Abstract

Barramundi seeds, Lates calcarifer are usually sourced from natural spawning using a limited number of broodstock. Therefore, the growth and survival rate of these seeds vary greatly between batches. The research was performed to determine the growth pattern and genetic variations of barramundi seed populations produced from domesticated broodstock sourced from Australia, Situbondo, and Lampung and reared in small-scale hatcheries. Ten individuals of barramundi from each population were used for microsatellite analysis using two microsatellite loci, namely: Lca 21 and Lca 32. The resulted microsatellite data was processed using the genetic analysis available in Excel software (GenAlEx 6.51b2). The results showed that the growth in length and body weight as well as the specific growth rate of barramundi seeds produced from Australia broodstock were higher than that of Situbondo and Lampung (P<0.05) while the later two were similar (P>0.05). The number of microsatellite alleles ranged from 2-8 and the highest heterozygosity was obtained by barramundi seeds produced by Situbondo (0.85), followed by Lampung (0.65), dan Australia (0.54) broodstock. From the results of the research, it can be concluded that the three populations of barramundi fish, which were domesticated and reared in the HSRT system, meet the criteria for use in hybridization program or for forming a synthetic population.

Abstract

Background: Indonesia’s ageing population demands proactive strategies to preserve older adults’ quality of life, particularly in maintaining occupational independence, which often declines due to physical limitations, social isolation, and inadequate access to community-based rehabilitation. This community service aimed to enhance the understanding and participation of older adults in occupational independence through a culturally grounded, meaningful activity program in Kersan Brintikan Hamlet, Tirtomartani Village, Kalasan Subdistrict, Sleman Regency. Methods: A pre–post-test counselling design was employed with 52 participants (46 older adults and 6 elderly health cadres). The intervention included health screening, morning walking, anti-hypertension exercise, occupational therapy services, and interactive health education using visual media. A structured questionnaire comprising 10 closed-ended items was administered before and after the session to assess changes in knowledge. Results: Most participants were in the pre-elderly age group (64.3%), nearly gender-balanced (48.1% male, 51.9% female), and moderately to highly educated (78.7% senior high school or above). The mean understanding score significantly increased from 4.75 (pre-test) to 9.33 (post-test), while the standard deviation decreased from 1.064 to 0.857, indicating a homogeneous and substantial improvement in comprehension across participants. Conclusion: A participatory, contextually relevant program combining health promotion and meaningful occupational activities effectively enhances awareness of occupational independence among older adults. For sustainability, follow-up sessions, cadre capacity building, and inclusive outreach to older and male elders are recommended to support active ageing in the community.

Abstract

The main objective of this study was to determine the profile of endogenous intestinal bacteria of yellowfin tuna, Thunnus albacares that can function as potential probiotics in the digestive system and used for the maintenance of yellowfin tuna larvae. Wild yellowfin tuna, T. albacares was collected from the waters of Gerokgak, Buleleng, North Bali, Indonesia. Intestinal bacteria were isolated, identified, characterized, and followed by enzymatic hydrolysis activity tests, antagonistic tests and pathogenicity tests to select candidate bacteria as probiotics. The results of the enzymatic hydrolysis activity test showed that there were 4 isolates of candidate probiotic bacteria that were active in enzymatic synthesis for amylase, lipase, casease, lecithinase, gelatinase. Identification through Sanger sequencing resulted in 4 potential bacterial isolates as probiotics, namely Bacillus subtillis strain T-A1, Bacillus amyloliquefaciens strain T-N2, Bacillus subtillis strain T-O4, and Paraclostridium bifermentans strain T-O6. These four bacteria were not antagonistic between varieties and were not pathogenic in Tuna fish. Yellowfin tuna fed probiotic supplements grew faster (5.5002 ± 0.2 mm) than the control (4.9002 ± 0.4 mm). The success of yellowfin tuna seed production is expected to be sustainable and its implementation in the private sector for aquaculture purposes.

Abstract

The objective of this study is to seek the challenges experienced by English teachers in applying deep learning, focusing on their classroom experiences and perceptions using the activity theory as a  framework that focuses on its concepts of contradiction, boundary objects, and boundary crossing. This study used a semi-structure interview to understand the three participants' experiences as teachers in implementing Deep Learning in the classroom practices. Key challenges to their implementation came in the form: limited understanding, workload and time constraints, implementation barrier, limited administrative preparation, curriculum adaptation gap, and government requirement. However, the participants were able to overcome the challenges that arose by treating it as a connecting tool to connect their teaching methods. These findings raise important theoretical and practical issues related to strategies for overcoming challenges encountered in implementing deep learning, as implemented by the participants by institutional support and teachers training, implementation experience, facility support, and fellow teachers support. One of the connections is understanding how teachers manage the challenges they encounter by connecting their various activity systems as objects to overcome the obstacles that occur in the deep learning implementation system they use in the classroom.

Abstract

This research aimed to investigate students’ perceptions, difficulties and learning experiences in a blended listening class situation. In the present qualitative study, thirty (30) English Language Teaching (ELT) undergraduate students were selected using a purposeful sampling to be the research participants. Open-ended questionnaires were distributed for data collection. Then, the data collected were analyzed through reflexive thematic analysis, so as to interpret the findings and further understand students’ experiences. Results indicated that students appreciated the independence that was afforded online, such as being able to pause audio and access transcripts, but they enjoyed face-to-face sessions for the feedback they could obtain and the questions they could ask. Though faced with issues like technical problems and struggling to focus on learning, it is concluded that a blended approach can be both effective and efficient if the online and offline components are well-integrated and supported by an appropriate system, which makes it also highly applicable in EFL.
