Abstract

Seed dormancy is one of the main factors that inhibit the process of germination and plant growth. This study aims to study the effectiveness of using gibberellic acid (GA3) in maintaining the dormancy of rice seeds (Oryza sativa L.) Mekongga variety to increase germination. In this study, Mekongga rice seeds that experienced dormancy were given GA3 treatment with various concentrations. 0.25 ppm, 0.50 ppm, and 0.75 ppm with 24- and 48-hour immersion. The parameters measured include germination percentage, maximum growing potential, dormancy intensity, and sprout length. This research uses a quantitative approach with experimental methods. This study was carried out using a factorial pattern Group Randomized Design (RAK), which consisted of two factors, namely four levels of soaking using three differences in GA3 concentration and one control (aquades) and two levels of seed immersion, namely 24 and 48 hours. The results showed that GA3 treatment in combination concentration treatment had an influence on observation parameters, and there was the best concentration, namely a concentration of 0.25 ppm in 48-hour soaking capable of producing the highest germination of 96.25%, maximum growth potential reaching 98.5% with a dormancy intensity value of 1.5%.

Abstract

This study aims to determine the composition and structure of vegetation and estimate the potential for carbon dioxide absorption in the Tangale Nature Reserve Area, Gorontalo Regency. The method used in this study is the determination of sampling locations carried out by purposive sampling, making multilevel plots, analyzing vegetation structure, and estimating tree biomass is done non-destructively based on measuring the height and diameter of trees at breast height (DBH), while for lower plants it is done destructively and litter is taken all in the plot area to be weighed wet and dry weight and then estimating biomass, C reserves, and CO2 uptake. The results showed that the vegetation composition consisted of 18 species dominated by the Magnoliopsida class. The highest Important Value Index is found at each observation station. Calculation of biomass value, C reserves, and CO2 uptake at each observation station showed varying results because they were influenced by vegetation type, vegetation density, vegetation composition and structure, and environmental factors. The total value of CO2 uptake by vegetation in the Tangale Nature Reserve Area is 5,304.393 tons of CO2/ha. This research provides a better understanding of carbon sequestration potential in conservation areas to support environmental conservation and climate change mitigation efforts.

Abstract

Huluduotamo village is one of the villages in Suwawa District, Bone Bolango Regency, Gorontalo Province. The village is situated in the middle of the countryside within the Suwawa district area. The potential of Huluduotamo village is the potential of natural resources; thus, the exploration of flower morphology and pollen types of various angiosperm families was carried out in the village. Therefore, this study aims to look at and figure out the shape of pollen from different plant families, mainly Malvaceae and Fabaceae, by looking at pollen units, aperture types, wall thickening, and exine decoration. We conducted the sampling in the Huluduotamo village, Gorontalo, using direct observation methods and descriptive analysis under a microscope. The study found that the pollen from the Hibiscus rosa-sinensis L. plant in the malvaceae family has a monad pollen unit, an apantoporate aperture type, thickening of the centrifugal wall, and decoration on the echinate exine. Some plants, like Caesalpinia pulcherrima L. in the Fabaceae family, have pollen that is made up of a single unit, an atricolporate aperture for the red variety and a parasyncolporate aperture for the yellow variety, and psilate axine decoration. The similarity of the monad pollen unit shows how angiosperm plants reproduce, while differences in aperture types and axine ornamentation show how the shape of the plant has changed to support reproduction. This research provides important information for the identification of plant species and supports flowering plants

Abstract

Momordica cochinchinensis, locally known as Dumbaya in Gorontalo Province, is a traditional medicinal plant with underutilized potential due to the extremely hard morphology of its seed coat. This physical barrier limits the penetration of water, air, and nutrients, thereby reducing the success rate of seed germination and plant propagation. The challenge of propagating plants that are difficult to reproduce sexually, such as dumbaya, can be addressed through various approaches, one of which is asexual reproduction using tissue culture techniques. This study aims to address propagation constraints by applying tissue culture techniques supplemented with Plant Growth Regulators (PGRs), specifically BAP (Benzyl Amino Purine) and NAA (Naphthalene Acetic Acid), which are critical factors in successful in vitro plant regeneration. The combination of Benzyl Amino Purine (BAP) and Naphthalene Acetic Acid (NAA) is a commonly used Plant Growth Regulator (PGR) for inducing callus formation and organogenesis, where BAP is effective in stimulating shoot formation, while NAA plays a role in root induction. The research employed an experimental method using a Completely Randomized Design (CRD) with five treatment levels: H0: MS + 0 ppm NAA + 0 ppm BAP; H1: MS + 1.5 ppm NAA + 0.5 ppm BAP; H2: MS + 3.0 ppm NAA + 0.5 ppm BAP; H3: MS + 1.5 ppm NAA + 1.0 ppm BAP; and H4: MS + 3.0 ppm NAA + 1.0 ppm BAP. Each treatment was replicated three times. The results showed that treatment H1 produced the earliest callus formation, with an average callus initiation time of 7 days after planting.

Abstract

Lemon cui (Citrus microcarpa Bunge) is widely utilized by the people of Gorontalo as a cooking ingredient, traditional medicine, cleaning agent, and cosmetic ingredient. This diversity of uses indicates the potential for further development of lemon cui. The initial stage of development that can be undertaken is the mass propagation of high-quality lemon cui seedlings. This study aims to produce lemon cui seedlings using in vitro culture techniques through seed germination. The experimental design used in this study is a Completely Randomized Design (CRD) with four treatments, namely:  A: MS0 (without the addition of plant growth regulators and young coconut water); B: MS + 50 mL young coconut water + 0.5 ppm 2,4-D; C: MS + 50 mL young coconut water + 1 ppm 2,4-D; and D: MS + 50 mL young coconut water + 1.5 ppm 2,4-D. The culture medium used was Murashige and Skoog (MS) supplemented with young coconut water and 2,4-dichlorophenoxyacetic acid (2,4-D). The study's results showed that using young coconut water and 2,4-D significantly accelerated the germination of lemon cui seeds. The best result was observed in treatment D, where the citrus seeds exhibited a maximum germination rate of 100%. In addition, the combination of young coconut water and 2,4-D is capable of inducing callus formation during the germination process. The combination of 50 mL of young coconut water and 1.5 ppm 2,4-D is the most effective concentration for inducing germination of lemon cui seeds and callus formation.
