Abstract

Abstract

The main purpose of this present study is to evaluate the ability of near infrared technology as an alternative method in determining and assessing quality parameters of meat product where in this case is frozen beef. At first, beef samples from chest and legs parts were sliced and taken at the amount of 100g per sample to be frozen. Then spectra data of beef samples were obtained using near infrared spectrophotometer (PSD IR i16) in wavelength range from 1000 to 2500 nm with optical gain 4x. Actual protein contents were obtained by Kjeldahl method and measured in triplicate. The near infrared spectra data were enhanced and improved by means of mean centering (MC) and baseline shift correction (BSC) methods. The results showed that protein content of frozen beef samples can be predicted rapidly with maximum correlation coefficient is 0.91. Heat properties of beef samples changes exponentially during freezing and thus, optimum freezing temperature and time can be predicted as well. Based on those obtained results, it may conclude that near infrared technology can assess frozen beef qualities rapidly and effectively.

Abstract

This study aims to determine the histomorphometry of the small intestine of turkeys (Meleagris gallopavo) at different age levels. This study used a completely randomized design. This study used 18 male turkeys, divided into 3 groups of ages 8, 16, and 24 weeks, each group comprising 6 turkeys. The necropsied turkey's small intestine was taken into histological preparations and stained with haematoxylin-eosin (HE). The results showed that the histological picture of the small intestine layer of turkeys at different age levels consisted of four layers: tunica mucosa, tunica submucosa, tunica muscular, and tunica serosa. Histomorphometrically, there was an increase in the thickness of all layers of the small intestine from 8 and 16 weeks of age (P0.05), and the height of the intestinal villi increased in all parts of the small intestine at 8, 16 and 24 weeks of age (P0.05 ). In conclusion, with histomorphometric measurements, it can be seen that there are differences in the three age groups.

Abstract

The aim of this research was to determine an effect of Newcastle disease virus (NDV) vaccine to histopathologic on Fabricius bursa of broiler chicken at various age levels. Eighteen commercial DOC (Day Old Chick) were divided into 2 groups namely a control group that was given distilled water and a treatment group that was given NDV vaccine namely ND Lasota vaccine ocularly as much one drop per eye at day 3, then a booster dose in drinking water at day 13 was given. The bursa of Fabricius was collected from these chickens on the age of 14 days, 21 days, and 28 days after sacrificing the chickens through necropsied, then histopathological preparations are made using hematoxylin-eosin staining. The parameters observed were measured the number, diameter, and thickness of cortical of lymphoid follicles. Data were analyzed descriptively and quantitatively by unpaired t-test. The results of this research showed the NDV vaccine effects were not significantly different (P0.05) to the number and diameter of lymphoid follicles. In the treatment group, 14 days old broiler chickens had a decreased number of lymphoid follicles, while its diameter tends to grow and the thickness of cortex was also seemed unchanged compared with the control group. Then at the age of 21 days, the number, diameter, and thickness of cortex of lymphoid follicles tend to increase, while at 28 days diameter of lymphoid follicles tend to be slightly reduced, but the number and thickness tend to grow out of control. It was concluded that the NDV vaccine had a good effect on the Fabricius bursa of broiler chickens with increasing age, then histopathological preparations are made using hematoxylin-eosin staining.
