated variable with the sputum conversion time (p = 0.016; OR = 2.604; 95% CI 1.195-5.674) compared to gender, age, TB resistance type, and anti-TB regimen. Conclusion: DM is associated with prolonged sputum conversion time in DR-TB patients at Dr. Soebandi Regional General Hospital, Jember. DM was also the most associated variable with the sputum conversion time compared to gender, age, TB resistance type, and anti-TB regimen.

Abstract

Cigarette smoke exposure is the major risk of chronic obstructive pulmonary disease, the third cause of death worldwide. Inhaled smoke triggers oxidative stress resulting in airway epithelium thickening and cilia shortening. Shallot (Allium cepa L.) peel contains flavonoid which can neutralize oxidative stress. This study aims to determine correlation between shallot peel infusion (SPI), bronchus epithelium thickness, and cilia length in cigarette smoke-induced rats; and establish maximum effective dose of SPI. Rats were divided into normal; cigarette; SPI 125, 250, 500, 1,000, 2,000 mg/kgBW groups. Two hours after administration, rats were exposed to cigarette smoke 2 cigarettes/day for 28 days. Hematoxylin-eosin-stained bronchus was observed and variable measurements were carried out. Comparation tests of epithelium thickness and cilia length between normal and cigarette group showed significant difference (p<0.05); Pearson coefficient between SPI dose and epithelium thickening was -0.614, Spearman coefficient between SPI dose and cilia length was 0.860; and maximum effective dose to prevent bronchus epithelium thickening and cilia shortening are 1,275.4 mg/kgBW and 1,325.8 mg/kgBW. In conclusion, the higher the SPI dose, the lower epithelium thickness, the higher cilia length. Maximum effective dose of SPI to prevent bronchus epithelium thickening and cilia shortening are 1,275.4 mg/kgBW and 1,325.8 mg/kgBW.

Abstract

Background: The pathogenesis of severe malaria involves the antigenic protein Plasmodium falciparum erythrocyte membrane protein-1 (PfEMP1), encoded by the var gene. One of the important domains in PfEMP1 is Duffy Binding Like 1α (DBL1α). To diagnose malaria, microscopic examination has low sensitivity and specificity, therefore, the development of molecular-based methods is needed. This study aimed to determine the potential of DBL1α-PfEMP1 as a diagnostic marker and clinical predictor for falciparum malaria. Methods: An exploratory descriptive study was conducted in 2019 on malaria patients at the Tiakur public health center, Southwest Maluku, Indonesia. Blood samples of patients infected with Plasmodium falciparum malaria were collected on filter paper for DNA isolation. Amplification by polymerase chain reaction (PCR) method used primers αAF [5'-GCA CG(A/C) AGT TTT GC-3'] and αBR [5'-GCC CAT TC(G/C) TCG AAC CA-3'] with cycles of denaturation 95oC 1-minute, annealing 42oC 1-minute, elongation 60oC 1-minute. PCR products were electrophoresed using 1% agarose gel. Amplicons were sequenced directly and analyzed using nucleotide BLAST-NCBI. Results: All patients showed mild malaria symptoms. PCR amplification yielded bands of 370 bp in all samples and 600 bp in 8 out of 10 samples, and 1 sample had a different pattern. Sequencing results confirmed that the amplicon was DBL1α, a var gene that had similarities to sequences from other regions. Conclusion: Positive amplification and sequencing results confirm the sensitivity of DBL1α-PfEMP1 as a diagnostic marker. The sequence variability of PCR product implies the presence of DBL1α variations, indicating a correlation with clinical outcomes and making it a clinical predictor.

Abstract

Background: In the malaria vaccine study, Duffy binding-like 2β Plasmodium falciparum erythrocyte membrane protein 1 (DBL2β-PfEMP1) could induce the  IgG and CD4+production. Antibody to DBL2β-PfEMP1 reduces the risk of developing severe malaria through the blockade of cytoadherence and destruction of rosette formation. During the process of antibody formation after immunization, the released peripheral cytokines have the potential to cause blood-brain barrier disruption resulting in the activation and proliferation of brain microglia as primary innate immune cells leading to neuroinflammation. Objective: This study aims to evaluate brain microglia proliferation as a response to recombinant protein DBL2β-PfEMP1 immunization in Wistar rats. Methods: Wistar rats were injected subcutaneously with recombinant protein DBL2β-PfEMP1 at doses of 100, 150, and 200 µg/kgBW on days 0, 21, and 42. Rats were euthanized on day 56. Brain histopathological slides were prepared and stained using hematoxylin-eosin. Histological examination was performed using a microscope at 400X magnification and documented using an AmScope microscope digital camera. Brain microglia were calculated using Fiji Image-J. The data were statistically analyzed using the Kruskal-Wallis test.  Results: The average number of brain microglia in both the control and treatment groups was 82–88. There was no significant difference in brain microglia number between the control and treatment groups (p>0.05) after recombinant protein DBL2β-PfEMP1 immunization. Conclusion: Recombinant protein DBL2β-PfEMP1 immunization does not provoke the proliferation of brain microglia in Wistar rats. This suggests that the protein does not have the potential to cause neuroinflammation.

Abstract

Diazinon, an organophosphate pesticide, is used extensively in agricultural sector. Consumption of agricultural products containing diazinon residue may lead to harmful health consequences. Among these is nephrotoxicity, which includes lipid peroxidation, that can damage the kidney. Flavonoids in shallot peel can scavenge free radicals, inhibit necrosis, and activate bone marrow-derived cells for cell regeneration. This study aimed to determine the correlation between shallot peel infusion (SPI) dose and kidney damage amelioration to establish the maximum effective dose of SPI to ameliorate kidney histopathological damage in diazinon-induced rats. This study was performed at the Pharmacology Laboratory, Faculty of Medicine, University of Jember, Indonesia, in April 2021, on 35 rats that were divided into 7 groups—normal, diazinon, and five treatment groups. Diazinon 40 mg/kgBW was administered on day 1-7, while SPI 125, 250, 500, 1,000, 2,000 mg/kgBW were administered on day 8–14 according to the treatment group. Kidney histopathological slides with hematoxylin-eosin (H.E.) staining were assessed using Kocoglu scoring and Kidney damage scores of the treatment groups were analyzed using Pearson test. The maximum effective dose was determined using regression test. The damages found in diazinon-induced rats were tubular degeneration, necrosis, and inflammation with a higher damage score than normal rats (p<0.05). Pearson test showed moderate correlation (coefficient -0.594). Higher SPI doses presented lower kidney damage scores, with 1,359 mg/kgBW being the maximum effective dose. SPI dose and the kidney damage amelioration are moderately correlated with a SPI maximum effective dose to ameliorate kidney damage in diazinon-induced rats of 1,359 mg/kgBW.
