Abstract

Spondias pinnata dengan suku Anacardiaceae merupakan salah satu tanaman yang daunnya dimanfaatkan sebagai obat batuk. Penelitian sebelumnya menunjukkan bahwa, ekstrak daun S. pinnata memiliki aktivitas antituberkulosis terhadap isolat Mycobacterium tuberculosis strain multidrug resistant. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak etanol daun S. pinnata terhadap organ hati pada mencit betina galur balb/c. Penelitian ini diawali dengan ekstraksi serbuk daun S. pinnata menggunakan metode digesti, kemudian diuji pengaruhnya pada 5 kelompok yang masing-masing terdiri dari 5 ekor mencit betinagalur balb/c. Kelompok kontrol negatif diberikan suspensi CMC Na, kelompok perlakuan diberikan ekstrak dosis tunggal 0,015; 0,15; 1,5; dan 15 g/kg BB secara per oral. Setelah 24 jam perlakuan, mencit dibedah dan diamati berat organ hatinya. Data yang diperoleh dianalisis secara statistik dengan One way Anova. Hasil penelitian menunjukkan adanya perbedaan bermakna pada berat organ hati antara kelompok kontrol dengan kelompok perlakuan ekstrak dosis 0,015 dan 0,15 g/kg BB. Perubahan berat organ merupakan indikator adanya perubahan pada sel-sel organ akibat senyawa kimia. Data yang diperoleh menunjukkan bahwa, pemberian ekstrak etanol dosis 0,015 dan 0,15 g/kg BB menyebabkan penurunan berat organ hati setelah 24 jam pemberian ekstrak.

Abstract

Newcastle disease is a pathogenic viral disease in poultry which is infectious and can cause massive economic losses. The disease is still endemic in Indonesia. To understand the pathogenesis and the distribution pattern of the virus in the tissues, pathological observation was evaluated using newly virulent isolate Newcastle disease virus (NDV) that was inoculated in embryonated chicken eggs. As many as seven embryonic chicken eggs aged 11 days and specific antibody negative against Newcastle disease, divided into two categories: inoculated with phosphate buffer saline and inoculated with isolates. Then the allantois fluid was tested using hemagglutination assay and hemagglutination inhibition tests to prove the infection serologically. The hearts, lungs, livers and small intestines of the inoculated products were collected and followed with the process of histopathological preparation using Hematoxylin and Eosin (HE) stain. The pathological analysis showed that all organs had necrosis, hemorrhages, inflammation, and congestion. Congestion and hemorrhages in the hearts only occurred at 60% of the samples. However, necrosis, hemorrhages, and inflammation that were observed in liver occurred at 60%, 40% and 60% of the samples, respectively. Furthermore, the hearts were edema, thinner in the heart muscle fibers; while in the lungs, proliferation of pneumocyte type II was founded. Our finding provided valuable insight into the pathology of a virulent isolate of NDV which is dominated by blood circulation disorders with necrosis and inflammation in the chicken’s embryos and have important implication for the future studies.

Abstract

A study was carried out on the molecular characteristics of gene encoding for nucleocapsid protein(NP) of Newcastle disease virus (NDV) Bali-1/07. A portion of the fragment gene was amplified by reversetranscriptase-polymerase chain reaction (RT-PCR). The RT-PCR product purified from the gel was treatedwith Taq polymerase and cloned into plasmid pT7blueT vector. The recombinant plasmid was tranfectedinto Nova blue singles strain of Escherichia coli-competent cells and selected using white and blue colorselection method. Good white colonies were subcultured and tested for presence of expected gene fragmentby polymerase chain reaction (PCR). The correct size PCR product was recloned and sequenced. The PCRproduct of 540 base pairs were sequenced and aligned with several cognates genes of world NDVisolates.using Cluster IW. The phylogenetic analysis was then performed using MEGA 4. Phylogeneticanalysis showed that the NP gene of NDV Bali-1/07 is closely related with virulent NDVs such asGuangxii-11/03, KBNP-4152 and Ast2755/01 with nucleotide sequence homology of 92%, 91.4% and91%respectively, whereas the nucleotide sequence homology with avirulent NDVs such as LaSota and B1were 77%.

Abstract

Rabies was a new emerging disease in Bali. After the first case of rabies in 2008 there were morethan one hundred of human victims in Bali. The aim of this study was to report the anatomical locationof human body bitten by rabies infected dogs in Bali, and also the day of victims death after the bitten. Aretrospective cross-sectional review of rabies incidences from September 2008 to the end of 2011 was usedin this study. A total of 122 rabies human victims data were used as a population sample. The data on theprofiles of individuals victim were colected , such as the age, gender, living place, anatomical bite site onhuman body, bite date, and type of biting animal. The obtained data were analyzed using descriptiveanalysis. The result showed that the anatomical location of human body that bitten by rabies infected dogwas occurred on leg (52%), hand (32%), body (6%), and head (4%). Of 122 victims, 61.5% were men and38.5% were woman. The death time of the human rabies victims in average occurred at day 95th after thebite date, and time of the victims death depended on the anatomical location of the bite. Bite that occurredon the head caused death in average on day 19th, on the body on day 83th, on the hand on day 122nd, and onleg on day 166th. In conclusion, the most victims of human rabies in Bali was men and the death was foundoccurred on day 95th after the bite date. The bite at the head caused death more quickly than the other partof human body.

Abstract

This study aims were to describe lesions and their intensities of histopathological changes in gallbladder mucosa of bali cattle that infected with Fasciola gigantica. As many as 42 gall bladers of balicattle were used in this studi, i.e: 35 samples were infected and sefen were non-infected with F.gigantica.The samples were randomly gathered from Pesanggaran slaughter house in Denpasar. The gall bladderswere evaluated macroscopically and prepared for microscopically examination using haematoxylin andeosin staining.The result showed that there were histopathological changes observed in the mucosa of thegall bladder:necrosis cells of gall bladder with intencity 100%, hemorrhage 68.5%, hyperplasia serousgland and mucous hypertrophy gland 100%, collagen in filtration 100%, fibroblast proliferation 91.4%,and infiltration of inflammation cells, 74.2%. Histopathological changes in gallbladders mucous of balicattle infected F.gigantica experiencing severe lesons with frequency percentage above 50%.

Abstract

Complex dermatitis is a disorder of the skin caused by the combination of several agents such as fungi, ectoparasites, bacteria, and metabolic diseases with combined clinical symptoms of primary lesions and secondary lesions. This study aims to determine the histopathology of dog skin with complex dermatitis. There were 15 samples of dogs with complex dermatitis, skin biopsy and histopathology. The results showed that the incidence of complex dermatitis in young dogs was 60%, long-haired 67%, male sex 73%, and domestic race 73%. Macroscopic examination found a change in hyperkeratosis. Microscopically observed that infiltration of polymorphonuclear cells, monomorphonuclear cells and macrofag from moderate to severe in the dermis. In hair follicles and epidermis occured akantosis, and keratin proliferation. There were segment fragments from infectious agents such as Demodex canis, Sarcoptes scabiei, and yeast from Malasezia sp. Other changes were observed in the presence of hydrophic degeneration, necrosis, and ulcers. Dogs suffering from complex dermatitis have chronic infections with more than one infectious agent. Disabilities based on histopathological observation of dogs with complex dermatitis with moderate severity of 33%, and severe 67%.

Abstract

Cases of ascariosis due to infection with Ascaris suum are still prevalent in the world, including Indonesia. Apart from causing economic losses, ascariosis can also threaten human health because it is zoonotic. Case studies of ascariosis with typical white spot lesions in the landrace pig liver is described in this paper. Pigs aged 60 days old, sick for 10 days, thin and coughing gathered from the village of Suwung, Denpasar City. After necropsy and post mortem examination were done, tens of A. suum worms were found in the intestine and white spot lesions observed on the livers. Tissues of small intestine, large intestine, liver, lungs, kidney, spleen, urinary vesicles and brain were taken and put into 10% of formalin neutral buffer before they then processed in making histopathological preparations. Histopathological preparation was carried out using paraffin block method and hematoxylin eosin (HE) staining technique. The results of histopathological examination of the small intestine, large intestine, liver and lungs were found to be infiltrated by eosinophil cells. Eosinophil cells infiltration indicates that there are parasites A. suum or the larvae migrant in the tissues. Some of the tissues in livers were also found to have fibrosis, which is suspected that the infection has been chronic. It can be concluded that cases of the landrace pigs were chronically infected by A.suum and accompanied by migration of the larvae to the livers and lungs. More attention is needed to prevent the increase of ascariosis in pigs in order to minimize economic losses and transmission to humans.

Abstract

In order to study the distribution of Newcastle disease virus (NDV) following infection, chickenswere experimentally infected with visceretropic velogenic NDV isolate. Monoclonal antibodies (mAbs)against the NDV LaSota vaccine strain were then produced to detect viral antigen in the infectedorgans. The mAbs were firstly tested for their specificity by enzyme linked immunosorbent assay(ELISA) using NDV and normal allantoic fluids as antigens. Eight mAbs specific against NDVwere isolated and two mAbs were used for immunodetection of NDV antigen in chicken’s tissues.By immunohistochemistry labeled streptavidin-biotin (LSAB) staining NDV–antigen was detectedin paraffin embedded tissues of NDV-infected chickens. NDV antigen was not detected in noninfected chickens. In the infected chickens, high intensity of NDV antigen was detected in thelymphoid tissues, lung and intestine. The NDV antigen with a lesser intensity was detected in thebrain, trachea, liver and myocardium. This study shows that although viscerotropic velogenicNDV isolate can infect almost all organs, the main target of infection are lung, intestine andlymphoids tissues

Abstract

This research aims were to measure the prevalence of Entamoeba in pigs in Bali and to identify thezoonotic potential species of Entamoeba. A total of 183 pig stool samples from Bali have been examined.The method being used in this study were combination between coproscopy and molecular techniques.Concentration sedimentation with Sodium Acetic Formaldehide (SAF) as a solution was used in thecoproscopy method, while the Polimerase Chain Reaction method was used to amplify DNA of Entamoeba.Extracted sample’s DNA examined by using primers that specifically for Entamoeba: Entam 1 (F) (5’-GTTGAT CCT GCC AGT ATT ATA TG-3’) and Entam 2 (R) (5’-CAC TAT TGG AGC TGG AAT TAC-3), and toidentify the zoonotic potential species of Entamoeba, samples that produce 550 bp in first amplificationcontinued by primers Epolecki1 (F) (5’-TCG ATA TTT ATA TTG ATT CAA ATG-3’) and Epolecki2 (R) (5’-CCT TTC TCC TTT TTT TAT ATT AG-3’). The results showed that 76.6% of samples were positive incoproscopical examination, but 84.7 % produced 550 bp bands on PCR amplification by using generalprimers. All positive samples on the first PCR continued to second PCR used specific primers for E.poleckii as a potential zoonotic disease and all of the samples showed negative results. This datademonstrated that the prevalence of Entamoeba in a traditional pig scavenging systems in Bali was 84.7%but no specific infection infection caused by E. polecki was found.

Abstract

This study aims to determine the pathological changes in dog suspected of being infected by Leptospira spp. This research used a retrospective study design. A total of 210 canines sample were examined at Faculty of Veterinary Medicine The Udayana of University between January 2008 to January 2010. Of the 210 specimens, five canines with clinical sign anorexia, fever, vomiting, polyurea, dyspnea and only one are noted icteric in the sclera. Macroscopically : the anemic to petechial haemorrhagis was find in pulmo, icterus in liver with gall blader distention, kidney swollen with black color in hillus, spleen rather swollen with black color on end and mild hemorrhagis on intentines. The infected organ is inserted into a pot filled with neutral buffer formalin 10%. Then stained with haematoxyllin-eosin for the microscopic preparation. Microscopic examimination revealed the presence of spiral bacterial cells on the necrotic liver and kidneys tissue. Infiltration neutrophils and macrophages was also find in pulmonary and spleen tissue. Besided that intertubuler edematous were also observed under microscopic examination. It can be concluded changes associated with Leptospira spp infection can be found in liver and kidney tissue with changes such as necrosis, proliferation of kuffer cells and intertubular edematous.
