Abstract

(The Pedological Features of Paddy Soils from a Toposequence on Volcanic Parent Materials in the Bogor – Jakarta Area)ABSTRACT. The pedological feature of paddy soils are different from the non paddy cultivated. The aim of the research were: to study the pedofeatures of paddy soils in a toposequence on volcanic materials in the Bogor – Jakarta area; to study the influence of paddy cultivated intensity of pedofeatures. Twelve pedons of different altitude (90 – 650 m above see level) and different paddy cultivated intensity (0/year, 1/year, 2/year) were investigated, each horizons were sampled for laboratory analysis to study the pedofeatures. The result of the research showed that on non paddy cultivated, pedofeature observed at B horizon is indicating rock weathering. On 1x paddy, at tillage layer nodules and hypocoating were found. On 2x paddy, at plow plan layer clay infilling dense complete was found.

Abstract

The objectives of this study were to characterize and evaluate the physicochemical properties of 10 cassava breeding genotypes developed by cassava crop improvement of (Institut Pertanian Bogor) IPB research group. The physicochemical characterization was performed by measuring water content, ash, protein, fat, carbohydrate, pasta (pasting properties), starch total, amylose content, and whiteness. The results showed that each genotype has different chemical content and gelatinization properties. The highest water content (11.48 ± 0.12 %) was found in genotype V4D0 (variant genotype Malang 4), ash (1.83 ± 0.02 %) in V5D2 (1) (A variant of genotype Malang 4-1), fat (1.62 ± 0.93 %) and protein (4.73 ± 0.19 %) in V3D1 (1) (variant UJ 5-1). Genotype V1D1-1(1) (A variant of genotype Ratim-1) has the highest amylose (23.33 ± 0.04 %) and the lowest ones (13.13 ± 0.48 %) was found in genotype V2D1-1(3). Genotype V2D0 has the highest total starch (88.67 %) and the lowest (74.3%) was obtained in V5D2 (1). The highest whiteness value was measured in genotype V1D0 i.e. 93.13 %. For pasting properties testing, it was measured that genotype V2D1-1 (3) had the lowest peak viscosity i.e 4,006 cP and the high of end of viscosity occurred 2,592 cP in genotype V4D2-1 (2) (A variant of genotype Adira 4-1), were correlated with high amylose content owned flour is 22.03 ± 0.25%. ABSTRAKPenelitian ini bertujuan untuk mengkarakterisasi dan mengevaluasi sifat fisikokimia 10 genotipe ubi kayu hasil pemuliaan team crop improvement ubi kayu Institut Pertanian Bogor (IPB). Karakterisasi fisikokimia dilakukan dengan cara melakukan analisis kadar air, abu, protein, lemak, karbohidrat, pasta (pasting properties), total pati, kadar amilosa, dan derajat putih. Dari hasil karakterisasi diperoleh bahwa setiap genotipe ubi kayu memiliki kandungan kimia dan sifat gelatinisasi yang berbeda-beda. Kadar air tertinggi dimiliki genotipe V4D0  (Genotipe Malang 4) 11,48 ± 0,12 % , kadar abu dan lemak tertinggi adalah genotipe (V5D2 (1) (Varian dari genotipe Malang 4-1) yaitu 1,83 % ± 0,02 dan 1,62 ± 0,93 % serta kadar protein tertinggi adalah V3D1 (1) (Varian UJ 5-1) 4,73 ± 0,19 %. Genotipe V1D1-1 (1) (Varian dari genotipe Jame-1) memiliki kadar amilosa tertinggi yaitu 23,33 ± 0,04 % sedangkan yang terendah dihasilkan oleh genotipe V2D1-1 (3) (Varian dari genotipe Ratim-1) yaitu 13,13 ± 0,48 %. Genotipe V2D1-1 (3) (Varian genotipe Ratim-1)  memiliki viskositas puncak terendah yaitu 4.006 cP berkorelasi positif dengan kandungan amilosa paling rendah yang dimilikinya. Viskositas akhir yang tinggi terjadi pada genotipe V4D2-1(2) (Varian genotipe Adira 4-1) yaitu 2.592 cP yang berkorelasi dengan tingginya kandungan amilosa yang dimiliki tepung tersebut yaitu 22,03 ± 0,25 %.

Abstract

Dieback disease on jabon seedlings is caused by Botryodiplodia theobromae decreased seedlings quality and economic benefits of nurseries. This is still sparse research for resistance test between white and red Jabon. This research aimed to test B. theobromae attacked with non wounded and wounded stem infection methods in Jabons and to obtain the most resistance of Jabons to B. theobromae attacked. The results showed that the disease incidence and the disease severity on both control of Jabon seedlings were 0% and 0%. But on treatment, white jabon seedlings non-wounded and wounded stem infection were 30% and 100%. For red jabon seedlings non-wounded and wounded stem infection were 80% and 100%. The disease severity on white jabon seedlings non-wounded and wounded stem infection were 12% and 62%. For red jabon seedlings non-wounded and wounded stem infection were 16% and 38%. The disease incidence on red Jabon seedlings were wider than white Jabon seedlings, but for the disease severity, white Jabon seedlings were worse than red Jabon seedlings. Botryodiplodia theobromae attacked non-wounded and wounded stem infection on Jabons.Key words: nursery, stem infection methods, the disease incidence, the disease severity

Abstract

Desa Payageli di Kabupaten Deli Serdang merupakan salah satu sentra penghasil handicraft berupa tas, topi dan dompet yang memanfaatkan bahan baku dai enceng gondok dan lidi sawit. Produk-produk tersebut telah dikenal oleh wisatawan mancanegara antara lain yang berasal Malaysia, SIngapura, Jepang, Belanda dan Perancis; sehingga berpotensi menjadi komoditas ekspor unggulan dari Kabupaten Deli Serdang. Selama ini kendala yang dihadapi oleh para pengrajin adalah kapasitas dan kualitas produk yang belum optimal sehingga produk handicraft tersebut sulit untuk menembus pasaran luar negeri. Kendala lainnya adalah masih lemahnya kompetensi sumber daya manusia baik khususnya dari sisi manajerial bagi pemenuhan mutu produk berstandar internasional yang menjadi hambatan dalam menjalin kerjasama dengan investor asing. Kegiatan ini bertujuan untuk meningkatkan ketrampilan para pengrajin, maupun manajemen produksi dan pemasaran sehingga dapat menjadikan produk handicraft dari Desa Payageli ini sebagai komoditas ekspor unggulan. Melalui kemitraan dengan Lembaga Pengabdian Kepada Masyarakat Universitas Negeri Medan (LPM UNIMED), optimalisasi manajemen produksi handicraft para pengrajin handicraft ini dicapai dengan pelaksanaan serangkaian pelatihan peningkatan mutu dan desain produk, aplikasi mesin pengerik sebagai teknologi tepat guna dalamproduksi handicraft, serta pelatihan pemasaran berbasis system informasi kepada pengrajin handtcraft di Desa Payageli Kabupaten Deli Serdang.

Abstract

Banyak instansi yang sekarang ini telah menggunakan platform teknologi informasi untuk mengetahui tingkat kehadiran pegawainya. Dalam contoh kasus pada salah satu instansi pemerintah di Garut, data kehadiran pegawai dari setiap unit kerja yang tersebar di beberapa kecamatan dikirimkan oleh bagian personalia melalui surat elektronik ke kantor pusat. Penelitian yang dilakukan dimaksudkan untuk pengembangan sebuah sistem yang bisa menyajikan informasi presensi pegawai di seluruh unit kerja kepada pengguna informasi di kantor pusat. Metode pengembangan yang digunakan pada penelitian ini yaitu Rational Unified Proccess dengan menggunakan model Unified Modeling Language. Dalam penelitian ini dibuat sebuah sistem informasi yang dapat menyajikan informasi presensi pegawai berdasarkan data presensi dari fingerprint unit kerja yang lokasinya saling berjauhan. Platform memberikan kemudahan dalam pengolahan dan peyajian informasi kehadiran pegawai.

Abstract

The problem of this research are (1) is there an influence of work motivation toward employees’ work of District General Services at Mayjen HM. Ryacudu Hospital of North Lampung; (2) is there an influence of work discipline toward employees’ work of District General Services at Mayjen HM Ryacudu Hospital of North Lampung; and (3) is there an influence of work motivation and discipline of work toward employees’ work of District General Services at Mayjen HM Ryacudu Hospital of North Lampung.            The objective of this research is to analyze and to know: (1) the influence of work motivation toward employees’ work of District General Services at Mayjen HM Ryacudu Hospital of North Lampung; (2) the influence of work discipline toward employees’ work of District General Services at Mayjen HM Ryacudu Hospital of North Lampung; and (3) the influence of work motivation and the discipline of work toward employees’ work of District General Service at Mayjen HM Ryacudu Hospital of North Lampung.            The hypothesis of this research are:  (1) motivation of work has a positive influence toward employees’ work of District General Services at Mayjen HM Ryacudu Hospital of North Lampung; (2) the discipline of work has a positive influence toward employees’ work of District General Services at Mayjen HM Ryacudu Hospital of North Lampung and (3) work motivation and the discipline of work influential work together toward employees’ work of District General Services at Mayjen HM Ryacudu Hospital of North Lampung.            This research uses explanatory Survey with analyze the influence of work motivation and the discipline of work as independent variables and employees’ work of District General Services at Mayjen HM Ryacudu Hospital of North Lampung as dependent variable. The population of this research consists of 575 employees and the sample of this research consists of 58 employees (10%). The technique of collecting data of this research uses questioner, interview, observation, and documentation. The technique of data analysis uses statistic Multiple Linear Regression.            The result of this research indicates that (1) work motivation has a positive influence and significant toward the employees’ work of District General Services at Mayjen HM Ryacudu Hospital of North Lampung; (2) the discipline of work has a positive and significant toward employees’ work of District General Services at Mayjen HM Ryacudu Hospital of North Lampung; and (3) work motivation and the discipline of work have a positive influence and significant toward employees’ work of District General Service at Mayjen HM Ryacudu Hospital of North Lampung simultaneously.

Abstract

Smooth Cayenne is the largest pineapple type cultivated in Indonesia, but its vegetative planting materials for mass propagation are limited. Somatic embryogenesis is a potential method to be applied. The aim of this study was to investigate the somatic embryogenesis regeneration under the effect of picloram and light. Callus formation was induced by picloram (21, 41 and 62 μM) added with 9 μM thidiazuron. The calli were transferred onto MS or Bac medium  enriched with N-organic compounds with or without addition of 21 μM picloram under dark or light condition. The compact calli were subcultured onto MS medium supplemented with 4.65 μM kinetin, while the friable calli were  transferred onto BIG medium (modified MS + 1.1 μM benzyl adenine + 0.9 μM indole butyric acid + 0.09 μM giberelic acid) or B medium (MS + 0.018 mM benzyl adenine). The results showed that the events of somatic embryogenesis were started from cell polarization, asymmetrical division, proembryo formation as  embryogenic tissues and friable embryogenic tissues, and embryo development. The best treatment for callus induction was 21 μM picloram. The addition of 21 μM picloram on N-organic enriched medium and the use of light condition  proliferated embryogenic calli. The N-organic enriched Bac medium and light condition yielded the highest number of mature somatic embryos (17 embryos perexplant in 2 months). The B medium was better than BIG medium to develop  somatic embryos from friable embryogenic tissues. The somatic embryogenesis method presented is potential for pineapple mass propagation and artificial seedproduction.Abstrak Bahasa IndonesiaSmooth Cayenne merupakan kultivar nenas yang banyak dibudidayakan di  Indonesia, namun ketersediaan benih untuk perbanyakan massal masih terbatas. Embriogenesis somatikadalah metode yang potensial untuk produksi bibit secara massal. Tujuan penelitian adalah untuk mempelajari pengaruh pikloram dan pencahayaan terhadap regenerasi embriogenesis somatik nenas. Kalus diinduksi menggunakan pikloram (21, 41, dan 62 μM) dan penambahan thidiazuron 9 μM. Selanjutnya, kalus dipindahkan ke media MS atau Bac yang diperkaya dengansenyawa N-organik dengan atau tanpa penambahan pikloram 21 μM dalam kondisi gelap atau dengan pencahayaan. Kalus kompak disubkultur pada media MS yang mengandung kinetin 4,65 μM, sedangkan kalus remah dipindahkan ke media BIG (MS modifikasi + bensil adenin 1.1 μM + indole butyric acid 0,9 μM + giberelic acid 0,09 μM) atau media B (MS + bensil adenin 0,018 μM). Hasil penelitian  menunjukkan bahwa tahapan embriogenesis somatik diawali dengan polarisasi sel, pembelahan asimetris, pembentukan proembrio sebagai jaringan embriogenik danjaringan embriogenik remah, serta perkembangan embrio. Perlakuan terbaik untuk induksi kalus adalah pikloram 21 μM. Penambahan pikloram 21 μM pada media yang diperkaya dengan senyawa N-organik mampu meningkatkan jumlah kalusembriogenik. Media Bac yang diperkaya senyawa N-organik dan kondisi pencahayaan menghasilkan jumlah embrio somatik dewasa terbanyak (17 embrio per eksplan dalam 2 bulan). Media B lebih baik daripada media BIG untuk regenerasi embrio somatik dari jaringan embriogenik remah. Metode embriogenesis somatik yang dihasilkan dari penelitian ini berpotensi diterapkan untukperbanyakan massal dan produksi benih nenas.

Abstract

Influence of Gamma Rays Irradiation on First Generation (M1) to Obtain New Promising Drought-Tolerance Soybean Genotype. Breeding to produce drought tolerant varieties is one of main strategies in the soybean production improvement. Some limitations in soybean conventional breeding are that it takes long time and that drought tolerant characters are rarely found. Therefore, the improvement of genetic variability is very important to assist drought tolerant. Mutation through gamma irradiation is one of techniques to improve genetic variability. The aims of this research were to obtain LD50, study the influence of irradiation, and genetic of mutation. To determine the LD50, seeds of soybean cv. Anjasmoro were irradiated with 0-800 Gy (100 interval) gamma rays. LD50 was calculated to be 202.5 Gy base on germination rate. Therefore, in further experiment 200-400 Gy (50 interval) dosage were used to irradiate seeds of soybean cv. Anjasmoro and Burangrang. All irradiation dosages (200-400 Gy) damage protoplast, decrease plant survival, prolong flowering dan harvest time, decrease plant height, number of total and filled pod, number of seeds, and improve the number of empty pod.

Abstract

Overexpression of alanine aminotransferase (AlaAT) gene can improve nitrogen use efficiency (NUE) in plants. The previous isolated AlaAT genes cannot be freely applied to generate NUE plants due to IPR restriction. Therefore, isolation of the gene from targeted monocot and dicot plants is necessary. The objectives of this study were to isolate AlaAT genes from barley, foxtail millet, cucumber, and tomato and analyze their homology to other isolated AlaAT genes in sequence databases (gene bank). Total RNA was isolated from roots of barley, foxtail millet, cucumber, and tomato, and then converted into cDNA using reverse transcription method. The cDNA was then cloned into pGEM®-T Easy plasmid and the verified clones were sequenced. The amino acid sequences were analyzed for their homologies using Clustal Omega software and phylogenetic tree was constructed. The results showed that the amino acids of AlaAT gene from barley was different from AlaAT genes of tomato and cucumber with homology level less than 80%. Phylogenetic tree predicted that AlaAT genes clustered into three groups with AlaAT genes of foxtail millet and barley clustered in one group together with other monocots in group I. AlaAT genes derived from dicots clustered into two groups in which AlaAT gene of tomato clustered in group II, while that of cucumber was in group III. The identity differences of AlaAT gene of tomato and that of cucumber as well as the estimates of the same enzymatic functions can open up enormous opportunities in genetic engineering research for the development of NUE rice.
