Abstract

The research characterization of cytotoxic fraction against P-388 leukemia murine cells from the extract honje (Etlingera elatior) seed have been reported. This research lead to isolated and characterization of cytotoxic compounds against P-388 leukemia murine cells from the extract E. elantior seed. The extract of E. elantior seed was maserated by methanol, n-hexane, and ethyl acetate, respectively and estimated their cytotoxic activity against P-388 leukemia murine cell with 3- (4, 5-dimetiltiazol-2-yl) -2,5-difeniltetrazolium bromide (MTT) assay guided toxicity test against of shrimp Artemia salina Leach. Brine shirmp Lethality Test (BSLT) method. The active extracts will be separated by fractionation using column chromatography, radial chromatography, and for analyzing the purity of isolate will estimate by HPLC. The chemical structure of pure isolate will be identified by spectroscopies data UV Vis, FTIR, NMR and MS. The ethyl acetate extract from honje seed have cytotoxic activity by leukemia P-388 cell  with IC50 19.21 µg/mL. The compound toxic as cytotoxicagainst P-388 leukemia murine cells is flavonoid compouds their is resveratrol, lapachol, apigenin, methylated chrysin, 6,2’-dihydroxyflavanone, 3-hydroxy-3,4’-dymethoxyflavone and 4’-hydroxy-5,7-dimethoxyflavanone.DOI: http://dx.doi.org/10.15408/jkv.v0i0.3640

Abstract

Dadih is naturally fermented buffalo milk in bamboo tubes and known to have antioxidant, antibacterial, and antihypertensive activity. Lactic acid bacteria in dadih can use as a starter to produce yogurt. The study aimed to produce yogurt of cow's milk with dadih as a starter, to determine the antioxidant activity and amino acid composition. Dadih is added with concentration variation of  2.5; 5; 7.5 and 10% (v/v) and it fermented for 48 hours at room temperature. Lactic acid bacteria cell counts of dadih were calculated by the total plate count method. Yogurt was tested by organoleptics with 33 panelists and proximate analysis water, ash, fat and protein contens based on the Association of Official Analytical Chemist (AOAC) in 2005. Antioxidant activities of yogurt were tested by DPPH method (1,1-diphenyl-2-picrylhydrazyl). Amino acid composition of yogurt was analyzed using ultra performance liquid chromatography (UPLC). The research result of amount of LAB on dadih are 1.01 x 1011 CFU/mL, has fulfilled the requirements of SNI 2981: 2009. Yogurt produced with the addition of dadih by 10% (v/v) is most accepted by panelists. The highest antioxidant activity was obtained by yogurt with 2.5% addition of dadih with IC50 value of 78.28 ppm. Yogurt almost contain all essential and non-essensial amino acids including tyrosine and phenylalanine as the antioxidants. Measurement of water, ash, protein and fat content in the sample meets the requirements of SNI 2981: 2009.

Abstract

Akway (Drimys beccariana Gibs) is an endemic plant of Papua. Ethnobotany is used as a medicinal plant, one of which is used as a malaria drug. This study aims to isolate, determine the chemical structure and evaluate the antimalarial activity of D. beccariana stem bark. Powder of D. beccariana bark is macerated with methanol and fractionated using n-hexane and ethyl acetate. Ethyl acetate extract was fractionated using gradient liquid chromatography (KCV) (n-hexane: ethyl acetate: methanol). The KCV fraction was separated by repeated gravity column chromatography and preparative thin layer chromatography (TLC-P) to obtain Isolate 1. Isolate 1 was determined by spectroscopic methods including UV-Vis, IR, 1D NMR, and 2D. Isolate 1 is determined by its compound structure which is 5,3'-dihydroxy-7,4'-dimethoxyflavone. 5,3'-dihydroxy-7,4'-dimethoxyflavone compounds evaluated antimalarial activity against Plasmodium falciparum in vitro with antimalarial activity better than artemisinin.

Abstract

Telah dilakukan penelitian untuk mengetahui struktur senyawa fenolik yang memiliki aktivitas antioksidan dari ekstrak etil asetat biji honje (E. elatior). Isolasi senyawa fenolik dilakukan dengan metode maserasi, fraksinasi dengan kromatografi kolom gravitasi (KKG), dan kromatografi lapis tipis (KLT). Uji aktivitas antioksidan menggunakan metode DPPH (2,2-diphenyl-1-picrylhydrazyl) dan penentuan struktur senyawa menggunakan spektroskopi UV-Vis, FTIR, NMR, dan MS. Isolat yang diperoleh berupa gum kuning sebanyak 18 mg dari 3 kg sampel kering. Hasil uji aktivitas antioksidan menunjukkan isolat dari ekstrak etil asetat memiliki aktivitas yang sangat kuat dengan IC50 1,32 µg/mL. Hasil analisis dengan spektroskopi UV-Vis, FTIR, NMR, dan MS menunjukkan isolat sesuai dengan rumus molekul C7H6O4 yang dikenal dengan asam protokatekuat (asam 3,4-dihidroksi benzoat).DOI:http://dx.doi.org/10.15408/jkv.v4i1.7225

Abstract

Soybeans contain proteins that have the potential to produce anticancer bioactive peptides. This study aims to determine the anticancer activity of soy protein hydrolysate against MCF-7 breast cancer cells. Soybean protein hydrolyzed by bromelain enzyme 0.5% (w/v) at the optimum temperature and pH for protein hydrolysis using the Bergmeyer and Grassl method. The degree of hydrolysis value of protein hydrolysate was determined by the Alder-Niesen method and the protein profile was analyzed by SDS-PAGE. The hydrolysate with the best degree of hydrolysis value was analyzed for anticancer activity against MCF-7 breast cancer cells by the Presto Blue assay method, and fractionation of protein hydrolysates by gel filtration chromatography (Sephadex G-15). The molecular weight of the peptide was characterized by LCMS/MS. Soy protein hydrolysis using 0.5% (w/v) bromelain enzyme was optimum at 65 ºC and pH 7.0 for 4 hours, with a hydrolysis degree value of 20.57%. The SDS-PAGE analysis showed that the protein hydrolysates had quite thick protein bands in the range of <35 kDa with an IC50 value of 70.37 mg/mL. Based on the LCMS/MS results, the peptide from fractionation has a molecular weight of 5.133 kDa.

Abstract

The impact of extraction methods on the potency of plant extracts is crucial. Bouea macrophylla Griff., a tropical fruit plant rich in phenolic and flavonoid compounds, exhibits strong antioxidant activity. This study aimed to evaluate the effect of maceration and soxhletation extraction techniques on B. macrophylla plants on antioxidant activity in vitro using the method of inhibiting DPPH radical compounds. Ethyl acetate and ethanol solvents were utilized to macerate and soxhlet all parts of the plant. Subsequently, each extract was subjected to quantitative analysis for antioxidant activity, total phenolic, and flavonoid content through UV-Vis spectrophotometry. Quantitative analysis revealed substantial antioxidant activity and notable levels of total phenolic and flavonoid content across all plant parts. The result indicated that although both maceration and soxhletation showed a significant impact on yield (p=0.009) and total flavonoids (p=0.038), significant differences in antioxidant activity (p=0.409) or total phenolics (p=0.874) within the plant extracts of B. macrophylla were not observed at a significance level of 5%.

Abstract

Alzheimer's is a chronic neurodegenerative disease characterized by low levels of acetylcholine and the accumulation of abnormal neuritic plaques, leading to rapid memory decline and cognitive impairment. Compounds found in the kedondong plant (Spondias dulcis) have been reported to exhibit in vitro activity as acetylcholinesterase inhibitors. This study examines the potential of active compounds in Spondias dulcis in their interaction with acetylcholinesterase, an enzyme implicated in the pathogenesis of Alzheimer's disease. The enzyme was obtained from the Protein Data Bank (PDB ID: 4EY7). The test ligands were screened based on Lipinski's rule and docked with the receptor. The results of molecular docking which yielded the five best affinity energy values were followed by ADMET testing (absorption, distribution, metabolism, excretion, and toxicity). The test ligand ellagic acid deoxyhexoside showed binding energy at -11.213 kcal/mol. Molecular dynamics simulations were performed using YASARA with AMBER14 force fields for 50 ns. The test ligand ellagic acid eoxyhexoside showed an MM-PBSA value of -51.277 kcal/mol and exhibited good complex stability with an average total RMSD value of 2 Å and low inter-residue fluctuation values. These findings are consistent with the results obtained from the comparator ligand, donepezil. Therefore, compounds in Spondias dulcis have the potential to act as acetylcholinesterase inhibitors and can be considered for the development of therapies for Alzheimer's disease.
