Abstract

Provinsi Gorontalo merupakan daerah penghasil jagung (Zea mays L.) di Indonesia, diantaranya jagung varietas unggul dan jagung lokal. Jagung lokal Gorontalo memiliki kenampakan morfologi yang hampir mirip, terutama jagung doti dan siropu. Perbedaan dari jagung lokal ini diantaranya dapat dilihat pada warna tongkol, warna batang dan bentuk biji. Secara anatomi memiliki perbedaan dalam ukuran berkas pengangkut batang, daun, dan jumlah stomata pada jagung doti dan siropu pada berbagai usia. Penelitian ini membandingkan berkas pengangkut batang, daun, tipe stomata dan jumlah stomata tanaman jagung doti dan siropu lokal Gorontalo pada berbagai usia. Tujuan dari penelitian ini adalah membandingkan berkas pengangkut batang,daun, tipe stomata dan jumlah stomata tanaman jagung doti dan siropu lokal Gorontalo pada berbagai usia. Indikator yang diamati dalam penelitian ini perbandingan berkas pengangkut batang,daun, tipe stomata dan jumlah stomata tanaman jagung doti dan siropu lokal Gorontalo pada berbagai usia. Hasil penelitian ini menunjukkan adanya perbedaan yang signifikan dalam struktur jaringan berkas pengangkut batang, daun jagung doti dan siropu pada berbagai umur yang berbeda menunjukkan adanya adaptasi dalam transportasi air, dan nutrisi. Stomata pada jagung doti dan siropu ditemukan di dikedua sisi daun adaksial (atas) dan abaksia (bawah) daun, dengan tipe stomata graminae.

Abstract

This study aimed to identify the relationship between floret length and the developmental stages of microspores in banana male inflorescence (Musa spp. cv. Tanduk), and to determine the optimal floret size range as an explant source for initiating microspore culture. Ten bract layers were analyzed, with five florets randomly selected from each layer and measured for their length. Anthers were extracted from each floret, crushed in sterile water, and observed microscopically using a wet mount preparation. The results revealed a strong correlation between floret length and microspore developmental stages. Microspores at the late uninucleate (vacuolated) to early bicellular stages were consistently found in florets ranging from 4.68 to 4.34 cm in length. Based on these findings, florets within the 4.60 to 4.00 cm range are recommended as the optimal size for microspore culture initiation, as they contain a higher proportion of developmentally competent microspores for embryogenesis induction. In conclusion, this study provides essential morphological and cytological criteria to support efficient explant selection for in vitro regeneration of banana through the microspore culture approach.

Abstract

The decline in seed quality due to storage beyond the shelf life is a major problem in rice cultivation. One effort that can be made to improve seed viability is through invigoration techniques using natural ingredients, such as bean sprout extract (Phaseolus radiatus), which contains growth hormones. This study aims to analyze the effect of bean sprout extract on the viability of Ciherang rice seeds that have exceeded their shelf life and to determine the best treatment. The study was conducted from August to November 2024 at the Biology Laboratory of the UPTD Seed Center, Supervision and Certification of Agricultural Seeds of Gorontalo Province. The study used a two-factor Randomized Block Design (RBD) with bean sprout age (3, 5, and 7 days after sowing) and bean sprout extract concentration (20 g/L, 40 g/L, and 60 g/L) as factors, with four replications. The parameters observed included germination rate, maximum growth potential, and sprout length. The data were analyzed using ANOVA and DMRT post-hoc test at a 5% level. The results showed that bean sprout extract had a significant effect on all observed parameters. The treatment of 5-day-old bean sprouts with a concentration of 20 g/L gave the best results with a germination rate of 95.5%, maximum growth potential of 98.5%, and the highest sprout length. Sprout extract has the potential to be used as a natural alternative to improve the quality of rice seeds that have passed their storage period.

Abstract

Biodiversity represents the condition and level of ecosystem complexity in a particular area, reflecting the presence of various living organisms and their interactions with the environment. Based on its ecological conditions, Mooat Lake in East Bolaang Mongondow Regency serves as a habitat for various plant species, one of which is moss (Bryophyta), which plays an important role in maintaining ecosystem balance. The presence of Bryophyta in this area has the potential to be utilized as a contextual learning resource in biology education. This study aims to determine the validity level of an interactive e-book based on Bryophyta research conducted at Mooat Lake, East Bolaang Mongondow Regency, as a biology learning material for tenth-grade senior high school students. This research employed a Research and Development (R&D) method using the ADDIE development model, which consists of the stages of analysis, design, development, implementation, and evaluation. The product developed in this study was an interactive e-book containing biodiversity material based on field research findings. The results showed that the material expert validation obtained a percentage of 94.2%, categorized as very valid, while the media expert validation obtained a percentage of 83.7%, also categorized as very valid. Based on these findings, it can be concluded that the interactive e-book based on moss research meets the criteria of being very valid and is suitable for use as a learning resource in biology subjects, particularly on biodiversity topics.

Abstract

Laboratorium merupakan salah satu sarana dan prasarana sekolah yang dapat menunjang kesuksesan kegiatan pembelajaran peserta didik. Laboratorium merupakan tempat dilakukannya berbagai kegiatan, seperti observasi, pengamatan, percobaan, latihan, dan pengujian terhadap suatu konsep pengetahuan ilmiah. Namun pada faktanya, pemanfaatan laboratorium sekolah masih sangat rendah. Rendahnya tingkat pemanfaatan fasilitas laboratorium yang ada di sekolah salah satu faktornya disebabkan oleh kurangnya kegiatan pelatihan terkait pengelolaan dan penggunaan alat-alat laboratorium yang sering digunakan disekolah. Tujuan dari kegiatan ini adalah untuk memberdayakan guru agar lebih terampil dan inovatif dalam memanfaatkan peralatan laboratorium IPA guna meningkatkan kualitas pembelajaran. Metode pelaksanaan kegiatan terdiri dari beberapa tahapan utama, yaitu koordinasi awal dengan dinas pendidikan untuk menentukan lokasi dan kebutuhan kegiatan, tahap pelaksanaan berupa kegiatan ceramah dan praktik, dan tahap akhir. Fokus kegiatan pengabdian ini adalah dengan kegiatan praktik langsung dan simulasi penggunaan alat-alat laboratorium. Hasil dari kegiatan ini menunjukkan adanya pemahaman guru dalam mengelola dan memanfaatkan peralatan laboratorium IPA serta inovasi dalam merancang percobaan. Kegiatan ini berhasil memberikan pemahaman guru terhadap fungsi dan pemeliharaan peralatan laboratorium IPA. Guru-guru menunjukkan antusiasme tinggi dalam mengikuti pelatihan dan praktik langsung. Evaluasi kegiatan menunjukkan adanya kemampuan dalam merancang dan melaksanakan praktikum sederhana. Selain itu, peserta memberikan tanggapan positif terhadap kegiatan, dengan menyatakan bahwa materi yang disampaikan sangat bermanfaat untuk diterapkan di sekolah masing-masing. Melalui kegiatan ini diharapkan dapat berkontribusi pada peningkatan kualitas pembelajaran IPA di SMP Kota Gorontalo. Untuk keberlanjutan, disarankan adanya pelatihan lanjutan serta pengadaan alat laboratorium yang lebih lengkap sesuai kebutuhan.

Abstract

Tuberkulosis (TB) paru merupakan penyakit menular yang memiliki hubungan erat dengan Diabetes Mellitus (DM), di mana penderita DM memiliki risiko lebih tinggi mengalami TB akibat penurunan fungsi imunitas. Kota Gorontalo menunjukkan peningkatan kasus komorbid TB-DM dari tahun ke tahun, sehingga diperlukan analisis ilmiah untuk mengidentifikasi faktor determinan yang berkontribusi terhadap kejadian TB pada penderita DM tipe II. Penelitian ini bertujuan untuk menganalisis faktor determinan kejadian TB paru pada penderita DM tipe II di Kota Gorontalo. Menggunakan metode penelitian kuantitatif dengan pendekatan analitik, jumlah sampel 90 responden dengan teknik purposive sampling. Data dikumpulkan melalui wawancara terstruktur, observasi, serta pemeriksaan kadar gula darah, kemudian dianalisis menggunakan uji Odds Ratio (OR) dan regresi logistik berganda. Hasil penelitian menunjukkan bahwa empat variabel yang terbukti berpengaruh signifikan, yaitu usia (p=0,003; Exp(B)=6,644), jenis kelamin (p=0,045; Exp(B)=0,008), pendidikan (p=0,006; Exp(B)=0,034), dan pekerjaan (p=0,013; Exp(B)=61,210) sebagai determinan paling dominan terhadap kejadian TB paru pada penderita DM tipe II. Pekerjaan, usia, jenis kelamin, dan pendidikan terbukti berpengaruh signifikan terhadap kejadian TB paru pada penderita diabetes mellitus tipe II di Kota Gorontalo, dengan pekerjaan dan usia muncul sebagai determinan paling dominan. Temuan ini menegaskan bahwa karakteristik sosiodemografis memiliki peran penting dalam meningkatkan risiko TB pada populasi dengan DM tipe II.

Abstract

Anthracnose disease is a plant disease caused by the fungus Colletotrichum capsici, the disease is one of the problems in the cultivation of cayenne pepper (Capsicum frutescens). This study aims to test the antagonistic ability of Trichoderma sp. against the fungus Colletotrichum capsici and observe the morphological changes in Colletotrichum capsici colonies exposed to Trichoderma sp in vitro on cayenne pepper plants Capsicum frutescens. The test method was carried out using the dual culture method to determine the percentage of inhibition by studying the interaction between Trichoderma sp. and Colletotrichum capsici colonies on Potato Dextrose Agar culture media. The results showed that Trichoderma sp. fungi can compete for space and resources against Colletotrichum capsici fungi macroscopically by controlling space and resources, and microscopically by producing antibiotics (antimicrobials) that can inhibit the growth of Colletotrichum capsici. The urgency of this research is the need for safe and environmentally friendly disease control efforts. One method that has been widely developed is by utilizing biological agents, such as Trichoderma sp. which is able to inhibit the growth of the pathogen Colletotrichum capsici.

Abstract

Callus induction is a critical stage in anther culture for supporting the propagation and genetic improvement of local banana cultivars. This study evaluated the effect of different combinations of 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzylaminopurine (BAP) on callus induction from goroho banana (Musa acuminata L.) anthers. Observations were conducted for 30 days after induction, focusing on callus emergence time and morphological characteristics, including color and texture. The results demonstrated distinct responses among treatments. The combination of 1.5 ml 2,4-D and 1.5 ml BAP produced the fastest callus initiation with an average of 13.17 days after induction and generated friable callus with a pale yellow coloration, indicating high cellular activity. Lower growth regulator concentrations delayed callus formation, while higher concentrations promoted color changes associated with early tissue differentiation. No callus formation was observed in the absence of exogenous growth regulators. These findings highlight the importance of balanced auxin and cytokinin concentrations in regulating anther-derived callus induction and provide a physiological basis for further development of goroho banana anther culture systems.

Abstract

Tepurang (Momordica cochinchinensis) memiliki potensi besar dalam bidang farmasi dan pangan, namun keterbatasan bibit dan rendahnya tingkat perkecambahan biji menghambat perbanyakannya. Kultur jaringan menjadi alternatif dalam pelestarian dan perbanyakan tanaman ini, terutama melalui induksi kalus. Penelitian ini bertujuan untuk mengevaluasi pengaruh kombinasi Benzylaminopurine (BAP) dan Indole Acetic Acid (IAA) terhadap induksi kalus dari batang muda M. cochinchinensis secara in vitro. Eksplan berupa batang muda ditanam pada media Murashige & Skoog (MS) dengan kombinasi BAP dan IAA dalam berbagai konsentrasi. Hasil penelitian menunjukkan bahwa perlakuan dengan kombinasi ZPT mampu menginduksi pembentukan kalus dengan warna putih dan tekstur friable setelah 2 minggu subkultur. Temuan ini diharapkan dapat berkontribusi dalam pengembangan metode perbanyakan M. cochinchinensis secara in vitro untuk mendukung konservasi dan pemanfaatannya dalam bidang bioteknologi.

Abstract

The integration of empathy values into science education has gained increasing scholarly attention as part of character education and holistic learning paradigms. In multicultural contexts such as Indonesia, local wisdom offers culturally grounded moral frameworks that can strengthen students' socio-emotional competencies, particularly empathy, within science learning. This study aims to synthesize and map global and national research trends on empathy-based local wisdom in science education through a hybrid review approach, combining a Systematic Literature Review (SLR) and bibliometric analysis. A systematic search was conducted across Scopus-indexed journals and SINTA-accredited Indonesian journals published between 2019 and 2025. Following PRISMA guidelines, 21 articles met the inclusion criteria and were analyzed qualitatively to identify conceptual frameworks, instructional models, learning outcomes, and assessment strategies related to empathy development in science education. The results indicate that embedding local wisdom—such as indigenous environmental ethics, communal values, and cultural narratives—into inquiry-based and project-based science learning significantly enhances students' empathy, environmental awareness, and ethical reasoning. However, empirical studies measuring empathy using validated psychometric instruments remain limited, particularly in experimental and longitudinal designs. Furthermore, international publications predominantly emphasize socio-scientific issues, while Indonesian studies focus more on contextual cultural integration. The findings suggest that future research should strengthen interdisciplinary designs, develop standardized empathy assessment tools, and expand cross-cultural comparative studies to advance globally relevant yet locally rooted science education practices.
