Abstract

Indonesia is highly susceptible to earthquakes, with the southern and western coasts of Java and Sumatra being the most vulnerable regions. Due to this vulnerability, it is necessary to establish a culture of disaster mitigation in the most fertile and heavily populated islands to reduce the number of fatalities and economic losses caused by earthquakes. One of the promising real-world seismic base isolation methods is using a rubber seismic bearing constructed of rubber and metal layers. This study aimed to examine the typical behavior of natural rubber compounds subjected to various sulfur-curing processes as basic designs in the manufacture of rubber seismic bearings. The experiment was carried out by arranging the sulfur curing system into three categories, namely an efficient which applied N-cyclohexyl-2-benzothiazole sulfonamide (CBS)/Sulfur (S) ratio as 1.4/0.15 and 1.4/0.25, semi-efficient with CBS/S ratio of 1.4/1.4 and 1.4/1.7, and conventional with CBS/S ratio of 1.4/3.0 and 1.4/3.5. The results showed that the ideal modification condition for NR macromolecule chain found in seismic-bearing rubber compound was a semi-efficient sulfur curing system with a CBS/S ratio of 1.4/1.4. It suggested that a semi-efficient sulfur curing system was appropriate for developing rubber compound formulation for seismic rubber bearings, mainly for low-damping types.

Abstract

Diabetes mellitus (DM) is a chronic metabolic disease distinguished by disrupted glucose metabolism, causing elevated blood sugar levels. One of the latest therapeutic strategies involves inhibiting dipeptidyl peptidase-4 (DPP-4) to regulate glucose metabolism. Emodin, a bioactive compound, has shown potential as a DPP-4 inhibitor, but its efficacy requires further research. This study aims to identify and assess emodin and its derivatives as potential DPP-4 inhibitors through a comprehensive in silico and in vitro analysis. Molecular docking analysis revealed that 3-ρ-toluoyl emodin (ρTE) had the lowest binding energy (-111.4 kcal/mol) among the tested compounds. Furthermore, in vitro testing showed consistent results in silico, indicating that ρTE had significant inhibitory activity with an IC50 value of 1.37 μM. Pharmacokinetic and physicochemical evaluations confirmed ρTE’s potential as a safe antidiabetic drug candidate. The research findings indicate that ρTE holds potential as a promising drug candidate for further development.

Abstract

This study aimed to investigate the effectivity of goat milk fermentation as tyrosinase inhibitory with Lactobacillus rhamnosus TW 2. The examination of fresh milk contained the density, protein, and fat content. The culture of Lactobacillus rhamnosus TW 2 acid lactid bacteria starter was added with the concentration of 3, 4, and 5%, inoculated in pasteurized goat milk, then incubated at 37 C for 24 hours. Fermented milk were extracted by centrifugation, then supernatant was collected and was used for inhibition of tyrosinase enzymes activity on L-tyrosin and L-dopa substrate. The result showed that the density, fat content, and protein content of Etawah crossbred goat milk are 1.028, 3.73, and 5.45%, respectively. Re-identification of lactic acid bacteria showed similar morphology, physiology, and bio-chemistry with the isolated lactic acid bacteria. The growth curve of TW 2 was observed in 12 hours. The 5 % of Lactobacillus rhamnosus TW 2 was the best concentration to inhibit tyrosinase activity in L-Tyrosin substrate. In conclusion, fermentation of goat milk using Lactobacillus rhamnosus TW 2 at concentration of 5% as starter is effective to inhibit tyrosinase activity significantly.Key words: goat milk, fermentation, tyrosinase inhibitory

Abstract

Penelitian ini bertujuan untuk membandingkan aktivitas antioksidan, antibakteri, dan uji toksisitas dari berbagai jenis ekstrak kulit batang pohon kesambi (Schleichera oleosa (Lour)Oken). Tahapan penelitian meliputi proses esktraksi kulit batang kesambi dari kecamatan Mancak Serang dengan teknik maserasi bertingkat menggunakan 3 pelarut : Metanol, n-heksan, dan etil asetat. Ekstrak tersebut kemudian diuji aktivitas antioksidan, antibakteri, dan toksisitasnya. Hasil pengujian antioksidan dengan metode DPPH, antibakteri dengan metode difusi agar, dan uji toksisitas dengan metode BSLT didapat hasil penelitian menunjukkan bahwa ekstrak etil asetat memiliki aktivitas antioksidan (IC50 7,723 ppm) yang lebih baik dibandingkan dengan ekstrak metanol (IC50 7,801 ppm), dan ekstrak n-heksan (IC50 8,568 ppm). Aktivitas antibakteri menunjukkan adanya kemampuan untuk menghambat pertumbuhan bakteri Streptococcus aureus dibandingkan bakteri Escherichia coli pada konsentrasi 10000 ppm, dan hasil uji toksisitas menunjukkan bahwa ekstrak etil asetat memiliki aktivitas yang lebih baik (LC50 305,17 ppm) dibandingkan ekstrak n-heksan (LC50 374,96 ppm) dan ekstrak metanol (LC50 431,26 ppm).

Abstract

Gelatin has been widely used as an additive in food industry pharmaceutical, and cosmetic. The similar physical appearance between bovine and porcine gelatin causes an issue for some communities like a Muslim due to awareness of halal food. This study aims to produce gelatin from femur bones of bovines with acid hydrolysis and their characteristics compared to standard gelatin of bovine and porcine. Bovine and porcine bones were soaked in 5% HCl for 10 days and every 2 days a HCl solution was replaced to get ossein. Ossein is hydrolyzed by gradual heating at 65, 75, and 85oC. Gelatin confirmed by the physico-chemical characters, FT-IR and analysis amino acid with HPLC.The results showed that the yield of bovine gelatin was 4.33%. The physico-chemical characters of bovine gelatin resulting from isolation and bovine gelatin standards are in conformity with porcine gelatin standards and meet the requirements of SNI 06-3735-1995 and GMIA. Therefore, bovine gelatin is specifically capable of substituting porcine gelatin for application in the pharmaceutical field. The FTIR spectrum of bovine gelatin shows the presence of amide A, amide I, amide II and amide III groups. The amino acid characters of gelatin were identified as glycine (13.57%) and proline (1.62%) for bovine gelatin and glycine (0.51%) and proline (0.09%) for porcine gelatin.

Abstract

Indonesia is one of the largest palm oil producers in the world. The availability of oil palm trunk as a waste of the final crop-harvesting is enormous, inexpensive, and can be a source of sustainable alternative wood for various applications if it can be utilized properly. The weaknesses of oil palm wood characteristics are low in specific gravity, as well as physical and mechanical properties compared to other biomaterial especially wood. This paper studies the oil palm wood quality improvement through compregnation technique using tannin recorcinol formaldehyde (TRF). The study was carried out by coating sample surface of oil palm wood (5 cm × 5 cm × 2 cm) with TRF adhesive composition 1:0.05:0.05 (v/v/v) and followed by cold pressing for 10 mins at 10 kg/cm2 pressure and then followed by hot pressing at 120oC and pressure of 12 kg/cm2 for 10 minutes. Density, hardness, thickness swelling as well as formaldehyde emission, were measured and compared to controls. Results showed that the compregnated wood significantly increased its density by 104.61%, and its hardness by six folds, and the thickness swelling decreased by 85.98%. The compregnated oil palm wood has also increased in strength quality, i.e from strength class V to III.

Abstract

Oil palm trunk is lower in dimensional stability, density, physical, and mechanical properties than other wood species. Efforts to overcome the weakness of the oil palm trunk properties could be conducted through densification with hot pressing and chemical adhesive addition. This paper studies optimum composition of Tannin Resorcinol Formaldehyde (TRF) in its application for improving the quality of oil palm trunk. The tannins were water extracted at 75°C and further copolymerized with resorcinol and formaldehyde. TRF was studied through functional group analysis using infrared spectroscopy and cristallinity test using X-ray diffraction. The TRF adhesive was applied through compregnation process. Results showed that reactivity of tannins in bark of Acacia mangium was 65.82%, the optimum adhesive composition of TRF (v/v) was = 1:0.05:0.05 with solid content of 8.33%, 0.09% free formaldehyde, and the degree of crystallinity was 10.92%. Tannin extract has a specific character on the wave number corresponding to the characteristics possessed by the imported tannin acacia standard. The presence of new functional groups and an increase in absorbance intensity of uptake at several wave numbers through infrared spectroscopy analysis on TRF as well as compregnated oil palm trunk indicate the occurrence bonds attributted by ether and methylene bridges to TRF and chemical compound on oil palm trunk. Oil palm trunk after compregnation significantly increased its density by 104.61%, hardness became 6 fold, and decreased the thickness swell to 85.98%. The oil palm trunk after compregnation also increased in quality from a wood strength class V to a wood strength class III, then, its potential for manufacturing exterior products.

Abstract

Biflavonoids are a distinctive class of dimeric flavonoids known for their diverse biological activities and chemotaxonomic significance. In this study, two biflavonoids were isolated from the acetone extracts of Araucaria hunsteinii twigs and Araucaria columnaris leaves collected from Bogor Botanical Garden, Indonesia. Chromatographic techniques, including Sephadex LH-20 column chromatography and preparative thin-layer chromatography, were employed for purification, followed by structural elucidation using LC-MS/MS and 1D/2D NMR spectroscopy. The compounds were identified as 4',4''',7,7''-tetra-O-methylcupressuflavone (1) and 7-O-methylcupressuflavone (2). Notably, this is the first report of 7-O-methylcupressuflavone isolated from A. columnaris leaves, providing new chemotaxonomic insights into the genus Araucaria. A literature-based pharmacological analysis revealed promising cytotoxic and α-glucosidase-inhibitory activities of the isolated compounds. These findings contribute to the phytochemical profiling and highlight the pharmaceutical potential of Araucaria-derived biflavonoids.
