Abstract

Anthracnose disease is a plant disease caused by the fungus Colletotrichum capsici, the disease is one of the problems in the cultivation of cayenne pepper (Capsicum frutescens). This study aims to test the antagonistic ability of Trichoderma sp. against the fungus Colletotrichum capsici and observe the morphological changes in Colletotrichum capsici colonies exposed to Trichoderma sp in vitro on cayenne pepper plants Capsicum frutescens. The test method was carried out using the dual culture method to determine the percentage of inhibition by studying the interaction between Trichoderma sp. and Colletotrichum capsici colonies on Potato Dextrose Agar culture media. The results showed that Trichoderma sp. fungi can compete for space and resources against Colletotrichum capsici fungi macroscopically by controlling space and resources, and microscopically by producing antibiotics (antimicrobials) that can inhibit the growth of Colletotrichum capsici. The urgency of this research is the need for safe and environmentally friendly disease control efforts. One method that has been widely developed is by utilizing biological agents, such as Trichoderma sp. which is able to inhibit the growth of the pathogen Colletotrichum capsici.

Abstract

Callus induction is a critical stage in anther culture for supporting the propagation and genetic improvement of local banana cultivars. This study evaluated the effect of different combinations of 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzylaminopurine (BAP) on callus induction from goroho banana (Musa acuminata L.) anthers. Observations were conducted for 30 days after induction, focusing on callus emergence time and morphological characteristics, including color and texture. The results demonstrated distinct responses among treatments. The combination of 1.5 ml 2,4-D and 1.5 ml BAP produced the fastest callus initiation with an average of 13.17 days after induction and generated friable callus with a pale yellow coloration, indicating high cellular activity. Lower growth regulator concentrations delayed callus formation, while higher concentrations promoted color changes associated with early tissue differentiation. No callus formation was observed in the absence of exogenous growth regulators. These findings highlight the importance of balanced auxin and cytokinin concentrations in regulating anther-derived callus induction and provide a physiological basis for further development of goroho banana anther culture systems.

Abstract

Banana (Musa spp) is a tropical fruit plant that has many benefits and important roles for the community. This study aims to provide information on the morphological characteristics of bananas in the Gorontalo Province and their phenetic relationships. This study uses a descriptive method to describe the diversity of morphological characters of bananas (Musa spp) with data collection using a survey study method (exploration) which is carried out by tracing or exploring the location of the Gorontalo Province, purposive sampling (Purposive Stratified Sampling) by seeing and observing directly the morphological characters of bananas (Musa spp). Data analysis using the Unweighted Pair Group with Mean of Arithmetic (UPGMA) method with the Gower Genelar Similarity Coeffiicient formula in the MVSP (Multi-Variate Statistical Package) v.3.1 computer program. The results of the study showed that there were 4 types of bananas found in the Gorontalo Province which showed unique morphological variations, namely Kepok Bananas have the characteristic of quite large leaf morphology, Abati Bananas have a unique yellowish color, Green Butota Bananas have morphological characteristics with very thick fruit and Wild Bananas have characteristics with a large number of stones. Based on the results of the clustering, 2 main groups were formed which separated at a cosiness of 0.643 (64.3%) found between Abati Bananas, Green Butota, and Wild Bananas, while Kepok Bananas showed a very distant relationship with a cosiness value of 0.482 (48.2%). The results of this study have the potential to support the preservation of local varieties and support the sustainable genetics management through the identification of varieties that are tolerant to salinity and drought.
