Abstract

ABSTRACTThe objectives of this research was to study the fruit production and quality of strawberry grown under different irrigation system. The trial was arranged in a completely randomized design. Four irrigation systems were examined: drip irrigation applied daily, drip irrigation applied every 2 days, manual irrigation applied daily and manual irrigation applied every 2 days . The result showed that the treatments gave significant effects on vegetative growth. Plants irrigated daily produced more runn er than those irrigated every 2 days. Plants irrigated manually every 2 days lowered flower production. The drip irrigation applied daily gave the highest response on growth and yield without any difference on fruit quality. The system could be considered for hydroponic strawberry production.Key words: strawberry, irrigation, fruit production, fruit quality

Abstract

Lactuca indica from Indonesia (Humbang Hasundutan) is a plant used to treat as antihyperlipidemia :Cholesterol, Trigliserida (TG), High Density Lippoprotein (HDL), Low Density Lippoprotein (LDL),on 28 male rats of the wistar strain (Rattus norvegicus) that have been given high-fat diets were given to3 control groups, namely rats given standard feed, groups of rats given standard and high-fat diets, andgroups of rats given simvastatin with treatment for 21 days. The groups with different doses were 100mg/kg bw, 200 mg/kg bw, 300 mg/kg bw and 400 mg/kg bw. Giving the ethanol extract of the leaves ofthe sijukkot plant has an effect on reducing total cholesterol levels. With the analysis obtained that theShapiro-Wilk Normality Test with a sig value > 0.05, the data is normally distributed. The results of theanalysis in the Test of Homogeneity of Variances table obtained a Sig value of 0.510. Cholesterol SigValue 0.510 > 0.05; TG 0.102>0.05; HDL 0.01 <0.05 and LDL 0.257> 0.05. Thus, the ethanolic extractof sijukkot leaves gave a positive effect on cholesterol levels in the serum lipid profile of High DensityLippoprotein (HDL and Low Density Lippoprotein (LDL).

Abstract

Saat ini termometer telah banyak digunakan oleh masyarakat. Pada umumnya termometer dirancanguntuk orang yang memiliki kondisi fisik normal terutama dalam kemampuan melihat. Orang cacat khususnyaorang buta, akan menemui kesulitan dalam menggunakan termometer yang ada. Berkaitan dengan masalahtersebut, makalah ini menjelaskan tentang perancangan termometer badan untuk mereka yang mengalamiketerbatasan dalam melihat.Secara umum termometer badan dengan output suara ini menggunakan sensor LM35D, ADC0804,mikrokontroller AT89C51 dan ISD 2590. Range pengukuran termometer ini adalah suhu 300C–420C. Hasilpengujian menunjukkan bahwa termometer yang telah didisain memiliki respon yang lebih cepat dalammencapai suhu tubuh dibandingkan dengan termometer air raksa. Waktu respon termometer ini adalah 5 menitdengan error output suhu adalah 0,190C. Output suara yang dihasilkan sangat jelas. Sehingga alat ini cukupbaik untuk digunakan sebagai termometer badan. Makalah ini akan menguraikan tentang rancangan sebuahTermometer badan dengan output suara, yang dapat digunakan untuk orang buta.Kata kunci: termometer, mikrokontroler MCS51, ISD 2590

Abstract

Banyak instansi yang sekarang ini telah menggunakan platform teknologi informasi untuk mengetahui tingkat kehadiran pegawainya. Dalam contoh kasus pada salah satu instansi pemerintah di Garut, data kehadiran pegawai dari setiap unit kerja yang tersebar di beberapa kecamatan dikirimkan oleh bagian personalia melalui surat elektronik ke kantor pusat. Penelitian yang dilakukan dimaksudkan untuk pengembangan sebuah sistem yang bisa menyajikan informasi presensi pegawai di seluruh unit kerja kepada pengguna informasi di kantor pusat. Metode pengembangan yang digunakan pada penelitian ini yaitu Rational Unified Proccess dengan menggunakan model Unified Modeling Language. Dalam penelitian ini dibuat sebuah sistem informasi yang dapat menyajikan informasi presensi pegawai berdasarkan data presensi dari fingerprint unit kerja yang lokasinya saling berjauhan. Platform memberikan kemudahan dalam pengolahan dan peyajian informasi kehadiran pegawai.

Abstract

Soil salinity could significantly reduce rice yield, therefore, varieties tolerant to salinity are urgent to be developed. Mutation induction could be used to create rice mutants tolerant to salinity. The study aimed to evaluate the tolerance of somaclonal mutants of rice to NaCl salinity in a greenhouse condition and characterize their tolerance mechanism. A total of 45 putative mutants were generated by a gamma ray mutation induction followed with in vitro selection in the growth media containing different NaCl concentrations in the greenhouse experiment. The study consisted of two-factor treatments, namely three levels of NaCl concentrations and 45 rice mutants suspected to be tolerant to salinity, arranged in a completely randomized design. Proline, cations (K, Na, Ca, and Mg) content, and stomata density were evaluated.  The results showed that eight mutants were tolerant to 150 mM NaCl, namely CH30, CH-4-2, II-13-42, II-13-7, II-13-10, II-13-13, II-13- 2, and IA-3-21. These tolerant mutants had a higher Na content compared to the check parent. The tolerant mutants had a high proline content, lower Na, and stable K, Mg and Ca cations as well as had a greater number of stomata and higher stomata length-width ratio. Some of the identified tolerant mutants demonstrated the tolerant mechanism against salinity stress. Further studies are required to evaluate these tolerant mutants in the field conditions under salinity stress.

Abstract

Delivering of Over-Expression Construct OsWRKY76Candidate Gene in Rice cv. Nipponbare throughAgrobacterium tumefaciens. Aniversari Apriana, AtmitriSisharmini, Wening Enggarini, Sudarsono, Nurul.Khumaida, and Kurniawan R. Trijatmiko. Plant geneticimprovement can be done through classical breeding orgenetic engineering. WRKY is a transcription factor involvedin regulating plant defense responses. OsWRKY76 gene islocated in a narrow segment of chromosome 9 which isidentified previously to be related to wide spectrumresistance in rice. A sequence of OsWRKY76 (+1.200 bp)has available in the gene bank and it makes possible toisolate, clone, and construct the gene into over-expressionvector. The aim of this research was to assemble an overexpressionconstruct of OsWRKY76 candidate gene andintroduce it into rice through Agrobacterium-mediatedtransformation. A construct of pCAMBIA-1301::35S::OsWRKY76 has been successfully assembled andtransformed into embryogenic calli of rice cv. Nipponbareusing A. tumefaciens strain Agl-1 and EHA 105. A number of126 independent lines has been produced, in which Agl-1showed 3.8 times more efficient than EHA 105. PCR analysisof randomly selected 25 independent lines showed that allof them positively contained hptII gene, a selectable markerused in the over-expression construct of the OsWRKY76candidate gene. Based on the result, it could be concludedthat the over-expression construct of OsWRKY76 candidategene have been successfully introduced into the tissue ofNipponbare.

Abstract

Effect of gamma irradiation on rice callus dependson the irradiation dose used. Irradiation dose is one of thefactors that affect the genetic changes in the cells of plants.High doses can result in tissue death, meanwhile low doseswill result in abnormal changes in plant phenotype. The levelof sensitivity of a plant tissue against gamma irradiation isdifferent. This study aimed to evaluate the growth andregeneration of callus in various irradiation doses of gammaray. The plant materials used were mature embryos of ricevar. Ciherang and Inpari 13. This study used a completelyrandomized design with gamma irradiation treatment atdoses of 10, 20, 30, 40, 50, 60, 70, 80, 90, and 100 Gy. Eachtreatment consisted of 10 replicates with 5 embryogenic calliper bottle. The results showed that the increasing doses ofgamma irradiation increased the percentage of dead callus,inhibited callus growth, and its ability to regenerate. Thehigh percentages of callus of Ciherang and Inpari 13 thatformed green spots and adventitious shoots were mostlyobtained from controls. The percentages decreased atirradiation doses of 10, 20, 30, and 40 Gy. Moreover, the calliof Ciherang and Inpari 13 were not able to form adventitiousshoots at irradiation doses higher than 40 Gy and 30 Gy,respectively.

Abstract

ABSTRAKKendala dalam pengembangan jahe di Indonesia adalah terbatasnyabenih bermutu. Secara konvensional, budidaya jahe dilakukan denganmenggunakan bibit dari potongan rimpang. Dengan cara ini diperlukanbibit dalam jumlah yang banyak, antara 2-3 t/ha untuk jahe yang dipanentua dan 5-6 t/ha untuk yang dipanen muda. Kendala lain adalah penyakittular benih layu bakteri yang disebabkan oleh Ralstonia solanacearum.Salah satu upaya yang dapat dilakukan untuk mendapatkan benih jahebebas penyakit adalah perbanyakan melalui kultur jaringan. Penelitianbertujuan untuk mengkaji sumber eksplan dari tingkat umur panenrimpang yang berbeda terhadap kapasitas pembentukan kalus embriogenikpada kultur meristem jahe putih besar. Penelitian dilakukan di BalaiPenelitian Tanaman Obat dan Aromatik dari September 2007 sampaiMaret 2008, menggunakan rancangan acak lengkap dengan 20 kaliulangan. Bahan tanaman yang digunakan adalah meristem jahe putih besaryang diambil dari rimpang panen muda dan tua. Peubah yang diamatimeliputi: histologi jaringan, persentase kalus embriogenik yang terbentuk,bobot segar kalus, diameter kalus, dan morfologi kalus. Hasil penelitianmenunjukkan adanya daerah meristematik pada sayatan eksplan meristemjahe putih besar ukuran ± 0,25 cm. Persentase kalus embriogenik (92,1%)dan diameter kalus (0,59 mm) dari rimpang yang dipanen tua lebih tinggidari yang dipanen muda. Berat kalus (1,18 g) dan jumlah embrio somatikglobular (29,34) asal eksplan panen tua nyata lebih tinggi dari yangdipanen muda. Kalus embriogenik yang berasal dari eksplan rimpang yangdipanen tua mampu berkembang membentuk embrio somatik danberkecambah menghasilkan planlet normal.Kata kunci : Zingiber officinale Rosc., umur rimpang, kalus embriogenik,embriogenesis somatikABSTRACTEffect of explants age on the success of embryogenic calliformation in meristem culture of ginger (Zingiberofficinale Rosc.)Constraint in ginger cultivation in Indonesia is the limited qualityof planting materials. In conventional cultivation, planting materials weretaken from a piece of rhizomes. By this technique, significant amount ofplanting materials is required, between 2-3 tons/ha for fully harvested and5-6 tons/ha for young harvested rhizomes. Another serious constraint isbacterial wilt disease infection caused by Ralstonia solanacearum. Effortfor obtaining free disease planting materials could be performed throughtissue culture mass propagation. In this study, different ages of rhizome asexplants sources was evaluated for their capacity in embryogenic calliformation on the meristem culture of ginger. The experiment wasconducted in Indonesian Medicinal and Aromatic Crops Research Institutefrom September 2007 to March 2008, using a completely random designwith 20 replicates. Plant material used was white ginger meristem takenfrom the fully and young harvested rhizomes. The observed variables wereexplant histology, percentage embryogenic calli formation (%), freshweight of calli, calli diameter, number of globular embryo, and callimorphology. The results showed a meristematic region at the incisionexplant big-white ginger meristem ± 0.25 cm in size. Percentage ofembryogenic calli formation from the fully harvested rhizome-explant(92.1%) and calli diameter (0.59 mm) were higher than that of the youngerone. Calli weight (1.18 g) and number of globular somatic embryos(29.34) from fully harvested rhizome-explants were significantly higherthan that of the younger one. Embriogenic calli derived from the oldharvested rhizome explants was able to grow well to form somaticembryos and then germinate to produce normal plantlet.Key words : Zingiber officinale Rosc, age of rhizome, embriogeniccalli, somatic embryogenesis

Abstract

ABSTRAKPenyakit layu bakteri yang disebabkan oleh Ralstonia solanacearummerupakan kendala utama budidaya jahe, yang menyebabkan kehilanganhasil lebih dari 90%. Upaya pengendalian yang dilakukan belum optimal,karena tidak tersedianya varietas jahe tahan patogen tersebut. Kendalautama untuk memperoleh varietas jahe yang tahan adalah terbatasnyasumber gen ketahanan dan adanya hambatan fisiologis pada prosespersilangan jahe karena sifat inkompatibilitas sendiri, serta rendahnyafertilitas polen menyebabkan persilangan jahe secara konvensional sulitdilakukan. Seleksi in vitro menggunakan medium selektif yangmengandung filtrat patogen merupakan salah satu metode inkonvensionaluntuk meningkatkan ketahanan tanaman. Penelitian ini dilakukan diLaboratorium Kultur Jaringan dan Laboratorium Penyakit Balai PenelitianTanaman Obat dan Aromatik (Balittro) dari bulan April 2008 sa,mpaiOktober 2008 dengan tujuan untuk mengetahui tingkat ketahanan jahepada stadia kalus terhadap filtrat R. solanacearum dan memperolehkonsentrasi filtrat yang tepat sehingga diperoleh varian kalus baru tahanterhadap filtrat patogen tersebut. Kalus embriogenik jahe putih besar asaleksplan meristem berumur 8 minggu, diseleksi selama 3 minggu di dalammedium proliferasi (MS + 3% manitol tanpa zat pengatur tumbuh),mengandung filtrat R. solanacearum. Seleksi bertingkat dilakukan denganmengaplikasikan filtrat R. solanacearum pada konsentrasi berbeda, yaitu:0; 0,1; 0,2; 0,3; 0,4; 0,5; 1; 2; 3; 4; dan 5%, pada tahap pertama. Padaseleksi tahap kedua, kalus disubkultur ke dalam media yang sama dengankonsentrasi filtrat dinaikkan 10 kali dari konsentrasi awal. Penelitianmenggunakan rancangan acak lengkap, diulang 10 kali. Hasil penelitianmemperlihatkan penggunaan filtrat R. solanacearum di dalam mediumkultur in vitro jahe pada seleksi tahap pertama dan kedua menyebabkanterjadinya perubahan warna kalus dari putih kekuningan menjadi kuningkecoklatan dan coklat kehitaman. Berat dan diameter kalus, jumlahembrio globular serta embrio torpedo berkurang secara nyata setelahperlakuan filtrat, pada seleksi tahap pertama maupun kedua seiring denganbertambah  tingginya  konsentrasi  filtrat.  Konsentrasi  filtrat  R.solanacearum  yang  mampu  menginduksi  dan  menyeleksi  kalusembriogenik berkisar antara 0,3 - 2% dari volume medium seleksi kaluspada seleksi tahap 1 dan 3 - 20% pada seleksi tahap 2.Kata kunci : Zingiber officinale Rosc., kalus, seleksi in vitro,ketahanan, filtrat R. solanacearumABSTRACTThe use of R. solanacearum filtrate in callus selection ofin vitro for ginger resistance to bacterial wilt diseaseBacterial wilt disease caused by Ralstonia solanacearum is the mainconstraint in ginger cultivation. It often causes significant yield loss ofmore than 90%. Various controlling techniques are not able to overcomethe disease, due to unavailability of resistant ginger cultivar. Limitation inobtaining resistant ginger variety is caused by several factors includingthe lack of resistant gene, physiological barrier due to the selfincompatibility, and low pollen fertility, these cause difficulty inconventional cross breeding. Therefore, genetic variability enhancementhas to be carried out unconventionally, to obtain ginger variety resistant tothe disease. In vitro selection using a selective medium containing filtrateof the pathogen is one of the potential unconventional method to improveginger plant resistance. The study was conducted at Meristem Culture andPlant Disease Laboratories of IMACRI from April to October 2008 aimingat determining the level of resistant ginger on stage of calli to the filtrateof R. solanacearum and to obtain an appropriate concentration of thefiltrate which induced calli variants resistant to the filtrate. Large whiteginger embryogenic calli meristems of 8 weeks old were selected for 3weeks in proliferation medium (MS + 3% mannitol without growthregulators), containing filtrate of R. solanacearum. For that purpose, twostages of in vitro selection were performed by applying differentconcentrations of R. solanacearum filtrate e.g; 0; 0.1; 0.2; 0.3; 0.4; 0.5; 1;2; 3; 4; and 5% at the first stage selection. Those concentrations were thenmultiplied 10 times at the second stage selection. Experiments werearranged in completely randomized design with 10 replicates. Resultsshowed that the use of R. solanacearum filtrate as selection agent in gingerin vitro culture medium has caused changes in calli color from theyellowish white into the blackish brown. In addition, increase of R.solanacearum filtrate concentration at the 1 st and 2 nd selection stages wasin line with the decreased of the calli weight and diameter, as well asnumber of globular and torpedo embryo. The concentration of R.solanacearum filtrate applied at 0.3 to 2% in the 1 st selection followed by3 to 20% in the 2 nd  selection induced resistant embryogenic calli of ginger.Key words : Zingiber officinale Rosc., calli, in vitro selection,resistance, R. solanacearum filtrate

Abstract

The Sequence of Horizons of Paddy Soils in a Toposequence of Latosol on Volcanic Parent Materials in the Bogor – Jakarta AreaABSTRACT. The sequence of horizons of paddy soils is different from non paddy cultivated. The aim of the research were: (1) to study the sequence of horizons of paddy soils in a toposequence of Latosol on Volcanic materials in the Bogor – Jakarta Area; (2) to study the sequence of paddy cultivated intensity to sequence of horizons. Twelve pedons of different altitude (90 – 650 m above sea level) and different paddy cultivated intensity (0/year, 1/year, 2/year) were investigated sequence of horizons in the field. The result of the research showed that the horizons sequence, on non paddy cultivated soils consist of: tillage layers and the original soil layers. On 1x paddy/year consist of: plow pan layer, and iron/manganese/nodule Mn illuviation layer. On 2x paddy /year consist of: tillage layer of surface reduction, plow pan layer, Fe/Mn illuviation layer, and reduce subsurface layer.
